Abstract
本发明涉及以贝壳为原料合成钙型干贝素——琥珀酸钙的新方法。主体工艺包括贝壳预处理、合成、结晶、烘干等工艺步骤。先将贝壳用0.05M盐酸浸泡、粉碎以及水飞法处理得钙源贝壳粉,按琥珀酸和贝壳粉(碳酸钙钙含量以95%计)摩尔比为1∶1.1~1.2配料,并加入200~250倍贝壳粉重量的蒸馏水,搅拌并加热至65~70℃,保温3.5~4.0小时,冷却,抽滤;将所得滤液减压浓缩至1/6~1/5体积,于4℃过夜、结晶;所得结晶用95%酒精洗涤1~2次,晾干;结晶在90℃烘箱中烘干即得琥珀酸钙产品。本发明工艺简便,可高值化利用多种贝壳,产品可作为鲜味剂、补钙剂等使用。
Description
技术领域:
本发明涉及一种利用贝类加工废弃物(蛤蜊壳、牡蛎壳、扇贝壳等)为钙源,经过高值化、清洁化工艺步骤,实现贝类加工废弃物综合利用的以贝壳为原料的钙型干贝素--琥珀酸钙合成方法。
背景技术:
琥珀酸二钠(Disodiumsuccinate),俗称干贝素,是近年来调味品中广泛应用的鲜味剂,在我国的肉制品、水产品、酱类、醋等家用调味料领域已经得到了广泛的应用,在日本琥珀酸二钠还被用到了酒类产品中。此外,琥珀酸作为体内三羧酸循环的中间代谢物,具有多种药理效应,在医药行业还被用作治疗抗癫痫的药物。
目前,作为鲜味剂使用的主要是钠盐,如谷氨酸一钠、琥珀酸二钠等。由于钠离子摄入过多存在引起高血压等心脑血管疾病的风险,无钠鲜调味剂开发是今后鲜味剂的发展方向。另一方面,目前国内外上市的各种维生素E类药品中,VE琥珀酸钙因稳定性好、熔点高、易贮存及具补钙功能等诸多优点,已经受到人们的重视。我国水产品资源丰富,但是贝类加工产生的大量废弃物(蛤蜊壳、牡蛎壳、扇贝壳等)尚有待开发应用。本发明着眼于贝类加工废弃物的高值化利用,利用其丰富钙质作为原料制备钙型干贝素--琥珀酸钙,不仅可作为鲜味剂,又可作为补钙使用,从而使其变废为宝,同时对保护环境也有重要意义。
本发明的目的在于提供一种以贝壳为原料合成琥珀酸钙的新方法。
为了实现上述目的,本发明的主体工艺包括贝壳预处理、合成、结晶、纯化、烘干工艺步骤,其中
贝壳预处理:将贝壳用0.05M盐酸浸泡1小时后刷洗去除贝壳表面泥沙等吸附物,湿法粉碎过60目筛,再经自来水水飞法处理、100℃烘干即得合成琥珀酸钙用钙源贝壳粉;
合成:按琥珀酸(食品级)和贝壳粉中碳酸钙摩尔比为1∶1.1~1.2配料,碳酸钙含量以占贝壳粉95%计,加200~250倍贝壳粉重量的蒸馏水,搅拌并加热至65~70℃,保温3.5~4.0小时,使琥珀酸完全反应,冷却,抽滤去除过量贝壳粉;
结晶:将上述抽滤所得清液减压浓缩至1/6~1/5体积,于4℃静置过夜、结晶,抽滤,所得结晶用95%酒精洗涤1~2次,晾干;
烘干:将上述所得结晶在90℃烘箱中烘干,即得琥珀酸钙产品。
本发明工艺过程简单,贝壳无需煅烧,合成过程无需人为调节反应液pH,所得产品质量好(白色,易溶),可高值化利用贝壳。
具体实施方式:
下面通过实例对本发明作进一步阐述:
实例1:将蛤蜊壳130克在0.05M盐酸溶液中浸泡1小时后刷洗去除贝壳表面泥沙等吸附物,湿法粉碎过60目筛,再经自来水水飞法处理、100℃烘干即得合成琥珀酸钙用钙源贝壳粉110克。将110克钙源贝壳粉与112.1克琥珀酸、22升蒸馏水混合、搅拌并加热至70℃,保温3.5小时,冷却,抽滤;将滤液减压浓缩至4.4升,于4℃静置过夜。抽滤,所得结晶用95%洗涤1次,晾干后在90℃烘箱中烘干,即得琥珀酸钙(C4H4CaO4·H2O)产品165.3克。
实例2:步骤与方法同实例1,原料选取牡蛎壳或扇贝壳,其制备琥珀酸钙产品的质量与蛤蜊壳相同。

1.以贝壳为原料的钙型干贝素——琥珀酸钙合成方法,包括如下步骤:
贝壳预处理:将贝壳用0.05M盐酸浸泡1小时后刷洗去除贝壳表面泥沙等吸附物,湿法粉碎过60目筛,再经自来水水飞法处理即得合成琥珀酸钙用钙源贝壳粉;
合成:按食品级琥珀酸和贝壳粉中碳酸钙摩尔比为1∶1.1~1.2配料,碳酸钙含量以占贝壳粉95%计,加200~250倍贝壳粉重量的蒸馏水,搅拌并加热至65~70℃,保温3.5~4.0小时,使琥珀酸完全反应,冷却,抽滤去除过量贝壳粉;
结晶:将上述抽滤所得清液减压浓缩至1/6~1/5体积,于4℃静置过夜、结晶,抽滤,所得结晶用95%酒精洗涤1~2次,晾干;
烘干:将上述所得结晶在90℃烘箱中烘干,即得琥珀酸钙产品;
贝壳包括蛤蜊壳、扇贝壳、牡蛎壳。
