本发明公开了一种用于皮肤美容的血小板生长因子制备及应用方法,包括制备步骤:1)将人体血液置入采血试管,在离心机中分离出血浆、血小板、血球;2)将所得血浆、血小板分离存放;3)在血浆、血小板内分别加入医疗专用氯化钙作为激活剂;4)将血浆、血小板置入离心机做二次分离;5)取出血浆、血小板,共同培养不超过6小时;应用方法包括:1)凸点滚筒来回于正常皮肤软组织或毛囊上滚动;2)光源照射皮肤软组织或毛囊;3)将采血试管内血小板立即涂抹于皮肤软组织或毛囊上;4)将采血试管内血浆涂抹于皮肤软组织或毛囊上。本发明制备过程简单、添加剂少,可高效利用血小板生长因子进行美容护肤。
Description
技术领域
本发明涉及美容护肤产品技术领域,尤其是涉及一种用于皮肤美容的血小板生长因子制备及应用方法。
背景技术
血小板(blood platelet),简称:PLT,是哺乳动物血液中的有形成分之一,是从骨髓成熟的巨核细胞胞质裂解脱落下来的具有生物活性的小块胞质,血小板具有特定的形态结构和生化组成,在正常血液中有较恒定的数量(如人的血小板数为每立方毫米10~30万),血小板除了可以促成凝血之外,它的功能远远不止是如此,除了形成血块之外,血小板在受伤的部位中受挤压后会破裂和慢慢释放出其中的生长因子,从而进行组织的修复,人体中主要储存大量活性的生长因子就是血小板,在人体受伤之后,局部释放出来的这些生长因子可以促进血管增生及组织的再生以及修复,而且是人体中最容易取得之生长因子。
中国专利文献公开了名为“可凝结的血小板生长因子浓厚液及其制备方法”的专利申请,其申请公布号为CN 101969985 A,申请公布日期为2011.02.09,主要发明内容是:本发明是关于一种可凝结的血小板生长因子浓厚液,其用于治疗性及/ 或美容性用途;前述可凝结的血小板生长因子浓厚液较佳是包含生长因子PDGF、TGF-β、IGF、EGF、CTGF、bFGF 及VEGF,在一较佳实施态样中,前述可凝结的血小板生长因子浓厚液不会诱发血液细胞相关的输血反应,本发明亦关于一种制备本发明的可凝结的血小板生长因子浓厚液的方法,且该方法包含下列步骤,使血小板浓厚液与溶剂及/ 或清洁剂接触;使前述血小板浓厚液与前述溶剂及/ 或清洁剂于维持在从约6.0 至约9.0 范围内的pH 值及在从2℃至50℃范围内的温度下共同培养至少5 分钟至6 小时的时间,较佳为在从25℃至45℃范围内的温度进行;以及通过油萃取及/ 或层析方法来移除前述溶剂及/ 或清洁剂。
上述公开技术中,在血小板生长因子的制备过程中使用了较多的溶剂、清洁剂等添加剂,而且针对血小板生长因子的美容护肤作用,也没有提出具体的操作使用方法。
发明内容
本发明是为了解决现有血小板生长因子制备过程复杂、添加剂过多,以及缺乏高效使用血小板生长因子进行美容护肤方法的问题,提供一种制备过程简单、添加剂少,可凝结的富血小板生长因子浓厚液,用于皮肤软组织上美容性用途,以及可高效利用血小板生长因子进行美容护肤的应用方法。
为了实现上述目的,本发明采用以下技术方案:一种用于皮肤美容的血小板生长因子制备方法,用采集自人体的血液来获得血小板生长因子,血小板生长因子包含生长因子PDGF、TGF-?,包括如下制备步骤:1)将人体血液置入采血试管,在离心 机中以rpm:1000~10000r/min转速,以RCF:1000~10000xg离心力,旋转3-15分钟,分离出血浆、血小板、血球;2)将所得血浆、血小板分别采用采血试管分离存放;3)在血浆、血小板采血试管内分别加入医疗专用氯化钙,作为血小板中生长因子或血浆中胶原蛋白的激活剂;4)将血浆、血小板采血试管置入离心机中以rpm:1000~10000r/min转速,RCF:1000~10000xg离心力,旋转3-15分钟,做二次分离;5)取出血浆、血小板采血试管,在10℃至35℃的温度下共同培养不超过6小时。血小板生长因子包含生长因子PDGF、TGF-β、IGF、EGF、CTGF、bFGF 及VEGF等,与皮肤软组织修复有关的因子主要是PDGF(Platelet-derived growth factors)和TGF-?(Transforming growth factor-beta,PDGF主要的功能是刺激细胞复制,血管生成,纤维细胞有丝分裂以及吸引白血球和间质干细胞到软组织修复处,TGF-?主要是促进细胞的有丝分裂,显著增加第一型胶原蛋白的生产,以及调节纤维化和肌肉细胞再生之间的平衡,在软组织修复时,需要胶原蛋白产生才会有弹性;采用离心机分离血液成分是常用技术,其中rpm为离心机转速,RCF为离心机所产生的离心力。
