该化合物具有很好的驱蠕虫效果,其治疗剂量比甲胺苯脒小5-10倍,具有相同疗效,药效高于现有商品药,且易于合成,成本低,无致畸,致突变作用,使用安全。

Description

抗蠕虫乙脒类化合物
本发明涉及一种抗蠕虫新药,特别是涉及一种名称为N′-[4(胡椒醛缩胺基)苯基]N、N-二甲基乙脒的抗蠕虫乙脒类化合物。
医用抗虫药种类很多,其中西德专利DE2346939题为AeyLamine-phengl-acetamidine(酰胺基苯基乙脒)公开了西德BayerAG药厂合成的对氨基苯二甲乙脒衍生物500余种,其中甲氨苯脒为同类化合物中疗效较高、毒性较低的一种,但对巴西日圆线虫驱虫效果较差,服用剂量较大。
本发明的目的是在于提供一种服用剂量低,驱虫效果好的其名称为N′-[4(胡椒醛缩胺基)苯基]N、N-二甲基乙脒的抗蠕虫乙脒类化合物,以下简称AH85012。
本发明抗蠕虫乙脒类化合物AH85012,分子式为C18H19N3O2、分子量309.368,化学结构式为:熔点124~126℃,核磁共振波谱分析H谱解析如下:δ(ppmm)溶剂为CDCl3

本发明抗蠕虫乙脒类化合物AH85012的制备方法是,在缩合剂及有机溶剂如苯系化合物存在下,将酰胺类与硝基苯胺加热回流,反应后得硝基脒类化合物,再将此化合物与还原剂进行还原反应得氨基脒类化合物,并用乙醚等进行分离提取,蒸馏回收溶剂得结晶物,最后将结晶化合物在催化剂和有机溶剂存在下与醛类化合物反应,回收溶剂得本发明产品,淡黄色结晶粉末。
本发明抗蠕虫乙脒类化合物AH85012药理活性筛选结果表明:驱除大鼠巴西日圆线虫的一次口服剂量10mg/kg时,虫体减少率为100%,5mg/kg时虫体减少率为99.3%,且虫体均在第一天内排出,在鼠粪中还查见鼠蛲虫,证明对鼠蛲虫有一定驱除作用。而一次口服甲胺苯脒剂量10mg/kg时,虫体减少率仅56.0%。药理活性筛选结果还表明:口服剂量1mg/kg时,对驱除犬钩虫即有效,剂量5mg/kg时虫体减少率为95.1%,剂量10mg/kg时虫体减少率为97.6%,剂量20mg/kg时虫体减少率为99.2%;口服甲胺苯脒剂量10mg/kg时,虫体减少率为95.0%;其阴性对照(不服药组)虫体减少率为0。本发明AH85012亦能驱除犬弓蛔虫,较大剂量时还可驱除犬绦虫。服用剂量2-10ng/kg时未见副反应,剂量20mg/kg时无明显副反应。
本发明抗蠕虫乙脒类化合物AH85012药理研究表明:剂量为160mg/kg以下时家犬心率、血压无明显影响;剂量为180mg/kg时出现呼吸改变,但可恢复;剂量为320mg/kg时有明显呼吸抑制和血压下降而死亡。高剂量时心电图除见T波倒置和室性期前收缩外,未有其它明显变化。AH85012长期毒性试验结果表明:采用两种动物试验,大白鼠给药剂量为300、600、800mg/kg及对照组连续给药14天,仅高剂量组(800mg/kg)有较明显食减和体重下降,停药即可恢复,血液及生化指标无明显变化,内脏病理切片无明显损害;实验犬15天长期毒性试验,给药剂量为9、30、90mg/kg,对家犬生长无影响,血象及生化检查证明对犬肝、肾无明显损害,病理切片无明显病变。
本发明抗蠕虫乙脒类化合物AH58012的优点在于治疗剂量比甲氨苯脒小5~10倍,具有相同疗效,驱虫快,且药效高于现有商品药、无致畸、无突变作用,使用安全,该化合物易于合成、成本低、原料易得,合成工艺成熟,产品质量收率稳定。
下面用实施例并结合附图详细说明本发明。

实施例1:将208.4g(1mol)五氯化磷和苯置于圆底烧瓶中加入122.0g(0.4mol)二甲基乙酰胺,再加入276.4g对硝基苯胺加热回流,回收溶剂得黄色沉淀物,干燥后取88.5g(0.43mol)与适量盐酸、锡粒加热进行还原反应,蒸干,用乙醚分离提取,蒸馏回收溶剂得结晶物,将该结晶物354g(2mol)和3.4-亚甲二氧基苯甲醛225g(约1.5mol)在适当的催化剂和甲苯中反应,回收溶剂后得本发明产品淡黄色结晶粉末。其熔点为124℃,核磁共振谱图如图1所示。
