呼吸道疾病是用于呼吸系统疾病的术语,包括上呼吸道和下呼吸道(例如肺、胸膜腔、支气管、气管、以及与呼吸有关的神经和肌肉)的疾病。呼吸道疾病可能很温和并且是自我限制的(例如普通感冒),所以通常不需要治疗便能好。然而,呼吸道疾病也可能威胁生命,例如细菌性肺炎或病毒性肺炎,所以对于更易受微生物感染影响的人来说可能需要额外的照顾和额外的治疗,例如年幼者、老人、业已存在肺部疾病的人、以及免疫系统弱的人。
呼吸道疾病的治疗取决于要治疗的具体疾病、疾病的严重性和患者。预防接种可以预防某些呼吸道疾病,也可以使用抗生素。然而,病毒和真菌感染的增长和人类细菌性病原体中抗微生物药物抗性的出现是一个日益加剧的世界性问题。此外,自从抗微生物剂的引入,抗药性的出现已变得越来越普遍,特别是对于重要的病原体来说,例如E.coli(大肠杆菌)和Staphylococcus spp(葡糖球菌)。因此,对这些微生物的有效治疗和对呼吸道疾病的控制成为更大的挑战。

本发明涉及用于治疗引起呼吸道感染的微生物感染的新型疗法的开发。本发明特别涉及用于治疗范围广泛的病毒感染(包括急性病毒感染)的新型疗法的开发以及由其引起的呼吸道疾病的治疗。急性病毒感染的特征为,快速发病、相对短暂的症状以及通常在几天内消退。其通常伴随传染性病毒体的早期生产和宿主免疫体系的消除感染。一般对于诸如流感病毒和鼻病毒的病原体观察到急性病毒感染。急性病毒感染可能很严重,一个显著的例子是H1N1流感病毒,其导致1918年西班牙流感大流行。
急性感染从潜伏期开始,在该时间段内病毒基因组复制,宿主的先天性应答被激发。早期在感染中产生的细胞因子导致急性感染的一般症状:疼痛(ache)、痛(pain)、发烧和恶心。某些潜伏期短至1天(流行性感冒、鼻病毒),表明这些症状是由接近入口处的局部病毒增殖产生。典型急性感染的例子是无并发的流感。打喷嚏或咳嗽时产生的病毒粒子以液滴形式吸入,然后开始在呼吸道的纤毛柱状上皮细胞中复制。当产生新的传染性病毒体时,它们传播到邻近的细胞中。感染1天到7天后,可以从喉拭子或鼻分泌物中分离出病毒。48小时内出现感染症状,这些症状将持续约3天,然后减轻。感染通常通过先天性和适应性应答在约7天内清除。然而,患者通常在数周内感觉不舒服,这是由感染期间产生的细胞因子对呼吸道上皮的损伤的结果。
急性病毒感染(例如流行性感冒、鼻病毒)是每年所涉及的数百万个体的流行病的原因。当疫苗不可得时,难以控制急性病毒感染。这就使得在大量人口和拥挤的地区中非常难以控制急性感染。诺如病毒性肠胃炎(一种典型的急性感染)的频繁爆发使该问题突出。不能使用抗病毒疗法,因为抗病毒疗法必须在感染提供才有效。因而用抗病毒药治疗大部分急性病毒感染的希望很小,除非快速诊断测试变得可用。然而,需要注意的是,目前没有用于大部分普通急性病毒疾病的抗病毒药。因此,该领域明显需要可用于病毒感染治疗(特别是急性病毒感染)的改进药物。
本发明人业已确定某些烷基取代或未取代的2-芳基乙酸或2-芳基,N-羟基乙酰胺衍生物具有可用于治疗微生物感染的性能。

因而,在另一方面,本发明涉及使用己酮可可碱、或其药物可接受的盐、溶剂化物、或盐的溶剂化物来治疗病毒感染。
