Abstract
本发明公开了一种利用重编程制备猪神经干细胞的方法,本发明提供了一种利用重编程制备哺乳动物神经干细胞的方法,包括如下步骤:1)将基因OCT3/4、SOX2、KLF4、LMyc和LIN28用非整合型附着体载体共同导入哺乳动物体细胞中,得到转基因细胞,2)用包含丁酸钠的诱导培养体系所述转基因细胞,即得到神经干细胞;本发明的实验证明,本发明利用非整合型附着体载体(episomal载体)将OCT3/4、SOX2、KLF4、LMyc和LIN28等因子导入猪体细胞,通过优化培养条件,将体细胞转分化为神经干细胞,进一步分化获得有生理活性的神经细胞,为以猪为模型的神经系统疾病奠定基础。

技术领域
本发明涉及生物技术领域,尤其涉及一种利用重编程制备猪神经干细胞的方法。
背景技术
神经干细胞在神经生物学基础研究和神经系统疾病细胞治疗中具有广阔的前景。长期以来,神经干细胞的来源始终局限于胎脑或经由多能干细胞的定向分化,严重限制了其进一步的应用。目前,一种新型的转分化技术为提供了新的神经干细胞来源。神经转分化技术是直接将成体终末分化细胞特定向转化为神经细胞的技术。该技术不需要经过多能干细胞阶段,就可快速直接的获得神经细胞材料,因此将在神经修复性治疗中得到广泛应用。目前神经细胞转分化已在小鼠和大鼠等生物成功实现。然而,小鼠和大鼠在体形特征、生理特性、寿命等方面与人存在很大的差异,因而利用其作为模式生物模拟人类神经系统疾病和治疗存在明显的缺陷。相对而言,猪在解剖、生理生化和代谢等方面与人更为近似,在器官大小、免疫学和物种演化上也与人接近,因此建立猪的神经干细胞系更有利于研究疾病机制和探索有效的治疗模式。
迄今为止,以体细胞为起始材料获得猪神经细胞均是通过重编程实现的,即通过病毒载体将重编程因子导入猪体细胞,诱导体细胞重新获得多能性;进一步对诱导多能干细胞进行神经定向诱导分化,产生具有重要科研价值和临床价值的神经细胞(Jeong-YehYang,JenniferL.Mumaw,YubingLiu,SteveL.SticeandFranklinD.West.SSEA4PositivePigInducedPluripotentStemCellsArePrimedforDifferentiationintoNeuralCells.CellTransplantation.2013;22(6):945-59.)。然而与其他物种的重编程不同,在对猪体细胞重编程过程中引入的外源重编程因子无法沉默,因而获得的细胞为持续表达外源干性基因和内源干性基因的非经典诱导多能干细胞(ToshihikoEzashi,BhanuPrakashV.L.Telugu,AndreiP.Alexenko,ShrikeshSachdev,SunilimaSinha,andR.MichaelRoberts.Derivationofinducedpluripotentstemcellsfrompigsomaticcells.PNAS.2009.106(27):10993-10998.)。在此基础上定向分化产生的神经干细胞,在应用于基础研究或临床研究时,具有不可避免的偏见性。
发明内容
本发明的一个目的是提供一种利用重编程制备哺乳动物神经干细胞的方法。
本发明提供的方法,包括如下步骤:
1)将基因OCT3/4、SOX2、KLF4、LMyc和LIN28用非整合型附着体载体共同导入离体的哺乳动物体细胞中,得到转基因细胞,
2)用丁酸钠诱导培养所述转基因细胞,即得到神经干细胞;
所述基因OCT3/4的核苷酸序列为序列表中的序列1;
所述基因SOX2的核苷酸序列为序列表中的序列2;
所述基因KLF4的核苷酸序列为序列表中的序列3;
所述基因LMyc的核苷酸序列为序列表中的序列4;
所述基因LIN28的核苷酸序列为序列表中的序列5。
上述方法中,所述哺乳动物体细胞为哺乳动物成纤维细胞,所述哺乳动物成纤维细胞具体为哺乳动物胚胎成纤维细胞。
上述方法中,所述非整合型附着体载体为Episomal质粒载体。
上述方法中,所述共同导入的方法包括如下步骤:将表达OCT3/4的Episomal载体、表达SOX2和KLF4的Episomal载体、表达LMyc和LIN28的Episomal载体和表达标记基因的载体共同导入哺乳动物体细胞中。
