Abstract

本发明提供了一种检测直肠渗液血红素的方法,包括在直肠部位稍加压力擦拭获取渗液;用PH为7.4的三羟甲基氨基甲烷-盐酸缓冲液处理渗液;再与显色试剂反应后与比色板比色记录检测结果;本发明简单实用,成本低廉,可快速的筛查出直肠渗液中不含血红素的样本、含血红素的样本和可能含有血红素的样本,为进一步筛查做准备,本发明同时还提供了一种检测直肠渗液血红素的检测盒。

Description

一种检测直肠渗液血红素的方法
技术领域
 本发明涉及一种检测方法,具体涉及一种检测直肠渗液血红素的方法,同时本发明还涉及一种检测直肠渗液血红素的专用检测盒。 
背景技术
直肠渗液中的血红素往往是由组织细胞氧感受器和FixL跨膜蛋白的血红素释放、肿瘤细胞血红素析出或微循环渗出引起的,其血红素的含量极少,肉眼不可得见,一般需借助先进的仪器和分析化学方法进行检测,费用高,周期长,操作繁复,所以发明一种操作简单,成本低廉的检测方法快速的确定直肠渗液中是否含有血红素,成为一个亟需解决的问题。 
发明内容
 为了解决现有技术检测直肠渗液血红素方法中费用高、周期长、操作繁复的问题,本发明提供了一种检测直肠渗液血红素的方法,本发明的灵敏度高,成本低廉,可快速的检测直肠渗液中的血红素;同时本发明还提供了一种检测直肠渗液中血红素的专用的检测盒。 
本发明的技术方案如下: 
本发明提供的检测直肠渗液中血红素的方法,包括以下检测步骤:
 1)  带上乳胶手套,在食指第一指节处带上指套,在直肠部位稍加压力擦拭获取渗液;其中,渗液量为0.5~1ml;
2)  把带有渗液的指套放入样品杯内,样品杯内置有三羟甲基氨基甲烷-盐酸缓冲液,缓冲液的量为3ml,混匀,得样品液;优选的,三羟甲基氨基甲烷-盐酸缓冲液的PH值为7.4 ;
3) 在测试池内顺序加入样品液1ml,试剂A 0.5ml,试剂B 0.5ml;混匀,2分钟内观察颜色变化,与比色板比对记录检测结果;所述试剂A的配制方法如下:取3,3',5,5'-四甲基联苯胺 50mg溶于PH为4.4柠檬酸缓冲液100ml混匀而成;所述试剂B为3%的过氧化氢溶液。
 本发明同时提供了一种检测直肠渗液血红素的检测盒,包括盒体和与盒体滑动连接的滑盖,所述滑盖上设有向外凸起的两个条形槽,条形槽内装有滴管,条形槽从底部密封,所述盒体内部中空,一侧设有比色板,盒体内底部还设有测试池和放置样品杯的样品杯放置区,测试池边放置有指套,所述测试池从盒体底部向内延伸,中部向下凹陷为碗状结构。 
进一步的,所述样品杯放置区设于比色板和测试池之间。 
进一步的,所述比色板与盒体底部平行,且位于盒体的中上部。 
进一步的,所述滴管带有刻度。 
进一步的,所述样品杯设有密封盖,样品杯的杯体呈圆筒状。 
进一步的,所述指套的材质为具吸附性的无纺布。 
 本发明提供的检测方法,简单实用,成本低廉,可快速的筛查出直肠渗液中不含血红素的样本、含血红素的样本和可能含有血红素的样本,为进一步检查做准备;本发明可检测出的最低血红素浓度可达0.5ng/L,灵敏度高;同时本发明还提供了一种检测盒,可专用来盛放检测过程中用到的器皿和试剂,本检测盒结构简单,只需一个检测盒便能完成整个检测步骤,节省空间和试剂,同时也减轻操作者劳动负担。

