Abstract

发明领域

发明背景

发明详细说明

I.i哺乳动物乳OPN的结构
根据一个第一实施例,本发明提供了用于增强对感染性疾病的免疫应答,并且具体地在哺乳动物中增强对感染性疾病的疫苗诱导的免疫抵抗力的哺乳动物乳OPN和/或其活性截短物。

I.iii哺乳动物乳OPN可增强哺乳动物的体液免疫应答

II疫苗

疫苗可以起预防性和治疗性作用,因而可以使用预防性或治疗性治疗来在哺乳动物中增加对感染性疾病的免疫抵抗力并且从而降低感染的风险或治疗现存的感染。

1.方案
1.1动物测试群体和研究设计

1.2动物膳食组和疫苗接种计划

1.3分析来源于血液样品的血清中的血清抗体浓度

使用在我们实验室中开发的ELISA来评价来源于所有取得的血液样品的血清中的流感病毒疫苗特异性IgG。简单地说,用涂布缓冲液[0.5MCarb/Bicarb缓冲液,pH9.6]中的以1:80稀释的透析FluzoneTM疫苗涂布平底板(能肯公司(Nunc),罗切斯特(Rochester),NY),并且在4℃下孵育过夜。在孵育之后,在室温(RT)下用磷酸盐缓冲盐水(PBS)中的10%胎牛血清(FBS)封闭孔,持续1小时。在添加稀释在PBS/10%FBS中的50μL血清之前,用PBS/0.05%吐温-20洗涤这些孔三次,并且在37℃下孵育1h。用PBS/吐温再次洗涤板,接着在37℃下添加PBS/10%FBS中的以1:400稀释的缀合到过氧化物酶上的山羊抗猪IgG(Bethyl实验室,蒙哥马利(Montgomery),TX),持续1h。在室温下将TMB(BD生物科技公司(BDBiosciences),圣何塞(SanJose),CA)孵育20分钟,接着添加50μL的2N硫酸。对于每个孔,使用SpectraMaxM2e(美国分子仪器公司(MolecularDevices),森尼韦尔(Sunnyvale),CA)测量450nm处的吸光度。在每个板上包括以从1:2,000-1:80,000范围内稀释的包含已知量的流感病毒疫苗特异性IgG的阳性储备血清的样品,并且用来对于流感病毒疫苗特异性IgG浓度提供标准曲线。以任意单位表达从该标准曲线的线性部分计算出的流感病毒疫苗特异性IgG。
使用商业上可获得的ELISA试剂盒(Bethyl实验室,蒙哥马利,TX)来测量来源于所取得的血液样品的血清中的总IgG和IgM浓度。
1.4血清抗体浓度的统计学分析
使用重复的测量分析来测试免疫球蛋白(FZ特异性IgG、总IgG和总IgM)的循环水平,其中多项式对比持续SAS(9.2版本,SAS软件研究所(SASInstituteInc.),卡瑞(Cary),NC)内的时间。在完整数据集中进行分析,并且分别地在接种疫苗的组和未接种疫苗的组中进行分析。使用具有较少作为随机变量的起点的混合效应线性模型(ProcMixed)分析来测试在第21天时取得的血液样品上所进行的测量。所分析的主要作用是膳食、疫苗接种以及膳食和疫苗接种的相互作用。当相互作用不显著时,从模型中去除该相互作用。数据被报告为平均值±SD。具有p<0.05的比较被认为是显著的,并且具有p<0.1的那些比较认为是趋势。
1.5从动物测试群体中进行组织样品收集。
在产后21天安乐死之前,用舒泰(Telazol)(7mg/kg体重,肌内(IM),富道动物保健公司(FortDodgeAnimalHealth),富道,IA)镇静小猪,并且通过心脏内穿刺将外周血收集在肝素束紧的真空管中。然后通过注射戊巴比妥钠(72mg/kg体重,FatalPlus,Vortech制药有限公司(VortechPharmaceuticals),迪尔伯恩(Dearborn),MI)使小猪安乐死。从幽门括约肌和回肠辦中切除小肠,并且测量总肠长度,并且在离近端10%和85%处切断肠以得到对应地相应于十二指肠、空肠和回肠的3个区段。还切除了脾和回肠肠系膜淋巴结(MLN)样品用于分离单核细胞。
1.6外周血单核细胞(PBMC)的分离
最初用RPMI-1640(2:1;美国生命技术公司(LifeTechnologies),格兰德岛(GrandIsland),NY)稀释外周血,然后铺层于Ficoll-PaquePlus(通用医疗公司(GEHealthcare),皮斯卡塔韦(Piscataway),NJ)上,并且在20℃下在400×g下旋转40min。从梯度界面中收集PBMC,并且用洗涤缓冲液(汉克斯缓冲盐溶液,无Ca++,无Mg++,美国生命技术公司)洗涤三次,该洗涤缓冲液用2%牛血清白蛋白(BSA;西格玛奥德里奇公司(Sigma-Aldrich),圣路易斯(St.Louis),MO)、0.01MEDTA(西格玛奥德里奇公司)、50μg/mL庆大霉素(美国生命技术公司)、以及1000U/mL青霉素(10000U/ml储液,西格玛奥德里奇公司)和100μg/ml链霉素(10mg/mL储液,西格玛奥德里奇公司)补充。用裂解缓冲液(0.15M的NH4Cl、10mMKHCO3、以及0.1mMNa2EDTA)裂解球粒中剩余的红细胞。将PBMC悬浮于用10%FBS、2mM谷氨酰胺、50μg/mL庆大霉素、1mM丙酮酸钠(美国生命技术公司)、20mMHEPES(美国生命技术公司)、以及20mM1000U/mL青霉素/100μg/ml链霉素补充的RPMI-1640中。使用自动细胞计数器(美国生命技术公司)评价活细胞数。然后通过流式细胞术或离体细胞刺激来将细胞用于表型细胞鉴定。
