Abstract

发明领域
本发明大体上涉及癌胚胎抗原/未成熟层粘连蛋白受体蛋白(OFA/iLRP)。更具体来说,本发明提供可用于检测和治疗OFA/iLRP相关疾病的抗体。
发明背景
癌胚胎抗原/未成熟层粘连蛋白受体蛋白(OFA/iLRP)的初始鉴定由研究癌胚胎抗原或层粘连蛋白受体的三个独立的组完成[1-3]。OFA/iLRP是高度保守的蛋白质,其在一系列不同癌症中过表达并具有作为核糖体蛋白p40的双重功能[4-27]。OFA/iLRP蛋白质包含295个氨基酸的单一多肽链并具有约37-44kDa的分子量。OFA/iLRP的结构最近被阐明到的水平[28]。层粘连蛋白受体(LPR)的成熟形式似乎为乙酰化的OFA/iLRP二聚体,其具有67kDa的分子量。结构显示OFA/iLRP的第112到140位氨基酸间的区域涉及形成LPR的OFA/iLRP二聚化[28]。尽管67kDa的LRP出现在许多正常细胞和肿瘤细胞中,OFA/iLRP似乎优选由胎儿和肿瘤细胞表达。因此,该表达模式使OFA/iLRP成为致敏免疫系统来治疗癌症和其他疾病的可能的候选蛋白质。OFA/iLRP的特异性抗体还可用于检测、诊断和治疗已知与OFA/iLRP错误表达相关的疾病。
关于OFA/iLRP抗体的初始工作分为两个单独的领域,癌胚胎抗原方面或层粘连蛋白受体方面。胚胎/胎儿抗原和层粘连蛋白受体的针对OFA/iLRP的单克隆抗体的初始报道发现于同一年[29,30]。针对胚胎或胎儿抗原所开发的抗体在变性条件下与44kDa的蛋白质反应[30]。以前开发的针对层粘连蛋白受体的抗体依据抗体结合位置具有不同的生物学活性[29]。单克隆IgM抗体识别一个具有阻断层粘连蛋白结合的生物学活性的区域。所述单克隆IgM抗体识别的表位是TEDWSAQPATEDWSA[26]。对44kDa OFA的研究表明,IgM单克隆抗体(MAb 115)可用于蛋白质印迹、流式细胞术并可能用于致癌性测试[31]。但是,由于该抗体不是专门针对OFA|OFA二聚化区域设计的,其与OFA/iLRP和LRP都反应[31]。该单克隆抗体用于OFA/iLRP或LRP的免疫组织化学及蛋白质纯化[12,32]。由检测67kDa层粘连蛋白受体并显示层粘连蛋白受体在乳腺癌中表达提高的肽开发了不同的抗体[5,33]。在寻找自身免疫抗体时,若干发表的文章描述了OFA/iLRP抗体的用途[6,9,10,16,19,20,25,27,34-36]。
由于OFA/iLRP与一系列不同的疾病相关,有需要开发诊断及临床抗体应用。至今,仅存在少数利用肽开发靶向抗体的尝试,并且其局限于一篇发表的报道中[5]。发表的报道中使用的区域产生报道为与OFA/iLRP和层粘连蛋白受体都反应的抗体[5,25]。因此,需要开发可单独地或联合其它抗体使用的OFA/iLRP特异性抗体来开发一系列不同的临床、诊断和/或兽医学应用。抗体的成对开发(一种可识别OFA/iLRP,一种可识别OFA/iLRP和LRP)使得可以开发若干测试,其可用于在所有物种中(由于蛋白的保守性质)治疗、诊断OFA/iLRP疾病或充当OFA/iLRP疾病的试剂。
发明概述
本发明一方面提供分离的抗体,其特异性结合癌胚胎抗原/层粘连蛋白受体蛋白(OFA/iLRP)的区域,其中所述区域是免疫原性的。在一个实施方案中,OFA/iLRP的免疫原性区域包括但不限于多肽序列(a)FFREPRLLWTDPR、(b)VTDPRADHQPLTE、(C)YRDPEEIEKEEQ或(d)FPTEDWSAQPATED。在另一个实施方案中,本发明的分离的抗体为多克隆或单克隆抗体,其包括但不限于,单克隆抗体2C6或3G7。在另外一个实施方案中,所述免疫原性区域位于OFA/iLRP的二聚化区域。
通过针对OFA/iLRP的免疫原性区域免疫产生本发明的抗体。所述抗体能够识别全长OFA/iLRP蛋白和用于产生本发明的抗体的OFA/iLRP的免疫原性区域。在一个实施方案中,抗体是OFA/iLRP特异性的。在另一个实施方案中,抗体识别OFA/iLRP和成熟LRP。
本发明的抗体可用于不同的检测OFA/iLRP相关蛋白质或癌症的方法中。
在一个实施方案中,抗体用于检测样品中OFA/iLRP的方法。该方法包括:
(a)使样品与第一和第二抗体接触,所述第一和第二抗体分别特异性结合癌胚胎抗原/未成熟的层粘连蛋白的区域,其中至少一种抗体对OFA/iLRP具有特异性;
(b)使抗体与OFA/iLRP结合并与OFA/iLRP形成夹心结构;和
(c)用OFA/iLRP特异性抗体检测样品中OFA/iLRP的表达。
在一个实施方案中,抗体的一种可以与OFA/iLRP和成熟LRP结合并作为捕获抗体。另一种抗体可以对OFA/iLRP具有特异性的并作为检测抗体。
在另一个实施方案中,本发明的抗体用于在样品中检测癌症的方法。该方法包括:
(a)使样品与OFA/iLRP特异性抗体接触;
(b)使样品与生物素标记的第二抗体接触;和
(c)用链霉抗生物素蛋白检测样品中的OFA/iLRP,
其中在样品中检测到OFA/iLRP代表存在癌症。
在另外的实施方案中,本发明提供测定样品中OFA/iLRP的量的方法,其包括:
(a)使OFA/iLRP特异性抗体与荧光团缀合;
(b)使缀合的抗体与样品接触;和
(c)用荧光偏振测定样品中OFA/iLRP的量。
本发明的抗体还可用于测定血液样品中OFA/iLRP阳性癌细胞的量的方法。该方法包括:

(b)用流式细胞术测定样品中OFA/iLRP的量。
本发明的抗体还可用于治疗OFA/iLRP阳性癌症的方法。该方法包括给患有OFA/iLRP阳性癌症的对象施用足够改善癌症相关症状的量的OFA/iLRP特异性抗体。在一个实施方案中,抗体连接于具有抗癌特性的胶体,或与化疗剂或蛋白质缀合。
本发明还提供了在对象中检测OFA/iLRP阳性癌症的方法。该方法包括:
(a)使OFA/iLRP特异性抗体与不透射线的染料缀合;
(b)给对象施用缀合的抗体;和
(c)用X射线检测缀合的抗体,
其中用X射线上检测到OFA/iLRP代表存在癌症。
本发明另一方面提供了包含本发明的抗体的药物组合物。所述组合物可以包括药物学载体。

发明详述
本发明令人惊奇地发现,免疫原性肽可以源自假定的OFA/iLRP表位区域,并用于产生以前未发现的针对OFA/iLRP特定区域的抗体。
因此,本发明一方面涉及特异性结合癌胚胎抗原/层粘连蛋白受体(OFA/iLRP)的区域的分离的抗体,其中所述区域是免疫原性的。为了本发明的目的,“分离的”抗体是已从其天然环境的组合物中鉴定并分离和/或回收的抗体。
