Abstract
本发明公开了一种常山胡柚鲜果的贮藏方法,所述方法为:将常山胡柚鲜果浸没在溶剂中,15~25℃浸泡20min,取出自然晾干后用保鲜膜包裹,20±2℃贮藏;成熟后采摘的鲜果在1.0%氯化钙水溶液浸泡20分钟后,用透明的保鲜膜包裹1层,在贮藏4个月后,鲜果外观较完好、无明显菌斑;在0.01%高锰酸钾水溶液中浸泡20分钟后,用透明的保鲜膜包裹1层,在贮藏6个月后,外观完好、无明显菌斑;在0.15g/L水杨酸水溶液中浸泡20分钟后,用透明的保鲜膜包裹1层,贮藏5个月后,外观完好、无明显菌斑;在1.0%CaCl2+0.01%KMnO4混合水溶液、0.01%KMnO4+0.15g/L水杨酸混合水溶液或1.0%CaCl2+0.01%KMnO4+0.15g/L水杨酸混合水溶液中浸泡20分钟后,再用透明的保鲜膜包裹1层,在贮藏8个月后,外观较完好、无明显菌斑。

(一)技术领域
本发明涉及一种水果的保鲜方法,特别涉及一种常山胡柚鲜果的长期贮藏方法。
(二)背景技术
常山胡柚是浙江省常山县特有的栽培种,至今已有上百年的种植历史(张韵冰.中国柑桔属植物一新种.植物研究,1991,11(2):5-7)。常山胡柚树体强壮,具有耐旱、耐寒、耐脊、抗病等特点。它对土壤要求不严,山地、平地均能良好生长;也可在肥水条件较差、25℃以下的红土低丘或油茶产区栽种;甚至在全是半风化石头,被称为“石壳山”的紫红砂土上也能生长(蒋柏宏.常山胡柚的若干特性.浙江柑桔,1994(2):14-15)。
常山胡柚果实品质优良,果形硕大、果肉色泽金黄、晶莹脆嫩、甜酸适度、甘中微苦,风味独特,是少有的凉性水果。常山胡柚果实品富含人体所需的16种必需氨基酸,大量的VC、V6、钙、镁、铁、锌、铜、锰等微量元素及类黄酮、柚皮苷、柠檬苦素等生物活性物质,具有清凉去火、止咳化痰、润喉醒酒、抗衰老、稳血糖等保健药用功效,深受广大消费者喜爱(徐小忠,赵四清,汪丽霞.常山胡柚果皮中所含功能性成分与作用.浙江柑橘,2014,31(1):10-12)。但常山胡柚在成熟采摘后的贮藏过程中,其所含有的水分及营养成份逐渐丧失,果品的营养价值和销售的品像都大打折扣(①仲山民,田荆祥,吴美春.常山胡柚果实贮藏期间营养成分含量的变化.浙江林学院学报,1994,11(1):53-57②陈昆松,张上隆,李方等.胡柚果实采后枯水的研究.园艺学报,1995,22(1):35-39)。特别是,胡柚果农采摘鲜果后,通常是将胡柚存放在家中,等待好的价钱才出售;而在贮藏过程中,特别是到采摘后的翌年的4、5月份,随着气温的升高,大量的胡柚果实堆积在一起极易腐烂。因此,胡柚采后贮藏保鲜技术非常重要。
(三)发明内容
本发明目的是提供一种常山胡柚鲜果长期保鲜贮藏的方法。
本发明采用的技术方案是:
本发明提供一种常山胡柚鲜果的贮藏方法,所述方法为:将常山胡柚鲜果浸没在溶剂中,15~25℃浸泡20min,取出自然晾干后,用保鲜膜包裹,20±2℃贮藏;所述溶剂为质量浓度0.5~2.0%氯化钙水溶液、质量浓度0.005~0.05%高锰酸钾水溶液、0.1~0.2g/L水杨酸水溶液中的一种或两种及两种以上的混合。
进一步,优选所述溶剂为质量浓度1.0%氯化钙水溶液、质量浓度0.01%高锰酸钾水溶液或0.15g/L水杨酸水溶液。
更进一步,优选所述溶剂为下列之一:(1)质量终浓度1.0%CaCl2+0.01%KMnO4混合水溶液;(2)质量终浓度0.01%KMnO4+0.15g/L水杨酸混合水溶液;(3)质量终浓度1.0%CaCl2+0.01%KMnO4+0.15g/L水杨酸混合水溶液。
