Abstract
本发明涉及化合物I和化合物II及其制备方法和应用,其中新合成的式I的化合物在大大提高利尿酸(EA)的抗肿瘤效果上,能够激发抗肿瘤的先天性免疫和细胞免疫,探索了抗肿瘤免疫协同一体化的药物设计;证明了式I的化合物抗黑色素瘤的免疫反应机制;由式I的化合物与ROR1共价制备的式II的化合物明显减慢了乳腺癌皮下移植瘤的生长,证明式II的化合物治疗乳腺癌的免疫反应机制,同时证明了式I的化合物不仅具有激发先天性免疫和适应性免疫的功能,还具有有免疫佐剂的作用。

技术领域
本发明涉及肿瘤癌症疾病预防和治疗的技术领域,更具体地说,涉及化合物I和化合物II及其制备方法和应用。
背景技术
恶性黑色素瘤是来源于黑色素细胞的高度恶性肿瘤,是皮肤癌中常见的恶性肿瘤,转移性强,目前主要的治疗方法是放疗、化疗和手术治疗,然而,传统的治疗并不能将其彻底治愈,随着肿瘤免疫学、分子生物学的发展,对肿瘤进行免疫治疗已成为人们研究的热点。Toll样受体(Toll like receptors,TLRs)是一类重要的模式识别受体,属于Ⅰ型跨膜蛋白,在各种免疫细胞上有表达,TLRs在激发先天性免疫和适应性免疫反应过程中发挥重要作用。TLRs的激动剂作用于抗原递呈细胞,比如树突状细胞(Dendritic Cell,DC),促进细胞因子的产生,从而更好的激发免疫反应。TLR7(Toll like receptor 7)是Toll样受体之一,TLR7的配体主要是病毒核酸成分,也可以识别一些合成的小分子激动剂,TLR7被激活后能够引发较强的免疫反应。T7(第一反应物)是深圳大学深圳合成生物学工程实验室合成的一种化学小分子TLR7受体的激动剂,据研究表明T7具有提高先天性免疫的能力,在体外作用于骨髓来源的树突状细胞能够迅速产生炎症介质TNF-α和IL-12以及作用于淋巴细胞迅速产生IFN-γ和IL-12,其中T7的来源和制备过程参考文献“Local administration of anovel Toll-like receptor 7agonist in combination with doxorubicin inducesdurable tumouricidal effects in a murine model of T cell lymphoma”Journal ofHematology&Oncology.2015,Mar 4;8(1):21.doi:10.1186/s13045-015-0121-9)”。利尿酸(Ethacrynic acid,EA,第二反应物)又叫依他尼酸,是一种利尿剂,有文献表明,利尿酸具有多种抗肿瘤效应,但无法激发特异性的抗肿瘤细胞免疫反应,使得其抗肿瘤效应差、副作用大,肿瘤细胞反复复发生长。
乳腺癌是发生在乳腺腺上皮组织的恶性肿瘤,患者大部分是女性,并且晚期患者易发生转移。目前,乳腺癌的治疗方式主要是手术治疗、放疗、化疗,然而放疗和化疗在杀伤癌细胞同时也会杀伤正常细胞,严重破坏了机体的免疫系统。肿瘤生物治疗是肿瘤治疗的第四种方式,肿瘤免疫治疗是肿瘤生物治疗中比较有应用前景的一种治疗方法,而这种治疗对乳腺癌也是比较好的一种治疗方法。肿瘤免疫治疗的目的是激发或调动机体的免疫系统,增强肿瘤微环境的抗肿瘤免疫力,诱导机体产生有效识别肿瘤相关抗原的免疫细胞,从而控制和杀伤肿瘤细胞。ROR1(Receptor-tyrosine-kinase-like orphan receptor,第三反应物)是一个典型的Ⅰ型孤儿受体酪氨酸激酶样表面蛋白,研究发现许多人源癌细胞系表达ROR1,ROR1在乳腺腺癌中高表达,但是正常乳腺组织不表达ROR1,ROR1的表达与肿瘤生长有关,ROR1表达越高,肿瘤生长越快。据文献报道,ROR1的表达与乳腺腺癌早期转移、复发及预后密切相关,ROR1表达越高转移、复发的概率越高并且预后越差,高表达ROR1的患者无转移生存期比低表达患者短,阻碍ROR1的表达能有效抑制癌细胞向肺部转移,因此ROR1可以作为肿瘤治疗的靶点,但是ROR1的免疫原性较弱,不能激发先天性免疫。