在本发明的一个实施方案中,抗体对OFA/iLRP具有特异性。如果在众所周知的标准抗体结合条件下,抗体优选结合于OFA/iLRP而不是67kDa形式的层粘连蛋白受体,则所述抗体对OFA/iLRP具有特异性。这样的肽的实例包括但不限于3G7。在一个不同的实施方案中,本发明的抗体可识别OFA/iLRP和67kDa形式的层粘连蛋白受体。这样的抗体的实施例包括但不限于2C6。在另外的实施方案中,本发明的抗体结合OFA/iLRP的二聚化区域。为了本发明的目的,所述二聚化区域是OFA/iLRP全长蛋白质的第112到140位氨基酸间的区域,其涉及OFA/iLRP的二聚化[28]。
本发明的抗体可识别全长OFA/iLRP蛋白质。它们也可以识别免疫原性的OFA/iLRP特定区域,特别是二聚化区域。为了本发明的目的,如果源自OFA/iLRP的区域的多肽可引发免疫应答并可用于产生本发明的抗体,则该区域是免疫原性的。例如,表2中列出的四种免疫原性肽产生的抗体可识别全长OFA/iLRP蛋白、它们各自的免疫原性肽和包含这些肽的OFA/iLRP区域。
为了本发明的目的,“抗体”是能够特异性结合于靶(如受体、碳水化合物、多核苷酸、脂质、多肽等)的免疫球蛋白分子,其通过至少一个位于免疫球蛋白分子可变区的抗原识别位点。如本文中所用的,该术语不仅包括完整的多克隆或单克隆抗体,还包括其片段(如Fab、Fab′、F(ab′)2、Fv)、单链(ScFv)、其突变体、包含抗体部分的融合蛋白质和包含抗原识别位点的免疫球蛋白分子的任何其它修改结构。抗体包括任何类别的抗体(如IgG、IgA或IgM(或其亚类)),以及抗体不需属于任何特定类别。根据其重链恒定结构域的抗体氨基酸序列,免疫球蛋白可分配至不同类别。有五个主要的免疫球蛋白类别:IgA、IgD、IgE、IgG和IgM,并且其中若干个可进一步分为亚类(同种型)如,IgGl、IgG2、IgG3、IgG4、IgAl和IgA2。对应于不同类别免疫球蛋白的重链恒定结构域分别叫做α、δ、ε、γ和μ。不同类别免疫球蛋白的亚基结构和三维构象是众所周知的。
本发明的抗体还旨在包括双特异性、多特异性、单链和嵌合和人源化的分子,其具有由至少一个该抗体的OFA/iLRP区域所赋予的多肽亲和力。本发明的抗体还包括单域抗体,其为抗体重链的可变区或为抗体轻链的可变区。制造包含抗体重链可变区或抗体轻链可变区,含有来自抗体六个天然出现的互补决定区中的三个的结构域抗体的方法也是本领域已知的。见,例如,Muyldermans,Rev.Mol.Biotechnol.,74:277-302,2001。
在一个实施方案中,针对OFA/iLRP开发的抗体是单克隆抗体。单克隆抗体,如2C6和3G7,也可识别全长OFA/iLRP蛋白、其各自的免疫原性肽和包含这些肽的OFA/iLRP区域。然而,单克隆抗体3G7不识别LRP的成熟形式(二聚体形式),而单克隆抗体2C6识别。但是,高剂量的3G7抗体可能破坏OFA/iLRP二聚体。
如本文中所用,术语“单克隆抗体”指基本上均质的抗体,即群体中的个体抗体都是相同的,除了可少量出现的可能天然发生的突变之外。单克隆抗体通常是高度特异地针对单个抗原位点的。此外,与多克隆抗体制备物(其一般包括针对不同决定簇(表位)的不同抗体)相比,每种单克隆抗体针对抗原上的单个决定簇。修饰语“单克隆”表明抗体的特征为从基本上均质的抗体群中所获得,并且不可解释为要求通过任何特定方法产生抗体。例如,依照本发明待使用的单克隆抗体可通过如在美国专利号4,816,567中所描述的重组DNA方法制造。单克隆抗体还可从噬菌体文库中分离,例如,所述噬菌体文库使用McCafferty等人,1990,Nature,348:552-554中描述的技术产生。
本发明的抗体基于选定的标准通过针对肽进行免疫而产生。肽的选择和选择标准在名为“用于癌症治疗的致敏树突状细胞的癌胚胎抗原/未成熟层粘连蛋白受体肽”的专利申请中完全公开,其与本申请同时提交,其中的有关内容完全并入本文。总的来说,包括OFA/iLRP氨基酸序列的至少一部分(即全部或部分)多肽或肽可用作抗原,只要其可诱导特异性的抗体应答,其中所产生的抗体可识别全长OFA/iLRP和本文中所用的免疫原性肽。根据本发明的实施方案,免疫原性肽基于OFA/iLRP不同的假定表位区域产生。为了提高可重复性和针对OFA/iLRP的多功能抗体单独地或联合针对OFA/iLRP二聚化区域的抗体使用的机会,优选地针对表1中列出的肽或表位而开发抗体。在一个实施方案中,下列四种肽用于产生本发明的抗体:1)FREPRLLWTDPR,2)VTDPRADHQPLTE,3)YRDPEEIEKEEQ,4)FPTEDWSAQPATED。
本发明的肽可通过化学合成或使用大肠杆菌等的生化合成制备。本领域技术人员熟知的方法可用于合成。
当本发明的肽为化学合成时,可应用肽合成领域众所周知的方法。例如,可例举这样的方法如叠氮化物法、酸氯法、酸酐法、混合酸酐法、DCC法、活性酯法、羰基二咪唑法和氧化还原法。可使用固相合成或液相合成。也可使用商业化的肽合成仪(如Shimadzu PSSM-8)。
反应之后,本发明的肽可通过组合常规纯化方法(如溶剂萃取、蒸馏、柱层析、液相色谱或重结晶)纯化。
可修饰本发明的肽以提高其免疫原性应答。在一个实施方案中,肽的改变可包括在任何一端的半胱氨酸,其使得可以与匙孔槭血蓝蛋白、卵清蛋白、血清蛋白或其它用于提高肽免疫原性应答的缀合物缀合。表2列出了用于缀合的修饰的肽的实例。
所述肽可用于在一系列不同的生物体中产生抗体。基于众所周知的蛋白质技术,还可以克隆抗体以产生一系列不同的重组抗体。本发明的目的之一是要通过针对本发明的肽进行免疫而不是针对全长蛋白质进行免疫来开发针对OFA/iLRP特定区域的抗体,并基于其对OFA/iLRP特定区域的免疫特异性选择抗体。
本发明的单克隆抗体可使用本领域已知的方法产生。例如,它们可通过培养杂交瘤细胞产生,并且由杂交瘤细胞分泌的抗体可进一步分离或纯化。抗体可从培养基或腹水中分离或纯化,其通过常规免疫球蛋白纯化步骤,例如,蛋白A-Sepharose.RTM.、羟基磷灰石层析、凝胶电泳、透析或亲和层析。