进一步,本发明所述常山胡柚鲜果为成熟后采摘10-20天的鲜果。
本发明所述百分浓度如无特别说明,均指质量浓度。
与现有技术相比,本发明有益效果主要体现在:本发明提供一种常山胡柚鲜果长期保鲜贮藏的方法,成熟后采摘的鲜果在1.0%氯化钙水溶液浸泡20分钟后,用透明的保鲜膜包裹1层,在贮藏4个月后,鲜果外观较完好、无明显菌斑。在0.01%高锰酸钾水溶液中浸泡20分钟后,用透明的保鲜膜包裹1层,在贮藏6个月后,外观完好、无明显菌斑。在0.15g/L水杨酸水溶液中浸泡20分钟后,用透明的保鲜膜包裹1层,贮藏5个月后,外观完好、无明显菌斑。在1.0%CaCl2+0.01%KMnO4混合水溶液,或者0.01%KMnO4+0.15g/L水杨酸混合水溶液,或者1.0%CaCl2+0.01%KMnO4+0.15g/L水杨酸混合水溶液中浸泡20分钟后,再用透明的保鲜膜包裹1层,在贮藏8个月后,外观较完好、无明显菌斑。
(四)附图说明

下面结合具体实施例对本发明进行进一步描述,但本发明的保护范围并不仅限于此:
实施例1
一、实验材料和方法
1、实验材料:常山胡柚鲜果(采摘15天左右,果实已经成熟、外观完整无破损、鲜果大小一致)、蒸馏水、无水乙醇、氯化钙、高锰酸钾、水杨酸、一甲基环丙烯(1-MCP)、MDA试剂盒、SOD试剂盒、POD试剂盒。试剂盒均购自上海信然生物技术有限公司。
2、实验仪器:烧杯、玻璃棒、天平、量筒、保鲜膜、研钵和研磨棒、移液枪、比色皿、紫外光分光光度计、酶标仪、离心机等。
3、不同保鲜剂对胡柚鲜果的处理
(1)不同氯化钙浓度处理
分别配成质量浓度0.5%、1%、1.5%、2%氯化钙水溶液。将常山胡柚鲜果分为9组,组1-组4分别放入0.5%、1%、1.5%、2%氯化钙水溶液中,15-25℃浸泡20min,取出自然晾干后分别用保鲜膜包裹;组5放入蒸馏水中,15-25℃浸泡20min后用保鲜膜包裹;组6-组8分别放入0.5%、1%、1.5%、2%氯化钙水溶液中,15-25℃浸泡20min后取出,不包裹保鲜膜;组9不做任何处理。组1-组9分别置于托盘上,放入20±2℃温室中贮藏。每天记录外观变化,结果见表1所示。
(2)不同高锰酸钾浓度处理
分别配成质量浓度0.005%、0.01%、0.03%、0.05%高锰酸钾水溶液。取常山胡柚鲜果为9组,组1-组4分别放入0.005%、0.01%、0.03%、0.05%高锰酸钾水溶液中,15-25℃浸泡20min,取出自然晾干后分别用保鲜膜包裹;组5放入蒸馏水中,15-25℃浸泡20min后用保鲜膜包裹;组6-组8分别放入0.005%、0.01%、0.03%、0.05%高锰酸钾水溶液中,15-25℃浸泡20min后取出,不包裹保鲜膜;组9不做任何处理。组1-组9分别置于托盘上,放入20±2℃温室中贮藏。每天记录外观变化,结果见表2所示。
(3)不同水杨酸浓度处理
分别配成0.1g/L、0.15g/L、0.2g/L的水杨酸水溶液。将常山胡柚鲜果为8组,组1-组3分别放入0.1g/L,0.15g/L,0.2g/L的水杨酸水溶液中,15-25℃浸泡20min,取出自然晾干后分别用保鲜膜包裹;组4放入蒸馏水中,15-25℃浸泡20min后用保鲜膜包裹;组5-组7分别放入0.1g/L,0.15g/L,0.2g/L的水杨酸水溶液中,15-25℃浸泡20min后取出,不包裹保鲜膜;组8不做任何处理。组1-组8分别置于托盘上,放入20±2℃温室中贮藏。每天记录外观变化,结果见表3。
(4)一甲基环丙烯(1-MCP)的处理