作为优选,步骤3)中所述的医疗专用氯化钙置入量为血浆或血小板重量的0.1%-30%。使血浆或血小板与医疗专用氯化钙接触,使二者维持在从约6.0 至约9.0 范围内的pH 值及在从2℃至50℃范围内的温度下共同培养至少5 分钟至6 小时的时间,较佳为在从25℃至45℃范围内的温度进行,通过分离纯化仪器方式来移除前述医疗专用钙质。
血小板生长因子的应用方法,包括如下使用步骤:1)采用表面带有凸点的滚筒来回于正常皮肤软组织或毛囊上滚动,凸点长度为0.1mm-10mm;2)采用光源照射皮肤软组织或毛囊1-30秒,光源波长为1500-11000nm,光源照射皮肤软组织或毛囊上相同点的能量密度20-150 mJ /cm2以内;3)将采血试管内血小板立即涂抹于皮肤软组织或毛囊上; 4)将采血试管内血浆涂抹于皮肤软组织或毛囊上。表面带有凸点滚筒来回在欲美容护肤的皮肤软组织或毛囊上滚动,可促进该处血液循环,待血液循环被充分促进后,采用1500-11000nm光源照射皮肤软组织或毛囊上1-30秒,能量密度20-150 mJ /cm2以内,皮肤软组织表皮与真皮层之间胶原蛋白会产生螺旋状洞体,此时立即将血小板涂抹上去,运用表皮与真皮层之间胶原蛋白会产生螺旋状洞体,将血小板直接快速的传送到真皮层;运用表皮与真皮层之间胶原蛋白会产生螺旋状洞体,最后将血浆涂抹上去,将血浆直接快速的传送到表皮与真皮层之间以便皮肤软组织或毛囊完全吸收生长因子。
作为优选,所述的采血试管内血浆可在10℃至35℃温度下,保存不超过100小时,涂抹于皮肤软组织上。
因此,本发明具有如下有益效果:(1)制备过程简单;(2)可高效利用血小板生长因子进行美容护肤;(3)运用不同的光线射频,直接快速传送血小板与血浆到真皮层与表皮层;(4) 使用医疗专用氯化钙作为血小板中生长因子或血浆中胶原蛋白的激活剂,添加剂少。
具体实施方式
一种用于皮肤美容的血小板生长因子制备方法,用采集自人体的血液来获得血小板生长因子,血小板生长因子包含生长因子PDGF、TGF-?,包括如下制备步骤:1)将人体血液置入采血试管,在离心 机中以rpm:2000r/min转速,RCF:2000xg离心力,旋转5分钟,分离出血浆、血小板、血球;2)将所得血浆、血小板分别采用采血试管分离存放;3)在血浆、血小板采血试管内分别加入医疗专用氯化钙,作为血小板中生长因子或血浆中胶原蛋白的激活剂,医疗专用氯化钙置入量为血浆或血小板重量的10%,使二者维持在7.0的pH 值及在25℃温度下共同培养2小时的时间;4)将血浆、血小板采血试管置入离心机中以rpm:8000r/min转速,RCF:8000xg离心力,旋转5分钟,做二次分离;5)取出血浆、血小板采血试管,在25℃的温度下共同培养不超过5小时。
具体实施过程是,包括如下使用步骤:1)采用带有凸点滚筒来回于正常皮肤软组织或毛囊上滚动,凸点长度为5mm;2)采用光源照射皮肤软组织或毛囊上3秒,光源波长为1550nm,光源照射皮肤软组织或毛囊上相同点的能量密度50mJ/cm2以内;3)将采血试管内血小板立即涂抹于皮肤软组织或毛囊上;4)将采血试管内血浆涂抹于皮肤软组织或毛囊上。如上使用后,皮肤美化率可达100%。
Claims (4)
1.一种用于皮肤美容的血小板生长因子制备方法,用采集自人体的血液来获得血小板生长因子,血小板生长因子包含生长因子PDGF、TGF-?,其特征在于,包括如下制备步骤:1)将人体血液置入采血试管,在离心机中以rpm:1000~10000r/min转速,RCF:1000~10000xg离心力,旋转3-15分钟,分离出血浆、血小板、血球;2)将所得血浆、血小板分别采用采血试管分离存放;3)在血浆、血小板采血试管内分别加入医疗专用氯化钙,作为血小板中生长因子或血浆中胶原蛋白的激活剂;4)将血浆、血小板采血试管置入离心机中以rpm:1000~10000r/min转速,以RCF:1000~10000xg离心力,旋转3-15分钟,做二次分离;5)取出血浆、血小板采血试管,在10℃至35℃的温度下共同培养不超过6小时。
2.根据权利要求1所述的一种用于皮肤美容的血小板生长因子制备方法,其特征是,步骤3)中所述的医疗专用氯化钙置入量为血浆或血小板重量的0.1%-30%。
3.根据权利要求1所得血小板生长因子的应用方法,其特征是,包括如下使用步骤:1)采用表面带有凸点的滚筒来回于正常皮肤软组织或毛囊上滚动,凸点长度为0.1mm-10mm;2)采用光源照射皮肤软组织或毛囊1-30秒,光源波长为1500-11000nm,光源照射皮肤软组织或毛囊上相同点的能量密度20-150 mJ /cm2以内;3)将采血试管内血小板立即涂抹于皮肤软组织或毛囊上;4)将采血试管内血浆涂抹于皮肤软组织或毛囊上。
4.根据权利要求3所述的血小板生长因子的应用方法,其特征是,所述的采血试管内血浆可在10℃至35℃温度下,保存不超过100小时,涂抹于皮肤软组织上。