本发明的抗体还可通过重组DNA方法制造,如在美国专利号4,816,567和6,331,415中所描述的,其通过引用并入本文,例如,编码本发明单克隆抗体的DNA可轻易地使用常规步骤分离并测序(如,通过使用能特异性结合于编码小鼠抗体重链和轻链的基因的寡核苷酸探针)。本发明的杂交瘤细胞是这样的DNA的优选来源。分离后,DNA可置于表达载体中,随后转染入宿主细胞如猴COS细胞、中国仓鼠卵巢(CHO)细胞或骨髓瘤细胞(不另外产生免疫球蛋白)中,在重组宿主细胞中获得单克隆抗体的合成。所述DNA还可以被修饰,例如,通过用人重链和轻链恒定结构域的编码序列取代同源小鼠序列(美国专利号4,816,567)或通过将非免疫球蛋白多肽的编码序列的全部或部分共价连接到免疫球蛋白编码序列。这样的非免疫球蛋白多肽可取代本发明抗体的恒定结构域,或可取代本发明抗体一个抗原结合位点的可变结构域以产生嵌合二价抗体。
本发明的抗体可用作一系列不同的功能。在一个实施方案中,本发明的抗体可通过与化学的、放射性的和核佐剂缀合而用于检测恶性组织。此外,这些抗体可用作治疗以提供被动免疫、提高的免疫应答以帮助靶向破坏恶性细胞,以其天然的或改变的形式或缀合:化学物质、放射性“种子”源、微电子装置、胶体银、胶体金、二氧化钛胶体、生物聚合物胶体(即淀粉、胶原蛋白、琼脂糖等)、肽序列或蛋白质序列。由于OFA/iLRP高度保守的性质,所述抗体也可用作药剂并应与所有表达该蛋白质的生物体交叉反应。
因此,本发明另一方面提供了包含本文中描述的抗体或多肽的药物组合物。组合物的抗体或多肽可单独地使用或缀合于化学物质、放射性的“种子”源、微电子装置、胶体银、金、二氧化钛胶体、生物聚合物胶体(即淀粉、胶原蛋白、琼脂糖等)、肽序列或蛋白质序列,其可帮助靶向破坏恶性细胞。
该组合物还可以包含药物学可接受的载体或赋形剂。药物学可接受的赋形剂为本领域已知的,为有利于药物学有效的物质施用的相对惰性的物质。例如,赋形剂可提供形状或稠度或起稀释剂的作用。合适的赋形剂包括但不限于,稳定剂、润湿和乳化剂、用于不同渗透压的盐、封装剂、缓冲液和皮肤穿透增强剂。赋形剂和用于胃肠外及非胃肠外药物输送的制剂阐述于Remington,The Science and Practice of Pharmacy 20th Ed.MackPublishing(2000)。
本发明的组合物配制成与其预定施用途径兼容。施用途径的实例包括胃肠外,如静脉、皮内、皮下,口服(如吸入),透皮(局部),穿粘膜和直肠施用。用于胃肠外、皮内或皮下应用的溶液或悬浮液可包含以下组分:无菌稀释剂如注射用水、盐水溶液、不挥发性油、聚乙二醇、甘油、丙二醇或其它合成溶剂;抗菌剂,如苯甲醇或对羟基苯甲酸甲酯;抗氧化剂,如抗坏血酸或亚硫酸氢钠;螯合剂,如乙二胺四乙酸;缓冲液,如醋酸盐、柠檬酸盐或磷酸盐;以及调整张力的物质,如氯化钠或右旋糖。可用酸或碱如盐酸或氢氧化钠调节pH。胃肠外制备物可封闭在安瓿瓶、一次性注射器或玻璃或塑料制成的多剂量小瓶中。
适合于注射用途的组合物包括无菌水溶液(如果可溶于水)或分散体和用于即时制备无菌注射溶液或分散体的无菌粉末。对静脉施用,合适的载体包括生理盐水、无菌水、Cremophor ELTM(BASF,Parsippany,NJ)或磷酸盐缓冲盐水(PBS)。在任何情况下,组合物必须是无菌的并应为流体的(达到易于注射的程度)。组合物在生产和储存的条件下应该是稳定的,并且必须防止微生物(如细菌和真菌)的污染作用。载体可以是溶剂或分散介质,其包含如水、乙醇、多元醇(例如甘油、丙二醇、液体聚乙二醇等)和其合适的混合物。通过例如使用涂层(如卵磷脂)、维持需要的颗粒尺寸(就分散体而言)以及使用表面活性剂可保持适当的流动性。微生物作用的预防可通过多种抗细菌剂和抗真菌剂实现,例如,对羟苯甲酸酯类、三氯叔丁醇、酚、抗坏血酸、硫柳汞等等。在许多情况下,优选在组合物中包含等渗剂,例如,糖或多元醇(如甘露醇、山梨醇)或氯化钠。通过在组合物中包含延迟吸收的物质(例如单硬脂酸铝和明胶)可使得注射组合物的吸收延长。
无菌注射溶液可通过在适当的溶剂中以所需的量引入化合物及一种或组合的上述列举成分而制备,根据需要,随后过滤消毒。一般来说,分散体可通过引入化合物到无菌容器中制备,其含有基本分散介质和所需的上述例举的其它成分。就用于制备无菌注射溶液的无菌粉末来说,优选的制备方法是真空干燥和冷冻干燥,其从预先无菌过滤的溶液产生活性成分加上任意额外的所需成分的粉末。
口服组合物一般包含惰性稀释剂或可食用的载体。为口服治疗施用的目的,组合物可掺入赋形剂并以片剂、锭剂或胶囊(如明胶胶囊)的形式使用。口服组合物也可以用流体载体制备,用作洗口药。可包含药物学兼容的结合剂和/或佐剂物质作为组合物的一部分。片剂、丸剂、胶囊、锭剂等可包含任意下述成分,或具类似性质的化合物:粘合剂,如微晶纤维素、黄芪胶或明胶;赋形剂,如淀粉或乳糖;崩解剂,如褐藻酸、Primogel或玉米淀粉;润滑剂,如硬脂酸镁或Sterotes;助流剂,如胶体二氧化硅;甜味剂,如蔗糖或糖精;或调味剂,如薄荷、水杨酸甲酯或柑橘香精。
对吸入施用,组合物以气雾喷雾的形式从包含合适的推进剂(例如,气体如二氧化碳)的压力容器或分配器或从喷雾器输送。
全身施用也可通过穿粘膜或透皮方法。为穿粘膜或透皮施用,在配方中使用了适合于待穿透的屏障的渗透剂。这样的渗透剂在本领域一般是已知的,并包括,例如对于穿粘膜施用而言,去污剂、胆盐、夫西地酸衍生物。穿粘膜施用可通过使用鼻腔喷雾或栓剂实现。对透皮施用,化合物配制成本领域一般已知的软膏、油膏、凝胶或乳膏。
本发明的制备物还可以制备为用于直肠输送的栓剂(例如,与常规栓剂基质如可可脂和其它甘油脂类一起)或保留灌肠的形式。
在一个实施方案中,组合物用保护化合物不会从身体迅速消除的载体制备,如控释制剂,包括植入物和微胶囊输送系统。可使用可生物降解的、生物兼容的聚合物,如乙烯醋酸乙烯酯、聚酸酐、聚乙醇酸、胶原蛋白、聚原酸酯和聚乳酸。用于制备这样的制剂的方法将会是本领域技术人员所熟悉的。所述物质还可以从Alza Corporation和NovaPharmaceuticals,Inc.商购获得。脂质体悬浮物(包含靶向被感染细胞的带有针对病毒抗原的单克隆抗体的脂质体)也可以用作药物学可接受的载体。
以易于施用和剂量一致的剂量单位形式配制口服或胃肠外组合物是有益的。本文中所用的“剂量单位形式”指物理上离散的单位,其适合作为用于待治疗的对象的单位剂量,每个单位含有经过计算以产生预期的治疗效果的预定量的活性化合物以及所需的药物学载体。
本发明的组合物,可包含在容器、包装或分配器中,与施用指南一起形成包装好的产品。其它活性化合物也可以并入组合物中。
