Abstract

技术领域
本发明涉及分子生物和生物医药领域。更具体而言,本发明涉及磷酸化途径相关因子、其激动剂或抑制剂通过调节FOXP3、IL-2、GITR和CTLA4的活性来调控调节性T细胞功能的用途。本发明尤其涉及调节FOXP3活性的负调节因子及其通过调控调节性T细胞产生的免疫抑制功能。

FOXP3是调节性T细胞(Treg,regulatory T cells)特异性转录因子,对Treg的发育和功能起着重要的调节作用,具有下调免疫应答的功能。FOXP3介导的Treg的免疫调节功能是通过FOXP3与若干转录共调节蛋白(例如转录因子、共抑制因子、共激活因子、组蛋白以及染色质重建因子)形成蛋白复合体以动态调控基因的特异性转录(参见参考文献3和4)。在不同刺激条件下,FOXP3蛋白复合体的不同状态及其转录活性对Treg及免疫系统调节都有不同的效果(参见参考文献3和4)。
FOXP3+调节性T细胞(FOXP3+Tregs)属于T淋巴细胞中表达CD4、CD25及转录因子FOXP3的一类T细胞亚型(即CD4+CD25+Treg),其正常功能对于人体免疫稳态的动态调节必不可少。调节性T细胞的发育及功能失调,与多种重大免疫相关性疾病,包括自身免疫性疾病、炎症反应、急性及慢性传染性疾病、肿瘤免疫耐受、移植排斥以及过敏性疾病的生理病变进程密切相关(参见参考文献1和参考文献2)。
尽管本研究领域近年来进展很快,但许多重大及根本性的问题仍然尚待回答,例如在细胞水平上调节性T细胞如何调节其它免疫细胞活性,以及在分子水平上FOXP3采用何种机制使得调节性T细胞获得免疫抑制活性。
不断积累的实验证据显示,FOXP3基因表达的水平及持续状态对于天然调节性T细胞的发育成熟及功能至关重要。此外,FOXP3结合多重转录因子以及具有酶学活性的组蛋白乙酰转移酶/脱乙酰基酶复合体,这对于抑制T细胞中促炎症类细胞因子的转录激活是必需的。并且,FOXP3蛋白翻译后修饰、转录复合体装配及其修饰酶类的活性受到T细胞受体及炎症细胞因子受体信号的动态调节。先前的研究发现FOXP3是一个赖氨酸乙酰化蛋白,并且FOXP3富含脯氨酸的N端可以直接招募组氨酸乙酰转移酶TIP60(Tat interaction protein,60kDa),从而介导FOXP3的转录抑制活性(参见参考文献6)。
原癌基因PIM1是一种组成性激活的Ser/Thr激酶,首先在淋巴瘤中作为原癌基因MYC的协同癌基因被发现。随后,在多种人癌组织中发现该原癌基因的表达,其中包括胰腺癌、急性骨髓性白血病及其它恶性肝癌。PIM1普遍表达于脾、胸腺、骨髓、胰腺、口腔内皮细胞等组织或细胞中,且高表达于肿瘤细胞中。PIM1具有许多重要作用,包括影响细胞程序性死亡、基因转译激活及细胞信号传导等。
IL-2是一种淋巴细胞因子,通常由凝集素或抗原激活的T淋巴细胞产生。可使细胞毒性T细胞、自然杀伤细胞和淋巴因子活化的杀伤细胞增殖,并使其杀伤活性增强,还可以促进淋巴细胞分泌抗体和干扰素,具有抗病毒、抗肿瘤和增强机体免疫功能等作用。人重组IL-2在肿瘤、炎症反应、急性传染性疾病或慢性传染性疾病的治疗中已有相应应用。
GITR是肿瘤坏死因子受体蛋白家族成员,又名TNFRSF18,通常在激活的T细胞中表达增加,同时也是调节性T细胞中重要的细胞因子,并在固有免疫中也起重要作用。小鼠体内基因敲除该基因表明GITR调控CD3驱动的T细胞激活(CD3-driven T-cell activation)及调控细胞程序性死亡,其它的研究表明用激活GITR抗体可抑制调节性T细胞功能或刺激调节性T细胞或效应性T细胞增殖。
CTLA4是免疫球蛋白家族成员。通常在辅助性T细胞表面表达并转导抑制信号到T细胞。CTLA4是T细胞共刺激因子CD28的类似物,可结合抗原呈递细胞表面的CD80和CD86。CTLA4也是调节性T细胞中重要的细胞因子,对调节性T细胞功能有重要作用。
因此,深入研究调节性T细胞及转录因子FOXP3、IL-2、GITR和CTLA4中的生化活性、生理功能及其调控的分子机理,对于治疗性控制人体内调节性T细胞的免疫活性以及基于调节性T细胞的免疫治疗皆至关重要。这为将基础免疫研究转化为临床研究,以及理解人类特有的重大传染性疾病提供了新的研究命题与挑战。
发明内容
本发明的主要目的之一是提供磷酸化途径相关因子及其激动剂或拮抗剂在调节FOXP3活性、IL-2活性、GITR和CTLA4活性中的用途。本发明的另一目的在于提供这些调节因子通过调节FOXP3、IL-2、GITR和CTLA4的活性来调控调节性T细胞功能的用途。本发明的其它重要目的还包括:提供原癌基因表达蛋白PIM1和/或其编码序列、其激动剂或拮抗剂在制备包含调节性T细胞的组合物或调控调节性T细胞活性的组合物中的用途;制备包含调节性T细胞的组合物方法;调控调节性T细胞活性的方法;以及一种用于制备包含调节性T细胞的组合物或调控调节性T细胞活性的组合物的试剂、试剂盒或药盒。
在本发明的第一方面中,提供了磷酸化途径相关因子、其激动剂或拮抗剂在制备调控调节性T细胞的免疫调节功能的组合物中的用途,其中所述磷酸化途径相关因子选自:原癌基因表达蛋白PIM1和/或其编码序列,且其中所述调控是通过调节选自下组的调节性T细胞调节因子的活性进行的:FOXP3、IL-2、GITR、CTLA4或它们的组合。
在本发明的一个实施方式中,对FOXP3、IL-2、GITR和CTLA4或它们的组合的活性调节是正调节或负调节,其中,所述磷酸化途径相关因子或其激动剂负调节FOXP3、GITR或CTLA4活性和正调节IL-2活性,所述磷酸化途径相关因子的拮抗剂正调节FOXP3、GITR或CTLA4活性和负调节IL-2活性。