已知在急性病毒感染(例如流行性感冒)期间,病毒占优势地与宿主先天性免疫系统和细胞介导的Th1应答斗争,随后被体液的抗体驱动的Th2应答抵抗。此外,本发明人认为,在易感染的个体(即,年轻和健康的个体),对流感感染的Th1应答可能极其强烈,可能引起所谓的“细胞因子风暴”,包括某些细胞因子(例如IFN-γ和TNF-α)浓度显著增大。“细胞因子风暴”可能导致受感染的肺部组织严重发炎、液体渗漏到肺中并且严重损害受感染个体的肺部。最终的结果可能是呼吸道疾病,例如肺水肿或继发性细菌感染,这最终可能使受感染的个体死亡,而不是病毒本身杀死该个体。
Baumgarth和Kelso(J.Virol.,1996,70,4411-4418)报导了Th1细胞因子(IFN-γ)的中和可以使受感染的肺部细胞中细胞浸润的量显著降低,并且提出IFN-γ可能涉及在发炎的肺部中控制增大的白细胞迁移的机理。他们还假定,IFN-γ影响在呼吸道中的局部细胞应答,以及对流感病毒感染的全身体液免疫应答。基于该研究的发现,本发明的发明人考虑了抑制细胞因子(IFN-γ和TNF-α)是否可用于治疗流感。
如实施例中所述,本发明人研究了烷基取代或未取代的2-芳基乙酸或2-芳基,N-羟基乙酰胺衍生物或己酮可可碱在体外对以反映出急性病毒感染的方式受刺激的血细胞的作用。作为病毒感染的模型,他们使用已用促细胞分裂剂(脂多糖或伴刀豆球蛋白A),触发信号转导途径(从而刺激存在于血细胞中的淋巴细胞开始有丝分裂)的化合物,刺激的血细胞样品。因而该模型非常接近地复制了由病毒感染引起的过程,能直接评估用测试化合物治疗之后淋巴细胞显示出的免疫应答。
如实施例中所述,本发明人发现,使用这种体外模型,异丁普生、苯恶洛芬羟肟酸盐或己酮可可碱有效地抑制两种细胞因子(IFN-γ和TNF-α)的产生。因此,本发明是基于对由IFN-γ驱动的Th1免疫系统的控制,IFN-γ是在易感染个体(例如年轻人和健康人)中引起呼吸衰竭的超免疫细胞介导应答的原因。

普通的病原体引起的呼吸道疾病或急性呼吸窘迫是医院内感染的肺炎和社会获得性肺炎。肺炎的特征为咳嗽、胸痛、发热、由于肺水肿引起的呼吸困难。这些症状出现在所有的患者中,不管引起肺炎的病原体如何,病原体可以为细菌(例如Streptococcus pneumonia)、病毒(例如流感病毒)和真菌(例如Histoplasma capsulatum)。不管引起肺炎的病原体如何,症状是相同的;不管刺激源是什么,发炎过程引起过大的炎性反应,导致潜在致命的肺水肿。在与流感感染(即,病毒病原体)有关的呼吸道疾病的动物模型中,终点被设计为测量与肺水肿有关的终点(即,感染后的生存)。对于本文所述的化合物来说,在流感试验中,对感染后生存的效果支持对由任意类型的病原体(病毒、细菌或真菌)引起的肺水肿的效果的可能性。

包含本发明的化合物的组合物可以多种方式使用。例如,当化合物包含在例如可以药片、胶囊或液体的形式口服吸收的组合物中时,可能需要口服给药。或者,组合物可以通过注射到血流中给药。注射可以是静脉(推注或注入)或皮下(推注或注入)。或者,包含本发明的化合物的组合物可以通过吸入(例如滴鼻或通过嘴)或直肠(例如栓剂)给药。
组合物也可以被配制成局部使用。例如,可以将软膏涂覆到皮肤、嘴或生殖器内和周围的区域,来治疗特定的病毒感染。局部涂覆到皮肤特别适用于治疗皮肤的病毒感染或作为透皮给药至其他组织的手段。
