鉴于大规模细胞培养过程的费用和对生物产物的更大量和更低成本的增长需求,提供重组蛋白生产的甚至增量改善的新细胞培养方法是有价值的。

发明详述
在重组蛋白生产期间,需要具有受控制的系统,其中使细胞生长到所需密度,并且然后将细胞的生理状态转换为生长受阻滞的高生产力状态,其中细胞使用能量和受质生产所关注的重组蛋白而非制造更多细胞。用于实现这一目标的方法(诸如温度变换和小分子诱导剂)并非总是成功的并且可能对产物品质具有非所需的作用。如本文所述,可以在生产期期间通过以暴露于低L-天冬酰胺条件在培养细胞中诱导细胞生长阻滞将压积细胞体积限于所需水平。可以通过使用含有有限浓度的L-天冬酰胺的灌注培养基并且在细胞培养物中维持低浓度的L-天冬酰胺(5mM或更小)来实现并且维持细胞生长阻滞。
还发现,当通过低L-天冬酰胺或通过L-天冬酰胺饥饿引发生长阻滞并且随后用细胞培养物和L-天冬酰胺浓度是5mM或更小的灌注培养基维持生长受阻滞的细胞时,生长受阻滞的细胞展示增加的生产力。
可以通过操纵L-天冬酰胺的浓度来实现生长受阻滞、高生产力的生产期。如本文所述,L-天冬酰胺的耗竭导致生长阻滞。在分批进料培养中,一旦细胞密度足够高(例如≥20×106个活细胞/mL),则尽管有重复的进料,培养物也由于L-天冬酰胺消耗和/或向L-天冬氨酸的转化而反复地缺乏L-天冬酰胺。在细胞培养物中,细胞外L-天冬酰胺可以转化L-天冬氨酸和氨。L-天冬酰胺耗竭导致细胞周期阻滞。在分批进料期间,L-天冬酰胺存在于培养物中的时期产生增加的生产力,并且L-天冬酰胺已经被耗竭的时期产生降低的生产力。在灌注系统中,连续地供应L-天冬酰胺,因此避免总耗竭,并且可以维持较高浓度的L-天冬酰胺,因此允许细胞继续繁殖并且不被暴露于L-天冬酰胺耗竭或有限的环境。将L-天冬酰胺的浓度控制在足够低浓度(诸如浓度5mM或更小)下可以将细胞保持在高生产力状态,同时维持活力和限制生长。在具有批式和灌注进料的系统中,进料培养基可以从在批式进料期间含有高(生长促进)水平的L-天冬酰胺的制剂转换为在灌注进料期间含有较低(生长受阻滞)水平的L-天冬酰胺的制剂。可以通过添加回低水平的L-天冬酰胺将已经通过限制L-天冬酰胺而被阻滞生长的细胞培养物刺激成高生产力状态。
对于商业规模细胞培养物和生物治疗剂的制造来说,将非常需要阻滞细胞生长和能够在生产期期间将细胞维持在生长受阻滞的状态的能力。还诱导细胞那样以增加生产力同时处于生长受阻滞的状态并且能够维持这一增加的生产力,出于制造目的是理想的。

还提供一种增加在表达重组蛋白的哺乳动物细胞培养物中的重组蛋白生产的方法,其通过在哺乳动物细胞培养物中诱导低天冬酰胺细胞生长阻滞。维持在低天冬酰胺生长受阻滞的状态的哺乳动物细胞展现比未经低天冬酰胺生长阻滞的细胞更大的生产力(g蛋白/细胞/天和g蛋白/细胞质量/天)。
该方法还适用于将哺乳动物细胞培养物限制在所需压积细胞体积下。可以通过降低生产培养基中的L-天冬酰胺水平来将在生产期期间的压积细胞体积限制在所需水平下。灌注培养基中5mM或更小的天冬酰胺浓度足以在培养期间控制细胞生长并且限于所需压积细胞体积。
本文所述的方法对于细胞生长提供较大控制以获得高产物滴度的细胞培养物;并且因此与高生物质灌注过程相比可以简化气体处理策略并且将在收获和下游加工期间的产物损失最小化。
该方法开始于在生产生物反应器中建立哺乳动物细胞培养物。优选地使用较小生产生物反应器,在一个实施方案中,生物反应器是500L到2000L。在一个优选实施方案中,使用1000L-2000L生物反应器。用以接种生物反应器的种子细胞密度可对所生产的重组蛋白的水平具有积极影响。在一个实施方案中,在无血清培养基中以至少0.5×106直到和超过3.0×106个活细胞/mL接种生物反应器。在一个优选实施方案中,接种是1.0×106个活细胞/mL。
哺乳动物细胞然后经历指数生长期。可以在无补充进料下维持细胞培养物直到达到所需细胞密度。在一个实施方案中,在无补充进料下维持细胞培养物多达3天,随后用L-天冬酰胺浓度是5mM或更小的无血清灌注培养基灌注以诱导并且维持低L-天冬酰胺生长阻滞。在另一实施方案中,可以在所需细胞密度下接种培养物以开始生产期而无短暂生长期,在接种时即刻引发细胞生长阻滞,这是通过用含有5mM或更小L-天冬酰胺的无血清灌注培养基灌注细胞培养物以诱导并且维持低L-天冬酰胺生长阻滞。在本文实施方案中任一者中,还可以通过L-天冬酰胺饥饿(使细胞经历0mM L-天冬酰胺环境)随后用L-天冬酰胺浓度等于或小于5mM的细胞培养基灌注并且将细胞培养物中的L-天冬酰胺的浓度维持在那个水平下来引发从生长期向生产期的转换。