发明内容
本发明的目的在于提供结构式为式I和式II的化合物及其制备方法和应用,即化合物I和化合物II及其制备方法和应用,具体是提供由T7(第一反应物)和EA(第二反应物)偶联得到的结构全新的式I的化合物,并由式I的化合物联合ROR1(第三反应物)共价得到全新的式II的化合物,解决了现有技术中的EA和ROR1无法诱导免疫反应而引起的治疗效果差及带来的不良反应等问题。
本发明解决技术问题所采用的技术方案是:一种式I的化合物,具有如下结构式:
上述式I化合物的制备方法,包括如下步骤:
S1、将第一反应物和第二反应物利尿酸溶于反应溶剂中,加入HBTU苯并三氮唑-N,N,N',N'-四甲基脲六氟磷酸盐、三乙胺和催化剂4-二甲氨基吡啶,室温下搅拌反应;
S2、反应完成后将反应液倒入水中,抽滤,滤渣经水洗、干燥得到式I的化合物。
在本发明的式I的化合物的制备方法中,在步骤S1中,所述第一反应物、第二反应物、苯并三氮唑-N,N,N',N'-四甲基脲六氟磷酸盐和三乙胺以下述摩尔比份数进行反应:第一反应物4-12份、第二反应物5-15份、HBTU苯并三氮唑-N,N,N',N'-四甲基脲六氟磷酸盐5-15份以及三乙胺18-30份。优选地,所述第一反应物、第二反应物、苯并三氮唑-N,N,N',N'-四甲基脲六氟磷酸盐和三乙胺以下述摩尔比份数进行反应:第一反应物6-10份、第二反应物7-12份、苯并三氮唑-N,N,N',N'-四甲基脲六氟磷酸盐7-12份以及三乙胺22-28份;更优选地,第一反应物和第二反应物之间的反应摩尔比为1:1。而催化剂4-二甲氨基吡啶的用量为催化量即可。
在本发明的式I的化合物的制备方法中,在步骤S1中,所述反应溶剂选自四氢呋喃、甲苯、苯、N,N-二甲基甲酰胺、N,N-二甲基乙酰胺和N-甲基吡咯烷酮中的任意一种或多种,优选N,N-二甲基甲酰胺。
在本发明的式I的化合物的制备方法中,步骤S1的反应时间为8-24小时,优选为10-15小时。
在本发明的式I的化合物的制备方法中,在步骤S2后还包括步骤S3:将得到的式I的化合物经柱层析分离进行纯化,其中柱层析分离所用溶剂为二氯甲烷和甲醇的混合溶剂,二氯甲烷与甲醇的体积比为20:1。
上述式I的化合物具有在制备用于预防和免疫治疗黑色素瘤引起的皮肤癌的药物中的用途。
本发明还提供了由式I的化合物制备得到的式II的化合物,具有如下结构式:
上述式II的化合物的制备方法是:将第三反应物溶于二甲基亚砜中,加入式I的化合物,室温搅拌8-24小时,优选搅拌12小时,利用液相色谱-质谱联用监测反应,制备液相色谱分离,得到式II的化合物;
其中,第三反应物为ROR1,其结构式如下:
第三反应物与式I的化合物的反应摩尔配比为1:1。
上述式II的化合物具有在制备用于预防和免疫治疗乳腺癌的药物中的用途。
实施本发明的式I和式II的化合物及其制备方法和应用,具有以下有益效果:本发明中的新合成的式I的化合物在大大提高利尿酸(EA)的抗肿瘤效果上,能够激发抗肿瘤的先天性免疫和细胞免疫,探索了抗肿瘤免疫协同一体化的药物设计;证明了式I的化合物抗黑色素瘤的免疫反应机制。本发明由式I的化合物与ROR1共价制备的式II的化合物明显减慢了乳腺癌皮下移植瘤的生长,证明式II的化合物治疗乳腺癌的免疫反应机制,同时证明了式I的化合物不仅具有激发先天性免疫和适应性免疫的功能,还具有有免疫佐剂的作用。