要理解的是,所需的化合物的量由其生物活性和生物利用度决定,这转而取决于给药模式、化合物的物理化学性质以及化合物是作为单一药物疗法使用或是以组合疗法使用。给药频率也将受到上述因素的影响,特别是化合物在要治疗的受试者体内的半衰期。

通常,根据所用的化合物,日剂量为0.001μg/kg体重到20mg/kg体重的化合物可用于预防和/或治疗微生物(例如病毒)感染。适宜地,日剂量介于0.01μg/kg体重和10mg/kg体重之间,更适宜地介于0.01μg/kg体重和1mg/kg体重之间,或介于0.1μg/kg和100μg/kg体重之间,最适宜地介于约0.1μg/kg和10μg/kg体重之间。
化合物的日剂量可以作为单次给药(例如,单次每日注射或单次吸入)给予。适合的日剂量可以介于0.07μg和700mg(即,假设体重为70kg)之间,或者介于0.70μg和500mg之间,或者介于10mg和450mg之间。可以在引起呼吸道疾病的病原体(例如病毒)感染之前或之后施用药物。可以在感染之后2、4、6、8、10或12小时内施用药物。可以在感染之后14、16、18、20、22或24内施用药物。可以在感染之后1、2、3、4、5或6天内施用药物,或者可以在其之间的任何时间段。
在其中要治疗的感染是流感感染的实施方式中,不论流感是否是大流行性流感,受试者是用包含本发明的化合物的药物进行治疗中,其出现呼吸困难的症状和/或其中细胞因子(上面提到的细胞因子中的任意一种,但通常为IFN-γ或TNF-α)水平在呼吸困难症状开始时增大。更优选地,受试者是其中出现呼吸困难和/或其中在流感症状开始之后的下列时间细胞因子增大的受试者:12、24、18或36小时或更长(更优选48小时或更长,60小时或更长,或72小时或更长;最优选36-96小时,48-96小时,60-96小时,或72-96小时)。或者,不论流感是否是大流行性流感,受试者是那些其中出现呼吸困难和/或其中细胞因子水平在适应性免疫系统招募到受感染的肺部中开始(或早期)时增大者。
如在实施例中的小鼠体内研究中所述,本发明人已表明,用不止一剂细胞因子抑制剂给药的小鼠显示出流感感染症状的改善。因此,预期了包含化合物(I)或己酮可可碱的药物可以向需要治疗的受试者施用不止一次。化合物可能每天需要施用两次或更多次。例如,化合物(I)可以两次(或更多次,这取决于要治疗的病毒感染的严重性)0.07g-700mg的日剂量(即,假设体重为70kg)施用。接受治疗的患者可以在起床后服下第一剂,然而在晚上服下第二剂(如果是双剂量方案),或之后每隔3小时或每隔4小时等。可想到的是,在病毒感染之后,化合物可以每天施用(如果需要可以不止一次)。
因此,优选地,本发明的化合物适合施用于如上所述的受试者,优选地适合在流感症状开始后的上述时间点施用。
或者,可以使用缓释装置以向患者提供本发明的化合物的最佳剂量,而不需要重复给药。
基于本发明人的发现,即本文所述的化合物可用于降低细胞因子(例如TNF-α和IFN-γ)水平,本发明人认为,化合物的这些作用可以被利用并用于生产临床可用的化合物。
因此,在第四方面中,提供了一种包含治疗有效量的如之前所定义的由通式1所代表的化合物或己酮可可碱、以及药物可接受的赋形剂的药物组合物,其用于治疗引起呼吸道疾病的病原体感染。
感染可以是急性的或慢性的。
“治疗有效量”的由式(I)所代表的化合物或己酮可可碱是当给药至受试者时,导致细胞因子(例如TNF-α和IFN-γ)水平降低,从而提供对微生物感染(例如急性病毒感染)的预防和/或治疗的任何量。
