Abstract
本发明涉及一种治疗骨质增生的中药组合物及其制备方法。该中药组合物是由以下原料制备而成:络石藤、枫荷梨、海风藤、扶芳藤、走马胎、黄水倒水莲、紫苏。该中药组合物具有祛风除湿、舒经通络、活血止痛的作用,对骨质增生有较好疗效。

技术领域
本发明涉及中药领域,具体涉及一种治疗骨质增生的中药组合物及其制备方法。
背景技术
骨质增生症又称为增生性骨关节炎、骨性关节炎等,是由于构成关节的软骨、椎间盘、韧带等软组织变性、退化,关节边缘形成骨刺,滑膜肥厚等变化,而出现骨破坏,引起继发性的骨质增生,导致关节变形,当受到异常载荷时,引起关节疼痛,活动受限等症状的一种疾病。骨质增生分原发性和继发性两种。
目前治疗骨质增生症的方法有很多,治疗都存着者局限:一、直流电药物离子导入法。直流电药物离子导入法充分发挥了药物的作用,但因导入药物为液体,易挥发,药效不持久,限制了该方法的治疗效果。二、紫外线疗法:其治疗作用主要表现在以下几个方面:(1)杀菌作用;(2)消炎作用;(3)止痛作用;(4)促进伤口愈合作用;(5)脱敏作用;(6)促进维生素D3的形成;(7)调解机体免疫功能等。三、西药治疗。目前西医对本症尚无有效的治疗药物,常采用对症处理,如疼痛时可服一些解热镇痛的药;麻木者可选用B族维生素类药物;关节肿胀有积液者可给予局部抽取积液或局部封闭等疗法。但实践证明这些治疗方法均不理想,只是治标而不治本,病情易复发。四、手术治疗:手术治疗不是骨质增生的首选疗法。当选用保守治疗无效且病情较重、严重影响患者生活时,可考虑手术治疗。
骨质增生症属于中医的“痹症”、“骨痹”的范畴,此病与外伤、劳损、风寒湿邪、淤血阻滞、痰湿内阻、肝肾亏虚等有关。
肝肾亏虚:就是说,肝肾不足,无力保护骨髓、充养骨髓,时间久了,关节在反复的活动过程中,渐渐地受到损害而过早过快地出现退化。
外伤与劳损:外伤和劳损引起受力最集中的关节局部发生气血逆乱,严重的导致筋损骨伤,血流不循常道而溢于脉外形成瘀血凝滞,导致关节骨骼结构受损,失去滋养,时间一久,退行性疾病便会出现。
外感风寒湿邪:是指感受风寒、着凉或久居潮湿之地、冒雨涉水等。这一条肯定不是引起李大爷骨质增生的原因。他家在北方,又住楼房的阳面房间,一点不潮湿,而且平时他也非常注意保暖。
痰湿内阻:一般阳虚多痰,容易患骨质增生。而体胖之人还加重了关节的负担,更容易造成关节局部血运不畅,筋骨失养,久则成痹。
中医学认为:肾主骨,肝主筋,筋附于骨。老年人由于肝肾功能衰退,肾虚不能主骨,肝虚不能养筋,再加上风寒湿邪的侵袭,或是外伤,导致气血失和,久而久之痰血加重,就会形成骨质增生,从而产生疼痛不适、肢体麻木、活动障碍等症状。因此,骨质增生的中医药治疗一般采用补肝肾、强筋骨,祛风除湿,活血的治疗原则。
发明内容
本发明的目的是提供一种治疗骨质增生的中药组合物;
本发明的另一目的是提供一种治疗骨质增生的中药组合物制备方法。
本发明的目的是通过下列的技术方案实现的:
本发明所述的治疗骨质增生的中药组合物,它是由包括如下重量份的原料制备而成的:
络石藤6-20份、枫荷梨25-50份、海风藤6-20份、扶芳藤6-20份、走马胎15-40份、黄水倒水莲20-40份、紫苏5-20份。
优选地,是由包括如下重量份的原料制备而成的:
络石藤10-15份、枫荷梨30-40份、海风藤10-15份、扶芳藤10-15份、走马胎20-30份、黄水倒水莲25-35份、紫苏10-15份。
更优的,是由包括如下重量份的原料制备而成的:
络石藤12份、枫荷梨35份、海风藤12份、扶芳藤12份、走马胎25份、黄水倒水莲30份、紫苏12份。