不考虑诱导多低L-天冬酰胺生长阻滞,在通过用低L-天冬酰胺培养基灌注并且将细胞培养物维持在5mM或更小的L-天冬酰胺水平下维持的生长受阻滞的细胞中可见更高生产力。
如本文所用,也可以称为“细胞生长阻滞”的“生长阻滞”是细胞停止数目增加或当细胞周期不再进展时所处的点。可以通过测定细胞培养物的活细胞密度来监测生长阻滞。生长受阻滞的状态下的一些细胞大小可以增加但数目不增加,因此生长阻滞培养物的压积细胞体积可增加。如果细胞不处于下降的健康,则可以通过添加额外的L-天冬酰胺到细胞培养物中在一定程度上逆转生长阻滞。
当培养物的细胞密度达到培养物中的L-天冬酰胺的浓度对于持续生长变得有限的水平时或当培养物缺乏L-天冬酰胺时,通过L-天冬酰胺引发生长阻滞。L-天冬酰胺饥饿在细胞培养基中的L-天冬酰胺浓度实际上是0mM时发生。饥饿可以在24小时内导致生长阻滞。饥饿持续长于48小时可能伤害细胞健康。为了将细胞维持在生长受阻滞的状态,必须将细胞培养物中的L-天冬酰胺浓度维持在5mM或更小。阻滞细胞生长所需的L-天冬酰胺的细胞培养基浓度取决于细胞制造其自身天冬酰胺的能力。对于细胞可以制造其自身天冬酰胺的培养物来说,生长阻滞可能需要L-天冬酰胺的浓度更低或甚至从培养基去除。对于不能制造其自身天冬酰胺的培养物(例如缺少活性天冬酰胺合成酶的细胞)来说,可以使用超过零直到5mM L-天冬酰胺的浓度来阻滞生长。
如本文所用,也称为“压积细胞体积百分比”(%PCV)的“压积细胞体积”(PCV)是由细胞占据的体积与细胞培养物总体积的以百分比表示的比率(参看Stettler等人,(2006)Biotechnol Bioeng.Dec20:95(6):1228-33)。压积细胞体积是细胞密度和细胞直径的函数;压积细胞体积增加可以起因于细胞密度或细胞直径或两者的增加。压积细胞体积是细胞培养物中的固体含量的量度。在收获和下游纯化期间去除固体。固体越多意味着在收获和下游纯化步骤期间从所需产物分离固体物质的努力越多。另外,所需产物在收获过程期间可能变得截留在固体中并且失去,导致产物产率降低。因为宿主细胞大小不同,并且细胞培养物还含有死亡和垂死的细胞和其他细胞碎片,所以压积细胞体积是一种比细胞密度或活细胞密度更准确的描述细胞培养物内的固体含量的方法。例如,2000L细胞密度是50×106个细胞/ml的培养物将取决于细胞大小而具有截然不同的压积细胞体积。另外,一些细胞当处于生长受阻滞的状态时将大小增加,因此生长阻滞之前与生长阻滞之后的压积细胞体积将很可能不同,这是由于生物质由于细胞大小增加而增加。
在生长期与生产期之间过渡时和在生产期期间,压积细胞体积百分比(%PCV)等于或小于35%。在生产期期间维持的所需压积细胞体积等于或小于35%。在一个优选实施方案中,压积细胞体积等于或小于30%。在另一个优选实施方案中,压积细胞体积等于或小于20%。在另一个优选实施方案中,压积细胞体积等于或小于15%。在又一个优选实施方案中,压积细胞体积等于或小于10%。

在生产期期间较低压积细胞体积有助于缓解可能阻碍较高细胞密度灌注培养物的溶解氧喷射问题。较低压积细胞体积还允许较小培养基体积,其允许使用较小培养基储存容器并且可以与较慢流速组合。与较高细胞生物质培养物相比,较低压积细胞体积还对收获和下游加工具有较小影响。其全部都降低与制造重组蛋白治疗剂相关的成本。
三种方法典型地用于由哺乳动物细胞培养物生产重组蛋白的商业过程:分批培养、分批进料培养和灌注培养。分批培养是一种不连续方法,其中使细胞在固定体积的培养基中生长短的时期随后完全收获。使用分批方法生长的培养物经历细胞密度增加,直到达到最大细胞密度,随后随着培养基组分被消耗并且代谢副产物(诸如乳酸和氨)水平累积,活细胞密度降低。收获典型地在达到最大细胞密度(典型地5-10×106个细胞/mL,取决于培养基配方、细胞系等)时进行。分批过程是最简单的培养方法,然而,活细胞密度受营养物可用性限制,并且一旦细胞处于最大密度,则培养下降并且生产减少。没有能力延长生产期,这是因为废产物累积和营养物耗竭快速地导致培养下降(典型地约3到7天)。