例如,所用的式(I)化合物或己酮可可碱的治疗有效量可以为约0.07μg到约700mg,优选地为约0.7μg到约70mg。式(I)的量为约7μg到约7mg,或为约7μg到约700μg。
“受试者”可以为脊椎动物、哺乳动物或驯养动物,优选地为人类。因此,根据本发明的药物可用于治疗任何哺乳动物(例如人)、家畜、宠物,或者可用于其他兽医应用。
本文中所称的“药物可接受的赋形剂”可以是本领域技术人员已知的用于配制药物组合物的已知化合物的任意组合。
在一个实施方式中,药物可接受的赋形剂可以是固体,该组合物可以是粉末或药片形式。固体药物可接受赋形剂可以包括一种或更多种也可充当下列试剂的物质:调味剂、润滑剂、增溶剂、悬浮剂、染料、填充剂、助流剂、压缩助剂、惰性粘合剂、甜味剂、防腐剂、染料、包衣、或药片崩解剂。赋形剂也可以是包封材料。在粉末中,赋形剂是与细碎的活性剂(即,根据本发明的式(I)化合物或己酮可可碱)混合的细碎固体。在药片中,活性剂可以与具有所需压缩性能的赋形剂以适当的比例混合并压成期望的形状和尺寸。优选地,粉末和药片包含至多99%的活性剂。合适的固体赋形剂包括例如磷酸钙、硬脂酸镁、滑石、糖、乳糖、糊精、淀粉、明胶、纤维素、聚乙烯基吡咯烷酮、低熔点蜡和离子交换树脂。
在另一个实施方式中,药物赋形剂可以是凝胶,并且组合物可以是乳膏或类似物的形式。在另一个实施方式中,药物赋形剂可以是液体,并且药物组合物可以是溶液的形式。液体赋形剂被用于制备溶液、悬浮液、乳液、糖浆、酏剂和加压组合物。活性化合物可以溶解或悬浮在药物可接受的液体赋形剂(例如水、有机溶剂、两者或药物可接受油或脂肪的混合物)中。液体赋形剂可以包含其他合适的药物添加剂,例如增溶剂、乳化剂、缓冲剂、防腐剂、甜味剂、调味剂、悬浮剂、增稠剂、颜料、粘度调节剂、稳定剂或渗透压调节剂。用于口服和肠胃外给药的液体赋形剂的合适例子包括水(部分包含上述添加剂,例如纤维素衍生物,优选羧甲基纤维素钠溶液)、醇(包括单羟基醇和多羟基醇,例如乙二醇)及其衍生物、以及油(例如分馏的椰子油和花生油)。对于肠胃外给药来说,赋形剂也可以是油状酯(例如油酸乙酯和肉豆蔻酸异丙酯)。无菌液体赋形剂可用在肠胃外给药的液体形式组合物中。用于加压组合物的液体赋形剂可以是卤代烷烃或其他药物可接受推进剂。
作为无菌溶液或悬浮液的液体药物组合物可以通过例如肌肉内注射、鞘内注射、硬膜外注射、腹膜内注射、静脉内注射并且特别地皮下注射来使用。根据本发明的组合物可以被制成无菌固体组合物,在给药时可以使用无菌的水、盐水或其他适当的无菌可注射介质溶解或悬浮。
化合物可以以包含其他溶质或悬浮剂(例如,足量的盐水或葡萄糖以使溶液等渗压)、胆盐、阿拉伯胶、明胶、失水山梨醇单油酸酯、聚山梨酯80(山梨醇的油酸酯及其酸酐与氧化乙烯共聚)等的无菌溶液或悬浮液的形式口服给药。化合物还可以液体或固体组合物形式口服给药。适用于口服给药的组合物包括固体形式(例如丸剂、胶囊、颗粒、药片和粉末)和液体形式(例如溶液、糖浆、酏剂和悬浮液)。可用于肠胃外给药的形式包括无菌的溶液、乳液和悬浮液。
本文(包括任何所附的权利要求、摘要和附图)中所述的所有特征、和/或所公开的任何方法或工艺的所有步骤可以与上述方面任意组合,但排除其中所述特征和/或步骤的至少一些相互排斥的组合。