1.6从脾和MLN中分离总免疫细胞
将脾和MLN样品放入收集缓冲液(汉克斯平衡盐溶液(HBSS),50μg/ml庆大霉素、0.01M4-(2-羟乙基)-1-哌嗪乙磺酸(HEPES)、1000U/ml青霉素和100μg/mL链霉素)中,并且用PBS(美国生命技术公司)+抗体(50μg/mL庆大霉素、1000U/ml青霉素和100μg/mL链霉素)洗涤三次。然后在HBSS中对组织进行均质化,并且使用GentleMACS(德国美天旎生物技术公司(MiltenyiBiotech),奥本(Auburn),CA)短切。使组织匀浆滤过100μm(美国BDFalcon公司(BDFalcon),圣何塞,CA)接着40μm的细胞滤过器(美国BDFalcon公司)。在裂解红细胞之后用洗涤缓冲液洗涤分离的细胞三次,并且悬浮于完全培养基(RPMI-1640,10%FBS、2mM谷氨酰胺、50μg/mL庆大霉素、1mM丙酮酸钠、20mMHEPES、以及20mM1000U/mL青霉素/100μg/mL链霉素)中。如上所述评价活细胞数。
1.7从MLN和脾中分离的PBMC和总免疫细胞的表型鉴定
通过流式细胞术(BDTMLSRII,生物科技公司),使用一系列荧光黄素(FITC)或藻红蛋白(PE)标记的mAb来监测来自外周血、MLN和脾的单核亚群的表型。通过小鼠抗猪CD4(FITC,克隆74-12-4)和小鼠抗猪CD8(PE,克隆76-2-11)抗体(BD生物科技公司)来鉴定T淋巴细胞。将10μl每种抗体添加到来自每个样品的1×106个细胞中。染色程序发生在冰上,并且当可能时将样品从光中移开。简单地说,用5%小鼠血清(南方生物技术公司(SouthernBiotec))和200μg/mL纯化的小鼠IgG(美国英杰生命技术有限公司(Invitrogen))封闭每个孔,各持续5min。在离心之后,将CD3添加到这些孔中并孵育20min(50μL:CD3:PE-Cy5),并且再次离心。添加CD4:FITC和CD8:PE(各10μL),并且孵育另外的15min直到离心。用PBS/1%BSA/0.1%叠氮化钠洗涤细胞,并且然后用2%低聚甲醛固定。使用LSRII流式细胞仪(BDTM,生物科技公司)评价细胞。使用FlowJo7.9软件(FlowJo,阿什兰(Ashland),OR)来确定T细胞亚群的百分比。CD3+事件被认为是T细胞。CD3+CD4+CD8-事件被认为是T辅助细胞,CD3+CD4-CD8+和CD3+CD4+CD8+被认为对应地是细胞毒性T细胞和记忆T细胞。CD3-CD4-CD8+事件被标记为自然杀伤细胞。
1.7离体刺激外周血单核细胞和脾细胞:
作为免疫系统功能能力的指示物而进行离体刺激测定。在37℃下,在5%CO2下将总共2×106个/毫升的来自血液的单核细胞和来自脾的细胞放在最终体积为200μL培养基(包含20%胎牛血清、2mML‐谷氨酰胺、100μg/mL青霉素以及100μg/mL链霉素的RPMI培养基)的96孔板中,持续72h。在存在或不存在OPN(10μL的10μg/mL)的情况下,将50μl的10μg/mL植物凝集素(PHA)溶液、50μL的0.8μg/mL脂多糖(LPS)溶液或18μL的180μg/mLFluzoneTM溶液添加到孔中。在72小时孵育周期之后,对板进行离心并且收集上清液用于细胞因子分泌的测量。
1.8测量由离体刺激的细胞产生的细胞因子分泌:
使用对于IL-10、IL-6和IL-12/IL-23p40的商业上可获得的试剂盒(安迪生物科技公司(R&DSystems),明尼阿波利斯(Minneapolis),MN)来测量细胞因子分泌。简单地说,在4℃下使用制造商所建议的浓度,用捕获抗体对96孔板进行涂布过夜。用PBS中的0.05%吐温洗涤板,并且然后在室温下使用PBS中的1%BSA封闭持续1小时。在用PBS中的0.05%吐温洗涤3次之后,将100μL的未稀释的上清液添加到这些孔中,并且在室温下孵育2小时。在添加以1:180稀释在PBS中的1%BSA中的检测抗体之前,再次洗涤孔,并且将板孵育2小时。将链霉抗生物素蛋白-辣根过氧化酶缀合物溶液添加到孔中并且孵育20min,接着添加TMB底物反应(OptEIA,BD生物科技公司)。在20min孵育之后,用50μL的2NH2SO4停止反应。在450nm波长处,用读板机测量吸光度。
