本发明公开了miR‑1307‑5p或其前体、上调剂及其衍生物在制备或筛选用于治疗Ⅱ型糖尿病药物中的应用,以及在作为DPPⅣ抑制剂方面的应用;所述miR‑1307‑5p的序列如SEQ ID NO:1所示。所述miR‑1307‑5p既指含有SEQ ID NO:1所示序列的微小RNA,也指体外合成的具有SEQ ID NO:1所示序列的微小RNA的上调剂或衍生物,包括其模拟物、激活剂及保护剂等。miR‑1307‑5p及其上调剂及衍生物等可作为DPPⅣ的专一性抑制剂,用于制备或筛选治疗Ⅱ型糖尿病的有效药物,为Ⅱ型糖尿病的诊断及制备或筛选治疗药物提供了新的靶点,对Ⅱ型糖尿病的治疗有重要意义。
Description
技术领域
本发明属于生物医学技术领域, 涉及一种小分子RNA在临床诊断、药物制备及药物筛选中的应用。更具体地,涉及miR-1307-5p作为DPPⅣ抑制剂在Ⅱ型糖尿病的诊断及治疗中的作用。
背景技术
糖尿病是以持续高血糖为基本生化特征的一种慢性全身性代谢性疾病,主要是由于体内胰岛素分泌绝对缺少、身体对胰岛素的需求量增多造成的胰岛素相对不足或胰岛素抵抗而导致的以糖代谢紊乱为主的一种综合病症。根据发病原因和机制的不同可以将其分为两种类型:Ⅰ型糖尿病(T1D)和Ⅱ型糖尿病(T2D)。其中,以胰岛素分泌缺陷或(和)周围组织的胰岛素抵抗为特征的Ⅱ型糖尿病所占的比例为90~95%。相关统计数据表明,糖尿病目前的患病人数已达2.2亿,其发病数量在全球已经达到了流行性疾病的规模。近年来,随着我国经济发展及人民生活方式的改变,糖尿病的患病人数呈急剧增长的趋势,糖尿病已经成为威胁我国居民生命和健康的重要疾病之一。然而,目前尚无能治愈糖尿病的药物,糖尿病患者的长期用药也使患者家庭面临着巨大的经济挑战。糖尿病已成为亟待解决的重大公共卫生问题,也是近年来科学研究的热点。
目前,传统降糖药物主要有胰岛素增敏剂、胰岛素促分泌剂、葡萄糖苷酶抑制剂、胰岛素 5 大类,这些药物在治疗的同时往往会带来如体重增加、耐药、低血糖、水肿和乳酸中毒等不良反应。根据英国前瞻性糖尿病研究(UKPDS)结果显示无论采取何种治疗手段,糖尿病患者胰岛β细胞功能均进行性丧失。传统的降糖药物无保护胰岛β细胞功能和促其新生的作用。因此,具有新型作用机制、毒副作用小的降糖药物便应运而生。近年来,在国内外糖尿病指南中,DPPⅣ抑制剂已跻身为治疗Ⅱ型糖尿病的二线药物。与传统的抗糖尿病药物相比,DPPⅣ抑制剂疗效确切、不良反应少,适合作为Ⅱ型糖尿病的治疗药物。
DPPⅣ也叫CD26,是一种跨膜丝氨酸蛋白水解酶,是脯氨酰寡肽酶家族中的一员,其能特异性裂解肽链N末端第二位的丙氨酸或脯氨酸残基,使其失去活性。目前,已经证实GLP-1是小肠和大肠黏膜L细胞中分泌的胰高血糖素样肽,GLP-1作为一种肠源性激素,除能促进胰岛素分泌及抑制胰高血糖素分泌外,还具有增加饱腻感、减缓胃排空、抑制胰岛β细胞凋亡和促进β细胞增殖等作用。肠抑胃肽(GIP)则是由排列于肠壁的内分泌K细胞释放的另一种胃肠激素。分泌到血液中的GIP快速作用于胰岛β细胞,刺激胰岛素的释放,被认为是肠-胰岛轴的另一种主要肠降血糖素因子。GLP-1和GIP都被普遍存在的DPPⅣ(二肽基肽酶)迅速灭活,其在体内的半衰期仅有几分钟。若能抑制 DPPⅣ的活性则可以增加GLP-1及GIP的半衰期,使其血浆中的浓度达到其受体激动剂的量,进而作用于胰岛β细胞,刺激胰岛素分泌,同时也可以抑制胰高血糖素的生成,从而有利于降低血糖水平。DPPⅣ抑制剂作为新型抗糖尿病药物具有高效价的优势,它不仅具有其他抗糖尿病药物的功能,还有独特的胰岛β细胞改善修复功能,为Ⅱ型糖尿病患者的治疗带来新的希望。
miRNA是一类大小约为22nt的内源非编码RNA,其通过下调蛋白编码基因的表达在生物学的各阶段如分化、发育等过程中起重要调控作用。有大量研究表明,miRNA的异常表达与癌症、心脏病等疾病的发生与发展密切相关。近年来,随着高通量测序技术的开展与完善,发现了越来越多未知功能作用的miRNA,miR-1307便是其中之一。目前尚无关于miR-1307的功能性报道。
发明内容
本发明要解决的技术问题是克服现有Ⅱ型糖尿病治疗药物的不足,提供一种小分子RNA在临床诊断、药物制备及药物筛选中的应用,主要涉及miR-1307-5p在Ⅱ型糖尿病治疗中的应用。miR-1307-5p能特异性靶向DPPⅣ的3’UTR区抑制其蛋白表达,从而抑制DPPⅣ水解GLP-1及GIP的能力,促进了胰岛素分泌及胰岛β细胞的增殖与分化,起到降低血糖的作用。
本发明上述目的通过以下技术方案实现:
本发明公开了miR-1307-5p或其前体、上调剂及其衍生物在制备或筛选用于治疗Ⅱ型糖尿病药物中的应用,所述miR-1307-5p的序列如SEQ ID NO:1所示。
具体地,本发明公开了上述miR-1307-5p或其前体、上调剂及其衍生物在作为DPPⅣ抑制剂方面的应用。
所述miR-1307-5p由miR-1307-5p前体在对象内经加工转化得到,所述miR-1307-5p前体的序列如SEQ ID NO:2所示。
所述miR-1307-5p的上调剂是miR-1307-5p的模拟物、保护剂或激活剂。
所述miR-1307-5p的衍生物是包括miR-1307-5p种子序列在内的微小RNA序列或其他能特异性靶向DPPⅣ mRNA序列的天然或人工合成的微小RNA序列;所述miR-1307-5p种子序列如SEQ ID NO:3所示。
本发明实验发现,上述的miR-1307-5p及其前体、上调剂、衍生物能通过靶向DPPⅣ,抑制其表达进而降低水解酶活性,延长体内GLP-1及GIP半衰期,促进胰岛素分泌及胰岛β细胞的增殖与分化,抑制胰高血糖素分泌,进而降低血糖。所述miR-1307-5p或其前体在Ⅱ型糖尿病中表达水平下降。
包含有效量的miR-1307-5p或其前体的治疗和/或预防Ⅱ型糖尿病的药物也在本发明的保护范围之内。
所述药物可为单一用药或药物组合物联合用药。
单一用药时,还包括药学上可接受的载体。
药物组合物联合用药,还包括治疗和/或预防Ⅱ型糖尿病的其他活性成分,如磺脲类药物、双胍类药物、α-葡萄糖苷酶抑制剂类药物、噻唑烷二酮类药物或肠促胰岛素类药物等。
本发明经过大量实验和研究,首次发现了miR-1307-5p作为DPPⅣ抑制剂的应用,所述miR-1307-5p既指含有5’UCGACCGGACCUCGACCGGCU3’(SEQ ID NO:1)的微小RNA(此序列来自于miRBase数据库(MI0006444)),也指体外合成的具有SEQ ID NO:1所示序列的微小RNA的上调剂或衍生物,包括其模拟物、激活剂及保护剂等,以及通过构建真核细胞表达载体进行表达而获得的miR-1307的有效序列,同时也指含有能够行使miR-1307-5p功能的核心序列(SEQ ID NO:3)的微小RNA分子及其衍生物。
发明人利用生物信息学分析miR-1307-5p的潜在下游靶标后,利用荧光素酶报告基因系统验证miR-1307-5p可以与DPPⅣ基因序列的3’UTR区直接作用调控其表达。而后,将合成的miR-1307-5p模拟物(mimics)及抑制物(inhibitors)转染细胞,检测DPPⅣ的变化情况,进一步说明miR-1307-5p可以特异性靶向DPPⅣ,抑制DPPⅣ的表达,降低DPPⅣ水解GLP-1及GIP的二肽基肽酶活性,延长体内GLP-1及GIP的半衰期,提高其作用于胰岛细胞的有效浓度,进而促进胰岛素分泌,降低血糖。
综上所述,本发明实验显示,miR-1307-5p通过靶向DPPⅣ,抑制DPPⅣ(人二肽基肽酶)水解人胰高血糖素样肽-1(GLP-1)及肠抑胃肽(GIP)的活性,延长GLP-1及GIP半衰期,促进胰岛素分泌及胰岛β细胞的增殖与分化,抑制胰高血糖素分泌,进而降低血糖。因此,miR-1307-5p的低表达可作为Ⅱ型糖尿病的诊断指标,该miR-1307-5p及其衍生物可用于制备治疗Ⅱ型糖尿病的药物。
本发明具有以下有益效果:
本发明首次发现了miR-1307-5p在预防及治疗Ⅱ型糖尿病中的作用。具体来说,鉴定了其下游靶基因是调控血糖平衡的关键蛋白DPPⅣ,miR-1307-5p通过靶向能催化肠降血糖素肽GLP-1和GIP降解的重要水解酶DPPⅣ,抑制其蛋白表达,提高GLP-1及GIP的有效浓度进而提高机体内胰岛素水平,保护胰岛β细胞功能及促其新生,从而在维持人体血糖稳定方面发挥十分重要的作用。miR-1307-5p及其上调剂及衍生物等可作为DPPⅣ的专一性抑制剂,用于制备或筛选治疗Ⅱ型糖尿病的有效药物,为Ⅱ型糖尿病的诊断及制备或筛选治疗药物提供了新的靶点,对Ⅱ型糖尿病的治疗有重要意义。
同时,miR-1307-5p作为新型的核酸类DPPⅣ抑制剂,具有靶向位点专一性强,体内易自行降解等优势,可用于研发预防及治疗Ⅱ型糖尿病的有效药物。
本发明首次阐明了这种通过微小RNA对血糖进行调控继而维持人体血糖平衡的的新机制。由此,miR-1307-5p的表达水平降低可作为Ⅱ型糖尿病的临床诊断指标。