实施例
本发明人进行了一系列体外和体内实验以确定式I所代表的各种化合物或己酮可可碱对细胞因子(IFN-γ和TNF-α)的产生的影响。在下面所述的结果中,本发明人业已证明,本发明的化合物令人惊讶地充当IFN-γ和TNF-α的抑制剂。此外,他们已证明,在小鼠体内模型中,所述化合物的给药导致小鼠的病毒感染症状减轻(即,重量损失降低、存活率增大、总的患病率降低)。
材料和方法
小鼠体内研究
方案:
50只(50)C57BL/6雌性鼠(6-7周大)被分为5个(5)实验组,每组包含10只(10)动物。在第1天,在氟烷诱导麻醉下,动物接受滴鼻致死剂量(总共50μl,每个鼻孔25μl)的流感病毒A/PR/8/34。在第3天,动物接受一次测试化合物(360μg异丁普生(BC1048),54μg奥沙美辛(BC1002),180ug苯恶洛芬(BC 1005),89μg苯恶洛芬羟肟酸盐(BC1006))的腹膜内注射(100-150μl)。
从第1天到至少第6天,每天评估所有动物的发病率、重量损失和存活率。根据下列严重程度记录发病率的变量(即,体况、姿势、活动性、立毛、呼吸、发声、运动失调、眼/鼻分泌物:正常(0),轻度(1),吃力(2)和严重/等外点(3)。
实施例—小鼠体内研究
使用如上所述的标准技术,用H1N1病毒使小鼠感染,并确定使每一个受试者受到感染。随后,在病毒感染后的第3天,用单次剂量的异丁普生(BC1048)、奥沙美辛(BC1002)、苯恶洛芬(BC1005)、苯恶洛芬羟肟酸盐(BC1006)或己酮可可碱治疗每一个测试小鼠。在对照组的小鼠中,没有施用异丁普生(BC1048)、奥沙美辛(BC1002)、苯恶洛芬(BC1005)、苯恶洛芬羟肟酸盐(BC1006)或己酮可可碱(BC1042)。然后测定治疗和未经治疗的小鼠的重量损失。
如图1、3和5所示,接受异丁普生(BC1048)、奥沙美辛(BC1002)、苯恶洛芬(BC1005)、苯恶洛芬羟肟酸盐(BC1006)或己酮可可碱(BC1042)的剂量的小鼠与对照组的小鼠相比,显示出重量损失的至少10%的降低。因此,尽管本发明人不想束缚于假设,但本发明人认为被H1N1感染的小鼠受到异丁普生、奥沙美辛、苯恶洛芬、苯恶洛芬羟肟酸盐或己酮可可碱作用之后,细胞因子(IFN-γ和TNF-α)水平的降低使得小鼠维持其体重。
参考图2、4和6,显示了用异丁普生、奥沙美辛、苯恶洛芬、苯恶洛芬羟肟酸盐或己酮可可碱治疗后小鼠的存活率的结果。可以看出,用异丁普生、奥沙美辛、苯恶洛芬、苯恶洛芬羟肟酸盐或己酮可可碱治疗的小鼠与对照组未经治疗的小鼠相比,显示出较高的存活率。
体外研究-使用促细胞分裂剂(LPS和Con A)的刺激实验
当浆细胞(B细胞)遇到与其免疫球蛋白相匹配的抗原时,它们可以进入有丝分裂。促细胞分裂剂是触发信号转导途径(其中涉及分裂原活化蛋白激酶)的化合物,从而促进细胞开始进行细胞分裂,导致有丝分裂。因而,促细胞分裂剂能有效地用于刺激淋巴细胞,从而评估免疫功能。通过刺激淋巴细胞,促细胞分裂剂可用于复制病毒感染的影响。