以下是本发明中药组合物的药材来源:
络石藤,为夹竹桃科植物络石Trachelospermum jasminoides (Lindl.)Lem. 的干燥带叶藤茎。苦,微寒。归心、肝、肾经。祛风通络,凉血消肿。用于风湿热痹,筋脉拘挛,腰膝酸痛,喉痹,痈肿,跌扑损伤。
枫荷梨,为五加科树参属植物树参Dendropanax chevalieri (Vig.) Merr. et Chun,以根或枝叶入药。甘,温。祛风除湿,舒筋活血。用于偏头痛,臂丛神经炎,风湿性关节炎,类风湿关节炎,腰肌劳损,慢性腰腿痛,半身不遂,跌打损伤,扭挫伤;外用治刀上出血。
海风藤,为胡椒科植物风藤Piper kadsura (Choisy) Ohwi的干燥藤茎。辛、苦,微温。归肝经。祛风湿,通经络,止痹痛。用于风寒湿痹,肢节疼痛,筋脉拘挛,屈伸不利。
扶芳藤,卫矛科卫矛属植物扶芳藤Euonymus fortunei (Turcz.) Hand.-Mazz.,以茎、叶入药。苦、甘,温。散瘀止血,舒筋活络。用于咯血,月经不调,功能性子宫出血,风湿性关节痛;外用治跌打损伤,骨折,创伤出血。
走马胎,为紫金牛科植物走马胎Ardisia gigantifolia stapf的干燥根。苦、微辛,温。祛风化湿,消肿止痛。根:用于风湿性关节炎,腰腿痛,跌打肿痛;叶:外用治扭伤,痈疖肿毒,慢性溃疡。
黄水倒水莲,毛茛科唐松草属植物峨眉唐松草Thalictrum omeiense W. T. Wang et S. H. Wang,以全草入药。苦、涩,寒。清热,燥湿,止痢。用于湿热黄疸,腹痛泻痢,疟疾寒热,目赤肿痛。
紫苏,唇形科紫苏属植物紫苏Perilla frutescens (L.)Britt.的带枝嫩叶。辛,温。散寒解表,理气宽中。用于风寒感冒,头痛,咳嗽,胸腹胀满。
本发明的另一方面是提供了本发明中药组合物的活性成分的制备方法,该方法是采用采用水提或40-80%体积百分浓度的乙醇提取制备而成。具体制备方法如下:
方案一:取全部7味药材,混合后加水提取2-3次,每次加水量相当于药材总重量的6-12倍,每次提取时间为1-3小时,合并提取液,滤过,滤液浓缩至70-80℃时相对密度为1.15-1.20的清膏,即得活性成分。
方案二:取全部7味药材,混合后用40-80%乙醇回流提取2-3次,每次乙醇用量为药材总量的4-10倍,提取时间为1-4小时,合并提取液,过滤,滤液浓缩至70-80℃时相对密度为1.15-1.20的清膏,即得活性成分。
方案三:取全部7味药材,混合后加水提取2-3次,每次加水量相当于药材总重量的6-12倍,每次提取时间为1-3小时,合并提取液,滤过,滤液浓缩至70-80℃时相对密度为1.15-1.20的清膏,加入乙醇,使含醇量为40-70%,静置12-24小时,滤过,滤液浓缩至70-80℃时相对密度为1.10-1.25的浸膏,即得活性成分。
本发明所述的治疗骨质增生的中药,还可以与医药学上可接受的载体配合,制成各种口服制剂,如片剂、胶囊剂、口服液、糖浆剂、合剂等。
本发明的药学上可接受的载体包括但不限于以下:
口服制剂:
稀释剂:淀粉、糖粉、乳糖、糊精、微晶纤维素、无机盐、糖醇类等。
润湿剂与粘合剂:纯化水、乙醇、明胶、聚乙二醇、纤维素衍生物等。
崩解剂:淀粉、羧甲基淀粉钠、纤维素衍生物、交联聚维酮等。
润滑剂:硬脂酸镁、微粉硅胶、滑石粉、聚乙二醇等。
助溶剂:水、乙醇、甘油、丙二醇、液状石蜡、植物油等。
矫味剂:蔗糖、单糖、芳香剂等。
防腐剂:苯甲酸、山梨酸、甲酯、乙酯、丙酯等。
方法1:片剂
取方案一至方案三所得活性成分之一,加入片剂常用辅料,按常规生产方法制备得本发明片剂。