分别用天平称取3份0.1克的1-MCP粉剂(粉剂的一甲基环丙烯的质量含量为3.3%)放入小烧杯中,分别在容器内加入1ml、10ml、100ml的蒸馏水,搅拌溶解,即稀释了10、100、1000倍,分别获得0.1g/mL、0.01g/mL、0.001g/mL一甲基环丙烯水溶液。将常山胡柚鲜果分为8组,每组3个;组1-组3分别用0.1g/mL、0.01g/mL、0.001g/mL一甲基环丙烯水溶液浸润;组4不浸润;组5-组7分别用0.1g/mL、0.01g/mL、0.001g/mL一甲基环丙烯水溶液浸润;组8不浸润。组1-组8分别置入密封的干燥器(体积5L)中,20±2℃℃放置24小时。24小时后,取出组1-组4并用透明保鲜膜包裹1层,组5-组8不包裹保鲜膜。分组置于托盘上,放入24±2℃温室中贮藏。每天记录外观变化,结果显示,用不同浓度一甲基环丙烯水溶液处理和未处理的鲜果之间未见明显差异,后期则不在进行该处理的相关实验。
(5)混合溶液处理
将常山胡柚鲜果为4组,组1放入质量终浓度1.0%CaCl2+0.01%KMnO4+0.15g/L水杨酸混合水溶液中,15-25℃浸泡20min;组2放入质量终浓度0.01%KMnO4+0.15g/L水杨酸混合水溶液中,15-25℃浸泡20min;组3放入质量终浓度1.0%CaCl2+0.01%KMnO4混合水溶液中,15-25℃浸泡20min;组1-组3取出后分别用保鲜膜包裹;组4不做任何处理;组1-组4分别置于托盘上,放入20±2℃温室中贮藏。每天记录外观变化,拍摄照片,结果见表4和图1所示。
4、鲜果中丙二醛(MDA)含量,超氧化物歧化酶(SOD)和过氧化物酶(POD)活性的测定
(1)果实处理:取15个大小相似、室温放置3个月左右的胡柚鲜果,平均分成5组,每组3个,分别置于质量浓度1%氯化钙水溶液、质量浓度0.01%高锰酸钾水溶液、0.15g/L水杨酸水溶液、蒸馏水(即图2-图4中纯水)中浸泡20分钟,取出用保鲜膜包裹,置于20±2℃温室中贮藏。
(2)酶液提取:以5天为一个间隔周期,每隔5天。分别从步骤(1)贮藏的胡柚上称取0.5克果皮,加入2ml0.05mol/L的PBS缓冲液(pH值7.0),研磨成匀浆。将匀浆转移到2ml的离心管中,在4000r/min下离心15min,所得的上清液即为粗酶液。
(3)利用MDA试剂盒、SOD试剂盒,POD试剂盒测试步骤(2)粗酶液中MDA含量、SOD含量以及POD酶活性的测定,操作按照试剂盒说明书进行,制备标准曲线,结果见图2-图4所示。
5、过氧化氢酶(CAT)和抗坏血酸过氧化物酶(APX)的活性的测定
(1)果实处理:自然放置3个月左右的胡柚鲜果,大小和成熟程度一致的常山胡柚12个,平均分成4组,每组3个,分别置于0.15g/L水杨酸水溶液,质量浓度0.01%高锰酸钾水溶液,质量浓度1%氯化钙水溶液、蒸馏水(即图5-图6中水)中,15-25℃浸泡20分钟,取出后分别用保鲜膜包裹,置于20±2℃温室中贮藏。
(2)粗酶液提取:以5天为一间隔周期,每隔5天,分别在步骤(1)贮藏的胡柚上割取0.3克果皮于研钵中,加入2ml0.05mol/L、pH7.0的PBS缓冲液,研磨成匀浆。将匀浆转移到2ml的离心管中,4000r/min下离心15min,所得的上清液即为粗酶液。
(3)CAT活性测定:取一支干净的试管,在试管中加入2800μlPBS(25mmol/L,pH7.0,含2mmol/LEDTA)和100μl粗酶液,轻轻摇匀后加入100μlH2O2,快速振荡后迅速转入比色皿,利用紫外光分光光度计法测定其活性,结果见图5所示。