本发明另外的方面提供了用本发明的抗体和多肽检测、诊断及监测与OFA/iLRP表位表达(相对于正常样本增加或减少,和/或不适当的表达,如出现在通常缺乏所述表位表达的组织和/或细胞中)相关的疾病、紊乱或病变的方法。
在一些实施方案中,该方法包括检测样品中表位的表达,所述样品获得自怀疑患有癌症或任何OFA/iLRP相关疾病的对象。优选地,检测方法包括在使得抗体与OFA/iLRP的表位区域结合的条件下使样品与本发明的抗体或多肽接触,并测定结合的水平是否不同于对照或比较样品。该方法也有助于确定本文中描述的抗体或多肽是否是对患者适当的治疗。
为了诊断的目的,多肽(包括抗体)可以用可检测的部分标记,所述可检测的部分包括但不限于,放射性同位素、荧光标记和多种本领域已知的酶-底物标记。使标记与抗体缀合的方法为本领域已知的。
在一些实施方案中,本发明的多肽(包括抗体)不需要被标记,其存在可用已标记的抗体(其可结合于本发明的抗体)检测。
本发明的抗体可在任何已知的测定方法中采用,如竞争性结合测定、直接和间接夹心测定和免疫沉淀测定。Zola,Monoclonal Antibodies:AManual of Techniques,pp.147-158(CRC Press,Inc.1987)。
所述抗体和多肽还可用于体内诊断测定,如体内成像。一般来说,抗体或多肽用放射性核素(如111In、99Tc、14C、131I、125I或3H)标记,因此可用免疫闪烁成像术定位感兴趣的细胞或组织。
抗体还可用作病理学染色试剂,其使用本领域众所周知的技术。
本发明将通过参考以下详细的实施方案和实施例进一步描述。提供这些实施方案和实施例仅用于说明的目的,而不是为了限制本文中所述发明的范围。
针对OFA/iLRP特定区域的抗体的开发
使用一系列不同的肽制造针对OFA/iLRP的抗体,所述肽针对一系列不同的OFA/iLRP假定表位区域。表1提供了这样的肽的列表。遵循标准方案使所述肽与匙孔槭血蓝蛋白(KLH)缀合[37-39]。该KLH-OFA/iLRP肽用于遵循标准方案免疫小鼠[37,39]。针对KLH-OFA/iLRP肽的第三次免疫后,使用间接ELISA技术使用标准方案,针对牛血清白蛋白(BSA)缀合的OFA/iLRP肽对分离自小鼠尾部采血样品的血清进行筛选(图1和表1)。简言之,OFA/iLRP在Nunc Star Immunosorp平板上4℃包被过夜。所述平板用水洗涤并封闭3次。通过尾部采血获得的小鼠血清在含BSA的PBS-t中稀释并室温温育1小时。洗涤平板并用生物素标记的二抗使用标准方法检测抗体抗原反应(图1A-B)。在Beckman DTX 880上于A620读取吸光度并用Prism Graph 5作图(图1C)。误差棒(bar)显示每种免疫肽的来自两个动物的两个独立的实验的平均。阳性对照用BSA和BAS抗体包被。阴性对照没有蛋白包被。
除了筛选肽,还针对使用标准方案分离的OFA/iLRP测试了尾部采血样品[1-3,5,26,29,30]。简言之,将全长OFA/iLRP包被于Nunc-ImmunoStar MaxiSorp平板并在含有叠氮化钠的PBS中稀释至5微克/毫升,37℃2小时。平板在18.2兆欧的水中洗涤三次并使用含5%干奶粉的PBS-t于25℃封闭1小时。平板用PBS-t洗涤3次。来自尾部采血样品的血清在(上述的)封闭缓冲液中稀释并4℃温育过夜。平板如上述用PBS-t洗涤。用稀释于封闭液的生物素羊抗鼠抗体检测抗体抗原复合物,在25℃温育1小时。平板用PBS-t洗涤三次。使用稀释于封闭液的链霉抗生物素蛋白-辣根过氧化物酶(HRP)检测二抗,在25℃温育30分钟。平板如上述用PBS-t洗涤。最终检测步骤遵循标准方案,其使用Sigma FASTTM OPD系统并在SpectraMax384上读取。
用于抗体-夹心ELISA检测OFA/iLRP的抗体
OFA/iLRP的RNA和蛋白质的表达已与更具侵略性的癌症形式相关联[2,3,5,6,8,10,13,15-17,20,21,23,25,26,29,40]。然而,蛋白质的工作主要集中于67kDa形式和免疫组织化学。本发明的目标之一是开发标准的夹心ELISA,其中使用针对不同区域设计的两种OFA/iLRP抗体。OFA/iLRP特异性捕获抗体可用于定量存在的37kDa OFA/iLRP的量,该区域外的捕获抗体可用于定量存在的总OFA/iLRP。
简而言之,抗体-夹心ELISA遵循如下的标准技术。捕获抗体包被于Nunc Star Immunosorb板(或类似的)上,然后在碳酸盐缓冲液中用一系列不同浓度的针对FREPRLLWTDRADHQPLT肽的捕获抗体包被,4℃温育过夜。捕获和检测抗体的最佳浓度用“十字交叉(criss-cross)”的稀释方法确定[39]。平板在水中洗涤三次并用PBS-t洗涤三次。洗涤后,平板用标准封闭缓冲液封闭1小时。封闭后的平板用PBS-t洗涤三次。可溶的蛋白裂解液用标准封闭缓冲液测试,OFA/iLRP室温温育1小时至4℃温育过夜。当完成初始温育后,平板用PBS-t洗3次。为检测OFA/iLRP捕获抗体的相互作用,二级/检测抗体在封闭缓冲液中稀释,并在室温下温育至少一小时。平板用PBS-t洗涤三次。为检测OFA/iLRP捕获抗体相互作用,二抗/检测抗体稀释于封闭缓冲液中并且室温温育至少一小时。检测抗体与生物素缀合(或其它方法)。用链霉抗生物素蛋白/辣根过氧化物酶(HRP)或等价物检测生物素缀合物。链霉抗生物素蛋白稀释于适当的封闭液并室温温育30分钟。洗涤平板并且遵循标准方案使用SIGMA FASTTM OPD系统检测与链霉抗生物素蛋白酶缀合的HRP。出现的OFA/iLRP的量在SpectraMax 384或等效仪器上读取。使用从纯化的OFA/iLRP生成的标准曲线反算OFA/iLRP的浓度,并遵循标准曲线拟合方法乘上稀释因子[37]。也可能通过使用非特异性捕获抗体检测单体形式的OFA/iLRP,其随后使用针对单体区域的检测抗体。针对二聚化区域外的区域的捕获和检测抗体的组合可用于测定存在于生物流体或组织裂解液中的OFA/iLRP总量。
为检测OFA/iLRP的天然二聚体形式,如上述使用标准ELISA。然而,不使用针对单体区域开发的抗体,所使用的抗体是在该区域外的,但距上述所使用的检测抗体具有足够大的距离以允许正确的结合。遵循标准方案纯化67kDa的OFA/iLRP[2,3,26,29]。使用非变性凝胶电泳或尺寸排阻层析如上述验证大小。进行标准ELISA,其中用针对不同于FREPRLLWTDRADHQPLT的表位的捕获抗体捕获二聚体,然后使用生物素标记的用于单体检测的检测抗体。ELISA遵循标准方案进行[39]。这将测定存在于生物流体和组织裂解液的OFA/iLRP二聚体的量。关于性质、转移负荷和其它临床相关信息的额外信息可通过单体OFA/iLRP与总OFA/iLRP之比而获得。