上述片剂常用辅料包括稀释剂、润湿剂、粘合剂、崩解剂、润滑剂之一或全部。
方法2:胶囊剂
取方案一至方案三所得活性成分之一,加入胶囊剂常用辅料,按常规生产方法制备得本发明胶囊剂。
上述胶囊剂常用辅料包括稀释剂、润湿剂、粘合剂、崩解剂、润滑剂之一或全部。
方法3:糖浆剂
取方案一至方案三所得活性成分之一,加入糖浆剂常用辅料,按常规生产方法制备得本发明糖浆剂。

方法4:合剂
取方案一至方案三所得活性成分之一,加入合剂常用辅料,按常规生产方法制备得本发明合剂。

本申请人在研究传统中医辨证治疗的基础上参考现代药理研究成果,经过多年的试验,将多种配方进行筛选,找到了能有效治疗骨质增生的中药配方。
本发明所公开的中药组合物有效成分是由络石藤、枫荷梨、海风藤、扶芳藤、走马胎、黄水倒水莲、紫苏制备而成。方中络石藤祛风通络,凉血消肿;枫荷梨祛风除湿,舒筋活血;海风藤祛风通络止痛;扶芳藤舒筋活络;走马胎祛风化湿,消肿止痛;黄水倒水莲清热燥湿;紫苏发散风寒。全方相互协同,具有祛风除湿、舒经通络、活血止痛的作用,对骨质增生有较好疗效。
本发明提供的一种治疗骨质增生的中药组合物具有以下优点:
1、本发明提供的组合物制剂安全有效,具有祛风除湿、舒经通络、活血止痛的作用,对骨质增生有较好疗效。
2、经临床试验证明,患者经本发明所述中药组合物治疗后,骨质增生患者的临床症状有明显改善,总有效率达91.1%。
3、本发明提供的中药组合物制剂,配方科学合理,工艺简单,口感好,成本低。
具体实施方式
下面通过实施例进一步说明本发明。应该理解的是,本发明的实施例是用于说明本发明而不是对本发明的限制。根据本发明的实质对本发明进行的简单改进都属于本发明要求保护的范围。除非另有说明,本发明中的乙醇量的百分数是体积百分数,v/v表示溶液的体积比。
实施例1 口服液
称取络石藤0.6kg、枫荷梨2.5kg、海风藤0.6kg、扶芳藤0.6kg、走马胎1.5kg、黄水倒水莲2kg、紫苏0.5kg。取7味药材,加水提取2次,两次加水量分别为药材总重量的10倍、8倍,两次提取时间分别为2小时、1小时,合并提取液,滤过,滤液浓缩至80℃时相对密度为1.10的浸膏,在浸膏中加入辅料甘露醇、山梨酸、水搅拌均匀,分装,制得口服液。
实施例2 片剂
称取络石藤2kg、枫荷梨5kg、海风藤2kg、扶芳藤2kg、走马胎4kg、黄水倒水莲4kg、紫苏2kg。取7味药材,用40%乙醇回流提取3次,每次乙醇用量分别为药材总量的10倍、8倍、4倍,提取时间分别3小时、2小时、1小时,合并提取液,过滤,滤液浓缩60℃时相对密度为1.25的浸膏,加入辅料淀粉、乙醇、滑石粉,制得片剂。
实施例3 颗粒剂
称取络石藤1kg、枫荷梨3kg、海风藤1kg、扶芳藤1kg、走马胎2kg、黄水倒水莲2.5kg、紫苏1kg。取7味药材,加水提取3次,三次加水量分别相当于药材总重量的12倍、8倍、6倍,每次提取时间分别3小时、2小时、1小时,合并提取液,滤过,滤液浓缩至70℃时相对密度为1.20的清膏,加入乙醇,使含醇量为40%,静置24小时,滤过,滤液浓缩至滤液浓缩至70℃时相对密度为1.10的浸膏,加入淀粉、糊精,制粒、干燥、整粒即得。
实施例4 胶囊剂
称取络石藤1.5kg、枫荷梨4kg、海风藤1.5kg、扶芳藤1.5kg、走马胎3kg、黄水倒水莲3.5kg、紫苏1.5kg。取7味药材,用乙醇回流提取2次,每次乙醇浓度分别为80%、60%,每次乙醇用量分别为药材总量的10倍、8倍,提取时间分别为4小时、3小时,合并提取液,过滤,滤液浓缩至80℃时相对密度为1.10的浸膏,在浸膏中加入辅料淀粉、乙醇、糊精、硬脂酸镁,按常规方法制成胶囊剂。