具体实施方式
发明人认为FOXP3会受到一系列时间和空间上的调控,包括转录后修饰、翻译或修饰以及蛋白相互作用等,其中有一类因子(包括酶、小分子化合物等)作为FOXP3的负调节因子,在不同的信号刺激下能够下调FOXP3的活性,从而调控整个免疫系统。通过长期而深入的研究,发明人发现了磷酸化途径相关因子可影响FOXP3与染色质的结合,进而调节FOXP3对其靶基因的转录调控活性。这些磷酸化途径相关因子为FOXP3负调节因子——包括PIM1蛋白及其炎症诱导信号。由此,发明人还在本申请中进一步提供了原癌基因表达蛋白PIM1和/或其编码序列、其激动剂或拮抗剂在制备包含调节性T细胞的组合物或调控调节性T细胞活性的组合物中的用途;制备包含调节性T细胞的组合物方法;调控调节性T细胞活性的方法;以及一种用于制备包含调节性T细胞的组合物或调控调节性T细胞活性的组合物的试剂、试剂盒或药盒。
具体而言,发明人在本发明之前的研究中构建了FOXP3稳定表达细胞株,并在不同信号刺激条件下纯化FOXP3蛋白,对纯化的蛋白质进行质谱分析,从而鉴定出FOXP3蛋白序列内多个Ser/Thr/Tyr磷酸化位点。在本研究中,发明人通过大量的筛选试验,发现FOXP3-S422位点是PIM1潜在的磷酸化位点。进一步的试验证明,PIM1蛋白可与FOXP3相互结合,并且能够在细胞内、细胞外使FOXP3在Ser422特异性位点磷酸化。并且,进一步研究还发现,在炎症因子刺激作用下,比如IL-6、IL-4等的刺激下,体外扩增的天然调节性T细胞(nTreg)能够迅速上调PIM1蛋白的表达水平。由此推断,PIM1有可能对nTreg的活性发挥重要的调节作用。
最近,研究发现PIM1在协同原癌基因编码蛋白MYC促进正常细胞向癌化细胞转化中发挥重要的调节作用。PIM1可以与MYC蛋白相互作用,并被其招募到转录因子MYC的靶基因上并使组蛋白H3第十位丝氨酸Ser10发生磷酸化,从而启动靶基因的转录。我们发现,PIM1在FOXP3序列上的磷酸化位点位于其叉头框结构域(Forkhead domain)中,而该结构与是决定其与DNA或染色质结合的重要结构域。我们由此认为,PIM1有可能影响FOXP3与基因组DNA的结合,从而调节nTreg的生理活性。试验结果与预期相符,过表达PIM1会显著抑制FOXP3与染色质的结合;
FOXP3+Treg中敲除PIM1能够显著增强FOXP3对其靶基因的转录调控活性:包括增强对FOXP3正向调节基因(CTLA4、GITR、CD25等)转录的促进和负向调节基因(IL-2)转录的抑制。利用PIM1激酶特异性抑制剂处理体外扩增的nTreg细胞,可以明显提高CD25+FOXP3+Treg及CTLA4+FOXP3+Treg的细胞水平。同时,体外细胞抑制试验显示,PIM1抑制剂可显著增强nTreg的免疫抑制活性。由此,我们确信,PIM1是通过对FOXP3的磷酸化修饰,从而调节FOXP3的转录调节活性并抑制FOXP3+Treg的免疫抑制活性。
已有的研究发现,单独提高FOXP3的表达水平并不能增强nTreg的免疫抑制活性,暗示FOXP3转录调控活性的发挥需要其它信号的调节。我们的研究发现为此提供了一条可能的途径,在调高FOXP3的表达水平的同时抑制PIM1的表达水平或其活性,有可能显著提高FOXP3+Treg的免疫抑制活性。
基于上述研究,本发明人在本发明中提供了首次揭示了FOXP3磷酸化途径相关因子――PIM1激酶及诱导其表达的炎症细胞因子对FOXP3+Treg免疫活性的调节机制以及对调节性T细胞的活性调控作用。这些调节因子的确定与应用,为通过调节FOXP3+Treg细胞的免疫抑制活性,治疗和/或预防与FOXP3+Treg细胞活性失调相关的疾病或症状(例如肿瘤、自身免疫性疾病等)、制备调节性T细胞或者调控其活性、以及筛选药物提供了新途径,同时也提供了利用这些调节因子作为疫苗佐剂,提高病毒感染的免疫原性的新方法。
在此基础上,发明人完成了本发明。
磷酸化途径相关因子、其激动剂或拮抗剂
如本文所用,术语“调节因子”或“活性调节因子”是指磷酸化途径相关因子或其激动剂或拮抗剂,它们可通过磷酸化FOXP3并影响FOXP3与染色质的结合,进而调节FOXP3对其靶基因的转录调控活性,调控Treg的免疫抑制活性,从而可用于治疗和/或预防与FOXP3+Treg细胞活性失调相关的疾病或症状(例如肿瘤、自身免疫性疾病)或作为疫苗佐剂提高病毒感染的免疫原性。
如本文所用,术语“磷酸化途径相关因子”是指可通过影响磷酸化途径来影响其下游因子或过程的活性(如FOXP3对其靶基因转录调控活性、调节性T细胞对免疫功能的调节活性)的活性因子,在本发明中,所述磷酸化途径相关因子包括:PIM1蛋白和/或其编码序列。
如本文所用,属于PIM1蛋白是指原癌基因PIM1,其为一种组成性激活的Ser/Thr激酶,首先在淋巴瘤中作为原癌基因MYC的协同癌基因被发现。本领域中已知的PIM1序列包括但不限于:Entrez Gene:5292。
如本文所用,磷酸化途径相关因子的“激动剂”是指能够促进磷酸化途径相关因子的表达或提高磷酸化途径相关因子的活性的物质,在本发明中也可称之为“促效剂”,例如PIM1的激动剂。可用于本发明中的PIM1激动剂包括但不限于:炎症细胞因子IL-6、IL-4;TCR;和/或其编码序列。广义而言,PIM1的促效剂还包括通过基因工程手段导入机体、组织或细胞以提高PIM1水平或活性的物质,例如PIM1转基因和/或其载体。
