Abstract

本发明公开一种配位金属的气味化合物的探测方法,利用以HEK293T以及其衍生细胞系为基础的哺乳动物嗅觉受体的异源表达系统,高效表达诸如小鼠嗅觉受体MOR244-3等一序列嗅觉受体,在加入气味化合物的同时在体系中另外加入铜离子,利用双荧光酶或Glosensor等测量受体活化后所产生的cAMP的方法检测嗅觉受体的激活,铜离子可以特异性地增强嗅觉受体的反应,达到高效的受体活化,且这种增强作用未见于其它金属离子。本发明中利用结合铜离子的细胞及其它平台可达到高灵敏度的探测及鉴别硫类及其它可螯合金属的化合物的目的,可被应用到工业及农业的一些领域,比如天然气泄漏检测、动物跟踪等。