OFA/iLRP的免疫组织化学检测
上面列出的抗体可用于免疫组织化学(IHC),作为原位检测癌症和可能的癌症的方法。类似的技术已用于鉴定癌症,但是,它们检测67kDa的蛋白质[5,17,41]。IHC方法遵循已定制用于OFA/iLRP检测的标准方案。该方案类似于以下列出的:
遵循用于对组织进行组织学分析的标准方案对切割的组织脱蜡,如二甲苯类,随后递减乙醇浓度,并至少转入I型实验室水。在0.01M的pH值调整至6.0的柠檬酸盐缓冲液中使用热诱导表位修复(HIER),遵循标准方案修复抗原。可以使用直接加热法(加热板)或间接加热法(微波)修复表位[37]。简言之,在>90℃将已脱蜡的组织切片放置于载玻片支持物并放置于装满柠檬酸盐缓冲液的染色盘中。加热组织并保持>90℃ 10约分钟。处理时间取决于组织类型、组织厚度以及一系列其它因素。适当处理后,遵循标准方法将组织在含0.1%吐温20的磷酸盐缓冲盐水(PBS-t)中洗涤3次。组织与标准IHC封闭液(用于减少非特异性抗体/蛋白质相互作用的含BSA或其它试剂的PBS-t)于室温下在潮湿室中温育。载玻片在PBS-t中洗涤三次。OFA/iLRP抗体在含BSA或其等价物的PBS-t中适当地稀释并与处理的组织一起温育。载玻片如上述洗涤三次(3×PBS-t)。组织与生物素标记的通用二体(或等价物)温育以使得用基于链霉抗生物素蛋白的系统进行检测。如上述洗涤载玻片并且用标准链霉抗生物素蛋白检测OFA/iLRAP|b|2°抗体复合物,所述链霉抗生物素蛋白缀合于:碱性磷酸酶(AP)、辣根过氧化物酶(HRP)、量子点、荧光团、放射性核苷酸或其它检测方法[37,39,42]。遵循标准方案使组织显影。
OFA/iLRP抗体用于荧光偏振定量的用途
所述抗体可用于荧光偏振(FP)实验以确定存在的OFA/iLRP的量。此外,单体对比二聚体的FP比例是可能的。FP也可用于确定细胞表面OFA/iLRP的移动性。抗体在荧光偏振实验中的用途的确定是基于以前的工作,但却是专门为OFA/iLRP定量而开发的[3-49]。为确定OFA/iLRP抗体的用途,使单克隆抗体与标准荧光团缀合,并且被标记的抗体可遵循SM-2生物珠方案从游离标记中移除[50]。纯化后,蛋白质浓度和荧光掺入可确定为蛋白与荧光团之比。缀合后,该抗体可适当地保存并以后使用。
为测试待用于FP实验的抗体的能力,所述抗体可以适当量混合,其类似于常用于ELISA技术的抗体浓度的确定[39]。简言之,使用了设计用于荧光的96孔板,其沿一轴具有两倍稀释的OFA/iLRP蛋白量并沿另一轴有两倍稀释的标记抗体量。稀释在标准FP缓冲液中进行,并可能含有一系列不同的添加剂以防止非特异性结合。典型的缓冲液可以包含:磷酸盐缓冲盐水、NP40、牛血清白蛋白、牛丙种球蛋白、甘油或其它试剂以降低背景噪音并提高特异性。一旦混合,平板放置于Beckman DTX880中,并使用其包含的FP(荧光素)过滤器,设定读取时间为每孔约1秒或其它,取决于所使用的荧光基团。偏振值以毫偏振单位(mP)测量,其使用以下公式计算:mP=[(Is-IsB)-(Ip-IpB)]/[(Is-IsB)+(Ip-IpB)]×1000[47-49]。初始实验基于抗体和OFA/iLRP的浓度生成剂量应答曲线。之后,过量的未标记抗体可用作竞争物来确定特异性和均衡性。此外,其生成的数据可用于反算结合分析图(即Scatchard或类似的),而且反过来,用作确定抗体效率的方法。确定初始条件后,FP方法可用于计算存在于不同的生物流体和组织裂解液中的OFA/iLRP量。
在流式细胞术分析中分离/分隔OFA/iLRP阳性细胞或测定血液中OFA/iLRP阳性癌细胞负荷的用途
为确定血液循环型癌症的总癌负荷,或为确定转移负荷,可使用流式细胞术。简言之,获得细胞悬浮液或外周血。细胞悬浮液于300×g离心8至10分钟,并弃去上清液。细胞在流式细胞术染色缓冲液(不含酚红的HBBS,0.1%叠氮化钠和1%牛血清白蛋白)中洗涤一次并以大约2×107细胞/ml重悬,或将50至100μl的细胞悬浮液置于96孔圆底板或其等价物的底部。向细胞悬浮液加入稀释于染色缓冲液的,荧光素异硫氰酸盐(FITC)(或其等价物)标记的OFA/iLRP抗体。作为背景对照,不用任何抗体、用未标记的同种型对照抗体或其等价物(过量的未标记抗体或竞争肽)对细胞进行测试。细胞悬浮液和适当稀释的抗体于冰上或4℃温和混合温育20到60分钟。已染色的细胞悬浮液用100μl染色缓冲洗涤三次,并于300至500×g离心3到10分钟。最终洗涤后,细胞悬浮于约400μl染色缓冲液中并储存于黑暗中,覆盖于4℃直至被分析。额外的步骤可能包括用碘化丙啶复染来检测死亡细胞或添加固定步骤。根据制造商说明遵照标准方案[30,39,51]收集和分析数据。预期直接缀合FITC或使用生物素/链霉抗生物素蛋白-FITC的检测可用于检测外周血、细胞悬浮液或其它来源中的OFA/iLRP阳性细胞数目。OFA/iLRP阳性细胞的整体数目可提供对OFA/iLRP阳性疾病的攻击性和/或进展的深入了解。
本方法另外的应用也可以通过分析前哨淋巴结而用于确定乳腺癌的转移负荷。简言之,遵循标准方案在5ml染色缓冲液中温和地破坏前哨淋巴结(或任何接近初始癌病变的淋巴结)。通过100μm网孔过滤细胞浆,并如上洗涤约两次。获得的细胞悬浮液用于定量局限性转移。
抗体可置换OFA/iLRP或结合于OFA/iLRP的其它蛋白质
FREPRLLWTDRADHQPLT抗体的位置和设计提供了抗体可置换相互作用的OFA/iLRP蛋白质的可能性。此外,针对其它区域设计的抗体可用于置换相互作用的蛋白质或预防它们结合。最初集中于置换或预防OFA/iLRP同源二聚体相互作用。
为测试抗体可以置换相互作用的OFA/iLRP分子的可能性,进行了两个独立的实验。首先,遵循纯化OFA/iLRP二聚体形式的标准方案纯化OFA/iLRP二聚体。二聚体形式的蛋白质与浓度递增的针对OFA/iLRP的抗体一起温育。在标准抗体结合缓冲液(含NP-40、吐温20、牛血清白蛋白、BGG或其他添加剂的PBS)中,37℃温育1小时后,通过抗体的二聚体向单体形式的转换通过遵循标准方案进行非变性聚丙烯酰胺凝胶测定[37,38]。
