一种基于流式细胞术的精子线粒体膜电位检测试剂
技术领域
本发明涉及精子线粒体膜电位检测试剂,具体涉及一种基于流式细胞术的精子线粒体膜电位检测试剂。
背景技术
传统的精液检测方法主要针对精子的运动力和形态,这些检测结果不能全面、准确地反映精子功能和状况。线粒体的主要功能是产生能量,为精子运动提供ATP,线粒体活性与精子的运动能力密切相关。线粒体膜电位又是启动细胞凋亡过程最早出现的因子,线粒体膜电位的测定对于精子质量和功能的测定意义尤为重要。因此,线粒体的功能状态是精子功能质量的一个关键性指标。通常对线粒体活性的评价主要通过检测线粒体的跨膜电位变化来分析。
传统的精子线粒体膜电位检测依赖于显微镜检测法,如罗丹明123(R123)染色法、DiOC6(3)碘化物染色法等,均可利用线粒体染色液对精子线粒体染色,此类方法存在诸多不足,检测结果过于依赖人为操作,检测结果主观性强;操作过程繁琐,检测效率低下,重复性差;染色检测时精子活力降低,部分染料在线粒体中有较多能量依赖型结合位点,敏感性较差。
发明内容
本发明所要解决的技术问题是提供一种染色敏感性好,检测结果准确的基于流式细胞术的精子线粒体膜电位检测试剂。
本发明解决上述技术问题所采用的技术方案为:一种基于流式细胞术的精子线粒体膜电位检测试剂,包括线粒体染色液,还包括染色缓冲液,所述染色缓冲液的pH值7.0~7.8,含有浓度为0.05~0.19mol/L的NaCl、浓度为0.5~2.0mmol/L的KH2PO4、浓度为2.0~10.5mmol/L的C6H6O6、浓度为2.3~7.8mmol/L的KCl、浓度为0.2~2.4mmol/L的MgSO4、浓度为0.8~3.4mmol/L的CaCl2、浓度为0.01~0.04mol/L的N-2-羟乙基哌嗪-N-2-乙磺酸二钠盐(Hepes-Na)、浓度为2.4~5.6mmol/L的NaHCO3、浓度为0.1~0.5mmol/LC3H3O3Na、体积百分浓度为0.15~0.75%的C3H5O3Na溶液。
线粒体染色液可以为JC-1(5,5′,6,6′-四氯-1,1′,3,3′-四乙基苯并咪唑基-羰化青碘溶于DMSO(二甲基亚砜)而成,JC-1浓度为0.1~1.5mmol/L。
与现有技术相比,本发明优点在于本发明解决上述技术问题所采用的技术方案为:一种基于流式细胞术的精子线粒体膜电位检测试剂,包括线粒体染色液,还包括染色缓冲液,染色缓冲液的pH值7.0~7.8,含有NaCl、KH2PO4、C6H6O6、KCl、MgSO4、CaCl2、N-2-羟乙基哌嗪-N-2-乙磺酸二钠盐、NaHCO3、C3H3O3Na、C3H5O3Na溶液。该染色缓液冲有助于精子线粒体染色液染色,染色敏感性好,因此本发明是一种染色敏感性好,检测结果准确的基于流式细胞术的精子线粒体膜电位检测试剂。
具体实施方式
以下结合实施例对本发明作进一步详细描述。
实施例1、一种基于流式细胞术的精子线粒体膜电位检测试剂,包括线粒体染色液和染色缓冲液,线粒体染色液为浓度为0.1~1.5mmol/L的JC-1液;染色缓冲液的pH值7.0~7.8,含有浓度为0.12mol/L的NaCl、浓度为1.2mmol/L的KH2PO4、浓度为6.0mmol/L的C6H6O6、浓度为5.0mmol/L的KCl、浓度为1.2mmol/L的MgSO4、浓度为2.0mmol/L的CaCl2、浓度为0.025mol/L的Hepes-Na、浓度为4mmol/L的NaHCO3、浓度为0.3mmol/LC3H3O3Na、体积百分浓度为0.45%的C3H5O3Na溶液。
实施例2、与实施例1基本相同,所不同的只是染色缓冲液含有浓度为0.05mol/L的NaCl、浓度为0.5mmol/L的KH2PO4、浓度为2.0mmol/L的C6H6O6、浓度为2.3mmol/L的KCl、浓度为0.2mmol/L的MgSO4、浓度为0.8mmol/L的CaCl2、浓度为0.01mol/L的Hepes-Na、浓度为2.4mmol/L的NaHCO3、浓度为0.1mmol/LC3H3O3Na、体积百分浓度为0.15%的C3H5O3Na溶液。
实施例3、与实施例1基本相同,所不同的只是染色缓冲液含有浓度为0.19mol/L的NaCl、浓度为2.0mmol/L的KH2PO4、浓度为10.5mmol/L的C6H6O6、浓度为7.8mmol/L的KCl、浓度为2.4mmol/L的MgSO4、浓度为3.4mmol/L的CaCl2、浓度为0.04mol/L的Hepes-Na、浓度为5.6mmol/L的NaHCO3、浓度为0.5mmol/LC3H3O3Na、体积百分浓度为0.75%的C3H5O3Na溶液。
检测例、取出的精液放置30~60分钟,使之完全液化。用染色缓冲液稀释精子浓度至1×106/mL,体积为1mL;加入10μL线粒体染色液,37℃避光孵育15min;然后流式细胞仪进行检测(488nm激发波长),检测红色荧光和红色荧光值,计数5000-10000个细胞;结果分析:精子线粒体膜电位正常率为37.22%,说明染色敏感性好。流式细胞仪检测时操作简单,检测效率高,重复性好,检测结果准确。

1.一种基于流式细胞术的精子线粒体膜电位检测试剂,包括线粒体染色液,其特征在于还包括染色缓冲液,所述染色缓冲液的pH值7.0~7.8,含有浓度为0.05~0.19mol/L的NaCl、浓度为0.5~2.0mmol/L的KH2PO4、浓度为2.0~10.5mmol/L的C6H6O6、浓度为2.3~7.8mmol/L的KCl、浓度为0.2~2.4mmol/L的MgSO4、浓度为0.8~3.4mmol/L的CaCl2、浓度为0.01~0.04mol/L的N-2-羟乙基哌嗪-N-2-乙磺酸二钠盐、浓度为2.4~5.6mmol/L的NaHCO3、浓度为0.1~0.5mmol/LC3H3O3Na、体积百分终浓度为0.15~0.75%的C3H5O3Na溶液。