下面结合附图和实施例,对本发明的化合物I和化合物II及其制备方法和应用作进一步说明:
本发明提供了由第一反应物(简称T7)和第二反应物(简称EA)合成的式I的化合物(简称T7-EA),式I的化合物(T7-EA)的结构式如下:

制备反应的具体过程包括如下步骤:将第一反应物(简称T7)和第二反应物利尿酸(简称EA)溶于反应溶剂中,加入苯并三氮唑-N,N,N',N'-四甲基脲六氟磷酸盐(HBTU)、三乙胺(TEA)和催化剂4-二甲氨基吡啶(DMAP),室温下搅拌反应;反应完成后将反应液倒入水中,抽滤,滤渣经水洗、干燥得到式I的化合物,然后进一步纯化,经柱层析分离,得白色固体状式I的化合物。其中反应溶剂选自四氢呋喃、甲苯、苯、N,N-二甲基甲酰胺、N,N-二甲基乙酰胺和N-甲基吡咯烷酮中的任意一种或多种,反应溶剂优选为N,N-二甲基甲酰胺(DMF)。

式I化合物制备实施例1:
称取344mg(1mmol)T7和341mg(1.125mmol)EA溶于8mL DMF中,加入HBTU(427mg,1.125mmol)、三乙胺(416μL,3mmol)和催化量的DMAP,室温下搅拌反应12小时。将反应液倒入100mL水中,抽滤,滤渣经水洗、干燥得粗品。再经柱层析分离(DCM:MeOH=20:1),得T7-EA白色固体475mg,产率75.5%。纯度:95.08%,ESI-MS:m/z=629.1[M+H]+.(如图1-2所示)
式I化合物制备实施例2:
称取172mg(0.5mmol)T7和189.4mg(0.625mmol)EA溶于6mL苯和甲苯的混合溶剂中且二者体积比2:1,加入HBTU(237.2mg,0.625mmol)、三乙胺(312μL,2.25mmol)和催化量的DMAP,室温下搅拌反应24小时。将反应液倒入100mL水中,抽滤,滤渣经水洗、干燥得粗品。再经柱层析分离(DCM:MeOH=20:1),得T7-EA白色固体236.8mg,产率73.2%。
式I化合物制备实施例3:
称取516mg(1.5mmol)T7和568.3mg(1.875mmol)EA溶于15mL四氢呋喃中,加入HBTU(711.7mg,1.875mmol)、三乙胺(520μL,3.75mmol)和催化量的DMAP,室温下搅拌反应18小时。将反应液倒入200mL水中,抽滤,滤渣经水洗、干燥得粗品。再经柱层析分离(DCM:MeOH=20:1),得T7-EA白色固体752mg,产率77.5%。
式I化合物制备实施例4:
称取258mg(0.75mmol)T7和265.2mg(0.875mmol)EA溶于10mL的N,N-二甲基乙酰胺中,加入HBTU(332.1mg,0.875mmol)、三乙胺(381μL,2.75mmol)和催化量的DMAP,室温下搅拌反应10小时。将反应液倒入100mL水中,抽滤,滤渣经水洗、干燥得粗品。再经柱层析分离(DCM:MeOH=20:1),得T7-EA白色固体391.6mg,产率80.7%。
式I化合物制备实施例5:
称取430mg(1.25mmol)T7和454.7mg(1.5mmol)EA溶于12mL的N-甲基吡咯烷酮中,加入HBTU(569.3mg,1.5mmol)、三乙胺(485μL,3.5mmol)和催化量的DMAP,室温下搅拌反应15小时。将反应液倒入150mL水中,抽滤,滤渣经水洗、干燥得粗品。再经柱层析分离(DCM:MeOH=20:1),得T7-EA白色固体645.5mg,产率79.8%。
关于式I的化合物的应用研究如下。需要说明的是,下述研究的统计学分析数据中,检测数据均用Mean±SEM来表示。Dunnet-t检验和One-way ANNOVA比较各处理组间的差异,P<0.05表示有统计学差异。