第二种可用的方法利用了依赖分子大小的荧光偏振假定的变化。在二聚体存在时,主要的FP是源自抗体自身旋转。如果抗体置换了二聚体,存在向单体的转变而荧光偏振应该改变。
由于邻近,荧光团还可以用于猝灭或修改发射光谱。可用两或多个抗体来修改发射光谱或可用猝灭来确定OFA/iLRP分子的状态和/或稳定性。
抗体用作癌症治疗
设计为靶向OFA/iLRP序列的抗体可用作治疗或可能的预防OFA/iLRP阳性疾病的形式。例如,可进行预防或治疗OFA/iLRP阳性癌症的测试,其使用类似的动物模型,使用标准的细胞系。为测试OFA/iLRP癌症的预防,抗体可先于癌症攻击(challenge)而注射入啮齿动物(小鼠或大鼠)。为测试治疗,OFA/iLRP抗体可在癌症攻击后注射。癌症攻击是一系列不同的已知癌细胞系,一些是粘附的而一些是非粘附的。培养细胞并将其导入啮齿动物模型系统。可以通过一系列不同的方法定量OFA/iLRP阳性癌症细胞的量。此外,通过尾静脉注射粘附细胞将会获得增加的肺部定殖。OFA/iLRP抗体的治疗和/或预防能力可以通过治疗与未经治疗的动物肺部存在的癌细胞数目的比值计算。额外的对照可以是与OFA/iLRP抗体相同的时间表注射非特异性同种型对照抗体。此外,OFA/iLRP抗体增强目前的癌症治疗。为减少可能的过敏反应,所述抗体在用于人体之前可被克隆并遵循标准方案进行人源化。
抗体能够改变生物学功能,导致改变的由表达OFA/iLRP的相关疾病引起
的影响
抗体可能改变整体侵袭力或结合于标准化底物的能力。可遵循标准方案测试标准细胞系。为确定对哺乳动物和非哺乳动物细胞可能的药理学影响而进行的一种测试是对生长速率的影响,例如,细胞在有或无一系列不同浓度的肽时的生长,以及测量对细胞凋亡、细胞坏死和细胞增殖的影响。OFA/iLRP阳性癌细胞可体外生长于具有一系列剂量的肽的一系列不同的基底膜上。肽的效果可由一系列不同的方法遵循标准方法测量,包括但不限于,DNA梯形条带、细胞死亡检测ELISA、半胱天冬蛋白酶测量、TUNEL测定、Annexin-V膜改变、DNA染色、FAS、P53、细胞毒性测定、细胞增殖和细胞活力。
所述肽可能拥有提高或降低OFA/iLRP阳性癌细胞侵袭力的能力。这可通过使用与若干涉及其它蛋白质的研究类似的,修改的Boyden-chamber,用或不用一系列不同浓度所述肽培养OFA/iLRP阳性细胞来测量[52-55]。所述肽还可能影响细胞粘附并可使用标准方法测量。粘附培养的OFA/iLRP阳性癌细胞在不同的细胞外基质蛋白(ECM)存在下和所述肽一起培养。然后遵循标准方法测定细胞以确定所述肽存在时细胞系的相对附着[56-59]。若干其它常用技术可应用于测定OFA/iLRP对细胞活力、细胞增殖、细胞死亡和凋亡的影响[37-39,42]。
抗体连接胶体以拥有抗癌活性或提供体内癌症标记物
抗体可附着或交联于一系列不同的胶体。例如,金或银胶体可用于通过共振光散射或替代方法鉴定OFA/iLRP阳性细胞。胶体另外的用途可能是可诱导氧自由基或类似物。连接于抗体的胶体剂提高所述可能性,并且OFA/iLRP阳性癌症细胞暴露于自由基。
抗体连接不透X射线的染料或类似类型的缀合物以和X射线一起使用
抗体与不透X射线的染料或类似物缀合[60]。及早地并准确地检测癌变状态的能力可以对患者治疗选择有帮助。有可能将抗体缀合于不透射线的染料或其等价物以提高目前基于X射线的技术的效率。此外,所述抗体可以连接于可以被“调”至特定共振频率以通过MRI来检测的分子。可能的缀合物包括但不限于基于钆的化学反应性系统,其可附着于与用于缀合荧光染料的抗体类似的抗体上。常用的技术是添加与抗体上的伯胺反应的四氟苯基(TFP)酯部分。可以使用尺寸排阻色谱去除未反应的TFP钆。为了在人类中临床使用,所述抗体可能需要人源化。上面描述的方法将遵循标准方案。
作为概念验证,动物模型不需要拥有活性免疫系统,其使得可以使用标准技术[61]。初始的工作并不需要使用X射线,但需要证明OFA/iLRP抗体在体内的结合能力。抗体或其等价物缀合于荧光团、胶体、酶缀合物或其它可容易检测的方法。缀合的抗体以一系列不同剂量在限定的时间点注射入动物,可对被诱导的肿瘤进行取样以寻找抗体的结合。这种结合可表明其可用于靶向肿瘤而用于诊断成像。其它方法包括使用纳米金颗粒,其可用环氧硅烷衍生物或其类似物包被以共价连接于抗体[62]。
抗体连接和缀合化疗剂并且被引导至OFA/iLRP阳性的癌细胞
为了减少与经典化疗相关的副作用数目,可采取靶向的方法。OFA/iLRP抗体用作抗癌药物靶向剂的用途可能会增加目前的治疗方法的安全性和效力。为了测试其效果并作为概念验证,可以将抗体掺入脂质体类型系统,其中脂质体含有化疗剂。为显示靶向能力,OFA/iLRP细胞、OFA/iLRP癌细胞和正常人体细胞的混合物可在细胞培养皿中混合。可添加所述混合物并与用类似剂量的化疗剂或脂质体(掺入同种型对照抗体)的细胞处理进行比较。预期靶向的方法将具有更高的OFA/iLRP阳性细胞死亡率,而对正常细胞没有剧烈的影响。所述影响可以通过一系列不同的标准技术确定。尽管可使用标准的化疗剂,另一种技术可以利用放射性分子对OFA/iLRP抗体的附着。例如,抗体遵循标准方案碘化。
除了直接连接于化疗剂、产生具有抗体物质的脂质体或浸泡于化疗剂的聚丙烯酰胺/琼脂糖珠,抗体还可附着于可选的输送方法。可能是可携带化疗剂的碳纳米管或其等价物。当与OFA/iLRP阳性细胞相互作用时,可提供信号以引起纳米管共振并释放所含的抗癌药物。触发可以为一系列不同的方法。所述抗体可以连接和缀合于纳米碳、纳米金和其它纳米管和纳米剂,并用微电子/无线电敏感装置或类似装置处理以靶向OFA/iLRP-阳性癌细胞。当活性接头分子直接连接抗体和抗癌剂时,活性接头分子被靶向而释放抗癌药物。这样的实例可以是抗体和抗癌药物之间的紫外线或X射线可裂解的接头。X射线存在时(放射治疗),可以裂解所述接头并释放抗癌剂。另一种方法使用带有蛋白酶裂解位点的肽接头,其在肿瘤微环境下将被裂解。
抗体可连接和并缀合于其它可改变靶细胞生物学功能的蛋白质
抗体只能引起某些类型的免疫应答。之前已用过两种不同序列的融合来产生DNA疫苗接种[63]。该分子是与可调节肿瘤或周边区域的生物学活性序列融合的OFA/iLRP抗体结合区域的杂合体。几种可能是克隆人源化的OPA/iLRP抗体活性区域并将其融合至免疫系统调节因子、细胞周期调节因子或诱导凋亡的蛋白质序列。为预防非特异性性事件,蛋白质设计成保持为非活性,直至处于肿瘤微环境中。OFA/iLRP抗体融合于蛋白酶裂解位点接头,然后所述接头附着于凋亡诱导肽或小分子以诱导针对肿瘤的免疫应答。两种不同分子的融合可提供多价抗体,其可以提高的特异性性靶向至癌细胞。
