本发明的癌症疫苗的特征在于,其含有由序列号1或序列号2组成的肽之一或这两种肽、表达序列号1或2的肽的表达载体、在细胞表面上呈递由序列号1或2的序列组成的肽的抗原呈递细胞、或上述T细胞中的至少一种。
此外,本发明的含有由序列号1或序列号2组成的肽之一或这两种肽的癌症疫苗也可含有这些肽以外的癌症抗原肽。
进而,本发明的癌症疫苗优选针对表达snail蛋白质的癌症细胞的癌症疫苗。
本发明的治疗和预防癌症的方法的特征在于,对人和人以外的脊椎动物使用本发明的癌症疫苗。
在此,本说明书中的术语“癌症”意指源自上皮细胞的癌、非源自上皮细胞的肿瘤、血液癌症等新生物(neoplasm),而与癌症的来源无关。
此外,在本说明书中,登记号为NM-005985(序列号3)的基因称为人snail基因,当述及无动物种类限定的snail基因时,不只限于人基因,也包括其他动物种类的同源和直系同源。另外,登记号为NP-005976(序列号4)的蛋白质称为人snail蛋白质,当述及无动物种类限定的snail蛋白质时,不只限于人蛋白质,也包括其他动物种类的同源和直系同源。

以下一边列举实施例一边详细说明基于上述见解而完成的本发明的实施方式。但是,本发明不限于下述实施例。

癌症疫苗
将由作为snail蛋白质具有的氨基酸序列的一部分的序列号1或2组成的肽添加至作为抗原呈递细胞的树突细胞时,通过结合HLA-I分子而呈递在细胞表面上,被细胞毒性T细胞识别,由此可诱导识别snail的细胞毒性T细胞。此外,通过各种肽的刺激而建立的细胞毒性T细胞可有效地识别表达snail蛋白质的癌症细胞,由此,序列号1和2二者或其中任意一种肽、在细胞表面上呈递所述肽的抗原呈递细胞、和由抗原呈递细胞诱导的识别表达snail抗原的癌症细胞的细胞毒性T细胞,可作为癌症疫苗用于癌症的治疗和预防。
癌症疫苗的给药方法
目前,开发了将肿瘤特异性癌症抗原、癌症抗原呈递细胞或癌症抗原反应性细胞毒性T细胞作为癌症疫苗给药至癌症患者的方法。成为使用snail蛋白质的部分肽的癌症疫苗的治疗和预防对象的癌症只要是表达snail蛋白质的癌症即可而没有特别限定,神经瘤、肾癌、肝癌、胰脏癌、肉瘤、结肠癌、黑色素瘤、肺癌、食道癌、子宫癌、睾丸癌、卵巢癌、白血病、淋巴瘤、骨髓瘤等实体癌和血液癌均可,但是优选胰脏癌、黑色素瘤、白血病、结肠癌。
通过本发明的癌症疫苗治疗和预防癌症的对象只要是罹患这种癌症的脊椎动物即可而没有限制,人和人以外均可。
另外,下文说明的癌症疫苗可分别单独给药、共同给药或与此处记载以外的癌症疫苗共同给药。
[含有肽的癌症疫苗]
本发明的癌症疫苗可含有序列号1和2的肽的二者或其中任意一种。此时,优选预先检查患者的HLA-I的类型。在此,当患者的HLA-I类型为A24时,优选给药序列号1和2的肽的二者或其中任意一种。另一方面,当HLA-I的类型为A02时,优选给药序列号1的肽。除序列号1和序列号2的肽以外,所述癌症疫苗也可含有作为治疗对象的癌症细胞表达的其他种类的癌症抗原肽。此外,给药时,肽也可与提高免疫诱导能力的辅药等共同给药。此外,给药的肽也可进行使之在体内变得难以分解之类的修饰。进而,也可使用非肽类而是嵌入了编码这些肽的DNA的表达载体等作为DNA疫苗给药。
关于给药部位,可考虑皮内给药、皮下给药、静脉内给药、腹腔内给药等而没有特别限制。
在此,序列号1和2的肽的获得方法没有特别限制,从表达肽的细胞中分离和纯化的肽、使用基因重组技术制造的重组肽、或用公知的方法化学合成的肽均可。
[含有抗原呈递细胞的癌症疫苗]
