Abstract

本发明描述了一种可用于细胞培养、组织工程和化妆品领域的水溶性胶原蛋白。属于医用生物材料领域。主要解决目前水溶性胶原蛋白存放过程中易交联变性,从而无法水溶的问题。我们制备的水溶性胶原是利用水溶性胶原蛋白及相应的抗氧剂相结合,制成的一种稳定的、具有抗交联作用的水溶性胶原蛋白,有助于水溶性胶原蛋白的保存和应用。

具有抗交联作用的水溶性胶原蛋白
技术领域
本发明涉及一种用于细胞培养、组织工程和化妆品领域的水溶性胶原蛋白,其主要成份是水溶性胶原蛋白和抗氧化剂组成,外观为冻干海绵。
背景技术
胶原蛋白是一种多细胞动物产生的细胞外基质成份之一。随着组织工程的发展,被广泛地用于组织培养、医用材料等领域,常见的I型胶原蛋白为非水溶性胶原,目前市场上常见的水溶性胶原其实是I型胶原降解为多肽后形成的胶原肽,人们在胎牛和犊牛皮中也提取出完全不同于I型胶原的未变性的水溶性胶原,但是由于其特殊的空间结构所产生的分子间力的作用极易产生交联而转化为不溶于水的胶原蛋白,保存期极短,以至于无法大量地应用于实验和生产过程中。本方法利用抗氧剂的独特作用,减小水溶性胶原分子结构中分子间力的作用,从而达到克服其产生交联的性质,延长保存期,改善保存效果,增加水溶性胶原的稳定性。
发明内容
本材料选用了市售的水溶性胶原与抗氧化剂复合而成:
1、水溶性胶原溶液的制备:取一定量市售的、未变性且存放期不超过一周的水溶性胶原,溶于去离子水中,搅拌溶解成均匀的质量体积比浓度为0.1%~0.5%的胶原胶液。
2、抗氧化剂水溶液的制备:市售维生素C、花青素、茶多酚、茄红素溶于去离子水,搅拌溶解成质量体积比浓度为5%~10%的抗氧化剂水溶液。
3、将(1)中所述溶液放入透析袋中,用(2)中的抗氧化剂水溶液做透析外液进行透析,透析温度2~8℃,每4小时换一次透析外液,透析72小时,透析物5000rpm,2~8℃条件下离心10min,取上清液冷冻干燥成型。
具体实施方式
实施例1、
取市售水溶性胶原溶液100mg,加入1000ml去离子水中,搅拌溶解成质量体积比浓度为0.1%的胶原溶液。维生素C50g溶于1000ml去离子水中,制成质量体积比浓度为5%的抗氧化剂溶液,该溶液现用现制,用抗氧化剂溶液做透析外液对胶原溶液进行透析,透析温度2~8℃,每4小时换一次透析外液,透析72小时,透析物5000rpm,2~8℃条件下离心10min,取上清液冷冻干燥成型。
实施例2、
取市售水溶性胶原溶液500mg,加入1000ml去离子水中,搅拌溶解成质量体积比浓度为0.5%的胶原溶液。花青素100g溶于1000ml去离子水中,制成质量体积比浓度为10%的抗氧化剂溶液,该溶液现用现制,用抗氧化剂溶液做透析外液对胶原溶液进行透析,透析温度2~8℃,每4小时换一次透析外液,透析72小时,透析物5000rpm,2~8℃条件下离心10min,取上清液冷冻干燥成型。
实施例3、
取市售水溶性胶原溶液200mg,加入1000ml去离子水中,搅拌溶解成质量体积比浓度为0.2%的胶原溶液。茶多酚70g溶于1000ml去离子水中,制成质量体积比浓度为7%的抗氧化剂溶液,该溶液现用现制,用抗氧化剂溶液做透析外液对胶原溶液进行透析,透析温度2~8℃,每4小时换一次透析外液,透析72小时,透析物5000rpm,2~8℃条件下离心10min,取上清液冷冻干燥成型。
实施例4、
取市售水溶性胶原溶液400mg,加入1000ml去离子水中,搅拌溶解成质量体积比浓度为0.4%的胶原溶液。茄红素80g溶于1000ml去离子水中,制成质量体积比浓度为8%的抗氧化剂溶液,该溶液现用现制,用抗氧化剂溶液做透析外液对胶原溶液进行透析,透析温度2~8℃,每4小时换一次透析外液,透析72小时,透析物5000rpm,2~8℃条件下离心10min,取上清液冷冻干燥成型。
实施例5、
取市售水溶性胶原溶液300mg,加入1000ml去离子水中,搅拌溶解成质量体积比浓度为0.3%的胶原溶液。花青素60g溶于1000ml去离子水中,制成质量体积比浓度为6%的抗氧化剂溶液,该溶液现用现制,用抗氧化剂溶液做透析外液对胶原溶液进行透析,透析温度2~8℃,每4小时换一次透析外液,透析72小时,透析物5000rpm,2~8℃条件下离心10min,取上清液冷冻干燥成型。

Claims (4)

1.一种用于细胞培养、组织工程和化妆品领域的水溶性胶原蛋白,其主要成份是水溶性胶原蛋白和抗氧化剂组成,外观为冻干海绵。
2.根据权利要求1所述的水溶性胶原,是指市售、未变性的可溶于水的胶原蛋白。
3.根据权利要求1所述的抗氧化剂,是指维生素C、花青素、茶多酚、茄红素。
4.根据权利要求1所述的冻干海绵,其特征在于制备过程包括下列各步骤:
(1)一定量的市售水溶性胶原加入去离子水中配制成质量体积比浓度为0.1~0.5%的胶原溶液;
(2)一定量的抗氧化剂溶于去离子水中配制成质量体积比浓度为5~10%的水溶液;
(3)将(1)中所述溶液放入透析袋中,用(2)中的抗氧化剂水溶液做透析外液进行透析,透析温度2~8℃,每4小时换一次透析外液,透析72小时,透析物5000rpm,2~8℃条件下离心10min,取上清液冷冻干燥成型。