如本文所用,磷酸化途径相关因子的“拮抗剂”是指能够抑制磷酸化途径相关因子的表达或降低磷酸化途径相关因子的活性的物质,在本发明中也可称之为“抑制剂”。可用于本发明中的拮抗剂包括但不限于如下的PIM1拮抗剂:抗PIM1抗体、shPIM1、针对PIM1的反义寡核苷酸、PIM1的化学抑制剂,例如见参考文献8-14,如:PIM1抑制剂:4-[3-(4-氯苯基)-2,1-苯并异噁唑-5-基]-2-嘧啶胺,即

本发明的上述蛋白质或多肽可以是天然纯化的产物,或是化学合成的产物,或使用重组技术从原核或真核宿主(例如,细菌、酵母、高等植物、昆虫和哺乳动物细胞)中产生。
应理解,本发明的上述术语定义中也包括所述蛋白质或多肽的保守性变异多肽、或其同源多肽。在本发明的一个实施方式中,所述蛋白质或多肽来自人或其它真核生物,例如小鼠、大鼠、牛或猴等,且它们之间具有高度的保守性。
本发明蛋白质或多肽的变异形式包括(但并不限于):一个或多个(通常为1-50个,较佳的1-30个,更佳的1-20个,最佳的1-10个,例如1、2、3、4、5、6、7、8、9或10个)氨基酸的缺失、插入和/或取代,以及在C末端和/或N末端添加一个或数个(通常为20个以内,较佳地为10个以内,更佳地为5个以内)氨基酸。例如,在本领域中,用性能相近或相似的氨基酸进行取代时,通常不会改变蛋白质或多肽的功能。又比如,在C末端和/或N末端添加一个或数个氨基酸通常也不会改变蛋白质或多肽的功能。本领域技术人员可根据本领域常识和/或常规试验很容易的确定这些变异方式,而不会影响蛋白或多肽的活性。
如本文所用,术语“编码序列”是指编码本发明所述的原癌基因PIM1的序列,或是它们的高度同源序列或在严格条件下与所述序列杂交的分子或与上述分子高度同源的家族基因分子。
如本文所用,术语“严格条件”是指:(1)在较低离子强度和较高温度下的杂交和洗脱,如0.2×SSC,0.1%SDS,60℃;或(2)杂交时加有变性剂,如50%(v/v)甲酰胺,0.1%小牛血清/0.1%Ficoll,42℃等;或(3)仅在两条序列之间的相同性至少在50%,优选55%以上、60%以上、65%以上、70%以上、75%以上、80%以上、85%以上或90%以上,更优选是95%以上时才发生杂交。
本发明的编码序列全长序列或其片段通常可以用PCR扩增法、重组法或人工合成的方法获得。对于PCR扩增法,可根据本发明所公开的有关核苷酸序列,尤其是开放阅读框序列来设计引物,并用市售的cDNA库或按本领域技术人员已知的常规方法所制备的cDNA库作为模板,扩增而得有关序列。当序列较长时,常常需要进行两次或多次PCR扩增,然后再将各次扩增出的片段按正确次序拼接在一起。
组合物以及疾病治疗或预防
本发明的调节因子还可用于制备治疗性或预防性的药物组合物、保健品组合物或疫苗组合物、或者佐剂组合物。
因此,另一方面,本发明还提供了一种组合物,它含有(a)安全有效量的本发明的调节因子;以及(b)药学上可接受的载体或赋形剂。本发明调节因子的数量通常为10微克-100毫克/剂,较佳的为100-1000微克/剂。
本文所用的术语“有效量”指治疗剂治疗、缓解或预防目标疾病或状况的量,或是表现出可检测的治疗或预防效果的量。对于某一对象的精确有效量取决于该对象的体型和健康状况、病症的性质和程度、以及选择给予的治疗剂和/或治疗剂的组合。因此,预先指定准确的有效量是没用的。然而,对于某给定的状况而言,可以用常规实验来确定该有效量,临床医师是能够判断出来的。
药物组合物还可含有药学上可接受的载体。术语“药学上可接受的载体”指用于治疗剂给药的载体。该术语指这样一些药剂载体:它们本身不诱导产生对接受该组合物的个体有害的抗体,且给药后没有过分的毒性。这些载体被本领域普通技术人员所熟知。在Remington's Pharmaceutical Sciences(Mack Pub.Co.,N.J.1991)中可找到关于药学上可接受的赋形剂的充分讨论。这类载体包括(但并不限于):盐水、缓冲液、葡萄糖、水、甘油、乙醇、佐剂及其组合。
治疗性组合物中药学上可接受的载体可含有液体,如水、盐水、甘油和乙醇。另外,这些载体中还可能存在辅助性的物质,如润湿剂或乳化剂、pH缓冲物质等。此外,免疫组合物中还可以含有免疫佐剂。
通常,可将治疗性组合物制成可注射剂,例如液体溶液或悬液;还可制成在注射前适合配入溶液或悬液中、液体载体的固体形式。
一旦配成本发明的组合物,可将其直接给予对象。待预防或治疗的对象可以是动物;尤其是人。
本发明的含本发明调节因子的治疗或预防性药物组合物(包括疫苗),可以经口服、皮下、皮内、静脉注射等方式应用。治疗剂量方案可以是单剂方案或多剂方案。
本发明的组合物可用于调控调节性T细胞,从而治疗或预防免疫失调引起的疾病。当需要下调患者的免疫应答时,可采用包含本发明磷酸化途径相关因子或其激动剂的组合物;当需要上调患者的免疫应答时,可采用包含磷酸化途径相关因子抑制剂的组合物。需要通过下调患者的免疫应答来治疗的疾病包括但不限于:自身免疫性疾病、器官移植。需要上调患者的免疫应答来治疗的疾病包括但不限于:肿瘤、病毒感染。所述上调可通过抑制调节性Treg细胞功能而实现。所述下调可通过增强调节性Treg细胞功能实现。
本发明的主要优点
(1)本发明中首次揭示了磷酸化途径相关因子——PIM1通过磷酸化FOXP3并影响FOXP3与染色质的结合,进而调节FOXP3对其靶基因的转录调控活性;