实施例5 片剂
称取络石藤1.2kg、枫荷梨3.5kg、海风藤1.2kg、扶芳藤1.2kg、走马胎2.5kg、黄水倒水莲3kg、紫苏1.2kg。取7味药材,三次加水量分别为药材总重量的12倍、8倍、6倍,三次提取时间分别为3小时、2小时、1小时,合并提取液,滤过,滤液浓缩至70℃时相对密度为1.25的清膏,加入辅料淀粉、乙醇、滑石粉,制得片剂。
实施例6 颗粒剂
称取络石藤1.5kg、枫荷梨3kg、海风藤1kg、扶芳藤1.5kg、走马胎3kg、黄水倒水莲2kg、紫苏1kg。取7味药材,每次加水量分别相当于药材总重量的12倍、10倍,每次提取时间分别为3小时、2小时,合并提取液,滤过,滤液浓缩至80℃下相对密度为1.05的清膏,加入乙醇,使含醇量为70%,静置12小时,滤过,滤液浓缩至70℃时相对密度为1.25的浸膏,加入淀粉、糊精,制粒、干燥、整粒即得。
实施例7 糖浆剂
称取络石藤1kg、枫荷梨2.5kg、海风藤1.5kg、扶芳藤1kg、走马胎2kg、黄水倒水莲3.5kg、紫苏1.5kg。取7味药材,两次加水量分别为药材总重量的10倍、8倍,两次提取时间分别为2小时、1小时,合并提取液,滤过,滤液浓缩至80℃时相对密度为1.10的浸膏。在浸膏中加入糖浆剂辅料蔗糖、山梨酸、水搅拌均匀,制得糖浆剂。
实施例8 合剂
称取络石藤1.4kg、枫荷梨4kg、海风藤0.8kg、扶芳藤0.7kg、走马胎2.3kg、黄水倒水莲3.2kg、紫苏1.3kg。取7味药材,三次加水量分别相当于药材总重量的12倍、8倍、6倍,每次提取时间分别3小时、2小时、1小时,合并提取液,滤过,滤液浓缩至总重量为药材总重量的3倍,灌装,加入合剂辅料蔗糖、乙酯、水、山梨酸搅拌均匀,制得合剂。
实施例9 粉剂
称取络石藤1.4kg、枫荷梨4kg、海风藤0.8kg、扶芳藤0.7kg、走马胎2.3kg、黄水倒水莲3.2kg、紫苏1.3kg。取7味药材,粉碎,过筛,制得粉剂。
临床试验
一、对骨质增生疾病的临床观察
1 一般资料
共90例患者,其中男42例,女48例。最大年龄67岁,最小年龄35岁,平均年龄47.4岁。最长病程11年,最短病程4个月,平均病程4.5年。临床表现:大部分患者发病部位均有慢性疼痛,部分颈椎增生患者,有向枕部的放射性疼痛,并伴有寒冷和麻木等异常感觉,少部分颈椎增生患者有植物神经功能紊乱,如恶心、头晕、耳鸣等症状。另外还有部分患者疼痛部位均有较明显的肌痉挛或肌紧张。
2 诊断依据
按照《中医病证诊断疗效标准》中骨痹的诊断依据判定。①病变部位出现受压迫和刺激症状,其中颈椎病变患者有长期颈肩背疼痛,头晕头痛,双手麻木或转侧不利等症状。②x线摄片中可见颈椎正、侧位片,左右斜位片,颈椎生理弯曲变直或反弓,腰椎间盘突出,椎间孔狭窄,椎体骨质增生;膝关节正侧可见:胫骨上端,股骨下端的内外侧髁骨赘形成,髌骨上下缘、髁间隆突,跟骨结节骨刺形成。患者诊断结果:膝关节骨质增生15例,腰椎骨质增生28例,颈椎骨质增生47例。
3 治疗方法
患者服用本发明实施例5片剂,每日2次,每次2片,以1个月为l疗程,持续4个疗程。
4 疗效评价
分为无效、有效、显著、痊愈。有效率+显著率+加痊愈率=总有效率。①无效:治疗前后症状和体征无任何变化,或者病情加重。②有效:病状和体征有限程度的好转,功能有限程度的恢复。③显著:病状和体征基本消失,功能恢复正常,但因天气变化或劳累等仍会出现不适感。④痊愈:病状和体征完全消失,功能完全恢复正常。
5 结果