具体实施方式
以下结合说明书附图和具体实施例来进一步说明本发明,但实施例并不对本发明做任何形式的限定。除非特别说明,本发明采用的试剂、方法和设备为本技术领域常规试剂、方法和设备。
除非特别说明,本发明所用试剂和材料均为市购。
实施例1 生物信息学筛选miR-1307-5p下游靶基因
1、利用网站预测:DIANA LAB、miRDB、miRanda和PicTar查找miR-1307-5p的靶基因,并依据各基因功能(与糖代谢中参与调控血糖平衡的关系)筛选出候选靶基因。
2、结果:从众多miR-1307-5p的潜在靶基因中筛选出在糖尿病中参与调控胰岛素分泌进而影响血糖水平的候选靶基因DPPⅣ,其3’UTR上存在8个与miR-1307-5p种子序列完全互补配对的碱基。
实施例2 荧光素酶双报告基因系统检测miR-1307-5p与DPPⅣ的相互作用
1、实验方法
(1)根据预测的DPPⅣ与miR-1307-5p的碱基互补配对区域序列设计并合成野生型及突变型DPPⅣ3’UTR序列。其中所使用的酶切位点为NotⅠ及XhoⅠ,合成序列时加入相应酶切位点及保护碱基,具体序列如下表1所示。