(2)本发明调节因子的确定与应用,为通过PIM1磷酸化FOXP3从而调节FOXP3对其靶基因的转录调控活性,从而治疗和/或预防与FOXP3、IL-2和/或IFN-γ活性失调相关的疾病或症状(例如肿瘤、自身免疫性疾病等)提供了新途径;
(3)本发明还提供了进一步提供了PIM1和/或其编码序列、其激动剂或拮抗剂在制备包含调节性T细胞的组合物或调控调节性T细胞活性的组合物中的用途;制备包含调节性T细胞的组合物方法;调控调节性T细胞活性的方法;以及用于制备包含调节性T细胞的组合物或调控调节性T细胞活性的组合物的试剂、试剂盒或药盒。

下面结合具体实施例,进一步阐述本发明。应理解,这些实施例仅用于说明本发明而不用于限制本发明的范围。本领域技术人员可对本发明做出适当的修改、变动,这些修改和变动都在本发明的范围之内。
下列实施例中未注明具体条件的实验方法,可采用本领域中的常规方法,例如参考《分子克隆实验指南》(第三版,纽约,冷泉港实验室出版社,New York:Cold Spring Harbor Laboratory Press,1989)或按照供应商所建议的条件。DNA的测序方法为本领域常规的方法,也可由商业公司提供测试。
除非另外说明,否则百分比和份数按重量计算。除非另行定义,文中所使用的所有专业与科学用语与本领域熟练人员所熟悉的意义相同。此外,任何与所记载内容相似或均等的方法及材料皆可应用于本发明方法中。文中所述的较佳实施方法与材料仅作示范之用。
实施例I.
材料与方法
1.质粒和载体:
参照参考文献4克隆构建N端FLAG-标签的FOXP3、PIM1及其它质粒(pIPFLAG2、pIPFHA2是真核表达载体;pET28a和pET21-MBP是原核表达载体)和慢病毒包装载体质粒(FUGW、dR8.9和VSV-G)。
PIM1基因从人类外周血单核细胞cDNA文库中扩增获得。根据Genbank下载的序列设计引物如下:
2.抗体:
Flag抗体(M2)购自Sigma公司。FOXP3抗体(hFOXY)购自eBioscience公司。PIM1抗体购自Santa cruz公司。pS422-FOXP3抗体从艾比玛特公司定制。HRP偶联的抗鼠二抗购自Promega公司。
3.细胞及其处理:
人HEK293T(购自中国科学院细胞库(目录号:GNHu17))在含有10%FBS、100单位/毫升青链霉素的DMEM(Dulbecco's modified Eagle's medium)中,于37°C,5%CO2条件下培养。
SZ4.03细胞(FOXP3高表达的淋巴细胞株,由本实验室保种;也可由本领域普通技术人员按照参考文献7所述方法常规制得)在含有10%FBS、100单位/毫升青链霉素、100单位/毫升非必需氨基酸和100单位/毫升丙酮酸钠的1640培养基中,于37°C,5%CO2条件下培养。
细胞转染:利用Lipofectamin2000(Invitrogen)进行脂质体转染(按操作说明书进行),转染48小时后收样分析。
用FUGW-TAP-FOXP3、dR8.9和VSV-G共同转染人胚肾细胞株HEK293,转染后48及72小时收集细胞培养基上清。
shPIM1引物序列设计如下:

将合成好的shRNA引物序列退火后连入shRNA慢病毒表达载体pLKO.1(购自Addgene公司)。按pLKO.1:dR8.9:VSVG=5:4:3的比例混合后,将混合物通过磷酸钙转染的方法转入HEK293T细胞,转染48小时后收集病毒,病毒悬液经低速离心和过滤后直接用于目的细胞的感染。
4.试剂及细胞核及染色质亚组分的分离:
PIM1抑制剂(4-[3-(4-氯苯基)-2,1-苯并异噁唑-5-基]-2-嘧啶胺)购自Tocris公司。
IL-4、IL-6因子购自Apotech公司。
MG132购自Merck公司。
蛋白质AG珠(Protein AG-beads)购自上海悦克生物技术有限公司。
细胞组分的分离:细胞用细胞质提取液(10mM Hepes,pH7.9,10mM KCl,0.1mM EDTA,1mM DTT,0.5mM PMSF,1′完全蛋白酶抑制剂混合试剂(complete protease inhibitor cocktails,Cat.No.1-697-498;Roche Biochem),1mMNa3VO4),放置冰上15分钟,然后加入终浓度0.6%的NP-40,经涡旋30秒裂解后,12000g离心30秒。所得上清为细胞质部分,沉淀即为细胞核组分。将细胞核组分用细胞核悬浮液(20mM Hepes pH7.9,400mM NaCl,1mM EDTA,1mMDTT,1mM PMSF,1′蛋白酶抑制剂混合试剂,1mM Na3VO4)溶解后,置于4°C匀速缓缓旋转30分钟,然后在16,060g,4°C离心15分钟。所得不溶沉淀物为染色质亚组分。
5.免疫沉淀:
用RIPA缓冲液(20mM Tris/HCL pH7.5,150mM NaCl,1%NP-40,0.5%Na-DOC,1mM EDTA以及蛋白酶抑制剂1mM PMSF,1×Cocktail,磷酸酶抑制剂1mM Na3VO4,1mM NaF)裂解细胞。在细胞裂解液中先加入一抗孵育1小时,再加入偶联ProteinA/G的琼脂糖珠作用1小时。用RIPA缓冲液裂解洗涤三次后,用SDS-PAGE检测所结合的蛋白质。
6.免疫印迹:
蛋白质样品通过SDS-PAGE蛋白电泳后,转膜至硝酸素纤维膜,再用5%脱脂奶粉TBST中封闭一小时,加入一抗孵育一小时,再加入HRP偶联的二抗孵育一小时,用ECL底物曝光显色。
7.基因定点突变:
通过突变PCR得到FOXP3突变体S422A、PIM1突变体K67M(参照参考文献5)。
8.蛋白质的表达及纯化:
将PIM1及PIM1K67M基因插入PET28a原核表达载体中后获得PET28a-PIM1及PET28a-PIM1K67M,随后转化BL21感受态细胞,IPTG诱导表达,收细胞,裂解,镍柱纯化(Qiagen公司),最后经快速蛋白液相色谱(GE公司)获得纯化蛋白。MBP-S422FOXP3和MBP-FOXP3蛋白按上述方法表达,收细胞,裂解,直链淀粉树脂法纯化(NEB公司),用标准缓冲液透析洗脱蛋白(25mMTris-HCl(pH7.5)、150mM氯化钠、10mMβ巯基乙醇和10%甘油)。最后用Brandford法测定所有蛋白浓度(上海碧云天生物技术有限公司)。
9.体外磷酸化实验:
将1μg6His-PIM1或6His-PIM1K67M和1μg MBP-FOXP3(共四组,分别为:6His-PIM1;MBP-FOXP3;MBP-FOXP3+6His-PIM1;MBP-FOXP3+6His-PIM1K67M)加入100μl含20mM MOPS缓冲液(pH7.4)、150mM氯化钠、12.5mM氯化镁、1mM氯化锰、1mM DTT、10μM ATP的反应液中,混合均匀,30°C孵育两小时后加入SDS上样缓冲液终止反应。对以上反应体系进行Western blotting,用抗pS422-FOXP3抗体分析各体系中的FOXP3S422位点磷酸化。
10.细胞增殖检测实验:
分别从人外周血单核细胞(购自上海市血液中心)中,通过抗体标记、磁珠分选得到调节性T细胞。如前所述,用1μM CFSE(Invitrogen公司)标记效应性T细胞后将调节性T细胞及效应性T细胞培养80小时,通过流式细胞仪检测效应性T细胞荧光强度变化,分析其增殖状况。
实施例I-1.原癌基因编码蛋白PIM1对FOXP3的影响
在HEK293T细胞中共转入HA-FOXP3和FLAG-PIM1,48h后收集细胞并经RIPA缓冲液裂解后以FLAG抗体作免疫共沉淀,然后用相应抗体作免疫印迹检测(结果如图1A所示)。该结果显示:FLAG-PIM1可与HA-FOXP3共沉淀。
在HEK293T细胞中共转入HA-FOXP3或其Ser422突变体与FLAG-PIM1或其酶活缺失突变K67M,48h收集细胞并裂解后以HA抗体作免疫共沉淀,然后用pS422-FOXP3抗体及其它相应抗体作免疫印迹分析(结果如图1B所示)。该结果显示:FLAG-PIM1可使HA-FOXP3中Ser422位点磷酸化。
原核表达并经FPLC纯化的MBP-FOXP3和His-PIM1及其K67M突变体,在ATP存在的条件下,体外反应后作免疫印迹分析。反应体系中的总蛋白经SDS-PAGE凝胶电泳,然后经LumiteinTM蛋白凝胶染色后在紫外光下显色(结果如图1C所示)。该结果显示:有且仅有MBP-FOXP3和His-PIM1的体外磷酸化实验体系可检测到FOXP3中Ser422位点磷酸化。
以上实验结果表明:原癌基因编码蛋白PIM1可以与FOXP3结合,并特异性催化FOXP3Ser422位点磷酸化。换言之,FOXP3可结合PIM1,并被其磷酸化Ser422位点。
实施例I-2.炎症细胞因子IL-6和IL-4可诱导PIM1的表达
SZ4.03细胞经IL-6(20ng/ml)或IL-4(30ng/ml)分别处理0h、1h、2h和4h后收集细胞并经SDS上样缓冲液直接煮沸裂解,然后对样品进行免疫印迹分析(一抗分别为PIM1抗体和FOXP3抗体)(结果如图2所示)。
结果显示:经IL-6或IL-4处理后,在FOXP3高表达的淋巴细胞株SZ4.03细胞中,PIM1的表达量随处理时间的延长逐步提高;而FOXP3的表达水平无明显差异。
该结果表明:IL-4和IL-6两种炎症细胞因子可刺激内源性PIM1表达。
实施例I-3.PIM1抑制FOXP3与染色质的结合
将HA-FOXP3和FLAG-PIM1共转入HEK293T细胞,48h后收集细胞并作染色质(Chr)、细胞质(Cy)及细胞核(Nu)分离,对分离后的样品进行免疫印迹分析(结果如图3所示)。
结果显示:当FOXP3和PIM1同时存在时,与仅存在其中一者时相比,与染色质结合的FOXP3的量显著减少;而细胞核与细胞质中FOXP3的量不变。
该结果表明:磷酸化修饰是FOXP3存在的翻译后修饰之一,PIM1可以抑制FOXP3与染色质的结合能力,进而抑制FOXP3靶基因的转录。
实施例I-4.PIM1负调节转录因子FOXP3的转录调节活性
用慢病毒包装系统包装病毒载体携带负对照shRNA(shCK)及PIM1shRNA(shPIM1),随后感染FOXP3高表达的SZ4.03细胞株,经嘌呤霉素(50ng/ml)筛选和DMSO或者PMA/肌霉素(P/I)处理4h后,提取总RNA并反转为cDNA,然后作定量PCR分析PIM1的mRNA表达水平(结果如图4A所示)、FOXP3正调节靶基因(GITR和CTLA4)的mRNA表达水平(结果如图4B和4C所示)、FOXP3负调节靶基因(IL-2)的mRNA表达水平(结果如图4D所示)以及非FOXP3靶基因(STUB1)的mRNA表达水平(结果如图4E所示)。
结果显示:与对照shCK相比,shPIM1能明显降低PIM1mRNA的表达水平。shPIM1增强FOXP3正调节靶基因(GITR和CTLA4)的mRNA表达水平,降低FOXP3负调节靶基因(IL-2)的mRNA表达水平,以及不影响非FOXP3靶基因(STUB1)的mRNA表达水平。