分批进料培养通过提供批式或连续培养基进料以补充已经被消耗的那些培养基组分来对分批过程加以改善。因为分批进料培养在整个运行期间都接收额外的营养物,所以当与分批方法相比时,其具有获得更高细胞密度(>10到30×106个细胞/m1,取决于培养基配方、细胞系等)和增加的产物滴度的潜力。不同于分批过程,通过操纵进料策略和培养基配方以将细胞增殖以获得所需细胞密度的时期(生长期)与暂停或缓慢细胞生长的时期(生产期)区分来产生并且维持二阶段培养。因此,分批进料培养与分批培养相比具有获得更高产物滴度的潜力。典型地,在生长期期间使用分批方法,并且在生产期期间使用分批进料方法,但在整个过程中可以使用分批进料的进料策略。然而,不同于分批过程,生物反应器体积是限制进料量的限制因素。另外,正如分批方法,代谢副产物累积将会导致培养下降,其限制生产期的持续时间,约1.5到3周。分批进料培养是不连续的,并且收获典型地在代谢副产物水平或培养物生存力达到预定水平时进行。
灌注方法通过添加新鲜培养基并且同时去除用过的培养基来对分批和分批进料方法提供潜在的改善。典型的大规模商业细胞培养策略努力达到高细胞密度60-90(+)×106个细胞/mL,其中反应器体积的几乎三分之一到超过二分之一是生物质。在灌注培养下,已经达到了>1×108个细胞/mL的极端细胞密度,并且预测甚至更高的密度。典型的灌注培养开始于持续一天或两天的分批培养启动,随后是连续、逐步和/或间歇添加新鲜进料培养基到培养物中并且同时去除用过的培养基,在培养的整个生长和生产期期间保留细胞和诸如蛋白的其他高分子量化合物(基于过滤器分子量筛截)。可以使用诸如沉降、离心或过滤的各种方法来去除用过的培养基同时维持细胞密度。已经报道了一工作体积/天达到许多多工作体积/天的部分的灌注流速。灌注过程的一个优点在于,可以将生产培养物维持比分批或分批进料培养方法更久的时期。然而,增加的培养基制备、使用、储存和处理为支持长期灌注培养、特别是具有高细胞密度的培养所需,其还需要甚至更多的营养物,并且这全部都驱使生产成本与分批和分批进料方法相比甚至更高。另外,较高细胞密度可能会在生产期间导致问题,诸如维持溶解氧水平和增加的气体处理的问题,包括供应更多氧气和去除更多二氧化碳,这将导致更多起泡和对消泡策略的变更的需要;以及在收获和下游加工期间导致问题,其中去除过量细胞物质所需的努力会导致产物损失,以致否定了由于细胞质量增加而增加的滴度的益处。
还提供一种大规模细胞培养策略,其将生长期期间的分批进料的进料随后是生产期期间的连续灌注组合。该方法靶向细胞培养物维持在小于或等于35%的压积细胞体积下的生产期。该方法还由于低天冬酰胺而提供细胞生长阻滞的引发和维持。
分批进料培养是一种用于由哺乳动物细胞大规模生产蛋白的广泛实践的培养方法。参看例如Chu和Robinson(2001),Current Opin.Biotechnol.12:180-87。哺乳动物细胞的分批进料培养是连续或定期用含有营养物的浓进料培养基对培养物进料的培养。进料可以根据例如每天、每两天一次、每三天一次等的预定时间表进行。当与分批培养(其中不进行进料)相比时,分批进料培养可以生产更大量的重组蛋白。参看例如美国专利第5,672,502号。
在一个实施方案中,在生长期期间使用具有批式进料的分批进料培养来维持细胞培养物。然后可以在生产期期间使用灌注进料。在一个实施方案中,灌注开始于所述细胞已经达到生产期时。在另一实施方案中,灌注开始于该细胞培养的第5天时或大约第5天时至第9天时或大约第9天时。在另一实施方案中,灌注开始于该细胞培养的第5天时或大约第5天时至第7天时或大约第7天时。
在另一实施方案中,可以通过使分批进料培养物经历L-天冬酰胺饥饿时期随后用L-天冬酰胺浓度是5mM或更小的无血清灌注培养基灌注来引发分批进料培养中细胞生长阻滞的开始。在一个实施方案中,在L-天冬酰胺饥饿之前和期间监测该细胞培养基的L-天冬酰胺浓度。在另一实施方案中,可以通过用L-天冬酰胺浓度是5mM或更小的无血清灌注培养基灌注来实现分批进料培养中细胞生长阻滞的引发。
在生长期期间使用批式进料允许细胞过渡到生产期,导致对作为引发和控制生产期的手段的温度变换依赖性较小,然而,36℃到31℃的温度变换可以在生长期与生产期之间发生。在一个优选实施方案中,变换是36℃到33℃。