(2)分别将合成的野生型/突变型DPPⅣ的两条单链退火形成互补双链,经酶切、连接、转化、筛选等一系列分子克隆技术,将野生型/突变型靶基因靶位点(target)序列克隆到psiCHECK-2载体(市购,本实验室保存)上的海肾荧光素酶(Renilla luciferase)之后,构建野生型/突变型靶基因DPPⅣ的3’UTR报告载体。由于psiCHECK-2载体同时表达萤火虫荧光素酶(Firefly luciferase)基因,因此将萤火虫荧光素酶的表达作为转染的内参。
将报告载体(野生型/突变型)与miR-1307-5p的模拟物(miR-1307-5p mimics)或者阴性对照(negative control mimics)共转染细胞,转染后48h裂解细胞,检测荧光信号的强度。其中,miR-1307-5p的模拟物(miR-1307-5p mimics)及阴性对照(negativecontrol mimics)序列如下表2所示。

2、结果
转染miR-1307-5p的模拟物后,DPPⅣ报告基因表达量下调63.4%,这种抑制作用可通过靶位点的突变得到回复。
实验表明了DPPⅣ是miR-1307-5p的直接靶基因。
实施例3 转染miR-1307-5p模拟物(mimics)或抑制物(inhibitors)作用48h后检测miR-1307-5p的表达效率
1、实验方法
(1)目前对于miRNA的qRT-PCR检测主要采用颈环结构的RT引物进行逆转录,然后采用实时PCR(real-time PCR)的方法进行定量检测。因此对于miR-1307-5p的检测所采用的RT引物、PCR引物的设计参照Applied Biosystems公司的方法设计合成。所用引物如下表3所示。