该结果表明:PIM1能负向调节FOXP3的转录调控活性;shPIM1能正向调节FOXP3的转录调控活性,从而影响FOXP3靶基因mRNA的表达水平。
实施例I-5.PIM1抑制剂能提高CLTA4+FOXP3+Treg及CD25+FOXP3+Treg细胞
水平
人CD4+CD25+CD127low天然Treg(nTreg)经BD FACS AriaⅡ流式细胞分选仪分选后,并经Bead-CD3/CD28和IL-2(500U/ml)扩增,加入DMSO或PIM1抑制剂(4-[3-(4-氯苯基)-2,1-苯并异噁唑-5-基]-2-嘧啶胺,100nM)处理24h,经APC-FOXP3及PE-CD25抗体染色后作流式细胞分析(结果如图5A所示)。扩增后的nTreg静息24h,然后加入DMSO或PIM1抑制剂(100nM)处理48h,经APC-FOXP3及PE-CD25抗体染色后作流式细胞分析(结果如图5B所示)。
结果显示:与DMSO对照组相比,PIM1抑制剂能显著提高CLTA4+FOXP3+Treg及CD25+FOXP3+Treg细胞水平。
该结果表明:PIM1抑制剂能正向调节FOXP3的表达水平及提高CLTA4+FOXP3+Treg及CD25+FOXP3+Treg细胞水平。
实施例I-6.PIM1抑制剂可增强nTreg的免疫抑制活性
将扩增后的nTreg与CFSE标记的人PBMC外周血单核细胞(2×105个/孔)按相应比例(即仅PBMC、Treg:PBMC=1:2或1:4、或者仅Treg)在96孔板U型孔内混合,每孔添加5000个Bead-CD3/CD28。两组细胞分别用DMSO及PIM1抑制剂处理,培养5天后检测细胞增殖水平(结果如图6所示)。
结果显示:与DMSO处理组相比,PIM1抑制剂处理组中的PBMC增殖情况受到更强的抑制。
该结果表明:PIM1抑制剂可增强nTreg的免疫抑制活性。
实施例I-7.PIM1抑制剂促进FOXP3+及CTLA4+nTreg细胞的扩增
取2×105个nTreg细胞,加入500U/ml的IL-2,以CD3/CD28T细胞扩增珠和细胞3:1的比例扩增nTreg细胞。两周后收集部分扩增nTreg细胞做FACS检测;剩余的细胞除去扩增珠,静息48小时后再次收集作FACS检测FOXP3及CTLA4。雷帕霉素(Rapamycin)及PIM1抑制剂的使用浓度均为100nM(结果如图11所示)。
该结果显示:经由PIM1抑制剂处理的实验组与对照组相比,PIM1抑制剂能促进FOXP3+及CTLA4+nTreg细胞的扩增。
实施例I-8.PIM1基因敲除可增强FOXP3的转录调节活性
取2×105个nTreg细胞,加入500U/ml的IL-2,以CD3/CD28T细胞扩增珠和细胞3:1的比例激活原代nTreg细胞4小时,然后分别感染编码PIM1、PIM2及PIM3shRNA的慢病毒过夜,不除去扩增珠继续培养感染的细胞1周,然后检测FOXP3及其靶基因CTLA4的mRNA水平(结果如图12所示)。
该结果显示:shPIM1基因敲除的nTreg细胞中CTLA4的mRNA水平增高,说明PIM1基因可调节FOXP3的转录活性。
实施例II.
本部分实施例的目标是筛选到能够特异性影响FOXP3+Treg细胞功能的小分子,开发影响FOXP3蛋白功能的激动剂;检测这些抑制剂对Treg细胞功能的影响;为治疗自身免疫性疾病、器官移植等提供新的治疗手段。
上海标准化研究中心提供中药小分子标准库,根据信息检索,其中有95种小分子与炎症相关。我们通过分子免疫学相关技术,我们首次发现山柰酚对FOXP3翻译后修饰的作用及对FOXP3稳定性的影响。

材料与方法
1.质粒和载体:
参照参考文献4克隆构建N端Myc-FOXP3、Flag-PIM1、Flag-PIM2、Flag-PIM3。
PIM1、PIM2、PIM3基因从人类外周血单核细胞cDNA文库中扩增获得。
根据Genbank下载的序列设计引物如下:
细胞转染:利用Lipofectamin2000(Invitrogen)进行脂质体转染(按操作说明书进行),转染48小时后收样分析。
用FUGW-TAP-FOXP3、dR8.9和VSV-G共同转染人胚肾细胞株HEK293,转染后48及72小时收集细胞培养基上清。
shPIM1引物序列设计如下:
shCK引物序列如下:
将合成好的shRNA引物序列退火后连入shRNA慢病毒表达载体pLKO.1(购自Addgene公司)。按pLKO.1:dR8.9:VSVG=5:4:3的比例混合后通过磷酸钙转染的方法转入HEK293T细胞,转染48小时后收集病毒,病毒悬液经低速离心和过滤后直接用于目的细胞的感染。
2.抗体:
Flag抗体(M2)购自Sigma公司。FOXP3抗体(hFOXY)购自eBioscience公司;磷酸化-组蛋白(Phospho-Histone)H3S10和组蛋白H3抗体购自Cell signaling公司;pS422-FOXP3抗体从艾比玛特公司定制;GAPDH抗体购自艾比玛特公司。β-微管蛋白购自天津三箭生物技术有限公司;HRP偶联的抗鼠二抗购自Promega公司。
3.细胞及其处理:
人HEK293T(购自中国科学院细胞库(目录号:GNHu17))在含有10%FBS、100单位/毫升青链霉素的DMEM(Dulbecco's modified Eagle's medium)中,于37°C,5%CO2条件下培养。