如本文所述,可以用至少0.5×106直到和超过3.0×106个活细胞/mL、优选地1.0×106个活细胞/mL在无血清培养基中接种生物反应器。
灌注培养物是细胞培养物接收新鲜灌注进料培养基同时去除用过的培养基的培养物。灌注可以是连续的、逐步的、间歇的或这些中的任何或所有的组合。灌注速率可以是小于一工作体积/天到多工作体积/天。优选地,将细胞保持在培养物中,并且被去除的用过的培养基基本上不含细胞或具有比培养物显著更少的细胞。也可以将由细胞培养物表达的重组蛋白保持在培养物中。可以通过多种手段实现灌注,包括离心、沉降或过滤,参看例如Voisard等人,(2003),Biotechnology and Bioengineering82:751-65。优选的过滤方法是交替切向流过滤。通过将培养基泵送通过中空纤维过滤模块来维持交替切向流。参看例如美国专利第6,544,424号;Furey(2002)Gen.Eng.News.22(7),62-63。
如本文所用,“灌注流速”是从生物反应器通过(添加或去除)的培养基的量,典型地以给定时间中工作体积的一些部分或多工作体积表示。“工作体积”是指用于细胞培养的生物反应器体积的量。在一个实施方案中,灌注流速是一工作体积或更少/天。可以配制灌注进料培养基以将灌注营养物浓度最大化以将灌注速率最小化。
“细胞培养”或“培养”意味着多细胞生物体或组织外部的细胞的生长和繁殖。哺乳动物细胞的适合培养条件在本领域中已知。参看例如Animal cell culture:A PracticalApproach,D.Rickwood编,Oxford University Press,New York(1992)。可以在悬浮液中或在连接到固体受质时培养哺乳动物细胞。可以使用具有或不具有微载体的流化床生物反应器、中空纤维生物反应器、滚瓶、摇瓶或搅拌槽生物反应器。在一个实施方案中,使用500L到2000L生物反应器。在一个优选实施方案中,使用1000L到2000L生物反应器。

根据本发明的方法可以用以改善多阶段培养过程中的重组蛋白生产。在多阶段过程中,在两个或更多个不同阶段中培养细胞。例如,细胞可以首先在将细胞增殖和生存力最大化的环境条件下在一个或多个生长期中进行培养,然后转移到将蛋白生产最大化的条件下的生产期。在由哺乳动物细胞生产蛋白的商业过程中,存在通常多个(例如至少约2、3、4、5、6、7、8、9或10个)在最终生产培养之前的不同培养容器中发生的生长期。生长和生产期可以前面是一个或多个过渡阶段,或由一个或多个过渡阶段分隔。在多阶段过程中,根据本发明的方法可以至少在商业细胞培养物的最终生产期的生长和生产期期间采用,但其也可以在前面的生长期中采用。可以大规模进行生产期。可以在至少约100、500、1000、2000、3000、5000、7000、8000、10,000、15,000、20,000升的体积中进行大规模过程。在一个优选实施方案中,在500L、1000L和/或2000L生物反应器中进行生产。生长期可以在比生产期更高的温度下发生。例如,生长期可以在约35℃到约38℃的第一温度下发生,并且生产期可以在约29℃到约37℃、任选地约30℃到约36℃或约30℃到约34℃的第二温度下发生。另外,可以与温度变换同时、在温度变换之前和/或之后添加蛋白生产的化学诱导剂(诸如咖啡因、丁酸盐和六亚甲基二乙酰胺(HMBA))。如果在温度变换之后添加诱导剂,那么可以在温度变换之后一小时到五天、任选地在温度变换之后一到两天添加其。可以维持细胞培养物数天或甚至数周同时细胞生产所需蛋白。
可以使用本领域中已知的任何分析技术来监测和评估来自细胞培养物的样品。可以在培养持续期间监测多种参数,包括重组蛋白和培养基品质和特征。可以所希望的频率间歇地取用和监测样品,包括连续监测、实时和接近实时。在一个实施方案中,在L-天冬酰胺饥饿之前和期间监测该细胞培养基的L-天冬酰胺浓度。
典型地,使用在最终生产培养物之前的细胞培养物(N-x到N-1)来产生将用于接种生产生物反应器的种子细胞(N-1培养物)。种子细胞密度可以对所生产的的重组蛋白水平具有积极影响。产物水平倾向于随种子密度增加而增加。滴度的改善不仅与较高种子密度相关,而且很可能受处于生产的细胞的代谢和细胞周期状态影响。
