Abstract

本发明属于分子生物学技术领域,涉及中药及中药材鉴定领域,特别涉及一种能够特异鉴定断肠草的PCR扩增方法及其特异引物。使用此方法鉴别断肠草与金银花,只通过简单的DNA提取、PCR特异性扩增、电泳检测即可完成对样品的鉴别。具有操作简单、特异性强、重复性好等优点。主要用于断肠草与金银花之间的快速鉴定。

Description

断肠草鉴别的引物及PCR方法
技术领域
本发明属于分子生物学技术领域,涉及中药及中药材鉴定领域,特别涉及一种能够特异鉴定断肠草的PCR扩增方法及其特异引物。 
背景技术
断肠草为马前科植物钩吻Gelsemium elegans(Gardn.el Champ)Benth.的茎藤。又名胡蔓藤、野葛、水莽草、大茶叶、烂肠草,是一种剧毒草药。有祛风攻毒、消肿、止痛之功效。可治湿疹、痈肿、风湿痹痛等。 
近年来由于误服断肠草而中毒的事故在各地不断发生,对其也进行了许多研究,证实断肠草的根、茎、叶子的主要成分为钩吻素子、钩吻素寅、钩吻素卯等。调查发现误将断肠草为金银花煮水当凉茶喝而中毒的事件时有发生,在断肠草中毒的78例临床事件中,发现43例为因误将断肠草为金银花煮水当凉茶喝而中毒,18例为将断肠草根为其他草药煮水喝而中毒(李文升.急性断肠草中毒78例临床分析,医学信息,2011,24(9):5872)。因此,建立一种用于鉴别钩吻和金银花的方法非常重要。 
黄伟雄等采用色谱的方法通过分析钩吻碱成分对断肠草中进行鉴定(黄伟雄,李汴生,范山湖,等.应用TLC法鉴定断肠草中钩吻碱,华南预防医学,2008,34(1):60),但这种方法比较繁琐,不利于推广。近年来发展的以分子生物学为基础的检测方法如DNA barcoding和位点特异性PCR等克服了药材传统鉴别方法准确度不高、主观性大等缺点。对断肠草与金银花之间的分子鉴别还尚未见报道,本发明通过特异性PCR的方法实现了断肠草与金银花的快速、准确鉴别。在本发明被公布之前,尚未有任何公开或报道过本专利申请中所提及的PCR方法及引物。 
发明内容
本发明的目的是提供一种断肠草的PCR引物及其鉴定方法,该PCR鉴定方法操作简单、样品量少,能快速、准确的将断肠草从其易混淆的金银花样品中鉴定出来,为断肠草鉴定提供了一种实用的技术。 
1、一种鉴定断肠草的特异引物F1、F2,其序列为: 
F1:5’-TCTCCGCCCTCTGCTCTA-3’, 
F2:5’-GGAAGAAAGGAGTTTTTATGAGG-3’。 
2、一种断肠草的PCR鉴定方法,其特征在于,包括下列步骤: 
a)从所述材料中提取到DNA样品; 
b)用权利要求1所述的引物进行PCR反应,扩增反应体系包括:10×buffer缓冲液2μL,dNTP1.6μL,引物各0.5μL,EX Tap酶0.2μL,DNA0.5μL,灭菌蒸馏水14.7μL。反应条件:解链温度95℃,时间5min,退火温度57℃,时间30s,复性温度72℃,时间为1min40s,35个循环后72℃延伸,时间为7min; 
c)电泳所述扩增产物,如果存在分子量约为300bp的单一条带,则确定所检材料为断肠草。

具体实施方式
1材料 
所用材料如表1所示,包括:

2鉴别引物设计 
选择通用引物psbA-trnH(t rnH:5’-CGC GCA TGG TGG ATT CAC AAT CC-3’psbA:5’-GTT ATG CAT GAA CGT AAT GCT C-3’)对断肠草样品进行扩增,搜索Genbank中金银花的psbA-trnH序列(Genbank登录号:GU1353313.1;JN045253.1),然后进行同源比对,根据其变异区设计一对特异引物F1、F2,序列为下: 
F1:5'-TCTCCGCCCTCTGCTCTA-3’,F2:5’-GGAAGAAAGGAGTTTTTATGAGG-3’。 
3DNA提取 
选取无霉变的干燥药材约0.03g,置于粉碎机中研磨粉碎,过40目筛。将粉末转移到2.0mL的微量离心管中,加入900μL已灭菌的CTAB提取液(2%CTAB,100mmol/L Tris-HCl pH=8.0,20mmol/L EDTA,1.4mol/L NaCl)、0.02g PVP40000、10μL β-巯基乙醇充分振荡混匀,65℃水浴1.5h-2h,期间轻摇2-3次。结束后取出冷却至室温,加入900μL氯仿-异戊醇(24:1),充分振荡混匀,12000×g离心10min。取上清,加入等体积氯仿-异戊醇(24:1),充分振荡混匀,12000×g离心10min。取上清,加入2/3体积预冷的异丙醇溶液,-20℃放置0.5h以上。取出,12000×g离心10min,弃上清,沉淀用70%乙醇洗涤两次,37℃挥干乙醇,用适量灭菌水溶解,-20℃保存。 
4PCR扩增 
PCR反应在200μLPCR反应管中进行,反应总体积20μL,包括以下试剂:2.0μL10×buffer缓冲液,1.6μL dNTP(2.5mM),0.25μL鉴别引物F1(5μM),0.25μL鉴别引物F2(5μM),0.25μL通用引物1F(5μM),0.25μL通用引物724R(5μM),0.2μLTaqDNA聚合酶(Takara公司,Extaq5UμL-1),0.5μL模板DNA,14.7μL无菌双蒸水。 
PCR反应液配制完后,轻轻震荡混匀,将PCR管放入PCR仪中,进行如下反应:解链温度95℃,时间5min,退火温度57℃,时间30s,复性温度72℃,时间为1min40s,35个循环后72℃延伸7min,获得扩增产物。取3μL扩增产物,采用1%的琼脂糖凝胶,在电压100-150V下电泳15分钟,凝胶成像系统下观察并拍照。 
断肠草与金银花的电泳结果如图1,结果可见,断肠草能够扩增出片段大小分别约为1000bp和300bp的两条带,而金银花则只扩增出片段大小约为300bp的单一条带,表明该体系能将断肠草与金银花准确的鉴别出来。 

Claims (2)

1.一种鉴定断肠草的特异引物F1、F2,其序列为:F1:5'-TCTCCGCCCTCTGCTCTA-3’,F2:5'-GGAAGAAAGGAGTTTTTATGAGG-3’。
2.一种断肠草的PCR鉴定方法,其特征在于,包括下列步骤:
a)从所述材料中提取到DNA样品;
b)用权利要求1所述的引物进行PCR反应,扩增反应体系包括:10×buffer缓冲液2μL,dNTP1.6μL,引物各0.5μL,EX Tap酶0.2μL,DNA0.5μL,灭菌蒸馏水14.7μL;反应条件:解链温度95℃,时间5min,退火温度57℃,时间30s,复性温度72℃,时间为1min40s,35个循环后72℃延伸,时间为7min;
c)电泳所述扩增产物,如果存在分子量约为300bp的单一条带,则确定所检材料为断肠草。