Jurkat E6.1 T细胞(购自中国科学院细胞库(目录号:TCHU123))在含有10%FBS,100单位/毫升青链霉素,100单位/毫升非必需氨基酸和100单位/毫升丙酮酸钠的1640培养基中,于37°C,5%CO2中培养。
细胞转染:利用Lipofectamin 2000(Invitrogen)进行脂质体转染,转染48小时后收样分析。
用FUGW-HA-FOXP3、del8.9和VSV-G共同转染人胚肾细胞株HEK293,转染后48及72小时收集细胞培养基上清,超速离心纯化病毒并感染Jurkat E6.1最终获得HA-FOXP3 Jurkat稳定表达细胞系。
4.试剂及细胞组分的分离:
PIM1抑制剂(4-[3-(4-氯苯基)-2,1-苯并异噁唑-5-基]-2-嘧啶胺)购自Tocris公司;PI3K-抑制剂(LY294002)购自碧云天生物技术研究所;PMA购自碧云天公司;LPS购自invivogen公司;山柰酚购自上海标准化研究中心。
细胞用细胞质提取液(10mM Hepes,pH7.9,10mM KCl,0.1mM EDTA,1mM DTT,0.5mM PMSF,1'完全蛋白酶抑制剂混合试剂(complete proteaseinhibitor cocktails,Cat.No.1-697-498;Roche Biochem),1mM Na3VO4),放置冰上30分钟裂解,12000g离心30秒。
5.免疫印迹:
蛋白质样品通过SDS-PAGE蛋白电泳后,转膜至硝酸素纤维膜,再用5%脱脂奶粉或3%牛血清白蛋白的TBST中封闭一小时,加入一抗孵育一小时,再加入HRP偶联的二抗孵育一小时,用ECL底物曝光显色。
6.原代细胞分选及细胞增殖检测实验:
分别从人外周血单核细胞(购自上海市血液中心)中,将PBMC同时染上CD4、CD25及CD127抗体,然后在FACSAriaII细胞分选仪上分选出CD4+CD25hiCD127low的细胞群。分选出的nTreg细胞在体外使用含有1%GlutaMax(GIBCO)、1%NaPyr(GIBCO)和500U IL2的X-VIVO(Lonza)培养基中培养。
扩增过程中需要CD3/CD28 T细胞扩增珠的存在(Dynabeads;Invitrogen;目录号:111-41D),扩增珠与细胞的比例为3:1。扩增细胞的nTreg表型通过检测FOXP3的表达水平加以鉴定。如前所述,用1μM CFSE(Invitrogen公司)标记效应性T细胞后将调节性T细胞及效应性T细胞培养80小时,通过流式细胞仪检测效应性T细胞荧光强度变化,分析其增殖状况。
实施例II-1.山柰酚剂量依赖抑制FOXP3s422位点的磷酸化
在HEK293T细胞中共转入MYC-FOXP3和FLAG-PIM1,36小时后,分别用50nM、10nM、2nM、0.4nM的PIM1抑制剂和50μM、10μM、2μM、0.4μM、0.08μM的山柰酚处理12小时。48h后收集细胞并经细胞裂解液裂解。用免疫印迹法直接检测ps422-FOXP3、FOXP3、β-微管蛋白水平(结果如图9A所示);用软件ImageJ软件分析图1A(结果如图9B所示)。
该结果显示:山柰酚剂量依赖抑制FOXP3 s422位点的磷酸化。
实施例II-2.山柰酚可特异性抑制PIM1引起的FOXP3s422位点磷酸化
在HEK293T细胞中分别共转入Myc-FOXP3+FLAG-PIM1、Myc-FOXP3+FLAG-PIM2、Myc-FOXP3+FLAG-PIM3。36小时后,用50μM的山柰酚处理12小时。48小时后收集细胞并经细胞裂解液裂解。用免疫印迹法直接检测ps422-FOXP3、FOXP3和β-微管蛋白水平(结果如图10A所示);用软件ImageJ软件分析图10A(结果如图10B所示)。
该结果显示:山柰酚不影响PIM2和PIM3引起的FOXP3磷酸化水平,但特异性地抑制PIM1引起的FOXP3s422位点磷酸化。
实施例II-3.山柰酚可抑制PIM1引起的组蛋白H3s10位点磷酸化
在HEK293T细胞中共转入MYC-FOXP3和FLAG-PIM1,36小时后用50μM的山柰酚处理12小时。细胞转染48h后收集细胞,用SDS-加样缓冲液(loadingbuffer)直接超声破碎,煮沸裂解。用免疫印迹法直接检测磷酸化-组蛋白(Phospho-Histone)H3S10、组蛋白H3和PIM1的水平(结果如图11所示)。
该结果显示:PIM1可被原癌基因MYC招募到其靶基因上并使组蛋白H3第十位丝氨酸Ser10发生磷酸化,从而启动靶基因的转录,故磷酸化-组蛋白H3S10是PIM1的底物之一。该实验结果显示山柰酚不影响PIM1的表达水平,但抑制PIM1引起的组蛋白H3 s10位点磷酸化。
实施例II-4.山柰酚仅抑制PIM1引起的FOXP3的S422位点磷酸化和组蛋白H3
S10位点的磷酸化
在HEK293T细胞中转入FLAG-PIM1、共转MYC-FOXP3和FLAG-PIM1,36小时后,设HEK293空白对照组,每组分别用50μM的山柰酚处理12小时。48h后收集细胞并经细胞裂解液裂解。用免疫印迹法直接检测ps422-FOXP3、FOXP3、磷酸化-组蛋白H3(Ser10)、组蛋白H3和GAPDH水平(结果如图12所示)。
结果显示:山柰酚处理能抑制FOXP3蛋白s422位点磷酸化及组蛋白H3 s10位点磷酸化;而空白对照组中组蛋白H3 s10位点磷酸化无任何影响。
该结果表明:山柰酚特异性抑制PIM1引起的组蛋白H3 s10位点磷酸化水平。
实施例II-5.山柰酚可抑制PMA/LPS共刺激引起的FOXP3蛋白降解