所用材料:动物和细胞株BALB/c小鼠,♀,5-6周龄,由广东省医学实验动物中心提供,鼠源黑色素瘤细胞株B16f10购于上海ATCC细胞库。
所用试剂:RPMI-1640培养基、胎牛血清均购自美国Gibco公司,0.25%胰酶购自美国Hyclone公司,IL-12、TNF-α、IFN-γELISA试剂盒均购自美国eBioscience公司;LDH非放射性细胞毒性检测试剂盒购自美国Promega公司,DMSO购自美国Sigma,小鼠淋巴细胞分离液购自北京达科为公司,PBS缓冲液购自Solarbio,CCK8试剂盒购自日本同仁公司。75%乙醇医用试剂购于国药集团化学试剂有限公司。
式I化合物研究实施例1:T7-EA体外刺激脾淋巴细胞ELISA检测IFN-γ和IL-12
将小鼠断颈处死并浸泡于75%的酒精中,5分钟后,将小鼠取出在操净台上无菌操作取小鼠的脾脏,在35mm的培养皿中研磨脾脏,培养皿中含有4-5ml的小鼠脾淋巴细胞分离液,将细胞悬液移到15ml的离心管中,沿壁轻轻加入500μl的RPMI-1640进行封闭,离心800G、30分钟,取含有淋巴细胞的那一层于含有10ml培养基的离心管中离心250G、10分钟,倒上清并将细胞沉淀用培养基吹散并计数,将脾淋巴细胞铺于24孔板,细胞密度为1×106/ml,向每孔中加入相应浓度的T7-EA(0、0.001、0.01、0.1、1、2、4、8、16、20、24μmol/L)在37℃、5%CO2饱和湿度细胞培养箱中培养24h,第2天收集上清,按照ELISA试剂盒说明书进行检测。不同浓度的T7-EA刺激脾淋巴细胞24h后,收集上清,ELISA测得的结果如图3A和3B所示,由图可知,T7-EA在低浓度(≤4μmol/L)时,活化淋巴细胞的能力较强,在高浓度时(>4μmol/L)时,T7-EA活化淋巴细胞的能力降低。
式I化合物研究实施例2:淋巴细胞增殖实验
检测T7-EA对淋巴细胞活化增殖的能力,将小鼠脱颈处死,将小鼠浸泡于75%的酒精中,在操净台上无菌取出小鼠的脾脏,在含有4-5ml小鼠脾淋巴细胞分离液的35mm培养皿中研磨脾脏,将含有淋巴细胞的细胞悬液移到15ml离心管,沿壁轻轻加入500μl RPMI-1640进行封闭,离心800G、30分钟,取含有淋巴细胞的那层于含有10ml培养基的离心管,离心250G、10分钟,倒上清用培养基将细胞吹散并进行计数,将脾淋巴细胞按每孔2×105个细胞铺于96孔板,将相应浓度的T7-EA(0、0.001、0.01、0.1、1、2、4、8、16、20、24μmol/L)加到96孔板,将96孔板放到37℃、5%CO2饱和湿度细胞培养箱中培养24h,24h后CCK8测T7-EA对淋巴细胞增殖的影响,按照CCK8试剂盒说明书进行操作。采用CCK8测T7-EA对淋巴细胞增殖的影响,实验结果如图4所示,由图可知,T7-EA在低浓度(≦4μmol/L)时促进淋巴细胞的增殖,在4μmol/L时,相对增值率在50%左右,然而,浓度较高(>4μmol/L)时对淋巴细胞几乎没有增殖的作用,但对淋巴细胞也几乎没有杀伤效应。
式I化合物研究实施例3:T7-EA体内抗肿瘤实验
24只4-5周龄的♀BALB/c小鼠在动物房SPF级检疫1周后进行荷瘤,细胞数量5×105/只,待瘤的直径达到5mm时,将长出瘤的小鼠随机分为75%乙醇对照组、T7组、EA组、T7-EA组,并对各组给予相应的药物进行治疗,各药物的用药量:T7-EA10mg/kg,T7 5mg/kg,EA12.5mg/kg,瘤周给药,每三天给药一次,共给药2周,治疗期间观察肿瘤的生长情况,每三天测量一次肿瘤大小,肿瘤面积大小按下列公式计算:S=πab(a为长径b为短径)。24只小鼠在SPF级动物房检疫7天后进行荷瘤,接种细胞个数为5×105/只,待肿瘤直径达到5mm左右时将小鼠随机分配为75%乙醇对照组、T7组、EA组、T7-EA组,3天给药一次,共给药2周,肿瘤大小每3天测量一次,测得的肿瘤面积如图5所示,由图可知,T7-EA组与对照组、T7组、EA组相比明显的抑制了肿瘤的增长,并具有统计学差异(P<0.01)。