上述方法中,所述表达OCT3/4的Episomal载体为pCXLE-hOCT3/4;
所述表达SOX2和KLF4的Episomal载体为pCXLE-hSK;

2)将所述哺乳动物体细胞培养基替换为含有50-200μM丁酸钠的多能干细胞培养基,继续培养;

4)将所述克隆样细胞在神经干细胞培养基中培养,得到哺乳动物神经干细胞。
上述方法中,步骤1)中,所述培养为连续培养,所述培养的时间为1-2天。
步骤2)中,所述培养的时间为3-4天。
步骤3)中,所述继续培养的时间为3-4周。
步骤4)中,所述继续培养的时间为4-5天。

本发明的另一个目的是提供一种由哺乳动物体细胞获得哺乳动物功能性神经细胞的方法。
本发明提供的方法,包括如下步骤:
1)按照上述的方法进行,得到哺乳动物神经干细胞;
2)将所述哺乳动物神经干细胞在神经分化培养基中培养,得到哺乳动物功能性神经细胞;
所述哺乳动物为人、大鼠、小鼠、猴、狗、猫、牛、兔、马或猪;所述哺乳动物具体为猪;
所述哺乳动物体细胞为哺乳动物成纤维细胞,所述哺乳动物成纤维细胞具体为哺乳动物胚胎成纤维细胞;

本发明的实验证明,本发明利用非整合型附着体载体(episomal载体)将OCT4,SOX2,KLF4、LMyc和LIN28等因子导入猪体细胞,利用丁酸钠诱导培养,将体细胞转分化为神经干细胞,进一步分化获得有生理活性的神经细胞,为以猪为模型的神经系统疾病奠定基础。
预期本发明所含技术将在转化医学研究和应用方面产生重要意义。与鼠等小动物不同,以猪为代表的大动物模型在神经医学转化研究中有着无可比拟的价值。目前已经产生了包括帕金森病、老年痴呆、自闭症等猪模型,很好的重现了神经系统疾病症状。因而本发明转分化产生的猪神经干细胞是用于评价神经细胞移植治疗效果和安全性的重要移植材料。同时,结合转基因猪疾病模型,还能够建立用于筛选神经系统疾病治疗药物的个性化药物筛选平台。
总之,通过表达载体直接从猪成纤维细胞转分化为神经干细胞及其衍生的神经细胞,为实验动物模型和临床提供大量用于疾病模拟和治疗性移植的细胞移植材料,并且应用于治疗神经退行性疾病的药物筛选和安全评估。

下述实施例中的慢病毒载体构建:
慢病毒载体包装质粒为pMDL、pCMV-VSVG和pRSV-REV(均从美国Addgene公司购买,货号:12251、35616和12253)。表达GFP的慢病毒质粒载体为pGreenZeo慢病毒载体(美国SystemBiosciences公司,货号:#SR500VA/PA)。病毒包装过程是利用Lipofectamine2000(购自美国Invitrogen公司,货号:11668019)将pMDL、pCMV-VSVG、pRSV-REV和pGreenZeo转染进入人胚胎肾细胞293T(购自美国ATCC,货号:CRL-11268),于转染48和72小时后收集培养上清混合,并用0.45μmPVDF膜(购自美国Millipore公司,货号:SLHV033)过滤。最后用超速离心的方法浓缩病毒载体),得到表达GFP的慢病毒载体(滴度为6×107Infectiousunits/ml)。
下述实施例中间接免疫荧光染色的方法如下:用4%多聚甲醛于4摄氏度固定细胞30分钟后,将细胞用0.1%PBS清洗3次,每次5分钟;之后用含有0.4%TritonX-100的0.1%PBS室温通透30分钟;用10%驴血清于4摄氏度封闭细胞1小时后,再用含有一抗的10%驴血清于4摄氏度孵育细胞过夜;将细胞用0.1%PBS清洗3次,每次10分钟;用含有二抗的10%驴血清室温孵育细胞2小时;将细胞用0.1%PBS清洗3次,每次5分钟;封片观察。。
下述实施例中qPCR检测方法如下:提取待测细胞RNA,并将其反转录为cDNA,再用对应分子标志物的扩增引物进行扩增,同时以GAPDH为内参。
下述实施例中电生理检测方法参照《实用膜片钳技术》,刘振伟,军事医科出版社,2006。
实施例1、利用重编程由猪胚胎成纤维细胞获得神经细胞
一、共导入
猪胚胎成纤维细胞(记载在如下文献中:LiuK.,etal.Generationofporcine-inducedpluripotentstemcellsbyusingOCT4andKlF4porcinefactors.CellReprogram.2012,14(6):505-513,公众可从中国科学院生物物理研究所获得)在猪体细胞培养基中进行体外培养。