本发明人用于刺激淋巴细胞并从而评估免疫功能的两种促细胞分裂剂是脂多糖(LPS)和伴刀豆球蛋白(Con A)。LPS对B细胞起作用,但对T细胞不起作用;而Con A对T细胞起作用,但对B细胞不起作用。在LPS和Con A刺激的试验中,研究式1所代表的化合物的两个实施方式(即异丁普生(在表中被称为BC1048)和苯恶洛芬羟肟酸盐(BC1006))对IFN-γ和TNF-α水平的影响。外周血单核细胞(PMBC)独立于各促细胞分裂剂(LPS或Con A)单独施用,然后用异丁普生、苯恶洛芬羟肟酸盐或己酮可可碱治疗。对照试验在不加入LPS或Con A的条件下进行,对IFN-γ和TNF-α水平的任何影响直接归因于测试化合物(异丁普生、苯恶洛芬羟肟酸盐或己酮可可碱)的存在。
材料和方法
外周血单核细胞(PBMC)的分离、培养和处理
在6ml采血管(绿盖)中采集血液。在采集2小时内处理血液。
所用材料:未凝固的血;FCS;补充L-Gln和P/S的RPMI-1640培养基;PBS;无菌尖嘴和吸管;无菌的15ml Falcon;无菌的有盖的V型底96孔板;Neubauer腔室;台盼蓝溶液;70%IPA溶液;Accuspin-Histopaque管(Sigma,A7054)。
过程:
1.在无菌PBS中稀释样品1∶1;
2.将30ml经稀释的血液加入Accuspin-Histopaque管(Sigma,A7054)中;
3.在室温(RT)下以800rcf的速度离心15分钟;
4.离心后,红细胞将留在筛板下的底部。单核细胞(PBMC)将出现在筛板上方的层中,血浆在顶层;
5.用吸管将PBMC层收集到新的15ml Falcon管中,并加满PBS至15ml;
6.在室温下以250rcf的速度离心;
7.丢弃上清液,轻拂球粒,并再次加入10ml PBS;
8.在室温下以250rcf的速度离心;
9.重复步骤7和8;
10.丢弃上清液,使球粒在1ml完全培养基(RPMI-1640 10%FCS)中再悬浮;
11.计算细胞数,并使得完全培养基中的浓度为4x106细胞/ml悬浮液。向每个V型底96孔板加入100μl细胞悬浮液。然后,将50μl刺激物或赋形剂加入完全培养基,然后将50μl药物或赋形剂加入完全培养基。在37℃和5%CO2下培养细胞24小时;
12.培育之后,取出60μl细胞上清液,根据制造商的说明(BD Biosciences)通过ELISA测量IFNγ和TNFα(OptEIA人IFNγ,商品目录号555142和人TNF,商品目录号555212)。
LPS刺激研究
LPS刺激实验的结果显示在表1中。表中的值表示相对于仅作为对照组的LPS的值的百分比。因此,在仅存在LPS时由PMBC细胞表达的细胞因子(IFN-γ或TNF-α)的最大浓度被认为为100%,而存在(i)LPS和(ii)异丁普生(BC1048)、苯恶洛芬羟肟酸盐(BC1006)或己酮可可碱(BC1042)时,由PMBC细胞表达的细胞因子浓度表示为相对于仅有LPS的100%对照组的百分比。在所表达出的FN-γ或TNF-α水平的各值下方给出了标准差的值(st error)。

参考表1所示的数据,本发明人出乎意料地观察到,在LPS刺激的细胞中,在异丁普生(BC1048)、苯恶洛芬羟肟酸盐(BC1006)或己酮可可碱(BC1042)的存在下IFN-γ和TNF-α的浓度降低。在最高的浓度(100μM)下刺激之后,异丁普生完全阻断IFN-γ的产生,而对TNF-α的影响较小。,苯恶洛芬羟肟酸盐在所用的所用浓度(1-100μM)下都一贯地抑制IFN-γ的产生(35%到21%),其对TNF-α的最大影响是在较高的浓度下(41%)。己酮可可碱以与异丁普生类似的方式抑制IFN-γ的产生,在100μM下有影响,尽管该影响不如异丁普生明显,并且同样,对TNF-α的影响较小。