式I化合物研究实施例4:CTL实验
细胞毒性T淋巴细胞(cytotoxic T lymphocyte,CTL)活性的测定采用乳酸脱氢酶(Lactic dehydrogenase,LDH)法,选用CytoTox96Promega试剂盒检测。将实验组的小鼠脱颈处死,取各组小鼠的脾淋巴细胞作为效应细胞,B16f10黑色素瘤细胞作为靶细胞,效应细胞和靶细胞按50:1混合培养4h,具体操作步骤按照说明书进行,杀伤率按下列公式计算:杀伤率(%)=(实验组OD值-效应细胞自发OD值-靶细胞自发OD值)/(靶细胞最大OD值-靶细胞自发OD值)×100%。给药两周后将实验小鼠脱颈处死,并取其脾脏提取淋巴细胞作为效应细胞,B16f10黑色素瘤细胞作为靶细胞,按效应细胞:靶细胞为50:1将其铺于96u型孔板中共培养4h,实验结果如图6所示,T7-EA组与PBS组相比有明显差异(P<0.05),T7-EA组与T7组、EA组相比也有明显差异(P<0.05),即表明T7-EA与对照组、T7组、EA组相比对黑色素瘤产生了较强的免疫杀伤效应。
上述研究实施例的体内实验数据表明,T7与EA偶联的化合物即T7-EA治疗黑色素瘤的效果比单独的T7、EA效果好。
本发明中新合成的式I的化合物T7-EA在大大提高EA的抗肿瘤效果上,能够激发抗肿瘤的先天性免疫和细胞免疫,达到抗肿瘤免疫协同一体化的药物设计效果,证明了式I的化合物T7-EA抗黑色素瘤的免疫反应机制。
本发明还提供另一种新合成的式II的化合物(简称T7-EA-ROR1),其结构式如下:
该式II的化合物是由式I的化合物(简称T7-EA)和第三反应物ROR1共价形成,其中第三反应物ROR1的结构式如下:

具体制备过程包括:将第三反应物溶于二甲基亚砜中,加入式I的化合物,室温搅拌8-24小时,利用液相色谱-质谱(LC-MS)联用监测反应,制备液相色谱分离,得到式II的化合物。
式II化合物的制备实施例1:
称取10mg ROR1溶于DMSO中,加入等摩尔量(1eq)的T7-EA,室温搅拌反应12h,LC-MS(液相色谱-质谱联用)监测反应,制备液相色谱分离,得T7-EA-ROR1,收率97%,MS(ESI)分子量与理论值相符(参见图7):实测值(1885.9x2)-1=3770.8;(1257.7x3)-2=3771.1;(943.5x4)-3=3771.0;理论值3771.0。
在其它实施例中,ROR1与T7-EA的搅拌反应时间还可以为8小时、15小时或24小时等,均在本发明的保护范围之内。
关于式II的化合物的应用研究如下。需要说明的是,下述研究的统计学分析数据中,数据的统计分析采用Graphpad Prism 6软件进行统计学分析,结果均以均数±标准差(X±S)表示,并采用配对t-test进行数据分析,P<0.05有统计学差异。