可以通过任何培养方法生产种子细胞。优选的方法是使用交替切向流过滤的灌注培养。可以使用交替切向流过滤运行N-1生物反应器以提供高密度的细胞以接种生产生物反应器。可以使用N-1阶段以使细胞生长到>90×106个细胞/mL的密度。N-1生物反应器可以用于产生批式种子培养物,或可以用作可以被维持以在高种子细胞密度下对多个生产生物反应器进行播种的滚动种子储备培养物。生产的生长阶段的持续时间可以从7到14天变动,并且可以经设计以便在接种生产生物反应器之前将细胞维持在指数生长。将灌注速率、培养基配方和时序最优化以使细胞生长并且将其以最有益于将其生产最优化的状态传递到生产生物反应器中。可以达到>15×106个细胞/mL的种子细胞密度以便对生产生物反应器进行播种。接种时的较高种子细胞密度可以减少或甚至消除达到所需生产密度所需的时间。

实施例
实施例1
本实验比较不同启动条件,使用分批或分批进料的进料方法,随后是使用交替切向流过滤的连续灌注。灌注在早期指数生长期期间早期开始(“分批”启动),在灌注之前无额外进料,或在指数期结束并且进入静止或生产期时开始(“分批进料”启动),在灌注之前接收数次无血清成分确定的进料培养基的批式进料。
在第0天,将表达重组抗体的CHO细胞以1×106个活细胞/mL接种到2L生产生物反应器中,工作体积对于分批进料起始来说是1500ml无血清成分确定的培养基并且对于分批起始来说是1800mL无血清成分确定的培养基。将培养物维持在36℃、30%溶解氧(DO)、215RPM搅动下。将葡萄糖维持在高于0g/L并且低于8g/L。
灌注对于分批培养物来说在第4天开始(0.25体积/天)并且对于分批进料培养物来说在第7天开始(0.75体积/天)。使用交替切向流灌注与过滤系统(RefineTechnologies,Hanover,NJ,50kDa中空纤维过滤器)实现灌注。在开始灌注之前,分批进料培养物在第4天(7.5%初始工作体积)和第6天(10%初始工作体积)接收浓无血清成分确定的进料培养基的批式进料。灌注速率提供于表1中。

在培养运行期间,取每天的样品评估培养物。使用Vi-Cell(Beckman Coulter,Brea,CA)测定活细胞密度(VCD)和生存力。通过HPLC分析测量滴度。使用VoluPAC(Sartorius,Goettingen,Germany)测定压积细胞体积。
当活细胞密度超过20×106个活细胞/mL(对于分批和分批进料起始条件来说分别是第7天和第11天)时,应用温度变换(36.0℃到33.0℃)。
对于分批启动条件来说,活细胞密度在灌注开始之后继续增加;对于分批进料起始条件来说,灌注在细胞培养物已经达到了几乎不生长的平台或静止期之后开始。在第15天,分批进料的活细胞密度在27.7×106与30.7×106个活细胞/mL之间,而分批培养物的VCD在22.5×106与27.4×106个活细胞/mL之间(图1A)。分批进料培养物的生存力在73.9%与77.5%之间,而分批培养物生存力在72.5%与83.1%之间(图1B)。分批进料培养物的滴度在15.3与16.1g/L之间,而分批培养物滴度在10.6与12.3g/L之间(图1C)。因为积分活细胞密度(IVCD)值在第15天对于所有四种培养物都类似(约230×106细胞天/mL),所以比生产力在分批进料启动条件下更高。使分批进料培养物持续到第24天。在20天时达到20g/L的滴度。
与分批起始方法相比,分批进料启动情况下的交替切向流灌注导致生产力增加,将细胞维持在生产力更大的状态下。
实施例2
在第0天,将表达重组抗体的CHO细胞以1×106个活细胞/mL接种到2L生产生物反应器中,工作体积对于分批起始来说是1500ml无血清成分确定的培养基并且对于分批进料起始来说是1300mL无血清成分确定的培养基。对于分批培养物来说,将培养物维持在36℃、30%DO、215RPM搅动下。在430RPM下搅动分批进料培养物。在灌注之前每天将分批进料培养物进料到7g/L葡萄糖,并且在灌注期间将所有培养物都维持在等于或高于4g/L葡萄糖。灌注(交替切向流)对于分批培养物来说在第4天开始(0.25体积/天)并且对于分批进料培养物来说在第8天开始(0.75体积/天)。在开始灌注之前,分批进料培养物在第4天(7.5%初始工作体积)和第6天(10%初始工作体积)接收浓无血清成分确定的进料培养基的批式进料。灌注流速设定提供于表2中。将培养物维持21天。