OFA/iLRP抗体与微电子流(microelectrofluidic)或其等价装置在诊断筛选
方法或持续筛选方法中的用途
将OFA/iLRP抗体或活性区域包被于微电子流装置上以测量血流中实时OFA/iLRP表达。该装置是自容式(self-contained)的,作为癌症确认的监测装置。抗体包被于膜或其它微装置上以进行癌症检测。有可能基于几种方法确定结合的量。首先,抗体可包被于有限定通过流量的分子悬臂。由于发生抗原和抗体之间的实时相互作用,在包被的悬臂上应该存在增大的的扭矩。这反过来可以通过电荷感应或阻抗的变化测量。使用了类似的技术,其具有包被有OFA/iLRP抗体的薄膜,并且当与抗原结合时,薄膜上的电荷或相互作用或张力的变化均可被测量并与OFA/iLRP表达的变化关联。这些技术并不限于OFA/iLRP,而是任何癌症分子可用作微型植入装置,其可被医生依需要读取以提醒他们复发性或转移性疾病的进展。
附着于放射性、荧光或等价分子的OFA/iLRP抗体在辅助医生切除肿瘤中的用途
作为在癌症切除期间外科医生的辅助,OFA/iLRP的用途是连接到荧光团、放射性标记物、染料或等价物,使得外科医生可以轻松地鉴定癌生长。此外,该技术可用于快速鉴定可能含有转移性疾病的淋巴结,例如,抗癌抗体连接于适当的缀合物来进行鉴定,如目前放射活性用于前哨淋巴结活组织检查。注射后,患者进行常规步骤的手术。然而,如果难以找到癌症或检查淋巴结,可使用盖革计数器、UV或其他光源,其使得快速鉴定癌区域。肿瘤用适当的手术处理切除。这种方法可极大地帮助鉴定/切除癌症。
抗体用于免疫沉淀的用途
OFA/iLRP抗体用于免疫沉淀以鉴定新的治疗靶。细胞裂解物遵循标准方案进行免疫沉淀[37,39]。获得的蛋白质可用SDS-PAGE、2D凝胶电泳、质谱或其他方法分析。结果信息可以提供对可能的OFA/iLRP相关机制的深入了解,其可用于临床诊断或治疗。
下面的实施例旨在说明而不是限制本发明的范围。虽然这样的实施例是可能使用的实施例中典型的,仍可选择性使用本领域技术人员公知的其它方法。事实上,基于本文中的教导,无需过多实验,本领域普通技术人员可轻易地设想并产生另外的实施方案。
实施例1
制造OFA/iLRP单克隆抗体2C6和3G7的方法
OFA/iLRP单克隆抗体2C6和3G7在Precision Antibody使用标准的方法和方案制造。肽FREPRLLWTDPRC用于产生单克隆抗体3G7。肽YRDPEEIEKEEQC用于产生单克隆抗体2C6。
合成的肽使用标准方案缀合于马来酰亚胺活化的KLH(Pierce/Thermo,Rockfork,IL)。缀合后,KLH-OFA肽用于每种肽免疫两只不同的小鼠。免疫后,通过尾部采血获得血清并且该血清用于筛选针对BSA缀合肽的抗体滴度(图1)。如果针对所述肽为阳性,进行使用重组人OPA/iLRP的间接ELISA验证该抗体会与该蛋白反应。这是为了确保该抗体针对小肽和全长蛋白都反应。在验证了针对肽和蛋白质的反应性后,融合脾细胞与骨髓瘤细胞系并在96孔培养板中选择。保持可存活的集落存活,然后针对肽和OFA/iLRP进行筛选(类似上述)。针对OFA/iLRP但不针对BSA-包被的孔的具高活性的杂交瘤组织培养上清被选择用于免疫球蛋白分类(IgG或IgM)。任何是IgM的克隆从筛选中排除而产生IgG的杂交瘤遵循标准方案培养。为进一步从培养基中纯化抗体,遵循标准方案进行蛋白G选择(Pierce/Thermo,Rockford,IL)。洗脱的抗体浓度用A260吸光度测定并且其用于2C6和3G7抗体生产。
实施例2
针对rHU OFA的活性对尾部采血样品的分析
在本研究中,分析了针对重组全长人OFA/iLRP的活性。Nunc-Immunostar MaxiSorp平板用含叠氮化钠的PBS中的浓度为10μg/ml的OFA/iLRP蛋白包被。平板于37℃温育2小时。用水洗涤平板3次,然后用含5%脱脂奶粉(NFDM)的PBS-t封闭至少一小时。平板用PBS-t尾部采血样品洗涤3次,而阳性对照加入5%NFDM。平板于4℃摇动温育过夜。如上述洗涤平板,并在1∶100稀释于5%NFDM的通用二抗中室温温育1小时。洗涤平板并与链霉抗生物素蛋白-HRP(1∶200R&D系统)室温温育30分钟。洗涤平板并每孔加入200μl SIGMA FASTTM OPD。观察平板的反应并且在仍存在低背景时用50μl终止液(2n H2SO4)终止平板。在SpectraMax 384上读取并分析平板,其遵循仪器上的用于基础终点ELISA w/OPD和酸终止的标准方案。
所有针对OFA/iLRP设计的免疫肽(表2)都产生了可与重组全长OFA蛋白反应的尾部采血血清。当比对于以前开发的单克隆抗体(由Coggin、Roher和Barsoum博士提供(阳性对照)),尾部采血血清表现类似或稍好(图2),尽管单克隆抗体为大于2mg/ml的浓度。本实验进一步证明了OFA/iLRP作为捕获抗原在确定患者和诊断样品的抗体滴度中的用途。
结果表明,肽来源的针对OFA/iLRP的抗体能够识别重组全长蛋白。另外,将重组全长OFA包被于平板上可用作开发另外的抗体或监测抗OFA的免疫应答的筛选方法。
实施例3
rOFA/iLRP的间接ELISA测定单克隆抗体的反应性
本项研究中,测定了针对OFA/iLRP特定区域设计的单克隆抗体相对重组OFA/iLRP的单免疫反应性。简言之,用PBS中浓度为2μg/ml的OFA/iLRP蛋白包被Immulon 4HBX平板。平板于4℃温育过夜。用水洗涤平板三次,然后用含1%BSA的PBS-t封闭至少一小时。平板用PBS-t洗涤三次并且沿8孔进行单克隆抗体的5倍稀释系列,其高浓度为1∶10,低浓度为1∶781,250。平板于4℃摇动温育过夜。如上述洗涤平板,并与1∶50,000的生物素标记的抗鼠IgG二抗温育1小时。洗涤平板并在室温下与链霉抗生物素蛋白-HRP(1∶250,000)温育30分钟。洗涤平板并每孔加入100μl TMB双组分HRP微孔底物(BioFX实验室,Owings Mills,MD)。观察平板的反应并且在仍存在低背景时用50μl终止液(2n H2SO4)终止平板。在SpectraMax上读取并分析平板,其遵循仪器上的用于基础终点ELISA w/HRP和TMB的标准方案。