2.结果2.1向配方喂养的小猪补充OPN对它们的体重增加没有作用
所有组中的小猪展现出正常的体重增加。用OPN或疫苗接种补充配方不影响小猪体重增加(图2)。从出生到第15天,SR组中的体重(图2中的插图)是与配方喂养组可比的,而到第16天它们的体重大于FF或OPN。
2.2配方喂养的小猪中的FluzoneTM特异性IgG通过膳食OPN补充剂而增强到大母猪养育的小猪中的水平。
通过ELISA测量来源于7天龄、14天龄和21天龄小猪的血清中的流感病毒疫苗特异性IgG效价。阳性对照样品被用作用于计算FZ特异性IgG相对量的标准曲线,并且值被报告为任意单位(图3)。总体重复测量统计学显示了疫苗接种(p=0.0005)和时间作用(p=0.0001),但没有膳食处理作用。时间作用的进一步多项式趋势分析显示了显著的线性和二次项(p<0.05)对比。未接种疫苗的组的事后统计学分析表明,循环的FZ特异性IgG通常较低并且不受膳食影响。然而,FZ特异性IgG浓度从第7天至第14天和第21天显著地(p<0.05)降低。疫苗接种对第一次剂量的FZ之后的FZ特异性IgG的血清水平不具有影响。到第21天,在第14天给出加强剂量之后,来自所有3个处理组的动物响应于FZ疫苗。OPNV小猪中的FZ特异性IgG浓度类似于SRV小猪,并且这两者都显著高于(p<0.05)FFV小猪(在这3个组中所测量的水平是对应地371±329、400±171和137±157)。
2.3接种疫苗的小猪和未接种疫苗的小猪的血清中的总IgG和IgM效价随时间推移而下降。
通过ELISA测量的血清中的总IgG水平(图4)不受膳食或疫苗接种影响。然而,所测量的总IgG水平随时间推移的平稳下降是统计学显著的(p<0.01),其中在重复的测量分析之后出现显著的线性变化(p<0.002)。在第21天时的混合效应线性模型分析表明,当与未接种疫苗(对应地7.5±2.5和5.9±2.7mg/mL)相比时,在接种疫苗的小猪中的总IgG水平趋向更高模式(p=0.09)。此外,疫苗接种使OPN组中的总IgG水平增加大约2倍(96%),但在FF和SR小猪中观察到的变化(对应地0%和10%)相当小。总IgG水平的这种增加反映了在接受膳食OPN补充剂的小猪中更好地产生适应性免疫应答的能力。
总IgM浓度不受膳食或疫苗接种影响,但最初在产后的周期过程中下降(p<0.001;其中线性和二次项对比在p<0.0001处)。图5指示了在合并来自每个膳食处理内的未接种疫苗的组和接种疫苗的组的数据之后的总IgM水平。
2.4膳食和疫苗接种影响淋巴细胞的表型分布
通过流式细胞术,使用一系列荧光黄素(FITC)或藻红蛋白(PE)标记的mAb来鉴定在第21天取得的脾、PBMC和MLN样品中的单核亚群的表型。如实例1.7中所述,细胞被鉴定为细胞毒性T细胞、T辅助细胞、双阳性记忆T细胞或自然杀伤细胞(NKC)(图6)。PBMC中的淋巴细胞的表型分布和统计学分析(其中当不显著时可去除膳食*疫苗接种相互作用)呈现于表1中。在适应性免疫应答中具有活性作用的T辅助(CD4+)细胞是对膳食有响应的,但对疫苗接种没有响应。在OPN中的T辅助细胞显著高于FF和SR动物(对应地49.4%与42.2%和41.3%,图7A)。
在对抗胞质病原体的宿主防御中重要的细胞毒性T细胞(CD8+)也不受疫苗接种影响,而膳食作用显示差异趋势。最小均方分析的差异显示,SR组中的T细胞毒性细胞的%显著高于OPN(p=0.018)和在趋势水平(p=0.06)高于FF(图7B)。为了更好地理解膳食对单核细胞群体的作用,计算了T辅助细胞与T细胞毒性细胞比率(图7C)。OPN(2.73±0.89)和FF(2.24±0.90)小猪的PBMC中的T辅助细胞与T细胞毒性细胞比率显著高于SR动物(1.71±0.48)的该比率。T辅助细胞与T细胞毒性细胞比率的这种增加表明,在接种疫苗的动物中免疫系统被引发制造疫苗特异性的抗体,具体地接受OPN补充的膳食的那些小猪。
记忆T细胞的群体(对于CD4+和CD8+双阳性)显著地受疫苗接种影响,而膳食仅显示一种趋势(p=0.052)。疫苗接种导致CD3+细胞与CD4+CD8+记忆细胞的%降低21%。
在疫苗接种之后PBMC中的自然杀伤细胞(CD4+CD3+CD8-)群体变化,其中从未接种疫苗的动物中的14.8%显著(p<0.05)增加到接种疫苗的动物中的23.7%,但不存在膳食的作用。