Con A刺激研究
Con A实验的结果被示于表2中。

参考表2所示的数据,本发明人观察到,在Con A的细胞中,在异丁普生(BC1048)、苯恶洛芬羟肟酸盐(BC1006)或己酮可可碱(BC1042)的存在下IFN-γ和TNF-α的浓度也降低。在该体外系统中,在最高的浓度(100μM)下,异丁普生对Con A刺激的IFN-γ和TNF-α的产生的影响适中。苯恶洛芬羟肟酸盐对IFN-γ和TNF-α的影响较大,在10和100μM下的影响比较明显。在这种刺激下,己酮可可碱对Con A诱导的TNF-α和IFN-γ的产生的影响很小。
总结
总而言之,本发明人出乎意料地观察到异丁普生、奥沙美辛、苯恶洛芬、苯恶洛芬羟肟酸盐和己酮可可碱改善了受到流感激发的小鼠的存活率。因此本发明人认为式(I)所代表的任何化合物或己酮可可碱可以用作IFN-γ和TNF-α的抑制剂,其可用于治疗引起呼吸道疾病的病原体感染(例如流感)。实施例中所描述的小鼠体内研究的令人鼓舞的成果清楚地表明,被H1N1病毒感染的小鼠可以通过施用单次剂量的异丁普生、奥沙美辛、苯恶洛芬、苯恶洛芬羟肟酸盐或己酮可可碱而被有效治疗。因此,很明显任意化合物(I)或己酮可可碱可用于治疗引起暴发性呼吸道疾病的病毒感染或其他病原体感染。
Claims (24)
1.一种式I的化合物、或其药物可接受的盐、溶剂化物、或盐的溶剂化物:
其中,Ar是芳基或取代的芳基,R1为C1-3烷基或氢,R2为OH或-NHOH,用于治疗引起呼吸道疾病、优选引起暴发性呼吸道疾病的病原体感染。
2.如权利要求1所述的化合物,其中Ar是取代的苯基。
3.如前面任何一项权利要求所述的化合物,其中,R1为氢或甲基。
4.如前面任何一项权利要求所述的化合物,其中,R2为-NHOH。
5.如前面任何一项权利要求所述的化合物,其中,当Ar为取代的苯基时,将其连接到式I所示结构的剩余部分的所述键直接延伸至苯环上的碳原子上。
6.如前面任何一项权利要求所述的化合物,其中,化合物(I)是2-芳基,N-羟基乙酰胺或2-芳基,2-甲基,N-羟基乙酰胺衍生物。
7.如前面任何一项权利要求所述的化合物,其中,化合物(I)是异丁普生、奥沙美辛、苯恶洛芬、或苯恶洛芬羟肟酸盐。
8.如权利要求1-6所述的化合物,其中,化合物(I)是

13.如前面任何一项权利要求所述的化合物,其中,所述化合物被用于治疗真菌感染。
14.如前面任何一项权利要求所述的化合物,其中,所述化合物被用于治疗病毒感染,优选急性病毒感染。
15.如权利要求14所述的化合物,其中,所述化合物被用于治疗副粘病毒感染或正粘病毒感染。
16.如权利要求14或15所述的化合物,其中,所述化合物被用于治疗由A型流感病毒、B型流感病毒、C型流感病毒、或其衍生物中的任意一种引起的感染。
17.如权利要求16所述的化合物,其中,所述化合物被用于治疗选自由下列组成的组中的任意血清型的A型流感病毒的感染:H1N1;H1N2;H2N2;H3N1;H3N2;H3N8;H5N1;H5N2;H5N3;H5N8;H5N9;H7N1;H7N2;H7N3;H7N4;H7N7;H9N2;和H10N7,或其衍生物。
