菊芋性味甘平,无毒,能利水去湿,和中益胃,有清热解毒的作用。野生菊芋广泛存在于我国境内,深入开发菊芋可使资源高值化,土地资源利用高效化。菊芋新鲜块茎富含菊芋多糖,是一种低热量,在人体内可延长碳水化合物供能时间,而又不显著提高血糖水平,不需要胰岛素代谢的碳水化合物。菊芋多糖可降低血糖调血脂等,具有多种保健功能和优良的食用价值,已被世界上多个国家批准为食品的营养增补剂,以其与其它药食两用物品及中药保健物品相配伍,可以增强其保健功效,益智健脑,更可以有效地减缓减少中老年疾病发病几率,提高生活质量。
本发明的特征是以菊芋块茎为原料,经过提取与纯化得到菊芋多糖,再按一定比例与其它原料相配伍制成以菊芋多糖为主的一种健脑益智辅助调节血糖的口服片剂即菊芋多糖益脑片,其制剂具有消渴降血糖等多种功效,实为绿色的保健用品,适宜于工业化生产,过程无三废。
发明内容
本发明通过对菊芋块茎提取、纯化得到菊芋多糖,再与五味子、天麻、葛根相配伍,制成了一种可供口服的能改善睡眠,健脑益智且辅助调节血糖的菊芋多糖益脑片。明确提出了是以菊芋多糖为主原料,适于中老年及糖尿病人服用的新食品保健制剂。其特征在于含菊芋多糖25-50%(wt),其它处方原料含量为50-75%(wt)。本发明为功能性食用保健品的新型制剂,可以口服的咀嚼片,具有降低血糖等多种辅助功效。
具体实施方式
实例1
1).菊芋多糖的提取与纯化:称取粉碎的菊芋颗粒100g,在室温下加入到2600-3000ml水中;放置数小时后,用超声室温20-25℃提取多糖,时间5-15min,或直接水提取温度80-90℃,回流时间30-45min,抽滤取滤液,残渣再以上述条件重复提取1-2次,合并提取液体积,得到菊芋多糖提取液密度1.004-1.007g/cm3;提取液用旋转蒸发仪浓缩,浓缩温度50-60℃,浓缩真空0.085-0.095MPa,浓缩比1∶6-1∶8,浓缩液密度1.033-1.047g/cm3;将其浓缩液放入恒温水浴中,加入天然澄清剂ZTC1+1 B组份的量为浓缩液体积份数的1-3%,在温度为50-60℃下保温时间30-40min;再降温到45-55℃,加入天然澄清剂ZTC1+1 A组份(A与B的体积比为1∶2-1∶3),加入A组份后再保温15-25min;之后经过滤得到纯化后的液体,按纯化液∶乙醇=1∶4的醇沉比(体积比)加入92%乙醇,放置24小时后抽滤,所得固体再经低温干燥,即得白色菊芋多糖粉末43-49g。
2).菊芋多糖益脑片的制备:
配方原料,辅料,配方比,制备方法,配方原理如下:
原料配比:菊芋多糖250-500g,五味子100-150g,天麻235-302g,葛根100-250g。
辅料配比:崩解剂为羧甲基淀粉钠48-65g;润滑剂硬脂酸镁占软材颗粒的1-1.3%(wt);黏合剂:PVPK30乙醇溶液浓度0.5-2.5%(wt),体积为666-867ml。
制备方法:按比例取原料及辅料菊芋多糖250g,五味子150g,天麻302g,葛根250g,羧甲基淀粉钠48g,分别粉碎研磨过120目筛备用;用等量递加内加法将其置于混合机内混合均匀,再按一定比例滴加黏合剂666ml制成软材;将软材置于颗粒机中过16目筛制成颗粒,低温下干燥;将干燥后的颗粒经40目筛整粒,加入硬脂酸镁质量分数为软材颗粒质量的1%(wt),于压片机内压1000片,即得。产品经检测符合2010版药典规定的制剂标准要求。
配方原理:
菊芋多糖--有降低血糖促进糖分分解使过剩糖分转化为热量的作用,改善体内脂肪平衡。