如实例1.7所述,从MLN中分离免疫细胞,并且细胞群体被鉴定为CD3+的%和CD3-的%(表2)。疫苗接种对所研究的任何MLN免疫细胞不具有显著的影响。OPN组中的T细胞毒性细胞数(13.7%)最为类似SR组的T细胞毒性细胞数(12.0%),并且它们都与FF动物(16.6%,图8A)显著不同。膳食没有使作为T辅助细胞的CD3+细胞%变化,但膳食显著地(p<0.05)改变了T细胞毒性细胞群体。类似地,OPN和SRT辅助细胞/T细胞毒性细胞比值是可比的并且对于FF显著更大(图8B)。PBMC中看到的CD4+/CD8+细胞的比率增加反映了,在接受膳食OPN的小猪中对疫苗接种的适应性体液应答增强。SR小猪中的自然杀伤细胞群体高于这两个配方组,但在趋势水平(p<0.06)处达到统计学显著性。

从脾中分离的单核细胞的分布示于表3中。脾中的T辅助细胞和T细胞毒性细胞受疫苗接种影响,但不受膳食处理影响。作为记忆细胞的CD3+的%受膳食和疫苗接种影响。疫苗接种增加了记忆细胞的群体,这些记忆细胞在建立适应性(体液)应答方面是重要的。值得注意地,这两个配方喂养的组(OPN组和FF组)都具有比SR显著更高的记忆细胞水平(图9)。当与所有其他处理组相比时,自然杀伤细胞在未接种疫苗的SR动物中显著更高。疫苗接种不影响脾中的自然杀伤水平。在接种疫苗的小猪的脾中,T辅助细胞(CD4+)/T细胞毒性细胞(CD8+)比率被认为是增加的,具体地以SR或OPN喂养的那些小猪,从而反映了适应性体液应答的诱导。