(4)APX活性测定:取一支干净的试管,在试管中加入2700μlPBS(25mmol/L,pH7.0,含2mmol/LEDTA)和100μl粗酶液,轻轻摇匀后加入100μl抗坏血酸和100μlH2O2,快速振荡后迅速转入比色皿,利用紫外光分光光度计法测定其活性,结果见图6所示。
二、实验结果
1、不同保鲜方法对胡柚鲜果的保鲜效果
不同浓度氯化钙溶液、高锰酸钾溶液、水杨酸溶液处理的保鲜结果如表1-3。

结果显示:1.0%及以上浓度的氯化钙溶液+保鲜膜包裹,贮藏4个月后,鲜果依然外观较完好、无明显菌斑。0.01%及以上浓度的高锰酸钾+保鲜膜包裹,贮藏6个月后,外观完好、无菌斑。0.15g/l及以上浓度的水杨酸+保鲜膜包裹,贮藏5个月后,外观较完好、无明显菌斑。而且,同样处理条件下,使用保鲜膜的保鲜效果明显优于不使用保鲜膜的。另外,用1-MCP处理24小时候的果实,在保存的开始1个月里果皮的颜色色泽更佳,但果实的保鲜贮藏效果与对照不处理的无明显的差异。
为了进一步提高保鲜效果,根据单因子的实验结果,进行不同组合的实验。

结果显示:1.0%CaCl2+0.01%KMnO4+0.15g/L水杨酸+保鲜膜包裹,0.01%KMnO4+0.15g/L水杨酸+保鲜膜包裹,1.0%CaCl2+0.01%KMnO4+保鲜膜包裹,贮藏8个月后,外观完好、无明显菌斑(图1)。
2、MDA、SOD、POD、ARX、CAT的测定结果
MDA、SOD、POD、ARX、CAT的测定结果分别见图2、图3、图4、图5、图6。
植物体内MDA含量体现细胞膜脂的过氧化程度,与对照相比,处理的鲜果MDA含量较低,因此,贮藏过程中,处理的鲜果细胞膜的伤害较低,有利于维持细胞膜的完整性。SOD和POD酶是细胞清除过剩氧自由基的抗氧化酶类,与对照相比,处理的鲜果SOD酶的活性均有所提高,但POD酶活性没有明显的差异。处理的鲜果可能是通过SOD酶的活性增高来保护细胞的完整结构。抗氧化系统的关键解毒酶还包括抗坏血酸过氧化物酶(APX)与过氧化氢酶(CAT),在胁迫下能起到保护细胞功能的作用。与对照相比,处理的鲜果APX活性均有所提高,尤其是高锰酸钾处理更为突出;而CAT酶的变化无明显的规律。因此,贮藏过程中,处理的鲜果可能是提高了鲜果的APX的活性,从而有利于保护细胞功能的完整性。
Claims (8)
1.一种常山胡柚鲜果的贮藏方法,其特征在于所述方法为:将常山胡柚鲜果浸没在溶剂中,15~25℃浸泡20min,取出自然晾干后用保鲜膜包裹,20±2℃贮藏;所述溶剂为质量浓度0.5~2.0%氯化钙水溶液、质量浓度0.005~0.05%高锰酸钾水溶液、0.1~0.2g/L水杨酸水溶液中的一种或两种及两种以上的混合。
2.如权利要求1所述常山胡柚鲜果的贮藏方法,其特征在于所述溶剂为质量浓度1.0%氯化钙水溶液。
3.如权利要求1所述常山胡柚鲜果的贮藏方法,其特征在于所述溶剂为质量浓度0.01%高锰酸钾水溶液。
4.如权利要求1所述常山胡柚鲜果的贮藏方法,其特征在于所述溶剂为0.15g/L水杨酸水溶液。
5.如权利要求1所述常山胡柚鲜果的贮藏方法,其特征在于所述溶剂为质量终浓度1.0%CaCl2+0.01%KMnO4混合水溶液。
6.如权利要求1所述常山胡柚鲜果的贮藏方法,其特征在于所述溶剂为质量终浓度0.01%KMnO4+0.15g/L水杨酸混合水溶液。
7.如权利要求1所述常山胡柚鲜果的贮藏方法,其特征在于所述溶剂为质量终浓度1.0%CaCl2+0.01%KMnO4+0.15g/L水杨酸混合水溶液。
8.如权利要求1所述常山胡柚鲜果的贮藏方法,其特征在于所述常山胡柚鲜果为成熟后采摘10-20天的鲜果。