五味子--收敛固涩,益气生津,补肾宁心。用于久嗽虚喘,梦遗滑精,遗尿尿频,久泻不止,自汗盗汗,津伤口渴,内热消渴,宁心安神,心悸失眠。
天麻--息风止痉,平抑肝阳,祛风通络。用于助阳气,行气活血,头痛眩晕,肢体麻木,明目增强记忆,临床用于改善睡眠,增氧,补脑益智等。
葛根--解肌退热,生津止渴,通经活络,解酒毒。用于清热解毒,项背强痛,口渴,消渴,眩晕头痛,酒毒伤中。
实例2
操作过程与实例1基本相同,不同的操作参数和工艺条件如下:
菊芋粉100g,一次性加入6000ml水中。加完后,在反应温度80-90℃的条件下水提取菊芋多糖,提取时间30min,提取结束减压过滤得提取液11200ml,并经负压浓缩后得浓缩液1600ml,在温度55℃条件下加入天然澄清剂ZTC1+1 B组份纯化35min,再于温度45℃条件下加入天然澄清剂ZTC1+1 A组份纯化15min,纯化结束减压过滤得滤液1100ml,并通过90%(wt)乙醇醇沉,低温干燥得白色菊芋多糖粉末51g。
按比例取原料及辅料菊芋多糖500g,五味子100g,天麻235g,葛根100g,羧甲基淀粉钠65g;分别经粉碎,研磨过120目筛备用;用等量递加内加法将其置于混合机内混合均匀,再按一定比例滴加黏合剂867ml制成软材;将软材置于颗粒机中过16目筛制成颗粒,低温下干燥;将干燥后的颗粒经40目筛整粒,加入硬脂酸镁质量分数为软材颗粒质量的1.3%(wt),于压片机内压1000片,即可。产品经检测符合2010版药典规定的制剂标准要求。

1.菊芋多糖益脑片的制备,其特征是由以下过程和步骤组成:
(1)菊芋多糖的提取
选取阴干或低温风干贮存的菊芋块茎或片进行粉碎成颗粒。准确称量粉碎后的菊芋颗粒100g,在室温下加入到2600-3000ml水中;放置数小时后用超声室温20-25℃提取多糖时间5-15min;抽滤取滤液,残渣再以上述条件重复提取1-2次,合并提取液体积,得到提取液密度1.004-1.007g/cm3;再用旋转蒸发仪浓缩至体积比1∶5-1∶7,浓缩温度50-60℃,浓缩真空0.085-0.095MPa,浓缩液密度1.033-1.047g/cm3。或采用菊芋粉操作条件同上述,超生提取菊芋多糖;或在温度50-60℃,时间30min用水回流提取菊芋多糖。
(2)菊芋多糖的纯化
将(1)中的浓缩液放入恒温水浴中,加入天然澄清剂ZTC1+1A、B组份纯化,纯化后经过滤得到的液体850-870ml,按纯化液∶乙醇=1∶4的醇沉比(体积比)加入92%乙醇放置24小时后抽滤,所得固体再经低温干燥,即得纯化后的白色菊芋多糖粉末43-49g。
(3)菊芋多糖益脑片是由植物菊芋多糖250-500g、五味子100-150g、天麻235-302g、葛根100-250g相配伍,羧甲基淀粉钠48-65g为崩解剂,以硬脂酸镁1-1.3%为润滑剂,PVPK30乙醇溶液666-867ml为黏合剂,压片制成。
2.根据权利要求1所述的菊芋多糖提取方法,可以采用超声或水对菊芋颗粒或粉进行菊芋多糖的提取。
3.根据权利要求1所述的得到的菊芋多糖,可分别经天然澄清剂ZTC 1+1A、B组份纯化,并经乙醇进行醇沉制得。

5.根据权利要求1所述的菊芋多糖益脑片,其特征在于该产品形状为片剂,稳定性好,贮存服用方便,无毒可辅助降低血糖作为食品保健使用。
6.根据权利要求1所述的菊芋多糖益脑片,其特征在于采用菊芋块茎提取,纯化得菊芋多糖;再经原料、辅料相配伍制得一种菊芋多糖益脑片;并经副产物回收工艺三个工序过程,使整个生产过程无三废产生,实现了成本最低化、效益最大化的绿色工艺。