所用材料:BALB/c小鼠,♀,5~6周龄,由广东省医学实验动物中心提供,4T1乳腺癌细胞株购于上海ATCC细胞库。ROR1(代表ROR1蛋白的表位)由上海强耀生物科技有限公司合成,RPMI-1640培养基、胎牛血清均购自美国Gibco公司,0.25%胰酶购自美国Hyclone公司,IL-12、TNF-α、IFN-γELISA试剂盒均购自美国eBioscience公司;CytoTox96非放射性细胞毒性检测试剂盒购自美国Promega公司,BCA蛋白定量试剂盒购自南京凯基生物科技发展有限公司,DMSO购自美国Sigma公司,小鼠淋巴细胞分离液购自北京达科为公司,PBS缓冲液购自Solarbio公司,全细胞蛋白裂解液购自碧云天,兔抗鼠抗体购自Cell signalingtechnology,X-VIVO购自Lonza,细胞因子:GM-CSF,IL-4购自PeproTech公司,T7-EA由深圳大学深圳合成生物学工程实验室合成并提供。
式II化合物研究实施例1:乳腺癌相关抗原ROR1细胞毒性T淋巴细胞(cytotoxic Tlymphocyte,CTL)表位的预测及合成
在美国国立生物技术信息中心(National Center for BiotechnologyInformation,NCBI)查找ROR1的氨基酸全长序列(M937)并采用德国海德堡大学生物中心提供的MHC预测软件(网址:http://www.syfpeithi.de/bin/MHC Server.dll/ EpitopePrediction.htm)预测CD8﹢T细胞表位,列出其中6条得分较高的多肽表位序列,选取其中1条序列作为该研究表位肽,ROR1多肽由上海强耀生物科技有限公司合成。
ROR1CTL表位预测结果如表1,从预测结果中我们选取其中1条得分较高的且含有巯基的序列即PYCDETSSV作为该实验的表位肽,该序列由上海强耀生物科技有限公司合成,肽的纯度在95%以上。

式II化合物研究实施例2:ELISA检测T7-EA刺激脾淋巴细胞产生的细胞因子IFN-γ和IL-12
脱颈处死小鼠,浸泡于75%乙醇,在超净台无菌取出小鼠脾脏,35mm培养皿中放入4~5mL Mouse1×淋巴细胞分离液,并在其中研磨脾脏,把悬有脾脏细胞的分离液转移到15mL离心管,覆盖500μL RPMI-1640,室温800G,离心30min,取中间白膜层于含有10mLRPMI-1640培养基的离心管,颠倒洗涤,室温250G,离心10min,倾倒上清液,细胞沉淀用培养基均匀吹散并计数,提取的脾淋巴细胞铺于24孔板,细胞终浓度为1×106/mL,加入相应浓度的T7-EA(0、0.01、0.1、0.5、1、2、4、8、16、20、24μmol/L),在37℃、5%CO2饱和湿度细胞培养箱培养24h,第2天收上清,上清细胞因子的测定按照ELISA试剂盒说明书进行操作。如图8A和8B所示,T7-EA活化淋巴细胞并诱导其产生细胞因子IFN-γ和IL-12,T7-EA在≤4μmol/L作用效果较好,4μmol/L作为T7-EA的有效浓度。
式II化合物研究实施例3:ELISA检测T7-EA联合ROR1刺激DC产生的细胞因子TNF-α
脱颈处死小鼠,浸泡于75%乙醇,将小鼠固定在蜡板上,在超净台无菌取出小鼠的下肢股骨,1mL注射器用PBS(2%FBS)冲洗骨髓腔,冲出骨髓,BD筛网过滤,将细胞收集于15mL离心管,离心去上清(1400rpm,5min,4℃),用1mL红细胞裂解液重悬,静置1min加入完全培养基3mL终止,离心(1400rpm,5min,4℃)去上清,用含Ca2+Mg2+的1640完全培养基重悬,种于100mm培养皿,贴壁6h,吹打收集悬浮细胞并计数,以X-VIVO完全培养基重悬(细胞因子的浓度分别是GM-CSF 20ng/mL、IL-4 10ng/mL),种于24孔板,细胞终浓度为1×106/mL,在37℃、5%CO2饱和湿度细胞培养箱培养,第3天和第5天半量换液,第6天加入T7-EA、ROR1、T7-EA-ROR1刺激树突状细胞24h,T7-EA终浓度为4μmol/L,T7-EA-ROR1组按照T7-EA与ROR1摩尔比1:1混合,第7天收细胞,1400rpm,5min,4℃离心,收上清测因子TNF-α,按照ELISA试剂盒说明书进行操作。结果如图9所示,从细胞因子TNF-α的量来看,T7-EA-ROR1组与对照组PBS相比有明显差异(P<0.01),T7-EA-ROR1组与ROR1组相比也有明显的差异(P<0.01)。