经过4个疗程的治疗,90例患者中痊愈33例,显著32例,有效l7例,无效8例,总有效率达到91.1%,具体统计结果见表1。

由表1可知,经过4个疗程的治疗后,本发明实施例5对膝关节骨质增生的有效率为93.3%,对腰椎骨质增生的有效率为89.3%,对颈椎骨质增生的有效率为91.5%,对骨质增生的总有效率为91.1%。结果显示:本发明中药组合物对骨质增生有较好的治疗作用。
典型病例
宋XX,女,56岁,主诉20天前颈项强痛,手指麻木,头晕时轻时重,轻时如一常人,重时天旋地转。诊断为颈椎骨质增生。给予服用本发明实施例6颗粒剂,第2周时症状明显好转,1个月后麻木消失,1个半月后症状完全消失,判为痊愈。
张XX,女,58岁。主诉双膝关节疼痛四年,活动受限,不能下蹲、上厕所、上下楼困难。经多方治疗,效果不明显,查:双侧膝关节部轻度肿胀,局部压痛(+),X线片示:双膝关节间隙变窄,关节边缘骨质增生,胫骨髁间隆突变尖。诊断为双侧膝关节骨质增生。服用本发明实施例4胶囊剂5天即感疼痛减轻,连续服用2个月后痊愈,电话随访四年未复发。
虽然,上文中已经用一般性说明、具体实施方式及试验,对本发明作了详尽的描述,但在本发明基础上,可以对之作一些修改或改进,这对本领域技术人员而言是显而易见的。因此,在不偏离本发明精神的基础上所做的这些修改或改进,均属于本发明要求保护的范围。
Claims (8)
1.一种治疗骨质增生的中药组合物,其特征在于它是由包括以下重量份的原料制成的:
络石藤6-20份、枫荷梨25-50份、海风藤6-20份、扶芳藤6-20份、走马胎15-40份、黄水倒水莲20-40份、紫苏5-20份。
2.如权利要求1所述的治疗骨质增生的中药组合物,其特征在于它是由包括以下重量份的原料制成的:
络石藤10-15份、枫荷梨30-40份、海风藤10-15份、扶芳藤10-15份、走马胎20-30份、黄水倒水莲25-35份、紫苏10-15份。
3.如权利要求1所述的治疗骨质增生的中药组合物,其特征在于它是由包括以下重量份的原料制成的:
络石藤12份、枫荷梨35份、海风藤12份、扶芳藤12份、走马胎25份、黄水倒水莲30份、紫苏12份。
4.如权利要求1-3任一项所述的治疗骨质增生的中药组合物,其特征在于它的活性成分是采用水提或40-80%体积百分浓度的乙醇提取制备而成。
5.如权利要求4所述的治疗骨质增生的中药组合物,其特征在于它的活性成分是通过以下方法制备而成的:取全部7味药材,混合后加水提取2-3次,每次加水量相当于药材总重量的6-12倍,每次提取时间为1-3小时,合并提取液,滤过,滤液浓缩至70-80℃时相对密度为1.15-1.20的清膏,即得活性成分。
6.如权利要求4所述的治疗骨质增生的中药组合物,其特征在于它的活性成分是通过以下方法制备而成的:取全部7味药材,混合后用40-80%乙醇回流提取2-3次,每次乙醇用量为药材总量的4-10倍,提取时间为1-4小时,合并提取液,过滤,滤液浓缩至70-80℃时相对密度为1.15-1.20的清膏,即得活性成分。
7.如权利要求4所述的治疗骨质增生的中药组合物,其特征在于它的活性成分是通过以下方法制备而成的:取全部7味药材,混合后加水提取2-3次,每次加水量相当于药材总重量的6-12倍,每次提取时间为1-3小时,合并提取液,滤过,滤液浓缩至70-80℃时相对密度为1.15-1.20的清膏,加入乙醇,使含醇量为40-70%,静置12-24小时,滤过,滤液浓缩至70-80℃时相对密度为1.10-1.25的浸膏,即得活性成分。
8.一种治疗骨质增生的药物组合物,由权利要求1-3中任一项所述的治疗骨质增生的中药组合物与医药学上可接受的载体配合制备而成。