首先弃去猪胚胎成纤维细胞的原有培养基,然后添加TrypLETMExpress(美国LifeTechnology公司,货号:12605010)于37摄氏度消化细胞,5分钟后使用猪体细胞培养基终止消化反应,1000rpm离心10分钟收集细胞,即为待导入猪胚胎成纤维细胞。
利用哺乳动物细胞电转仪将表达基因OCT3/4的Episomal质粒载体pCXLE-hOCT3/4(OCT3/4的核苷酸序列为序列表中的序列1),表达基因SOX2和KLF4的Episomal质粒载体pCXLE-hSK(SOX2的核苷酸序列为序列表中的序列2,KLF4的核苷酸序列为序列表中的序列3)、表达基因LMyc和LIN28的Episomal质粒载体pCXLE-hUL(LMyc的核苷酸序列为序列表中的序列4,LIN28的核苷酸序列为序列表中的序列5)和表达报告基因EGFP的载体pCXLE-EGFP(报告基因EGFP核苷酸序列为序列表中的序列6)各1.5μg共同电转导入待导入猪胚胎成纤维细胞,电转后将细胞在猪体细胞培养基中连续培养4天,得到转基因细胞。
二、丁酸钠诱导培养
1、猪神经干细胞的获得
丝裂霉素(购自美国Sigma公司,货号:M0503)灭活小鼠胚胎成纤维细胞(MEFs,购自美国LifeTechnology公司,货号:S1520-100),得到小鼠胚胎成纤维细胞饲养层。
1)将上述1得到的转基因细胞用TrypLETMExpress(美国LifeTechnology公司,货号:12605010)消化,进而接种至小鼠胚胎成纤维细胞饲养层上,在猪体细胞培养基中培养2天;
2)次日将猪体细胞培养基替换为含有50μM(工作浓度)丁酸钠(NaBT,为组蛋白去乙酰化抑制剂)的cDF12培养基继续培养3天;
3)将含有50μM丁酸钠的cDF12培养基将培养基更换为无NaBT的cDF12培养基再次培养至克隆样细胞出现(3周)(转分化流程详见图1所示);
4)将克隆样细胞消化为单细胞,并接种于Matrigel(购自美国BDBiosciences公司,货号:354277)包被的培养板上,用神经干细胞培养基继续培养4天,即为猪神经干细胞。
2、检测
1)克隆样细胞验证
为了验证产生的克隆样细胞的细胞特性,本发明进一步鉴定了其细胞标志物的表达。
将克隆样细胞进行间接免疫荧光染色,结果如图2所示,克隆样细胞不表达多能干细胞标志物OCT3/4和Nanog蛋白(图2A),却表达神经干细胞标志物Nestin蛋白(图2B),同时细胞克隆的形态也表现出神经上皮细胞典型的玫瑰花环(Rosette)结构。
2)神经干细胞鉴定
(1)细胞标志物鉴定
将上述1得到的猪神经干细胞进行间接免疫荧光染色,结果如图2C所示,发现其能够表达神经干细胞的另一种标志物Pax6。
将上述1得到猪神经干细胞进行qPCR,结果如图2D(横坐标中PEF代表猪胚胎成纤维细胞,ipNPC代表猪神经干细胞)所示,与猪胚胎成纤维细胞相比,克隆样细胞中的神经干细胞的分子标志物NCAM、Nestin、Pax6以及Sox2的基因表达均显著升高,说明其具有神经干细胞的特性,即为猪神经干细胞。
(2)猪神经干细胞具有体内存活和神经整合能力
为了鉴定本发明制备的猪神经干细胞是否具有体内存活和整合的能力,进行如下实验:
将表达GFP的慢病毒载体标记猪神经干细胞,然后将其定向移植进入小鼠(免疫缺陷小鼠NODSCID,专业名称为NOD.CB17-Prkdcscid/NcrCrlVr,品系代码为406,购自北京维通利华实验动物技术有限公司)大脑的海马区域,移植5周后分离小鼠大脑,并实施冰冻切片术观察移植部位。
用间接免疫荧光染色方法检测,结果如图4所示,脑内移植猪神经干细胞4-5周后,在小鼠大脑的海马齿状回区域,仍有大量表达绿色荧光蛋白的细胞存活,并且已表现出明显的神经细胞分化状态,同时其神经突起也表现出向周边脑区伸展的形态,说明本发明制备的猪诱导神经干细胞具有良好的体内存活和神经整合能力。
实施例2、猪神经干细胞定向分化为功能性神经细胞
1、功能性神经细胞的获得
将由实施例1制备的猪神经干细胞接种于Magrigel(购自美国BDBiosciences公司,货号:354277)包被的培养板中,添加神经分化培养基连续培养3周,得到猪功能性神经细胞。