(2)用lipo2000分别转染50nM miR-1307-5p模拟物(miR-1307-5p mimics)/阴性对照(negative control mimics)及抑制物(miR-1307-5p inhibitors)/阴性对照(negative control inhibitors),作用48h后用Trizol提RNA。提取的RNA经逆转录后用SYBR green的方法进行荧光定量PCR检测miR-1307-5p的表达,其中,miR-1307-5p的抑制物(miR-1307-5p inhibitors)及阴性对照(negative control inhibitors)序列如下表4所示。

与阴性对照相比,转染miR-1307-5p模拟物(miR-1307-5p mimics)作用48h后,miR-1307-5p的表达量显著上调,而转染miR-1307-5p抑制物(miR-1307-5p inhibitors)后,其表达量有所下调,说明转染效率较高。
实施例4 实时定量PCR及免疫印迹检测(Western Blot)miR-1307-5p对内源靶基因的作用
1、实验方法
(1)根据DPPⅣ序列设计引物,引物序列如下表5所示。

(2)用lipo2000分别转染50nM miR-1307-5p模拟物(miR-1307-5p mimics)/阴性对照(negative control mimics)及miR-1307-5p抑制物(miR-1307-5p inhibitors)/阴性对照(negative control inhibitors)。
(3)转染48h后用Trizol提RNA及蛋白。
(4)提取的RNA经逆转录后用SYBR green的方法进行荧光定量PCR检测DPPⅣ的表达。
(5)提取的蛋白经Western Blot检测转染mir-1307-5p的模拟物及抑制物后,DPPⅣ蛋白水平的变化情况。
2、结果
转染miR-1307-5p模拟物能有效抑制DPPⅣ的表达;相反的,转染miR-1307-5p抑制物能促进DPPⅣ的表达。
实施例5 转染miR-1307-5p模拟物/抑制物后分别作用24、36、48h,检测DPPⅣ蛋白表达水平变化的趋势
1、实验方法
(1)用lipo2000分别转染50nM miR-1307-5p模拟物(miR-1307-5p mimics)/阴性对照(negative control mimics)及miR-1307-5p抑制物(miR-1307-5p inhibitors)/阴性对照(negative control inhibitors)。
(2)根据不同的作用时间点(24、36、48h)用Trizol提蛋白。
(3)提取的蛋白经Western Blot检测转染mir-1307-5p模拟物/抑制物作用不同时间后,DPPⅣ在蛋白水平的变化情况。
2、结果
转染miR-1307-5p模拟物作用36h后,其抑制DPPⅣ蛋白表达的效果最明显;而转染miR-1307-5p 抑制物对DPPⅣ蛋白表达的促进作用具有时间依赖性,随着时间的推移,其促进作用越来越显著。
综上所述,实验结果表明了miR-1307-5p能特异性靶向具有水解酶功能的二肽基肽酶DPPⅣ,抑制其水解肠降血糖素肽GLP-1和GIP的能力,延长GLP-1和GIP的半衰期。机体内较高水平的GLP-1和GIP能促进胰岛素分泌及胰岛β细胞的增殖与分化,抑制胰高血糖素分泌,进而降低血糖。miR-1307-5p作为新型的DPPⅣ抑制剂,可用于制备预防和治疗Ⅱ型糖尿病的有效药物。

1.miR-1307-5p或其前体、上调剂在制备或筛选用于治疗Ⅱ型糖尿病药物中的应用,其特征在于,所述miR-1307-5p的序列如SEQ ID NO:1所示,所述miR-1307-5p的上调剂是miR-1307-5p的模拟物、保护剂或激活剂。
2.权利要求1所述miR-1307-5p或其前体、上调剂在作为DPPⅣ抑制剂方面的应用。
3.根据权利要求1或2所述应用,其特征在于,所述miR-1307-5p由miR-1307-5p前体在对象内经加工转化得到,所述miR-1307-5p前体的序列如SEQ ID NO:2所示。
4.一种治疗和/或预防Ⅱ型糖尿病的药物,其特征在于,包含有效量的miR-1307-5p或其前体。
5.根据权利要求4所述药物,其特征在于,还包括药学上可接受的载体。
6.一种治疗和/或预防Ⅱ型糖尿病的药物组合物,其特征在于,包含有效量的miR-1307-5p或其前体。
7.根据权利要求6所述药物组合物,其特征在于,还包括一种或几种治疗或预防Ⅱ型糖尿病的其他活性成分。
8.根据权利要求7所述药物组合物,其特征在于,所述其他活性成分为磺脲类药物、双胍类药物、α-葡萄糖苷酶抑制剂类药物、噻唑烷二酮类药物或肠促胰岛素类药物。
