技术领域
本发明涉及一种玉米须浸提浓缩液及其制备方法,具体涉及一种以乳熟期玉米须和纯净水作为原料制备浓缩液,本发明属于保健饮料加工领域。
背景技术
玉米须为禾本科植物玉蜀黍ZeamaysL.的花柱和柱头,最早药用记载见于1476年的《滇南本草》。玉米须分离及鉴定的化合物主要有黄酮及其苷类、甾醇、生物碱、糖类、有机酸、挥发油、微量元素及多种维生素等。美国FDA将玉米须和玉米须浸提物作为一般公认为安全(GRAS)的可直接加入食品中的物质种类;我国卫生部《关于玉米须有关问题的批复》(卫监督函〔2012〕306号):玉米须在我国有一定的食用历史,未发现安全性问题,可作为普通食品管理。
现代医学研究证明,玉米须具有如下功效:利尿、抑制结石;降血糖、血压、血脂;提高免疫力;利胆、保肝;止血;抗氧化及抑制亚硝化反应、抗肿瘤等作用。药理学研究发现玉米须中起降糖作用的物质主要是多糖和皂苷类,玉米须水提液和粗多糖均有提高免疫力的作用,玉米须煎剂在治疗糖尿病、非酒精性脂肪肝、泌尿系结石、高血压等方面均有较好的临床观察结果。
玉米在不同生长阶段,玉米须多糖含量不同,乳熟期玉米多糖含量明显高于成熟期,使用乳熟期的鲜玉米须制备的浸提浓缩液,含有更多的多糖成分。各品种玉米乳熟期和成熟期玉米须多糖含量差别较大,农大610、东单213、郑单958和啃粘乳熟期玉米须中多糖含量分别是成熟期的9.79、10.28、16.98和9.60倍(吴华,梁清,张劲松,侯丽丽,刘春娟,张守勤,不同生长阶段不同玉米品种玉米须多糖含量的变化,安徽农业科学,2011,39(32):19716-19717);不同寒地品种玉米须成熟期玉米须多糖含量较乳熟期玉米须多糖含量降低,经单向配对检验,差异非常显著(刘玮,李佳梅,孙丽华,李兰红,北方寒地不同品种及生长期玉米须多糖含量比较研究,中国食物与营养2013,19(4):67-69)。
随着消费者对玉米须功效认知程度的提高,开发以玉米须为原料的饮料来满足消费者需求,例如中国专利号201010256252.2公开了“一种玉米须水及其制备方法”,其玉米须与纯净水的重量体积比为0.1-1kg:100L,参照玉米须(干须)在《全国中草药汇编》中,日用量为25-100g,每日玉米须水摄入量最低要2500mL,该产品无法满足消费者对玉米须功效的需求。
目前玉米须的工业最佳提取方法为热水浸提法,且制备方法简单。鉴于玉米须松散的特性,以及玉米须与水的浸提比例直接影响到总多糖的提取率,通过浓缩方法制备的浸提浓缩液,可以提高玉米须添加量,并保证浸提浓缩液中产品总多糖含量。
发明内容
本发明以乳熟期玉米须和纯净水作为原料,提供了一种玉米须浸提浓缩液,本发明针对生产中玉米须添加量过多不易提取和功效成分提取率低的问题,采用浓缩工艺制备出总多糖含量为2.0-3.5mg/mL,可作为代茶饮的玉米须浸提浓缩液,不仅可替代饮用水补充水分,而且有利尿、降血糖、降血压、降血脂、保肝护胆等需要的人群可将本发明作为日常饮品长期饮用。
本发明采用的技术方案为:
一种玉米须浸提浓缩液,其特征在于,它是由乳熟期玉米须和纯净水作为原料制成,其中,所述玉米须浸提浓缩液总多糖的含量为2.0-3.5mg/mL;
制备上述玉米须浸提浓缩液的方法,包括以下步骤:
1、取乳熟期鲜玉米须,或在-35℃速冷库中进行速冻,并在-18℃以下冷冻库中储存的乳熟期玉米须;
2、对玉米须进行除杂后,称取玉米须,使用纯净水进行清洗玉米须,后剪切至5-10mm;
3、将纯净水预热至90-95℃转至浸提罐中,将剪切后的玉米须放入浸提罐中,按照玉米须和纯净水比例(kg:L)为100:1000在90±2℃条件下浸泡1-2小时;
4、将浸泡溶液经浸提罐出口60目过滤及双联过滤器180目过滤得到滤液A;
5、将纯净水预热至90-95℃转至浸提罐,对步骤3中的玉米须进行二次浸泡,按照玉米须和纯净水比例(kg:L)为100:400-500在90±2℃条件下浸泡0.5-1.5小时;
6、将浸泡溶液经浸提罐出口60目过滤及双联过滤器180目过滤得到滤液B,并将滤液B与滤液A混合;
7、将A与B的混合滤液在双效降膜蒸发器中浓缩2.5-3.5倍,得到滤液浓缩液;
8、对玉米须浓缩液进行检测,总多糖含量为2.0-3.5mg/mL;
9、将浸提浓缩液在65-80℃进行热灌装,使用自动灌装封口机进行灌装封合,在杀菌釜中进行121℃、20分钟灭菌,产品通过包装后,即为玉米须浸提浓缩液。
本发明的技术特点:
1、玉米须浸提浓缩液未添加任何其他成分,且玉米须水无毒副作用,适合健康人群作为日常饮品长期饮用;
2、本发明采用乳熟期的玉米须,该时期的玉米须中多糖含量位于顶峰期,远高于成熟期玉米须中多糖含量;
3、本发明将玉米须剪切至5-10mm的小段,利于功效成分的溶出的同时,还可以保证后期过滤工艺的简便操作,提高产品总多糖含量。
4、本发明按照玉米须和纯净水比例(kg:L)为100:1000进行初次浸提,按照玉米须和纯净水比例(kg:L)为100:400-500进行二次浸提,最大限度的提取玉米须中的总多糖;
5、本发明的玉米须浸提浓缩液中,总多糖的含量为2.0-3.5mg/mL,适合有利尿、降血糖、降血压、降血脂、保肝护胆等需要的人群作为日常饮品长期饮用。
本发明制备方法简单易行,适用于工业化生产。

实施例1鲜玉米须浸提浓缩液的制备
1、取垦粘1号乳熟期玉米鲜须进行除杂后称取100kg;
2、使用纯净水清洗玉米须,后剪切至5-10mm;
3、将纯净水预热至90℃转至浸提罐中,将剪切后的玉米须放入浸提罐中,按照鲜玉米须和纯净水比例(kg:L)为100:1000在90±2℃条件下浸泡1.5小时;
4、将浸泡溶液经过滤得到滤液A;
5、将纯净水预热至90℃转至浸提罐,对步骤3中的玉米须进行二次浸泡,按照鲜玉米须和纯净水比例(kg:L)为100:400在90℃条件下浸泡1小时;
6、将第二次浸泡溶液经过滤得到滤液B,并与滤液A混合;
7、将混合滤液在双效降膜蒸发器中浓缩3倍,得到浸提浓缩液;
8、对鲜玉米须浸提浓缩液进行检测,总多糖含量为2.63mg/mL;
9、将浸提浓缩液在75℃进行热灌装,在杀菌釜中进行121℃、20分钟灭菌,产品通过包装后,即为鲜玉米须浸提浓缩液。
试验例1玉米须浸提浓缩液(以下简称玉米须液)总多糖含量检测方法
1仪器与试剂
1.1仪器:可见分光光度计、电子分析天平等。
1.2试剂:葡萄糖(分析纯)、浓硫酸(分析纯)、蒽酮(分析纯)、实施例1制备的玉米须液样品。
1.3试剂配制:
葡萄糖:准确称取105℃烘干至恒重的0.1000g葡萄糖,用蒸馏水溶解,容量瓶定容至1000mL,摇匀,即得0.1mg/mL葡萄糖对照溶液。
体积浓度为80%的硫酸:浓硫酸浓度按98%计,取204mL浓硫酸,沿壁加到46mL蒸馏水中,然后冷却至室温,摇匀备用。
蒽酮试剂:准确称取0.1000g蒽酮,加入100mL体积浓度为80%的硫酸,搅拌溶解蒽酮,摇匀,转入棕色瓶中,避光保存,当天有效。
2标准曲线绘制
2.1葡萄糖标准反应液制备
按表1配制标准样品,加塞,摇匀,置水浴(98℃-100℃)中加热15min,取出,放入冰浴(0℃)中暗处冷却15min,以0号管样为空白,在625nm波长处测定吸光度,测定结果见表1。

2.2标准曲线绘制
浓度C(ug/mL)与吸光度A标准方程为:C=26.853×A+0.1303,R2=0.9992,浓度C线性范围为2.5ug/mL-15ug/mL。蒽酮硫酸法葡萄糖标准曲线图见图1。
3样品检测
玉米须液摇匀,使用移液管准确吸取1mL样品,置100mL容量瓶中定容至100mL,然后准确吸取2mL放入试管,空白样加2mL蒸馏水,再分别吸取6mL蒽酮试剂,沿管壁加入,加塞,摇匀,放入100℃水浴中15min,取出,先用自来水快速冷却至室温,然后置冰水浴中、暗处冷却15min,以空白为对照,在625nm下检测吸光度。
计算公式:总多糖(mg/mL)=(26.853×吸光度+0.1303)×8÷2×100÷1000
实施例1制备的鲜玉米须浸提浓缩液检测结果:吸光度为0.240;
总多糖=(26.853×0.240+0.1303)×8÷2×100÷1000=2.63mg/mL
试验例2剪切与不剪切玉米须总多糖含量
1、采用垦粘1号乳熟期玉米须,玉米须经干燥处理
2、玉米须提取工艺:第一次90±2℃条件下浸泡60分钟
第二次90±2℃条件下浸泡40分钟
3、干燥玉米须:纯净水=1:20
4、检测结果见表2:

Claims (1)
1.一种制备玉米须浸提浓缩液的方法,其特征在于,包括以下步骤:
(1)取乳熟期鲜玉米须,或在-35℃速冷库中进行速冻,并在-18℃以下冷冻库中储存的乳熟期玉米须;
(2)对玉米须进行除杂后,称取玉米须,使用纯净水进行清洗玉米须,后剪切至5-10mm;
(3)将纯净水预热至90-95℃转至浸提罐中,将剪切后的玉米须放入浸提罐中,按照玉米须和纯净水比例(kg:L)为100:1000在90±2℃条件下浸泡1-2小时;
(4)将浸泡溶液经浸提罐出口60目过滤及双联过滤器180目过滤得到滤液A;
(5)将纯净水预热至90-95℃转至浸提罐,对步骤(3)中的玉米须进行二次浸泡,按照玉米须和纯净水比例(kg:L)为100:400-500在90±2℃条件下浸泡0.5-1.5小时;
(6)将浸泡溶液经浸提罐出口60目过滤及双联过滤器180目过滤得到滤液B,并将滤液B与滤液A混合;
(7)将A与B的混合滤液在双效降膜蒸发器中浓缩2.5-3.5倍,得到滤液浓缩液;
(8)对玉米须浓缩液进行检测,总多糖含量为2.0-3.5mg/mL;
(9)将浸提浓缩液在65-80℃进行热灌装,使用自动灌装封口机进行灌装封合,在杀菌釜中进行121℃、20分钟灭菌,产品通过包装后,即为玉米须浸提浓缩液。