2、功能性神经细胞的检测
检测上述1中得到的猪功能性神经细胞的细胞形态,产生的神经细胞的形态都是用间接免疫荧光检测其是否为神经元或神经胶质细胞。
将上述1得到的猪功能性神经细胞进行间接免疫荧光染色,结果如图3A所示,可以看出,猪诱导神经干细胞具有神经分化潜能,能够成功分化为Tuj1阳性的神经元和GFAP阳性的神经胶质细胞。
将功能性神经细胞进行qPCR检测,结果如图3B所示,证明在由猪诱导神经干细胞定向分化形成的功能性神经细胞群体中神经元分子标志物Tuj1和神经胶质细胞分子标志物GFAP的表达水平均显著升高,进一步证明,得到的功能性神经细胞为Tuj1阳性的神经元和GFAP阳性的神经胶质细胞。
由于电生理特性是神经细胞的重要生理表征,因此本发明进一步使用全细胞膜片钳技术对由猪诱导神经干细胞定向分化形成的功能性神经细胞实施了全细胞电流记录。
结果如图3C所示,显示定向分化获得的神经细胞能够产生正常的电压依赖的内向钠电流和外向钾电流。同时,通过电流钳记录,能够检测到神经细胞产生的动作电位和自发动作电位,更进一步地说明由猪诱导神经干细胞定向分化形成的神经细胞是功能性神经元。
Claims (8)
1.一种利用重编程制备哺乳动物神经干细胞的方法,包括如下步骤:
1)将基因OCT3/4、SOX2、KLF4、LMyc和LIN28用非整合型附着体载体共同导入离体的哺乳动物体细胞中,得到转基因细胞,
2)用丁酸钠诱导培养所述转基因细胞,即得到神经干细胞;
所述基因OCT3/4的核苷酸序列为序列表中的序列1;
所述基因SOX2的核苷酸序列为序列表中的序列2;
所述基因KLF4的核苷酸序列为序列表中的序列3;
所述基因LMyc的核苷酸序列为序列表中的序列4;
所述基因LIN28的核苷酸序列为序列表中的序列5;
所述用丁酸钠诱导培养所述转基因细胞包括如下步骤:
a将所述转基因细胞接种在小鼠胚胎成纤维细胞饲养层上,在哺乳动物体细胞培养基中培养;
b将所述哺乳动物体细胞培养基替换为含有50-200μM丁酸钠的多能干细胞培养基,继续培养;
c将所述含有50-200μM丁酸钠的多能干细胞培养基替换为无丁酸钠的多能干细胞培养基,再次培养至得到克隆样细胞;
d将所述克隆样细胞在神经干细胞培养基中培养,得到哺乳动物神经干细胞;
所述哺乳动物体细胞为猪胚胎成纤维细胞。
2.根据权利要求1所述的方法,其特征在于:
所述非整合型附着体载体为Episomal质粒载体。
3.根据权利要求1或2所述的方法,其特征在于:
所述共同导入的方法包括如下步骤:将表达OCT3/4的Episomal载体、表达SOX2和KLF4的Episomal载体、表达LMyc和LIN28的Episomal载体和表达标记基因的载体共同导入哺乳动物体细胞中。
4.根据权利要求3所述的方法,其特征在于:
所述表达OCT3/4的Episomal载体为pCXLE-hOCT3/4;
所述表达SOX2和KLF4的Episomal载体为pCXLE-hSK;
所述表达LMyc和LIN28的Episomal载体为pCXLE-hUL;
所述标记基因为EGFP,所述EGFP的核苷酸序列为序列表中的序列6;
所述表达标记基因的载体为pCXLE-EGFP。
5.根据权利要求3或4所述的方法,其特征在于:所述表达OCT3/4的Episomal载体、表达SOX2和KLF4的Episomal载体、表达LMyc和LIN28的Episomal载体和表达标记基因的载体等质量导入哺乳动物体细胞中。
6.根据权利要求1所述的方法,其特征在于:
步骤a中,所述培养为连续培养,所述培养的时间为1-2天;
步骤b中,所述继续培养的时间为3-4天;
步骤c中,所述再次培养的时间为3-4周;
步骤d中,所述培养的时间为4-5天。
7.根据权利要求1或2所述的方法,其特征在于:
所述多能干细胞培养基为cDF12培养基。
8.一种由哺乳动物体细胞获得哺乳动物功能性神经细胞的方法,包括如下步骤:
1)按照权利要求1-7中任一所述的方法进行,得到哺乳动物神经干细胞;
2)将所述哺乳动物神经干细胞在神经分化培养基中培养,得到哺乳动物功能性神经细胞;所述哺乳动物体细胞为猪胚胎成纤维细胞;
所述功能性神经细胞为Tuj1阳性的神经元或GFAP阳性的神经胶质细胞。
