按照质量份将人参皂苷Rb125份、稻米天然生物活性蛋白10份、荷叶黄酮8份、鞣花单宁3份、原花青素4份、茯苓提取物5份混合均匀,最后制成冲剂。
实施例3
动物功能性试验:利用大鼠高脂肪营养饲养肥胖模型,对受试样品(实施例1)的减肥功能进行了研究。方法如下:选取体重为100-180g范围内的雄性大鼠50只,按体重体脂随机分成一个普通对照组、一个营养对照组和三个剂量实验组,普通对照组小鼠饲喂基础饲料,营养对照组饲喂营养饲料,三个剂量试验组大鼠在饲喂营养饲料的基础上分别同时服用5倍、10倍、20倍人体推荐用量的受试样品,连续喂养30d,试验期间记录每只大鼠的给食量,定期称体重(每周2次),试验结束时称体重、剖腹取体脂并称重,计算脂/体比。
试验结果表明,营养对照组、普通对照组和试验组小鼠生长发育均正常;与普通对照组相比,营养对照组小鼠体重显著(P<0.05)增加,表明营养性肥胖模型建模成功;与营养对照组大鼠相比,10倍、20倍人体推荐量试食试验组大鼠生殖器周围脂肪重量、脂/体比均显著(P<0.05)降低,5倍人体推荐量试食组大鼠体重显著((P<0.05)降低,试验组大鼠体重增长速度明显低于对照组。根据判定原则,该受试样品动物减肥功能结果为阳性。
安全性毒理学评价:是按照卫生部《保健食品检验与评价技术规范》(2003年版)进行实验,方法及结果如下:a.用该饮料105倍人体剂量一次性经口灌胃后,无明显的中毒表现或不良反应,2周内无动物死亡,表明本品属无毒类。b.遗传毒性试验中,Ames试验、小鼠骨髓细胞微核试验和小鼠精子畸形实验3项遗传毒理性试验均为阴性结果,表明本品无致突变作用。c.大鼠30d喂养实验,未引起大鼠整体健康状况、生理生化功能和器官组织形态学各重要指标的异常变化。
人体试食试验中,按照受试者纳入标准,选择120例肥胖症人群作为受试对象,按受试人的体重、体脂重量随机分成试食组和对照组,试食组按人体推荐方法服用受试样品,对照组服用等量的安慰剂,连续35天,试验期间正常饮食、作息。试验结束后测定受试人体的体重、腰围、臀围、体内脂肪含量。据试验结果统计表明,实验前后受试者健康状态不受影响,体重显著(P<0.05)减轻;臀围显著(P<0.05)减少;4点测定位点中右三角肌下缘臂外侧正中点、右肩脚下角、右脐旁3cm等3个位点处皮下组织厚度显著(P<0.05)减少,由此可判定该受试样品具有减肥功能作用。
兴奋剂试验:结果为阴性。
本发明是以人参皂甙Rb1、稻米天然生物活性蛋白、荷叶黄酮、鞣花单宁、原花青素、茯苓提取物为主要原料制成的保健食品,经动物功能和人体试食试验证明,具有减肥的保健功能。适宜人群:单纯性肥胖人群;不适宜人群:孕期及哺乳期妇女;食用方法及食用量:口服,每日1次,每次150mg;保质期24个月;贮藏方法:阴凉、通风、干燥处;本发明不能代替药物。
