一种应用于药物分析领域中的检测含有氨基药物中铵盐的方法，该检测方法步骤如下：取供试品1.0-1.25g，置分液漏斗中，加浓度为20%碱性溶液4ml，振摇，溶液中呈现油状物，加与水不相溶的有机相2ml，继续振摇，静置约5分钟，分出有机相层；再加2ml有机相重复萃取一次去弃有机相层，取水层备用；取水层，置试管中，加无氨蒸馏水至50ml，加碱性碘化汞钾试液2ml，摇匀，放置15分钟，如显色，与标准氯化铵溶液2ml按上述方法制成的对照溶液比较，不得更深；所述与水不互溶的有机溶剂为二氯甲烷、三氯甲烷、正己烷、环己烷中的一种或几种；所述碱性溶液为氢氧化钠或氢氧化钾。该发明在通过该方法检测铵盐，其重现性、灵敏度、准确度均能符合要求。

Description

一种检测含有氨基药物中铵盐的方法
技术领域
本发明涉及药物分析领域中的一种检测含有氨基药物中铵盐的方法。
背景技术
在结构中含有氨基的药物，成盐工艺中可能产生铵盐，铵盐无挥发性，因此很容易存在于药品中，无法去除。随着药品服用后，进入生物体内。研究表明铵盐在生物体内代谢过程则具有很大危害性。为保证药品的质量及其安全性，需建立检测方法对铵盐的含量进行控制。
目前铵盐的检测方法有：1、《中国药典》2010版二部附录Ⅷ K中铵盐检测法2、铵盐的纳氏试剂光度法铵盐检测法。因待检测药物结构中含有氨基，故上述两种方法均有一定干扰，无法对铵盐进行有效检测。因此，研制开发一种检测含有氨基药物中铵盐的方法一直是亟待解决的新课题。
发明内容
本发明的目的在于提供一种检测含有氨基药物中铵盐的方法，建立一种可以对结构中含有氨基的药物进行铵盐检测的方法，该种铵盐检测方法解决了主成分中含有氨基的药物不能通过现有方法检测样品中无机杂质铵盐的问题，通过该方法检测铵盐，重现性、灵敏度、准确度均能符合要求，极大的解决了可以有效检测铵盐测定存在干扰的问题。
本发明的目的是这样实现的：一种检测含有氨基药物中铵盐的方法，该检测方法步骤如下：取供试品1.0-1.25g，置分液漏斗中，加浓度为20%碱性溶液4ml，振摇，溶液中呈现油状物，加与水不相溶的有机相2ml，继续振摇，静置约5分钟，分出有机相层；再加2ml有机相重复萃取一次去弃有机相层，取水层备用；取水层，置试管中，加无氨蒸馏水至50ml，加碱性碘化汞钾试液2ml，摇匀，放置15分钟，如显色，与标准氯化铵溶液2ml按上述方法制成的对照溶液比较，不得更深；所述与水不互溶的有机溶剂为二氯甲烷、三氯甲烷、正己烷、环己烷中的一种或几种；所述碱性溶液为氢氧化钠或氢氧化钾。
本发明的要点在于检测含有氨基药物中铵盐的方法。其原理是：因为结构中含有氨基的需要检测铵盐方法，采用相似相溶的原理，先通过预处理的方式使用与水不互溶的有机溶剂将铵盐与主成分的氨基分离，排除待检药物结构中的氨基的干扰，为检出含有氨基的药物中无机杂质铵盐提供了有效的检测方法。
一种检测含有氨基药物中铵盐的方法与现有技术相比，具有在通过该方法检测铵盐，其重现性、灵敏度、准确度均能符合要求等优点，将广泛地应用于药物分析领域中。
下面结合实施例对本发明进行详细说明。
具体实施方式
以下实施例将有助于对本发明的了解，但这些实施例仅为了对本发明加以说明，本发明并不限于这些内容。
实施例一
本实例以盐酸金刚乙胺为例，
1、取盐酸金刚乙胺1.25g，置100ml量瓶中，加水溶解并稀释至刻度，摇匀，量取2ml于分液漏斗中，加4ml氢氧化钠试液，振摇，使样品成油状物，加二氯甲烷2ml溶液，继续振摇，放置约5分钟，分出二氯甲烷层；再加2ml二氯甲烷重复萃取一次去弃二氯甲烷层，取水层备用。
2、取水层，置试管中，加无氨蒸馏水至50ml，加碱性碘化汞钾试液2ml，摇匀，放置15分钟，如显色，与标准氯化铵溶液2ml按上述方法制成的对照溶液比较，不得更深。（检测限度为0.08%）
3、标准氯化铵溶液的制备，称取氯化铵31.5mg，置1000ml量瓶中，加水适量使溶解并稀释至刻度，摇匀，即得（每1ml相当于10μg的NH4）
取三批样品按上述方法检测，结果如下：

与《中国药典》2010版二部 附录Ⅷ k铵盐测定比较：
取盐酸金刚乙胺1.0g，置蒸馏瓶中，加无氨蒸馏水200ml，加氧化镁1g，加热蒸馏，馏出液导入加有稀硫酸1滴与无氨蒸馏水5ml的50ml纳氏比色管中，俟流出液达40ml时，随着流出液增加，溶液变浑浊，使试验无法进行。

4、方法验证
①取无氨蒸馏水2ml代替样品，置试管中，以下按1.2.项下操作，结果不显色。说明空白不干扰。
②分别取标准氯化铵溶液1ml、2ml、4ml，置试管中，以下按1.2.项下操作，结果溶液颜色随着标准氯化铵溶液浓度增加而颜色逐渐加深。说明灵敏度高、准确度符合要求。
③取盐酸金刚乙胺1.0g三个，分别置试管中，按1.2项下操作，结果一致。说明重现性好。
实施例二
1、取盐酸美金刚1.0g，置分液漏斗中，加4ml氢氧化钾试液，振摇，使样品成油状物，加环己烷2ml溶液，继续振摇，放置约5分钟，分出环己烷层；再加2ml环己烷重复萃取一次去弃环己烷层，取水层备用。
2、取水层，置试管中，加无氨蒸馏水至50ml，加碱性碘化汞钾试液2ml，摇匀，放置15分钟，如显色，与标准氯化铵溶液2ml按上述方法制成的对照溶液比较，不得更深。（检测限度为0.02%）

1.一种检测含有氨基药物中铵盐的方法，其特征在于：该检测方法步骤如下：取供试品1.0-1.25g，置分液漏斗中，加浓度为20％碱性溶液4ml，振摇，溶液中呈现油状物，加与水不相溶的有机相2ml，继续振摇，静置约5分钟，分出有机相层；再加2ml有机相重复萃取一次去弃有机相层，取水层备用；取水层，置试管中，加无氨蒸馏水至50ml，加碱性碘化汞钾试液2ml，摇匀，放置15分钟，如显色，与标准氯化铵溶液2ml按上述方法制成的对照溶液比较，不得更深。
2.根据权利要求1所述的一种检测含有氨基药物中铵盐的方法，其特征在于：所述与水不互溶的有机相为二氯甲烷、三氯甲烷、正己烷、环己烷中的一种或几种。
3.根据权利要求1所述的一种检测含有氨基药物中铵盐的方法，其特征在于：所述碱性溶液为氢氧化钠或氢氧化钾。