呈递由序列号1或2组成的肽的抗原呈递细胞也可作为癌症疫苗使用。在此,在细胞表面上呈递的肽可未经改性、或经糖或磷酸等改性。作为抗原呈递细胞,可示例树突细胞、巨噬细胞、B细胞、通过转基因等而强制表达B7或4-1BBL等T细胞刺激因子等的肿瘤细胞(假抗原呈递细胞)等,但如果考虑抗原呈递能力的高低等,则优选树突细胞。以下记载了树突细胞的分离方法的实例,但其他细胞也可通过公知的方法容易地获得。
首先,分离来自脊椎动物个体的外周血的单核细胞(以下也称为PBMC),检查HLA-I的类型以确认是A24还是A02。所述PBMC优选从作为治疗对象的个体本身分离,但也可从其他个体分离。此外,所述PBMC优选为CD14阳性或CD11c阳性。PBMC的分离方法无特别限制,本领域技术人员可根据要分离的PBMC的种类适当地决定。例如,可通过聚蔗糖离心分离法等分离全部PBMC(PBMC级分),也可通过抗体结合磁珠分离法等分离CD14阳性PBMC或CD11c阳性PBMC。通过对分离的PBMC用添加GM-CSF和IL-4的培养基培养5-7天,可分化诱导为树突细胞前体细胞。如此分化诱导的树突细胞前体细胞的HLA-I类型为A24时,添加序列号1或2的肽。另一方面,当HLA-I类型为A02时,添加序列号1的肽。这样得到的树突细胞为呈递序列号1或2的肽的抗原呈递细胞,将其给药至罹患癌症的个体。
给药部位可考虑皮内给药、皮下给药、静脉内给药、淋巴结给药、腹腔内给药等而无特别限制。但是,如果考虑包括生理性树突细胞的抗原呈递的生理性抗癌免疫反应在癌症组织内和树突细胞给药部位的所属淋巴结附近发生,则优选向癌症组织内或淋巴结内直接给药。
[含有T细胞的癌症疫苗]
此外,通过刺激呈递由序列号1或2组成的肽的抗原呈递细胞而建立的T细胞也可作为癌症疫苗使用。所述T细胞通过在血清的存在下与呈递由序列号1或2组成的肽的抗原呈递细胞一起共同培养纯洁T细胞,分化为CD8阳性细胞毒性T细胞(CTL)或CD4阳性辅助性T细胞等而得到。还可对罹患癌症的个体给药以此方式建立的T细胞。
另外,纯洁T细胞的来源没有特别限制,例如,可来自脊椎动物的外周血。所用的纯洁T细胞可为从PBMC级分中分离的CD8阳性细胞或CD4阳性细胞,但是如果考虑CTL的诱导效率,则优选为不从PBMC级分中分离、而是与其他种类的细胞或成分混合在一起的状态下的CD8阳性细胞或CD4阳性细胞。例如,当PBMC级分的细胞用添加了由序列号1或2组成的肽和血清的培养基进行培养时,PBMC分化为树突细胞前体细胞,树突细胞前体细胞进一步与肽结合从而分化为树突细胞,成为呈递所述肽的抗原呈递细胞。所述抗原呈递细胞刺激含有PBMC的CD8阳性T细胞,分化诱导为CTL。这样,可得到识别添加的肽的CTL。在这种情况下,培养时间可在得到CTL的范围内由本领域技术人员合适地设定,但在37℃下优选4-10天,更优选6天。
由此得到的CTL可以在分离后以其本身作为癌症疫苗使用,但也可在IL-2等白介素、抗原呈递细胞和由序列1或2组成的肽的存在下进一步培养后作为癌症疫苗使用。通过所述操作,可提高CTL的细胞毒性。
给药部位可示例皮内给药、皮下给药、静脉内给药、肿瘤内给药等而无特别限制,但对于细胞毒性T细胞,为了可直接进攻表达抗原的细胞,优选肿瘤内给药。
实施例
[实施例1]snail基因的表达
本实施例表明,snail基因在正常组织中几乎不表达,但在癌症细胞株和癌症组织中具有高表达。
通过RT-PCR的基因表达分析方法
使用RNeasykit(Qiagen公司),从健康人的正常组织、对结肠癌患者的癌症组织进行肉眼判断而获得的结肠正常部位和各种人肿瘤细胞株(胰脏癌、黑色素瘤、白血病、结肠癌)(参见图1、2)中提取出RNA,用AMV反转录得到cDNA。然后,使用下述引物,用iCycler(Biorad公司)扩增基因,通过电泳检测基因表达。另外,使用GAPDH基因的表达作为内标。
