本发明揭示一种2，4-二羟基苯基－苄基酮的新合成法，包括将间苯二酚和苯乙腈在无水氯化锌的存在下，在溶剂中反应，其特征在于所述的溶剂为1，2-二氯乙烷以及催化剂乙二醇二甲醚。

2，4-二羟基苯基-苄基酮的新合成法
本发明涉及2，4-二羟基苯基-苄基酮的新合成法。
2，4-二羟基苯基-苄基酮系制造治疗骨质疏松药依普黄酮的主要中间体。传统的2，4-二羟基苯基-苄基酮的合成方法是以间苯二酚和苯乙腈为原料，在无水氯化锌存在下，以乙醚为溶剂，通入干燥的氯化氢气体，反应完全后，分离乙醚后，经过水解、过滤、干燥，再经重结晶后制得，产率约50％。
上述传统的合成方法必须采用低沸点(b.b34℃)的乙醚，大量使用时危险性极大，安全得不到保障，而且收率偏低(仅50％左右)，因此，寻找一种安全可靠的溶剂和催化剂得以提高收率的合成方法是当务之急。
于是，本发明的目的在于提供一种采用安全溶剂和提高2，4-二羟基苯基-苄基酮收率的新合成法。
本发明的上述目的是通过以下方式实现的：一种2，4-二羟基苯基-苄基酮的合成方法，包括将55克间苯二酚和58.5克苯乙腈以及67.6克无水氯化锌加至400毫升1，2-二氯乙烷和催化剂乙二醇二甲醚中，所述的催化剂的体积与1，2-二氯乙烷的体积比为1/40-1/4，在搅拌和冷却下通入干燥氯化氢气体，于内温5°-6℃下反应，直到出气口有氯化氢气体逸出时，停止通气，在0℃环境下放置过夜，除去溶剂，加入425毫升水，加热沸腾二小时后，冷却、过滤、水洗、干燥，干燥物用苯重结晶，得到目的物93-95克，收率81.25-83％，熔点106°-108℃。
本发明的优点是明显的：以1，2-二氯乙烷加上一定量的催化剂取代了危险性很大的乙醚，使大规模生产2，4-二羟基苯基-苄基酮有了安全保障，而且本发明比使用乙醚时的产率大大提高，从而降低了生产成本。
以下，将通过具体实施例，对本发明作进一步的详细说明，以使本发明的优点更为突出。实施例1：
将经磨细的间苯二酚55克、苯乙腈58.5克和无水氯化锌67.6克加至400毫升1，2-二氯乙烷和催化剂25毫升乙二醇二甲醚中，在搅拌和冷却下，从液面下通入干燥氯化氢气体，直至出气口的干燥管有氯化氢气体逸出，停止反应，在0℃环境下放置过夜，除去溶剂，加入425毫升水，加热沸腾二小时，冷却后过滤、水洗、干燥，干燥物用苯重结晶，得到95克2，4-二羟基苯基-苄基酮，m.p108℃。实施例2：
重复实施例1的过程，除了用400毫升1，2-二氯乙烷和10毫升乙二醇二甲醚外，得到93克2，4-二羟基苯基-苄基酮，m.p.106℃。实施例3：
重复实施例1的过程，除了用400毫升1，2-二氯乙烷和100毫升乙二醇二甲醚外，得到95克2，4-二羟基苯基-苄基酮，m.p.106℃。

Claims (3)

1.一种2，4-二羟基苯基-苄基酮的合成方法，包括将间苯二酚和苯乙腈在无水氯化锌的存在下，在溶剂中反应，其特征在于所述的溶剂为1，2-二氯乙烷以及催化剂乙二醇二甲醚。
2.如权利要求1所述的方法，其特征在于所述的催化剂的体积与1，2-二氯乙烷的体积比为1/40-1/4。
3.如权利要求1或2所述的方法，其特征在于所述的催化剂的体积与1，2-二氯乙烷的体积比为1/16。