具体实施方式
下面结合实施例对本发明做进一步的说明。 
实施例1 
       一种检测直肠渗液血红素的方法,方法如下:滑开检测盒的滑盖1,从盒体8内取出指套6,带上乳胶手套,再把指套6在食指第一关节处戴上,在直肠部位稍加压力擦拭获取渗液,其中,渗液量为0.5-1 ml;取样品杯放置区3的样品杯7,置入PH值为7.4的三羟甲基氨基甲烷-盐酸缓冲液3 ml,把带有渗液的指套6放入样品杯7中,混匀;从两个条形槽2内取出两支滴管,两支滴管分别预装有试剂A和试剂B,在测试池4内顺序加入样品液1ml,试剂A 0.5ml,试剂B 0.5ml,混匀,2分钟内观察颜色变化,与比色板5比对记录检测结果;所述试剂A的配制方法如下:取3,3',5,5'-四甲基联苯胺 50mg溶于PH4.4柠檬酸缓冲液100ml混匀而成;所述试剂B为3%的过氧化氢溶液。
检测结果判定如下: 
若不显色则为阴性,说明渗液中无血红素;若显示浅绿蓝色至浅蓝色,可能含有血红素,需进一步检测确定;若显示蓝色至深蓝色则说明渗液中含有血红素。
为确定本发明提供的检测方法的灵敏度,对显示不同颜色的样品用质谱仪进行进一步的验证,方法如下: 
    分别取检测为不显色、显示浅绿蓝色至浅蓝色、蓝色至深蓝色的样品各三例,用质谱分析仪进行血红素的检测,检测结果如下:不显色的均未测及血色素;浅绿色至浅蓝色为:0.5 ng/L 、1.7ng/L 、4.3 ng/L;蓝色至深蓝色为  20ng/L、2.5 μg/L、52μg/L 。
通过以上数据可以看出,本发明提供的检测方法可使血红素含量小于1ng/L的渗液显色,对血红素含量在20ng/L以上的渗液就可以确定渗液中含有血红素,所以本发明提供的检测方法的灵敏度较高,完全可以用来对直肠渗液中的血红素进行定性检测。 
实施例2 
    下面结合图1至图4对本发明专用检测盒做进一步的介绍。
本发明提供的检测直肠渗液血红素的检测盒,包括盒体8和与盒体8滑动连接的滑盖1,滑盖1可沿盒体8上表面方向滑动,所述滑盖1上设有向外凸起的两个条形槽2,条形槽2内装有一支滴管,条形槽2从底部密封,盒体8内部中空,盒体8的一侧设有比色板5,盒体8内底部还设有测试池4和放置样品杯7的样品杯放置区3,测试池4边放置有指套6,所述测试池4从盒体8底部向内延伸,中部向下凹陷为碗状结构,所述样品杯放置区3设于比色板5和测试池4之间,所述比色板5与盒体8底部平行,且位于盒体8的中上部,所述滴管带有刻度,所述样品杯7设有密封盖,样品杯7的杯体呈圆筒状,指套6的材质可为具吸附性的无纺布。 

Claims (1)

1.一种检测直肠渗液血红素的方法,其特征在于,包括以下检测步骤:
 1) 取直肠渗液;
2)  把带有渗液的指套放入样品杯内,样品杯内置有0.05mol/L三羟甲基氨基甲烷-盐酸缓冲液,缓冲液的量为3ml,混匀,得样品液;
3) 在测试池内顺序加入样品液1ml,试剂A 0.5ml,试剂B 0.5ml;混匀,2分钟内观察颜色变化,与比色板比对记录检测结果;所述试剂A的配制方法如下:取3,3',5,5'-四甲基联苯胺 50mg溶于PH4.4柠檬酸缓冲液100ml混匀而成;所述试剂B为3%的过氧化氢溶液;
所述的渗液的量为0.5-1ml;
所述的三羟甲基氨基甲烷-盐酸缓冲液的PH值为7.4 。