如实施例1中在第6天当活细胞密度超过20×106个活细胞/mL时,将温度变换(36.0℃到33.0℃)应用于分批培养物。将分批进料培养物维持在36.0℃下持续培养的持续时间。
分批启动方法培养物具有类似于上文所述结果的结果,在第10天之后细胞达到约20×106到25×106个活细胞/mL,不生长。分批进料培养物在大多数培养持续时间低于20×106个活细胞/mL之后,在第20天达到近30×106个活细胞/mL,参看图2A。生存力都保持高于80%直到第10天,并且然后在第20天下降到分批起始培养物的约40%和分批进料培养物的60%,参看图2B。滴度对于分批起始培养物来说在近15g/L下是峰值,但其对于高搅动分批进料培养物来说达到超过20g/L,参看图2C。观测到,分批起始培养物在第3天的L-天冬酰胺浓度是约3到4mM,并且并不经历天冬酰胺有限的培养环境。然而,分批进料灌注起始培养物在第7天开始灌注之前在第6天经历L-天冬酰胺有限的环境。然后用含有浓度是2.0g/L(或13.3mM)的L-天冬酰胺的培养基灌注培养物,这导致在第8天之后不再有L-天冬酰胺限制(图2D)。将葡萄糖浓度主要维持在4与10g/L之间。
高搅动灌注培养情况下的分批进料启动在20天中达到最高滴度(超过20g/L),比分批启动培养物高出大于5g/L,这类似于上文所述结果。未观测到较高搅动速率的消极作用。维持恒定温度并未消极地影响分批进料培养物。
实施例3
本实验表征灌注体积和温度变换对如上文所述的分批进料启动情况下的交替切向流灌注的作用。对所有培养物进行分批进料开始操作,灌注在第7天开始。测试从四分之三工作体积移动到完整体积或从完整工作体积移动到四分之三工作体积的灌注流速。还测试在第14天从36℃到33℃的温度变换。
在第0天,将表达重组抗体的CHO细胞以1×106个细胞/mL接种到2L生产生物反应器中,工作体积是1200ml无血清成分确定的培养基。将培养物维持在36℃、30%DO下。在灌注之前,每天将葡萄糖进料到7g/L,并且在灌注期间将葡萄糖维持在高于1g/L。将培养物维持20天。
培养物在第4天(7.5%初始工作体积)和第6天(10%初始工作体积)接收浓无血清成分确定的进料培养基的批式进料。灌注在第8天开始。灌注速率提供于表3中。每组中一个培养物在第15天具有从36℃到33℃的温度变换,其他培养物维持在36℃下持续实验的持续时间。

温度变换和灌注速率不影响活细胞密度,参看图3A。然而,温度变换似乎有助于在培养的后期时间点保持生存力。似乎在第15天起开始的温度变换条件之间存在突发。温度变换的培养物的生存力比保持在36℃下的培养物更缓慢地下降,参看图3B。对于滴度来说,三种培养物在第15天(17.1-17.9g/L)以及在第20天(22-24g/L)展示非常类似的滴度,但一种培养物在第15天(21.58g/L)以及在第20天(28.33g/L)具有较高滴度(参看图3C)。温度和灌注速率似乎对滴度生产都不具有任何影响,表明可以不同灌注速率维持培养物。
实施例4
设计本实验以研究灌注培养基天冬酰胺浓度和灌注起始条件(L-天冬酰胺有限的或非有限的培养环境)对在生产期期间活细胞密度的作用。
在第0天,将表达重组抗体的CHO细胞以1×106个细胞/mL接种到2L生产生物反应器中,工作体积对于分批和分批进料起始方法来说都是1500ml。将培养物维持在36℃、30%溶解氧(DO)、400RPM搅动下。使用钻管或烧结喷射器进行喷射。将葡萄糖维持在高于0g/L并且低于8g/L。
灌注(交替切向流)对于分批起始“非天冬酰胺有限的培养物”来说在第3天(0.29体积/天)开始并且对于分批进料“天冬酰胺有限的培养物”来说在第7天(0.48体积/天)开始。分批培养基含有10mM L-天冬酰胺。在开始灌注之前,分批进料培养物在第3和6天(7%初始工作体积)接收含有113.4mM L-天冬酰胺的浓无血清成分确定的进料培养基的批式进料。灌注培养基天冬酰胺浓度是对照浓度(17.3mMAsn于无血清成分确定的灌注培养基中)或低浓度(5mM Asn于无血清成分确定的灌注培养基中)。如上文所述进行灌注。灌注速率提供于表4中。

