Abstract

本发明提供了一种检测前列腺液游离血红素的方法,包括获取前列腺液渗液;用PH为7.4的三羟甲基氨基甲烷-盐酸缓冲液处理渗液;后再与显色试剂反应再与比色板比对记录检测结果;本发明简单实用,成本低廉,可快速的筛查出宫颈渗液中不含血红素的样本、含血红素的样本和可能含有血红素的样本,为进一步筛查做准备。同时本发明还涉及一种检测前列腺液渗液的检测盒。

一种检测前列腺液游离血红素的方法
技术领域
本发明涉及一种检测方法,具体涉及一种检测前列腺液游离血红素的方法,同时本发明还涉及一种检测前列腺液游离血红素的专用检测盒。 
背景技术
前列腺液中的游离血红素往往是由组织肿瘤细胞的细胞色素、氧感受器和FixL跨膜蛋白的游离血红素释放、析出或肿瘤组织微循环渗出引起的,其游离血红素的含量极少,肉眼不可得见,一般需借助先进的仪器和分析化学方法进行检测,费用高,周期长,所以发明一种操作简单,成本低廉的检测方法快速的确定前列腺液中是否含有游离血红素,成为一个亟需解决的问题。 
发明内容
 为了解决现有技术中检测前列腺液中游离血红素的方法费用高、周期长的问题,本发明提供了一种检测前列腺液游离血红素的方法,本发明提供的方法其灵敏度高,成本低廉,可快速的检测前列腺液中的游离血红素;同时本发明还提供了一种检测前列腺液中游离血红素的带有专用取样套的检测盒。 
本发明的技术方案如下: 
    本发明提供的检测前列腺液中游离血红素的方法,包括以下检测步骤:
     1) 使受检者取膝胸位,打开检测盒的外包装盒,从盒体内取出前列腺液取样套,套在受检者阴茎龟头部位超过冠状沟,检查者左手扶持患者左肩或臀部,右手戴手套或指套,指端涂凡士林或液体石蜡;食指先在肛门口轻轻按摩,然后将手指徐徐插入肛门,当指端进入距肛门门约5cm直肠前壁处即可触及前列腺,以手指末节作向内、向前徐徐按摩,每侧约4-5次,然后再将手移至腺体的上部顺正中沟向下挤压,这样前列腺液即可由尿道排出至前列腺液取样套上,留取标本送检;其中,所取前列腺液量为0.5-1 ml;
    2) 把带有前列腺液的前列腺液取样套放入样品杯内,样品杯内预装有三羟甲基氨基甲烷-盐酸缓冲液,缓冲液的量为3ml,混匀,得样品液;优选的,三羟甲基氨基甲烷-盐酸缓冲液的PH值为7.4 ;
3) 在测试池内顺序加入样品液1ml,试剂A 0.5ml,试剂B 0.5ml;混匀,2分钟内观察颜色变化,与比色板比对记录检测结果;
    所述试剂A的配制方法如下:取3,3',5,5'-四甲基联苯胺 50mg溶于PH为7.4柠檬酸缓冲液100ml混匀而成;所述试剂B为3%的过氧化氢溶液。
本发明同时提供了一种检测前列腺液游离血红素的检测盒。     
一种检测前列腺游离血红素的检测盒,包括盒体和盒体外的外包装盒,所述外包装盒的外表面设有比色板,所述盒体设有向内凹陷而成的镊子存放区、样品杯存放区、滴管存放区和测试池,所述测试池内存放有前列腺液取样套,所述前列腺液取样套呈底端封闭的圆筒状,所述底端还设有一凸起的小囊。
进一步的,所述前列腺取样套材质为吸附性无纺布。 
进一步的,所述前列腺液取样套的直径3.5cm,长度5cm,所述小囊直径1cm,深度0.5cm。 
进一步的,所述测试池设于测试盒中上方,直径1.8cm,深度1.2cm。 
进一步的,所述滴管存放区设于测试池两侧,所述样品杯存放区设于测试池左侧,所述镊子放置区设于样品杯存放区左侧。 
进一步的,所述滴管存放区内设有滴管,滴管上带有刻度。 
进一步的,所述样品杯放置区内设有样品杯,所述样品杯设有密封盖,样品杯的杯体呈圆筒状。 
进一步的,所述镊子存放区内设有镊子,所述镊子为塑料镊。 
本发明提供的检测方法,简单实用,成本低廉,可快速的筛查出前列腺液中不含游离血红素的样本、含游离血红素的样本和可能含有游离血红素的样本,为进一步检查做准备;本发明可检测出的最低游离血红素浓度可达0.5ng/L,,灵敏度高;同时本发明还提供了一种前列腺液取样套,可专用于采集前列腺液;同时本发明还提供了一种检测盒,可专用来盛放检测过程中用到的器皿和试剂,本试剂盒结构简单,只需一个试剂盒便能完成整个检测步骤,节省空间和试剂,同时也减轻操作者劳动负担。