Snail用引物:
Forward 5'-CAGATGAGGACAGTGGGAAAGG-3'(序列号5)
Reverse 5'-ACTCTTGGTGCTTGTGGAGCAG-3'(序列号6)
GAPDH用引物:
Forward 5'-GTCAACGGATTTGGTCGTATT-3'(序列号7)
Reverse 5'-ATCACTGCCACCCAGAAGACT-3'(序列号8)
如图1A所示,健康人的正常组织中,在胎盘(Placenta)、睾丸(Testis)、黑色素细胞(Melanocyte)和结肠(colon)中检测出弱的snail基因表达,而在脑(Brain)、心脏(Heart)、肾脏(Kidney)、脾脏(Spleen)、肝脏(Liver)、胰脏(Pancreas)、胸腺(Thymus)、肌肉(Muscle)、骨髓(Bone marrow)、外周血单核细胞(PBMC)中未能检测出表达。
此外,从进展程度不同的结肠癌患者中采集的结肠癌的癌症组织(Tu)和同一患者的结肠的正常组织(目视判断)(N)中的snail基因表达示于图1B。在基于世界范围内通用的“美国癌症联合委员会(AJCC)”所规定的癌症患病期诊断基准进行分类的I期、IIB期、IIIB期全部进展程度的结肠癌组织中,与正常组织相比,检测出高的snail基因表达。
进而,如图2所示,胰脏癌、黑色素瘤、白血病和结肠癌的各细胞株分别与正常的胰脏组织、黑色素细胞、外周血单核细胞和结肠组织相比,snail基因表达低。
如此,snail基因可癌症组织特异性地进行表达。因此,可将snail基因的表达用于癌症的诊断,与此同时,将snail作为癌症抗原靶标的治疗方法对于各种脏器的副作用小,可适用于广泛癌症种类的患者。
[实施例2]使用HLA-A24阳性健康人PBMC的CTL诱导
本实施例表明,通过使用snail 1和snail 3肽,由健康人的HLA-A24阳性PBMC可诱导识别各种肽而活化的CTL。
首先,从HLA-A24阳性健康人(A、B)中如以下所述分离PBMC。首先,在采集的外周血中,加入1/10量的4%柠檬酸钠,在Ficoll-Paque(Amersham公司)上分层并离心(1500rpm,20分钟,室温)。分离出作为PBMC级分的包含PBMC的中间层。将2.5×107个PBMC漂浮在含有20ml 10%的胎牛血清(FCS)的RPMI1640培养基(Invitrogen公司制造)中,加入10μg/ml下述snail 1-3的各肽,在37℃、5%CO2环境下刺激培养6天。通过所述刺激培养,分化对各抗原肽反应的CTL。通过Myltenyi公司的抗体结合MACS磁珠方法,对所述CTL进行分离。
另外,使用HERV-Henv肽(序列号10)和NY-ESO-1肽(序列号11)作为阳性对照。HERV-Henv肽和NY-ESO-1肽在HLA-A24阳性抗原呈递细胞上呈递而由CD8阳性T细胞诱导CTL、以及所述CTL识别HERV-Henv肽、NY-ESO-1肽而产生γ-干扰素是已知的。

HERV-Henv:SYLHHTINL(序列号10)
NY-ESO-1:LLMWITQCF(序列号11)