式II化合物研究实施例4:ELISA检测T7-EA-ROR1刺激脾淋巴细胞产生的细胞因子IFN-γ、IL-12
提取小鼠脾淋巴细胞具体操作方法同式II化合物的研究实施例2,T7-EA终浓度为4μmol/L,T7-EA-ROR1组按照T7-EA与ROR1摩尔比1:1混合,第2天收上清测因子IFN-γ、IL-12按照ELISA试剂盒说明书进行操作。结果如图10A和10B所示,对照组PBS和ROR1组不能诱导脾淋巴细胞产生细胞因子IFN-γ和IL-12,T7-EA组、T7-EA-ROR1组诱导脾淋巴细胞产生了细胞因子IFN-γ和IL-12与对照组PBS、ROR1组相比有统计学差异(P<0.01)。
式II化合物研究实施例5:抗肿瘤动物实验
36只4~5周龄♀BALB/c小鼠在动物房SPF级检疫1周后进行荷瘤,接种乳腺癌4T1细胞数量为2×105/只,长出瘤的小鼠随机分为PBS对照组、T7组、EA组、T7-EA组、ROR1组、T7-EA-ROR1组,待瘤直径达到5mm左右,腹腔给药进行免疫治疗,每周一次,共免疫治疗4次,最后一次免疫治疗后的第6天处死小鼠取其脾脏和肿瘤。肿瘤体积大小按下列公式计算:V=ab2/2(a为长径b为短径)。实验结果如图11A-11C所示,T7-EA-ROR1组与ROR1组和对照组PBS相比肿瘤增长速度明显减慢,差异具有统计学意义(P<0.01),与T7-EA组相比,T7-EA-ROR1组肿瘤增长速度也明显减慢(P<0.05)。肿瘤重量大小与肿瘤生长情况相符,T7-EA-ROR1组肿瘤的重量明显小于PBS组(P<0.01),与T7-EA组相比,T7-EA-ROR1组肿瘤的瘤重明显减小(P<0.05),T7-EA-ROR1组肿瘤重量也明显小于ROR1组(P<0.05)。
式II化合物研究实施例6:全细胞抗体的测定
按照BCA蛋白定量试剂盒说明书操作提取乳腺癌4T1细胞的蛋白并测其浓度,提取的蛋白作为包被抗原,每次免疫治疗后的第6天采集小鼠的静脉血,并取其上清,共采集4次,血清抗体的测定按照ELISA试剂盒说明书进行操作。结果如图12所示,T7-EA组和T7-EA-ROR1组小鼠的血清中IgG抗体的水平随着免疫次数的增加逐渐升高。从抗体IgG产生的量来看,T7-EA-ROR1组与PBS组相比有显著性差异(P<0.01),与ROR1组相比T7-EA-ROR1组产生的IgG抗体有明显差异(P<0.05),由以上结果可知,T7-EA提高了ROR1的免疫原性,T7-EA-ROR1诱导机体产生了体液免疫反应。
式II化合物研究实施例7:细胞毒性T淋巴细胞(cytotoxic T lymphocyte,CTL)活性检测
CTL活性测定采用乳酸脱氢酶(lactic dehydrogenase,LDH)法,具体操作按照LDH试剂盒说明书进行。小鼠脾淋巴细胞提取的具体操作方法同式II化合物研究实施例2,提取的脾淋巴细胞为效应细胞,乳腺癌4T1细胞为靶细胞,效应细胞:靶细胞=25:1,共孵育4h,杀伤率按下列公式计算:杀伤率(%)=(实验组OD值-效应细胞自发OD值-靶细胞自发OD值)/(靶细胞最大OD值-靶细胞自发OD值)×100%。结果如图13所示,从对4T1的杀伤率来看,T7-EA-ROR1组与对照组相比效应细胞对靶细胞4T1有明显的杀伤作用(P<0.01),与T7-EA组和ROR1组相比,T7-EA-ROR1组的杀伤率明显较高(P<0.05),这说明T7-EA与ROR1相结合可以更好的激发细胞免疫,从而增强疫苗对靶细胞的杀伤性。