2.5由分离的免疫细胞产生的离体刺激和细胞因子分泌:
为了评价PBMC和脾细胞的细胞免疫应答,与PHA、LPS或流感病毒疫苗一起孵育分离的细胞持续72小时。植物凝集素(PHA)(一种植物外源凝集素)和脂多糖(LPS)(一种细菌细胞壁组分)是对应地活化T细胞和B细胞的促分裂原。免疫细胞的活化引起细胞因子的分泌。白细胞介素6(IL-6)(又称为干扰素-β2)是对从急性炎症转变成获得性免疫或者慢性炎症疾病所必要的多效性α-螺旋细胞因子。白细胞介素10(IL-10)是一种抗炎性Th2细胞因子,而白细胞介素-12(IL-12)是又称为自然杀伤细胞刺激因子(NKSF)或细胞毒性淋巴细胞成熟因子的一种促炎性Th1细胞因子。在培养基中,在存在或不存在10μg/mLOPN的情况下进行离体细胞培养。添加OPN对所测定的细胞因子的分泌不具有显著影响,因此合并来自OPN处理的细胞和未处理的细胞的数据。对于PBMC和脾的所有处理的具有细胞因子浓度(pg/mL)的数据对应地概述于表4和表5中。然后基于统计学差异合并统计学显著的数据,并且示于图10-17中。
外周血单核细胞:在未刺激的PBMC中,IL-6和IL-10的浓度在检测水平以下(表4)。在未刺激的细胞的上清液中检测到IL-12们并且膳食和疫苗接种这两者的作用都是统计学显著的(p<0.05)(图10)。在OPNV组的PBMC中,IL-12相对于所有其他处理组是最高的。在PBMC中PHA刺激细胞因子不受疫苗接种影响。然而,膳食对IL-12分泌的影响是统计学显著的,其中在OPNV中观察到最高分泌(图11A)。IL-10分泌趋向于在膳食组之中不同,其中获自OPN组的细胞趋向于高于来自来自SR和FF小猪的细胞(图11B)。
在来源于OPN喂养的小猪的LPS刺激的PBMC中,IL-6和IL-12的浓度相对于SR组和FF组显著更高(p<0.05),无论是否疫苗接种(对应地图12A和图12B)。在LPS刺激IL-10分泌中观察到类似的模式,其中暴露于OPN引起IL-10的浓度更高。另外,疫苗接种在OPN组和SR组中引起更高的IL-10水平(图12C)。
流感病毒疫苗对IL-12的作用是疫苗接种依赖性的,其中在膳食与疫苗接种之间具有统计学显著的相互作用(图13A)。疫苗接种在SRV和FFV中导致IL-12分泌减少,而OPN组保持不变。与SR和FF相比,在流感病毒疫苗刺激的细胞中的IL-10分泌高于在OPN喂养的组中,而接种疫苗的小猪具有更低的IL-10浓度(图13B)。
脾免疫细胞:用PHA、LPS和流感病毒疫苗刺激从脾中分离的细胞,并且在72小时孵育之后所收集的上清液中测量细胞因子产生(表5)。脾细胞不产生对用于本研究的刺激物有响应的任何IL-6。另一方面,在未刺激的细胞的上清液中发现IL-12(图14A)。来自SR组和FF组的细胞分泌更高量的IL-12,而膳食OPN和疫苗接种趋向于(p=0.07)降低IL-12浓度。未刺激的细胞的上清液中的IL-10浓度在SRV组中最高(图14B)。对PHA刺激有响应的由脾细胞产生的IL-12和IL-10分泌是类似的。当与在未接种疫苗的组中发现的水平相比时,疫苗接种降低IL-12(图15A)和IL-10(图15B)的浓度。此外,OPN组具有比SR组和FF组显著更低的这两种细胞因子的水平。类似地,对LPS有响应的IL-12分泌在来源于未接种疫苗的小猪的细胞上清液中显著高于接种疫苗的小猪(p<0.05)(图16A)。获自OPN组的细胞相对于SR组和FF组分泌最低量的IL-12。在LPS刺激的细胞中的IL-10分泌不受疫苗接种影响,但在SR组中高于FF组和OPN组(图16B)。当流感病毒疫苗刺激时,IL-12和IL-10分泌在接种疫苗的组中最低(p<0.05),并且从OPN动物中分离的细胞比SR和FF分泌更少的IL-12(图17)。
总之,当小猪接受用OPN补充的配方膳食时,它们的胃细胞暴露于恒定的OPN浓度中。这与大母猪养育的小猪相反,在这些大母猪养育的小猪中,它们接受的OPN水平将随着用大母猪乳替代大母猪初乳的供应而下降,并且将低于在OPN补充的配方膳食中提供的140mg/L。当在不存在和存在免疫刺激剂这两种情况下离体孵育时,当与来源于配方喂养的小猪或大母猪养育的小猪的细胞相比时,接受OPN补充的配方的小猪特征在于分泌更多IL-12和IL-10的免疫细胞(PBMC)。这提供了以下证据:膳食OPN具有引发PBMC细胞分泌IL-12和IL-10的作用。当用PHA(T细胞活化)和LPS(B细胞活化)刺激时,来自OPN-配方喂养的小猪的PBMC分泌IL-12(促炎性)和IL-10(抗炎性)的能力表明免疫机制推向免疫平衡。
实例2
使用向婴儿给予的OPN-10进行的临床试验
2.1试验设计
在中国上海进行双盲随机临床试验以便评估将牛OPN添加到配方中的作用。母亲选择母乳喂养或者配方喂养她们的从1至6月龄的婴儿。分组如下(n=60/组):
1)母乳喂养的婴儿
2)用没有OPN添加的常规配方(RF)(F0)喂养的婴儿
3)用添加大约65mgOPN/L的牛OPN的RF(F65)
4)用添加大约130mgOPN/L的牛OPN的RF(F130)
*在(未补充的)常规配方中发现的OPN的基础水平是大约15mgOPN/L。
按月登记人体测量学,并且在1月龄、4月龄和6月龄时通过静脉穿刺取得静脉血液样品。分析血液学、免疫参数、血浆氨基酸类和血液尿素氮(BUN)。
2.2试验结果
与母乳喂养的婴儿相比,在接受常规配方(F0)的婴儿中,对感染(诸如病毒、细菌、真菌或变形虫感染)有响应的婴儿中的发热发病率显著增加(图18)。以65mgOPN/L或130mgOPN/L的量将OPN添加到常规配方中可使在用常规配方喂养时所见到的高发热发病率降到极为接近在母乳喂养的婴儿中见到的低发病率水平。当与母乳喂养的婴儿相比时,接受常规配方(F0)的婴儿组是仅有的显示发热发病率统计学显著增加的组。