通过刺激培养1×106个得到的CTL,在IL-2(100U/ml,Peprotech公司)和用于分化诱导CTL的相同的肽(0.1、1或10μg/ml)的存在下,在含有10%FCS的RPMI1640培养基中与抗原呈递细胞共同培养,由此用肽刺激CTL。抗原呈递细胞通过以下方法制备:将从用于分化诱导CTL的PBMC相同的健康人中得到的PBMC(完整细胞)在添加了10μg/ml丝裂霉素C的含有10ml的10%FCS的RPMI1640培养基中漂浮培养2小时(37℃,5%CO2)而钝化,然后用RPMI1640培养基洗涤。当呈递各肽的抗原呈递细胞以HLA-A24限制性的方式刺激CTL时,CTL产生γ-干扰素。使用人Cytometric Bead Array试剂盒(BDBiosciences公司)测定包含在培养上清液中的γ-干扰素值,检查CTL是否可识别在抗原呈递细胞上呈递的肽。
图3示出添加0.1-10μg/ml的各浓度的肽来共同培养CTL和抗原呈递细胞时,由健康人A和B的CTL所产生的γ-干扰素的产生量。另外,健康人A的图表中最左边的点为不添加任何肽的无添加组,健康人B的图表中的最左边的点为添加10μg/ml的阴性对照肽(KSPWFTTL,小鼠反转录病毒抗原p15e肽,序列号12)的阴性对照组。在无添加组和阴性对照组中,仅产生100-500pg/ml的γ-干扰素。另一方面,在用snail 1肽、snail 3肽、作为阳性对照的HERV-Henv肽和NY-ESO-1肽刺激的组中,γ-干扰素的产生量随着肽浓度的上升而显著增加(P<0.01,t检验)
如图3所示,γ-干扰素的产生量在肽浓度为10μg/ml时显示出最高值。明显的是,健康人A或B中γ-干扰素的产生量在添加10μg/ml的snail 1肽或snail 3肽的组中,与无添加组或阴性对照组相比显著地高(P<0.01,t检验),与作为阳性对照的HERV-Henv肽和NY-ESO-1肽相比程度相当或更高。
由此,使用健康人的HLA-A24阳性PMBC,可得到在细胞表面上呈递snail 1肽(序列号1)或snail 3肽(序列号2)的抗原呈递细胞。此外,用所述抗原呈递细胞刺激CD8阳性T细胞,可分化诱导能识别在HLA-A24阳性抗原呈递细胞上呈递的各肽的CTL。
[实施例3]识别snail 3的CTL的肿瘤细胞毒活性
本实施例表明,通过使用snail 3肽刺激培养PMBC而得到的CTL以HLA依赖性的方式对snail抗原阳性的肿瘤细胞具有毒活性。
将从HLA-A24阳性健康人的血液中采集的PBMC用snail 3肽刺激培养而制备的CTL(参见实施例2,识别snail 3的CTL)和作为靶标细胞的人结肠癌细胞株COLO320(HLA-A24阳性,snail阳性,NY-ESO-1阳性)以CTL:COLO320=6.25:1、12.5:1、25:1或50:1的比例混合,在含有10%FCS的RPMI1640培养液中在37℃、5%CO2的条件下培养6小时。使用Immunocyto Cytotoxity Detection Kit(MBL公司)检测杀伤的肿瘤细胞,按照所附说明书计算出肿瘤特异性毒性率。另外,在本实施例中,作为阳性对照,使用NY-ESO-1肽代替snail 3肽来进行PMBC的刺激培养。
如图4所示,由识别snail 3的CTL引起的肿瘤特异性毒性率与识别作为阳性对照的NY-ESO-1的CTL的程度相当。此外,所述肿瘤特异性毒性率随着CTL混合比的增加而增加。由此,通过使用snail 3肽刺激培养PMBC而得到的CTL表现出对表达snail的肿瘤细胞的毒活性(参见图4A、B,白圈的图示)。
此外,为了说明由识别snail 3的CTL引起的肿瘤细胞上的snail 1抗原的识别是HLA依赖性的,在抗HLA抗体(HLA中和抗体,BioLegend公司,最终浓度10μg/ml)的存在下对识别snail 3的CTL和COLO320细胞进行共同培养。