通过限制培养基中的天冬酰胺在生产期期间实现细胞生长减少和生产力改善。在第15天,最大活细胞密度对于具有低天冬酰胺的分批进料起始培养物来说是约17.0×106个活细胞/mL(图4A)。对照水平的天冬酰胺培养物达到超过40×106个活细胞/mL(>30%压积细胞体积)的活细胞密度。低天冬酰胺分批进料培养物的生存力是67.1%,而分批培养物生存力是55.1%,并且对照物是69%(图4B)。低天冬酰胺分批进料培养物的压积细胞体积调节滴度是17.0g/L(针对压积细胞体积调节),而分批培养滴度是15.4g/L(图4C)。对照物具有10.2到12.9g/L(分批起始)和14.2到15.9g/L(分批进料起始)的滴度。
在生产期间将天冬酰胺水平维持在5mM或更小导致生长阻滞,刺激生产力,并且在生产期期间维持生存力。
实施例5
本实验比较灌注期间的培养基条件。在这个2L生物反应器实验中,将细胞接种到化学成分确定的分批培养基中,工作体积是1.5L,培养3天,并且然后使用含有17.3mM L-天冬酰胺和4.6mM L-谷氨酰胺或5mM L-天冬酰胺和10mM L-谷氨酰胺的化学成分确定的灌注培养基灌注12天。使用具有30kDa中空纤维过滤器(GE Healthcare,Little Chalfont,UK)的交替切向流灌注与过滤系统(Refine Technologies,Hanover,NJ)实现灌注。灌注在第3天以0.3培养物体积/天的速率开始。如下表6中所指示,在第4、9和11天增加灌注速率。将培养物维持在36℃、30%DO、pH7.0和400rpm搅动下。

天冬酰胺限制导致较少的细胞累积并且改善生产力。用含有5mM天冬酰胺的培养基灌注的培养物达到8.16×107-8.54×107个细胞/mL的最大VCD,而用含有17.3mM天冬酰胺的培养基灌注的培养物达到11.9×107-12.2×107个细胞/mL(图5A)。尽管17.3mM天冬酰胺下的培养物具有更多细胞,但5mM天冬酰胺下的培养物制造更多产物。与用含有5mM天冬酰胺的培养基灌注的培养物的7.59-8.15g/L(5.01-5.38g/L压积细胞体积调节的)相比,用含有17.3mM天冬酰胺的培养基灌注的培养物给出6.89-7.18g/L(4.33-4.67g/L压积细胞体积调节的)(图5B和5D)。5mM天冬酰胺培养物的最终压积细胞体积(PCV)倾向于略低于17.3mM天冬酰胺培养物(图5C),并且培养物生存力方面不存在差异(图5E)。
有趣的是,在这个实施例中,增加低天冬酰胺条件下的谷氨酰胺浓度多于两倍(4.6mM相较于10mM谷氨酰胺)不干扰低天冬酰胺培养基阻滞培养物生长的能力。
实施例6
本实施例比较在实验室与中试规模下的在使用天冬酰胺限制控制生长的ATF灌注过程中培养的克隆抗体表达CHO细胞系的性能。实验室规模模型利用2L生物反应器,并且中试规模是500L。在实验室规模下,将细胞接种到化学成分确定的分批培养物中,工作体积是1.5L,并且在中试规模下,工作体积是约378L。将细胞在分批培养基中培养3天,并且然后使用含有5mM L-天冬酰胺和10mM L-谷氨酰胺的化学成分确定的灌注培养基灌注12天。使用具有30kDa中空纤维过滤器(GE Healthcare,Little Chalfont,UK)的交替切向流灌注与过滤系统(Refine Technologies,Hanover,NJ)实现灌注。灌注在第3天以0.3培养物体积/天的速率开始。如下表7中所指示,在第4、9和11天增加灌注速率。将培养物维持在36℃、30%DO和pH6.9下。

提供四种实验室规模培养物和两种中试规模培养物的数据。VCD曲线在两种规模下类似,实现生长控制(图6A),并且总细胞质量(压积细胞体积)在两种规模下都维持在低于30%(图6C)。尽管VCD在约第10天或第11天达到平台,但压积细胞体积继续增加直到约第13或14天(图6C)。在各规模之间生产力也类似。与500L中试规模下的15.0-17.3g/L(10.6-12.8g/L压积细胞体积调节的)相比,在2L实验室规模下用含有5mM天冬酰胺的培养基灌注的培养物给出14.2-15.7g/L(10.7-11.4g/L压积细胞体积调节的)(图6B和6D)。生存力在中试规模下倾向于略低(图6E)。
Claims (34)
1.一种培养表达重组蛋白的哺乳动物细胞的方法,其包括
在生物反应器中在无血清培养基中建立哺乳动物细胞培养物;
使所述哺乳动物细胞在生长期期间生长并且用无血清进料培养基的批式进料(bolusfeed)补充所述培养基;和