T7-EA刺激脾淋巴细胞产生细胞因子IFN-γ和IL-12(图8A和8B),这些数据表明,T7-EA具有激发先天性免疫的功能。其实验的结果(图9、图10A和10B)表明ROR1的免疫原性较弱不能激发先天性免疫,然而,T7-EA与ROR1结合后使疫苗更好的激发先天性免疫。肿瘤生长情况(图11A、图11C)和肿瘤重量(图11B)的结果表明,T7-EA与ROR1结合后明显减慢了肿瘤的增长。
TLR7在免疫细胞中有表达,比如巨噬细胞、树突状细胞和B淋巴细胞,TLR7与其激动剂结合后,TLR7依赖的MyD88信号通路被激活产生细胞因子,诱导活化树突状细胞,从而更好的活化B细胞、T辅助细胞,激发机体产生免疫效应。该实验的CTL结果(图13)表明T7-EA与ROR1结合后在激发先天性免疫的同时促进T细胞的活化,从而更好杀伤靶细胞,全细胞抗体效价的结果(图12)表明T7-EA-ROR1诱发机体产生了乳腺癌特异性IgG抗体。这些数据说明T7-EA与ROR1结合后,使疫苗更好激发细胞免疫和体液免疫。
综上所述,式II的化合物T7-EA-ROR1、明显减慢了4T1乳腺癌皮下移植瘤的生长,该实验证明了T7-EA-ROR1具有抗乳腺癌的作用且与激发机体的先天性免疫和适应性免疫有关,同时证明了T7-EA具有激发先天性免疫和适应性免疫的功能,也有免疫佐剂的作用。
应当理解的是,对本领域普通技术人员来说,可以根据上述说明加以改进或变换,所有这些改进或变换都应属于本发明所附权利要求的保护范围之内。
Claims (10)
1.一种式I的化合物,具有如下结构式:
2.权利要求1所述的式I化合物的制备方法,其特征在于,包括如下步骤:
S1、将第一反应物和第二反应物利尿酸溶于反应溶剂中,加入HBTU苯并三氮唑-N,N,N',N'-四甲基脲六氟磷酸盐、三乙胺和催化剂4-二甲氨基吡啶,室温下搅拌反应;
S2、反应完成后将反应液倒入水中,抽滤,滤渣经水洗、干燥得到式I的化合物。
3.根据权利要求2所述的式I的化合物的制备方法,其特征在于,在步骤S1中,所述第一反应物、第二反应物、HBTU苯并三氮唑-N,N,N',N'-四甲基脲六氟磷酸盐和三乙胺以下述摩尔比份数进行反应:第一反应物4-12份、第二反应物5-15份、苯并三氮唑-N,N,N',N'-四甲基脲六氟磷酸盐5-15份以及三乙胺18-30份。
4.根据权利要求2所述的式I的化合物的制备方法,其特征在于,在步骤S1中,所述反应溶剂选自四氢呋喃、甲苯、苯、N,N-二甲基甲酰胺、N,N-二甲基乙酰胺和N-甲基吡咯烷酮中的任意一种或多种。
5.根据权利要求2所述的式I的化合物的制备方法,其特征在于,步骤S1的反应时间为8-24小时。
6.根据权利要求2所述的式I的化合物的制备方法,其特征在于,在步骤S2后还包括步骤S3:将得到的式I的化合物经柱层析分离进行纯化,其中柱层析分离所用溶剂为二氯甲烷和甲醇的混合溶剂,二氯甲烷与甲醇的体积比为20:1。
7.权利要求1所述的式I的化合物在制备用于预防和免疫治疗黑色素瘤引起的皮肤癌的药物中的用途。
8.由权利要求1所述的式I的化合物制备得到的式II的化合物,具有如下结构式:
9.权利要求8所述的式II的化合物的制备方法,其特征在于,将第三反应物溶于二甲基亚砜中,加入式I的化合物,室温搅拌8-24小时,利用液相色谱-质谱联用监测反应,制备液相色谱分离,得到式II的化合物;
其中,第三反应物为ROR1,其结构式如下:
第三反应物与式I的化合物的反应摩尔配比为1:1。
10.权利要求8所述的式II的化合物在制备用于预防和免疫治疗乳腺癌的药物中的用途。