如图4所示,与不添加抗HLA抗体的情况(参见图4A、B,白圈的图示)相比,在添加抗HLA抗体的组中,由识别snail 3的CTL引起的肿瘤特异性毒性率显著降低(参见图4A、B,黑圈的图示)。其结果表明,由识别snail 3的CTL引起的肿瘤特异性毒性以HLA依赖性的方式识别肿瘤细胞上的snail抗原。
这些结果表明,通过使用snail 3肽刺激正常人的HLA-A24阳性的PMBC,可诱导对snail抗原阳性的靶标细胞具有细胞毒性的CTL,另外,由所述CTL引起的靶标细胞的snail抗原识别是HLA依赖性的。
[实施例4]使用HLA-A02阳性健康人PBMC的CTL的分化诱导
本实施例表明,使用snail1肽分化诱导的CTL可以以HLA-A02依赖性的方式识别呈递snail 1肽的细胞。
首先,按照实施例2记载的方法从HLA-A02阳性健康人(D、E)中采血并分离PBMC,使用snail 1肽进行一次刺激培养。本实施例使用HERV-Henv肽作为阳性对照,使用NY-ESO-1肽作为阴性对照。另外,已悉知,HERV-Henv肽通过在HLA-A02阳性抗原呈递细胞上呈递而由CD8阳性T细胞诱导CTL,所述CTL识别HERV-Henv肽而产生γ-干扰素。此外,还悉知,NY-ESO-1肽不与HLA-A02阳性抗原呈递细胞结合,不发生CTL的诱导。由此得到的CTL,在与实施例2相同的方式制备的HLA-A02阳性抗原呈递细胞和IL-2的存在下,在包含各肽(10μg/ml)的含有10%FCS的RPMI1640培养基中刺激24小时。当呈递肽的抗原呈递细胞以HLA-A02限制性的方式刺激CTL时,CTL产生γ-干扰素。用人Cytometric Bead Array试剂盒(BD Biosciences公司)测定培养上清液中含有的γ-干扰素值,检查CTL是否可识别呈递肽的抗原呈递细胞。
图5示出由健康人D、E的PBMC制备的CTL所产生的γ-干扰素的产生量。在健康人D和E中,用snail 1肽和HERV-Henv肽(阳性对照)刺激的组,与用作为阴性对照的NY-ESO-1肽刺激的组相比,观察到显著更高的γ-干扰素的产生(P<0.01,t检验)。另一方面,用snail2肽或snail 3肽刺激的组,与阴性对照相比,γ-干扰素的产生量降低。
由此,snail 1肽(序列号1)在来自健康人的HLA-A02阳性PBMC的抗原呈递细胞上呈递,由CD8阳性T细胞诱导识别肽的CTL。进而,所述CTL可识别HLA-A02阳性呈递细胞上的snail 1肽。
工业适用性
通过本发明,可提供可有效诱导癌症免疫的肽、在细胞表面上呈递所述肽的抗原呈递细胞、由所述抗原呈递细胞诱导的T细胞,以及含有这些肽、表达这些肽的表达载体、呈递这些肽的抗原呈递细胞或由所述抗原呈递细胞诱导的T细胞的癌症疫苗,还可提供使用所述癌症疫苗治疗和预防癌症的方法。
Claims (9)
1.肽,其由序列号1(KMHIRSHTL)或序列号2(RTFSRMSLL)的序列组成。
2.抗原呈递细胞,其在细胞表面上呈递由序列号1或2的序列组成的肽。
3.T细胞,其由权利要求2的抗原呈递细胞诱导、并识别表达snail抗原的癌症细胞。
4.权利要求3的T细胞,其特征在于,其为细胞毒性T细胞。
5.权利要求3或4的T细胞,其特征在于,所述癌症细胞为胰脏癌细胞、黑色素瘤细胞、白血病细胞或结肠癌细胞。
6.癌症疫苗,其含有选自序列号1、2中的一种以上的肽、表达所述肽的表达载体、权利要求2的抗原呈递细胞或权利要求3-5之一的T细胞。
7.权利要求6的癌症疫苗,其特征在于,其为针对表达Snail蛋白质的癌症细胞的癌症疫苗。
8.权利要求6或7的癌症疫苗,其特征在于,所述癌症为胰脏癌、黑色素瘤、白血病或结肠癌。
9.治疗或预防癌症的方法,其对人以外的脊椎动物使用权利要求8的癌症疫苗。