通过用无血清灌注培养基灌注来在生产期期间维持所述哺乳动物细胞,其中在所述生产期期间的所述压积细胞体积小于或等于35%,
其中所述哺乳动物细胞是中国仓鼠卵巢(CHO)细胞。
2.根据权利要求1所述的方法,其中灌注开始于所述细胞培养的第5天时或第5天时至第9天时或第9天时。
3.根据权利要求1或2所述的方法,其中灌注开始于所述细胞培养的第5天时或第5天时至第7天时或第7天时。
4.根据权利要求1所述的方法,其中灌注开始于所述细胞已经达到生产期时。
5.根据权利要求1所述的方法,其进一步包括通过L-天冬酰胺饥饿随后用L-天冬酰胺浓度是5 mM或更小的无血清灌注培养基灌注来诱导细胞生长阻滞。
6.根据权利要求1所述的方法,其进一步包括通过用L-天冬酰胺浓度是5 mM或更小的无血清灌注培养基灌注来诱导细胞生长阻滞。
7.根据权利要求5-6中任一项所述的方法,其中所述无血清灌注培养基中的L-天冬酰胺浓度小于或等于4.0 mM。
8.根据权利要求5或6中任一项所述的方法,其中所述无血清灌注培养基中的L-天冬酰胺浓度小于或等于3.0 mM。
9.根据权利要求5或6中任一项所述的方法,其中所述无血清灌注培养基中的L-天冬酰胺浓度小于或等于2.0 mM。
10.根据权利要求5或6中任一项所述的方法,其中所述无血清灌注培养基中的L-天冬酰胺浓度小于或等于1.0 mM。
11.根据权利要求5或6中任一项所述的方法,其中所述无血清灌注培养基中的L-天冬酰胺浓度是0 mM。
12.根据权利要求5所述的方法,其中在L-天冬酰胺饥饿之前和期间监测所述细胞培养基的L-天冬酰胺浓度。
13.根据权利要求1所述的方法,其中所述压积细胞体积小于或等于30%。
14.根据权利要求1所述的方法,其中所述哺乳动物细胞培养物在小于或等于35%的压积细胞体积下的活细胞密度是10×106个活细胞/ml到80×106个活细胞/ml。
15.根据权利要求14所述的方法,其中所述哺乳动物细胞培养物的活细胞密度是20×106个活细胞/ml到30×106个活细胞/ml。
16.根据权利要求1所述的方法,其中灌注包括连续灌注。
17.根据权利要求1所述的方法,其中灌注速率是恒定的。
18.根据权利要求1所述的方法,其中灌注以小于或等于1.0工作体积/天的速率进行。
19.根据权利要求1所述的方法,其中灌注以在所述生产期期间从0.25工作体积/天增加到所述细胞培养期间的1.0工作体积/天的速率进行。
20.根据权利要求1所述的方法,其中灌注以在所述细胞培养的第9天到第11天达到1.0工作体积/天的速率进行。
21.根据权利要求1或20所述的方法,其中灌注以在所述细胞培养的第10天达到1.0工作体积/天的速率进行。
22.根据权利要求1所述的方法,其中无血清进料培养基的所述批式进料开始于所述细胞培养的第3天或第4天。
23.根据权利要求1所述的方法,其中通过以至少0.5×106到3.0×106个细胞/mL在无血清培养基中接种所述生物反应器来建立所述哺乳动物细胞培养物。
24.根据权利要求1和23中任一项所述的方法,其中通过以至少0.5×106到1.5×106个细胞/mL在无血清培养基中接种所述生物反应器来建立所述哺乳动物细胞培养物。
25.根据权利要求1所述的方法,其进一步包括从36℃到31℃的温度变换,其中所述温度变换在所述生长期与生产期之间过渡时发生,或者在所述生产期期间发生。
26.根据权利要求1所述的方法,其进一步包括从36℃到33℃的温度变换,其中所述温度变换在所述生长期与生产期之间过渡时发生,或者在所述生产期期间发生。
27.根据权利要求1所述的方法,其中所述灌注通过交替切向流来实现。
28.根据权利要求1所述的方法,其中所述生物反应器具有至少500 L的容量。
29.根据权利要求1和28中任一项所述的方法,其中所述生物反应器具有500 L到2000L的容量。
30.根据权利要求1或28中任一项所述的方法,其中所述生物反应器具有1000 L到2000L的容量。
31.根据权利要求1所述的方法,其中所述重组蛋白选自:人类抗体、人源化抗体、重组融合蛋白或细胞因子。
32.根据权利要求1所述的方法,其中所述重组蛋白是嵌合抗体。
33.根据权利要求1所述的方法,其进一步包括收获由所述细胞培养物生产的所述重组蛋白的步骤。
34.根据权利要求1所述的方法,其中将由所述细胞培养物生产的所述重组蛋白纯化并且配制成药学上可接受的制剂。