6.用于根据权利要求1至5中任一项所述使用的哺乳动物乳和/或其活性截短物,其中在哺乳动物中对感染性疾病的免疫抵抗力是通过疫苗接种来诱导的。
7.用于根据权利要求1至5中任一项所述使用的哺乳动物乳和/或其活性截短物,其中该感染性疾病是选自以下各项之中:流感、白喉、破伤风、百日咳、脊髓灰质炎、麻疹、腮腺炎和风疹、结核病、乙型肝炎、丙型脑膜炎、轮状病毒、人乳头瘤病毒、甲型流感、乙型流感、肺炎球菌感染以及带状疱疹。
8.一种营养组合物,包含根据权利要求1至7中任一项所述的哺乳动物乳和/或其一种或多种活性截短物。
9.一种用于在哺乳动物中预防性或治疗性治疗感染性疾病的疫苗系统,该疫苗系统包含一种疫苗和一种哺乳动物乳骨桥蛋白和/或其一种或多种活性截短物,其中该骨桥蛋白和/或其活性截短物是用于口服给予的;其中给予该骨桥蛋白和/或其活性截短物增强由该疫苗诱导的免疫抵抗力。
10.用于根据权利要求9所述使用的疫苗系统,其中该哺乳动物乳骨桥蛋白和/或其活性截短物在该哺乳动物中加强体液免疫。
11.用于根据权利要求9或10所述使用的疫苗系统,其中该骨桥蛋白和/或其活性截短物是选自以下各项之中:牛、山羊、绵羊、骆驼、水牛、单峰骆驼、美洲驼、以及其任何组合。
12.用于根据权利要求9至11中任一项所述使用的疫苗系统,其中该骨桥蛋白和/或其活性截短物是用于在对该哺乳动物进行疫苗接种之前、同时、或之后、或其组合进行口服给予的。
13.用于根据权利要求9至12中任一项所述使用的疫苗系统,其中该哺乳动物是人。
14.用于根据权利要求9至13中任一项所述使用的疫苗系统,其中该感染性疾病是选自以下各项之中:流感;白喉、破伤风、百日咳、脊髓灰质炎、麻疹、腮腺炎和风疹、结核病、乙型肝炎、丙型脑膜炎;轮状病毒、人乳头瘤病毒;甲型流感、乙型流感、肺炎球菌感染以及带状疱疹。
15.用于根据权利要求9至14中任一项所述使用的疫苗系统,其中该人属于一个年龄组,该年龄组选自以下各项之中:0–5岁、6–11岁、12–18岁、19–34岁、35–44岁、45–54岁、55–64岁、65–74岁、75–84岁、以及大于84岁。