具体实施方式
下面结合实施例对本发明作进一步的说明。 
实施例1
  使受检者取膝胸位,打开检测盒的外包装盒1,从盒体3内取出前列腺液取样套8,套在受检者阴茎龟头部位超过冠状沟。检查者左手扶持患者左肩或臀部,右手戴手套或指套,指端涂凡士林或液体石蜡,食指先在肛门口轻轻按摩,然后将手指徐徐插入肛门,当指端进入距肛门门约5cm直肠前壁处即可触及前列腺,以手指末节作向内、向前徐徐按摩,每侧约4—5次,然后再将手移至腺体的上部顺正中沟向下挤压,这样前列腺液即可由尿道排出至前列腺液取样套8上,留取标本送检。其中,所取前列腺液量为0.5-1 ml;取样品杯放置区5的样品杯内预装有PH值为7.4的三羟甲基氨基甲烷-盐酸缓冲液3 ml,把带有前列腺液的前列腺取样套8放入样品杯中,混匀;从滴管存放区6内取出两支滴管,两支滴管分别预装有试剂A和试剂B,在测试池7内顺序加入样品液1ml,试剂A 0.5ml,试剂B 0.5ml,混匀,2分钟内观察颜色变化,与检测盒外包装的比色板2比对记录检测结果;所述试剂A的配制方法如下:取3,3',5,5'-四甲基联苯胺 50mg溶于PH4.4柠檬酸缓冲液100ml混匀而成;所述试剂B为3%的过氧化氢溶液。
检测结果判定如下: 
若不显色则为阴性,说明渗液中无游离血红素;若显示浅绿蓝色至浅蓝色,可能含有游离血红素,需定期进行复查;若显示蓝色至深蓝色则说明渗液中含有游离血红素。若即刻显示深蓝色且瞬间转为深黄色或棕红色,预示有肿瘤组织出血,亦应视为强阳性。
实验资料: 
分别取检测为不显色、显示浅绿蓝色至浅蓝色、蓝色至深蓝色的样品各三例,用质谱分析仪进行血红素的检测,检测结果如下:不显色均未测及血红素;浅绿色至浅蓝色为:0.5 ng/L ,1.7ng/L 、4.3 ng/L;蓝色至深蓝色为  20ng/L、2.5 μg/L、52μg/L 。
通过以上数据可以看出,对游离血红素含量在0.5ng/L以上的渗液本检测方法就可以初步的判断,游离血红素含量在20ng/L以上的渗液本检测方法就可以确定为渗液中含有游离血红素,所以本检测方法的灵敏度高。 
实施例2 
一种检测前列腺游离血红素的检测盒,包括盒体3和盒体外的外包装盒1,所述外包装盒1的外表面设有比色板2,所述盒体设有向内凹陷而成的镊子存放区4、样品杯存放区5、滴管存放区6和测试池7,所述测试池7内存放有前列腺液取样套8,所述前列腺液取样套8呈底端封闭的圆筒状,所述底端还设有一凸起的小囊9,所述前列腺液取样套8材质为吸附性无纺布,所述前列腺液取样套8的直径3.5cm,长度5cm,所述小囊9直径1cm,深度0.5cm,所述测试池7设于盒体3中上方,直径1.8cm,深度1.2cm,所述滴管存放区6设于测试池7两侧,所述样品杯存放区5设于测试池7左侧,所述镊子放置区4设于样品杯存放区5左侧,所述滴管存放区6内设有滴管,滴管上带有刻度,所述样品杯放置区5内设有样品杯,所述样品杯设有密封盖,样品杯的杯体呈圆筒状,所述镊子存放区4内设有镊子,所述镊子为塑料镊。

Claims (1)

1.  一种检测前列腺液游离血红素的方法,包括以下检测步骤:
    1) 用取样套取前列腺液0.5-1 ml;
    2) 把带有前列腺液的前列腺液取样套放入样品杯内,样品杯内预装有三羟甲基氨基甲烷-盐酸缓冲液,缓冲液的量为3ml,混匀,得样品液; 3) 在测试池内顺序加入样品液1ml,试剂A 0.5ml,试剂B 0.5ml;混匀,2分钟内观察颜色变化,与比色板比对记录检测结果;
所述试剂A的配制方法如下:取3,3',5,5'-四甲基联苯胺 50mg溶于PH为4.4柠檬酸缓冲液100ml混匀而成;所述试剂B为3%的过氧化氢溶液,
所述的三羟甲基氨基甲烷-盐酸缓冲液的PH值为7.4 。
