一种原花青素的提取工艺属于药物加工领域,更具体地说,是涉及一种原花青素的提取工艺。本发明提供了一种低成本、高效、无污染的原花青素提取工艺。本发明的工艺步骤为:将荔枝果皮清洗、干燥,干燥后的荔枝皮用粉碎机粉碎成粉末,将荔枝皮粉末加到55~75%的乙醇溶液中进行水浴提取,提取温度为60~80℃,提取时间为75~140min,然后将提取液过滤后,用旋转蒸发仪减压回收乙醇,水相上AB-8大孔吸附树脂,采用8倍量蒸馏水和60%乙醇分布洗脱,洗脱液在45℃真空浓缩,冷冻干燥成粉末。

技术领域
本发明属于药物加工领域,更具体地说,是涉及一种原花青素的提取工艺。
背景技术
荔枝是中国南方特产水果,因其营养丰富,味甜肉嫩,品质极优而被誉为“中华之珍品”。荔枝含有丰富的糖分、蛋白质、多种维生素、脂肪、柠檬酸、果胶以及磷、铁等,对大脑组织有补养作用,能明显改善失眠、健忘、神疲等症,有助于增强机体免疫功能,提高抗病能力,是对人体有益的水果。在荔枝的工业化生产中,大量荔枝皮作为副产品没有得到有效的开发和利用,常常当作废弃物丢弃,其实荔枝皮也具有很高的生化活性,富含丰富的花色素类、原花色素类、脂肪族化合物及多种营养成分。
原花青素是一种有着特殊分子结构的生物类黄酮,是目前国际上公认的清除人体内自由基最有效的天然抗氧化剂。一般为红棕色粉末,气微、味涩,溶于水和大多有机溶剂。
原花青素是一种新型高效抗氧化剂,是目前为止所发现的最强效的自由基清除剂,具有非常强的体内活性,它能够保护人体的血液循环系统;阻抑胶原酶和弹性蛋白酶对取缔组织的降解作用,因而有利于保持皮肤的弹性,发挥抗皮肤衰老的功效;可以增强维生素C 的功效,延长维生素C 在人体内发生功效的时间。
发明内容
本发明就是针对上述问题,提供了一种高效、无污染的原花青素提取工艺。
为了实现本发明的上述目的,本发明的工艺步骤为:将荔枝果皮清洗、干燥,干燥后的荔枝皮用粉碎机粉碎成粉末,将荔枝皮粉末加到55~75%的乙醇溶液中进行水浴提取,提取温度为60~80℃,提取时间为75~140min,然后将提取液过滤后,用旋转蒸发仪减压回收乙醇,水相上AB-8 大孔吸附树脂,采用8倍量蒸馏水和60%乙醇分布洗脱,洗脱液在45℃真空浓缩,冷冻干燥成粉末。
本发明的有益效果:
1. 本发明对荔枝皮进行了再利用,既减少了环境的污染,又实现了对资源的全面利用;
2. 本发明所用的原料为荔枝皮,大大降低了生产成本;
3. 本发明对原花青素的提取率达到了1.0%。
具体实施方式
本发明的工艺步骤为:将荔枝果皮清洗、干燥,干燥后的荔枝皮用粉碎机粉碎成粉末,将荔枝皮粉末加到55~75%的乙醇溶液中进行水浴提取,提取温度为60~80℃,提取时间为75~140min,然后将提取液过滤后,用旋转蒸发仪减压回收乙醇,水相上AB-8 大孔吸附树脂,采用8倍量蒸馏水和60%乙醇分布洗脱,洗脱液在45℃真空浓缩,冷冻干燥成粉末。
作为一种优选方案,工艺参数为:乙醇浓度72%,提取温度58℃,提取时间为110min。
原花青素由不同数目的儿茶素、表儿茶素及其衍生物聚合而成,可采用甲醇、乙醇和丙酮来提取荔枝皮原花青素,三种溶剂均可不同程度地渗透植物细胞,与原花青素相结合,将其溶出。三种溶剂的提取效果相差不大,但由于甲醇和丙酮的毒性比较高,提取后如果除去不完全,会对人体造成伤害,因此本发明这里选用乙醇作为荔枝皮原花青素的提取溶剂。
乙醇浓度、提取温度和提取时间均对原花青素的提取效果有影响,可见表1。

1. 一种原花青素额提取工艺,其特征在于本发明的工艺步骤为:将荔枝果皮清洗、干燥,干燥后的荔枝皮用粉碎机粉碎成粉末,将荔枝皮粉末加到55~75%的乙醇溶液中进行水浴提取,提取温度为60~80℃,提取时间为75~140min,然后将提取液过滤后,用旋转蒸发仪减压回收乙醇,水相上AB-8 大孔吸附树脂,采用8倍量蒸馏水和60%乙醇分布洗脱,洗脱液在45℃真空浓缩,冷冻干燥成粉末。
2. 根据权利要求1 所述的一种原花青素的提取工艺,其特征在于工艺参数为:乙醇浓度72%,提取温度58℃,提取时间为110min。