HA-FOXP3-Jurkat稳定表达细胞用PMA(25ng/ml)和LPS(1ng/ml)共处理,每份样品再分别加入50μM山柰酚、100nM PIM1抑制剂(即(4-[3-(4-氯苯基)-2,1-苯并异噁唑-5-基]-2-嘧啶胺))及10μg/ml PI3K抑制剂(LY294002),DMSO作为对照。24小时后收集细胞。用免疫印迹法直接检测FOXP3及GAPDH的水平(结果如图13所示)。
该结果显示:PMA和LPS共刺激能引起HA-FOXP3-Jurkat稳定表达细胞中FOXP3蛋白降解。山柰酚与PIM1抑制剂及PI3K抑制剂能抑制FOXP3蛋白降解。
实施例II-6.山柰酚增强诱导的Treg中FOXP3和CTLA4的表达
人CD4+CD45RA+天然T细胞(即天然T细胞而非Treg细胞)经BD FACS AriaⅡ流式细胞分选仪分选,并经Bead-CD3/CD28(2:1),IL-2(100U/ml)和TGF-β(1ng/ml、3ng/ml、5ng/ml)扩增5天后得到诱导的Treg细胞,加入DMSO或山奈酚(50uM)处理48h经PE-FOXP3及APC-CTLA4抗体染色后作流式细胞分析(结果如图14a和14b所示)。
结果显示:与DMSO对照组相比,山奈酚能显著提高FOXP3的表达水平和CLTA4+FOXP3+Treg细胞水平。
该结果表明:山奈酚能正向调节FOXP3的表达水平及提高CLTA4+FOXP3+Treg细胞水平。

Claims (17)
1.磷酸化途径相关因子、其激动剂或拮抗剂在制备包含调节性T细胞的组合物或者调控调节性T细胞活性的组合物中的用途,其中所述磷酸化途径相关因子选自:原癌基因表达蛋白PIM1和/或其编码序列。
2.如权利要求1所述的用途,其特征在于,其中所述制备是通过用所述磷酸化途径相关因子、其激动剂或拮抗剂调节选自下组的调节性T细胞调节因子的活性进行的:FOXP3、IL-2、GITR、CTLA4或它们的组合。
3.如权利要求2所述的用途,其特征在于,对FOXP3、IL-2、GITR、CTLA4或它们的组合的活性调节是正调节或负调节,其中,所述磷酸化途径相关因子或其激动剂负调节FOXP3、GITR或CTLA4活性和正调节IL-2活性,所述磷酸化途径相关因子的拮抗剂正调节FOXP3、GITR或CTLA4活性和负调节IL-2活性。
4.如权利要求1所述的用途,其特征在于,所述激动剂选自:诱导PIM1表达的信号因子和/或其编码序列;所述拮抗剂选自:抗PIM1抗体、shPIM1、针对PIM1的反义寡核苷酸、PIM1的化学抑制剂,优选4-[3-(4-氯苯基)-2,1-苯并异噁唑-5-基]-2-嘧啶胺、3-氰基-4-苯基-6-(3-溴-6-羟基)苯基-2(1H)-吡啶酮、2-羟基-3-氰基-4-苯基-6-(3-溴-6-羟基苯基)吡啶、吡啶并咔唑-环戊二烯钌聚合物的消旋混合物。
5.如权利要求4所述的用途,其特征在于,所述激动剂选自炎性细胞因子,更优选IL-4、IL-6、TCR或其编码序列。
6.如权利要求1所述的用途,其特征在于,所述拮抗剂是山奈酚或其药学上可接受的盐。
7.如权利要求1所述的用途,其特征在于,制备所述调节性T细胞的起始物中包含CD4阳性的T细胞,优选CD4+CD45RA+天然T细胞。
8.如权利要求1-7中任一项所述的用途,其特征在于,所述组合物用于治疗和/或预防与调节性T细胞的免疫调节功能失调相关的疾病或症状。
9.如权利要求8所述的用途,其特征在于,所述与调节性T细胞的免疫调节功能失调相关的疾病或症状是与调节性T细胞免疫抑制功能过高或过低相关的疾病或症状。
10.如权利要求1所述的用途,其特征在于,所述组合物是药物组合物、保健品组合物、疫苗组合物或佐剂组合物。
11.一种制备包含调节性T细胞的组合物方法,所述方法包括:
(a)提供包含CD4阳性T细胞的初始物;
(b)使所述初始物接触磷酸化途径相关因子、其激动剂或拮抗剂以制得调节性T细胞或包含调节性T细胞的组合物,其中所述磷酸化途径相关因子选自:原癌基因表达蛋白PIM1和/或其编码序列。
12.如权利要求11所述的方法,其特征在于,所述方法还任选包括选自下组的一个或多个步骤:
(a')在步骤(b)之前,对所述初始物进行纯化或分离,优选纯化或分离CD4阳性T细胞;
(c)在步骤(b)之后,对所述调节性T细胞或包含调节性T细胞的组合物中的细胞进行分离或纯化。
13.一种调控调节性T细胞活性的方法,所述方法包括:
使磷酸化途径相关因子、其激动剂或拮抗剂、或者用权利要求11或12所述的方法制备的组合物接触包含CD4阳性T细胞的样品或对象。
14.一种用于制备包含调节性T细胞的组合物或调控调节性T细胞活性的组合物的试剂、试剂盒或药盒,其包含:
(a)一种或多种选自下组的磷酸化途径相关因子:原癌基因表达蛋白PIM1和/或其编码序列;和/或所述磷酸途径相关因子的激动剂或拮抗剂;
(b)药学上、免疫学上或保健品学上可接受的载体。
15.如权利要求14所述的试剂、试剂盒或药盒,其中,所述调控是通过调节选自下组的调节性T细胞调节因子的活性进行的:FOXP3、IL-2、GITR、CTLA4或它们的组合。
16.如权利要求14所述的用途,其特征在于,所述拮抗剂是山奈酚或其药学上可接受的盐。
17.一种筛选调控调节性T细胞活性的物质的方法,所述方法包括检测FOXP3蛋白的稳定性或翻译后修饰酶类(如PIM1)的活性的步骤,即通过检测PIM1诱导的FOXP3的磷酸化或FOXP3蛋白的稳定性筛选调控调节性T细胞活性的物质。