图3中的结果显示了在针对rHu OFA/iLRP的间接ELISA上进行的单克隆抗体2C6和3G7的稀释曲线(n=4)。起始于1∶10并范围降低至1∶781,250的5倍稀释的8点稀释系列显示,单克隆2C6对OFA有强烈的反应,并且在本测定中使用2C6抗体时存在动态检测范围。
图3还显示了针对OFA/iLRP的间接ELISA上进行的单克隆抗体3G7的稀释曲线。与上述相同的8点稀释系列显示,单克隆3G7对OFA有强烈的反应,并且在本测定中使用3G7抗体时存在动态检测范围(n=4)。
该数据表明,这些单克隆抗体(2C6和3G7)都有能力识别并结合全长的OFA/iLRP。该数据表明,工程抗体可用于以特异性的方式检测OFA,其可用于广泛的下游诊断测试。它们还可以用作在一系列生物流体中寻找抗OFA的抗体存在的标准。
实施例4
rOFA/iLRP的夹心ELISA测定单克隆抗体的反应性
用PBS中的浓度10μg/ml的2C6单克隆抗体包被Immulon 4HBX平板。平板于室温温育过夜。用水洗涤平板三次,然后用含1%BSA的PBS-t封闭一小时。用PBS-t洗涤平板三次并且沿8孔进行OFA/iLRP的2倍稀释系列,其高浓度为5000ng/ml,低浓度为78.125ng/ml。于5%BSA的PBS中制备OFA/iLRP标准物以模拟血清浓度。平板于室温摇动温育2小时。如上述洗涤平板,并与生物素标记的3G7单克隆抗体温育2小时。洗涤平板并于室温与链霉抗生物素蛋白-HRP(1∶200)(R&D)温育30分钟。洗涤平板并每孔加入100μl TMB双组分HRP微孔底物(BioFX实验室,Owings Mills,MD)。观察平板的反应并且在仍存在低背景时用50μl终止液(2n H2SO4)终止平板。在SpectraMax上于450nm读取并分析平板,其遵循仪器上的用于基础终点ELISA w/HRP和TMB的标准方案。
图4显示了宽范围的OFA/iLRP浓度标准曲线以及我们的单克隆抗体用夹心ELISA法检测OFA/iLRP的能力。该数据显示,这些单克隆抗体(2C6和3G7)都有能力以使用夹心ELISA可检测的方式识别并结合全长的OFA/iLRP。当反过来时(即,3G7捕获/2C6检测),看到了类似的结果,但反应吸光度显著低于使用2C6作为捕获抗体(数据未显示)。同样,该数据表明,工程抗体可用于以特异性的方式检测OFA,其可用于广泛的下游诊断测试。该类型的ELISA可用于在一系列生物流体及组织裂解液中寻找OFA/iLRP的存在。另外,该技术可用作包含/排除标准,其用于基于OFA/iLRP的治疗或用作癌症诊断。所述抗体为针对蛋白质的保守区域设计的而应与所有表达OFA/iLRP的物种交叉反应。
实施例5
使用肽来源单克隆抗体的癌胚胎抗原未成熟层粘连蛋白受体的荧光偏振
本实验表明OFA/iLRP的2C6克隆在荧光偏振中的可用性。为模拟血清样品,所有标准物稀释于含5%牛血清白蛋白(BSA)的磷酸盐缓冲盐水。该实验部分基于对HSP 90的工作,但是荧光素-5-马来酰亚胺-标记的抗体用于代替格尔德霉素-BODIPY(结合HSP 90的小荧光分子)[49,45]。
OFA/iLRP单克隆抗体2C6和3G7遵循标准方案用荧光素-5-马来酸亚胺(Pierce/Thermo,Rockford,IL)标记。遵循制造商方案使用标准染料去除柱去除未结合的染料。抗体标记后,其用于荧光偏振实验。
OFA/iLRP在含5%BSA的PBS中稀释至40000ng/ml并且将50μl添加到顶部孔,并且在96孔黑色板(Cliniplate,Thermo Scientific)中于含5%BSA的PBS中稀释6倍。高浓度的血清白蛋白(>5%血清白蛋白)用于模拟血清浓度。标记的抗体在含0.1%正常人血浆和0.01%Tween-20的PBS中稀释至20μg/ml。稀释后的抗体溶液(50μl)分装入孔、混合、密封、遮盖避光并在4℃温育过夜。使得平板调整至室温并在DTX-880(Beckman Instruments,Palo Alto,Ca)上测量荧光偏振。使用了制造商荧光方案,除了荧光积分时间为0.0001秒。数据输出至excel并且mP按先前介绍的计算[49]。原始数据使用Prism 5.0(GraphPad软件公司)绘制于对数x轴(图5A),使用单因素ANOVA分析来比较背景与计算的mP值,并且进行转换以进行四参数逻辑斯蒂分析(图5B)(r2=0.975)n=2。
OFA在5%BSA/PBS中的浓度范围为40000至0.85ng/ml,并显示了低于1000ng/ml(由于饱和度)到低于30.86ng/ml(图5A)的范围。在超过1000ng/ml的浓度,信号趋于饱和,而低于30.6ng/ml(5.1ng/ml),信号与背景差异不显著(数据被转换并且进行四参数逻辑逻辑斯蒂分析并且r2>0.95)(图5B)。
结果表明,肽来源的OFA/iLRP抗体可用于使用荧光偏振测定OFA/iLRP浓度。由标准曲线提供的此数据具有可接受的r2值,其可用于计算来自包含OFA/iLRP的血清、血浆、组织裂解液或任何可溶性来源的未知量。
实施例6
OFA/iLRP的免疫组织化学染色
本研究的目的是分析我们的单克隆抗体在福尔马林固定石蜡包埋(FFPE)的,6微米厚的肿瘤切片中检测OFA/iLRP的能力,其在相邻的正常组织中有限反应。
遵循与MaxTag Histo试剂盒一起提供的方案进行实验,其与小鼠一抗(Rockland Immunochemicals,Gilbertsville,PA)一起使用。简而言之,FFPE6微米肿瘤载玻片用二甲苯脱蜡,然后用浓度递减的乙醇(EtOH)复水,然后用水稀释,并最终置于PBS中。所有温育步骤在湿润室中进行。载玻片上含组织的区域用PAP笔标记,然后在1%双氧水和含1%正常山羊血清的PBS中封闭5分钟以去除内源性过氧化物酶的活性。然后载玻片在PBS中洗涤三次5分钟。一抗按1∶250稀释于PBS+1%正常山羊血清中并在载玻片上4℃温育过夜。以前的实验在1∶10和1∶50的一抗稀释液中进行,具有太高的背景染色(数据未显示)。然后载玻片在PBS中洗涤三次五分钟,然后加入试剂盒中提供的二抗,并在室温温育30分钟。载玻片再次洗涤并且加入来自试剂盒的稀释的链霉抗生物素蛋白过氧化物酶试剂并在室温下温育30分钟。载玻片再次洗涤并且加入提供的DAB试剂并温育15分钟,同时监测颜色显影。使DAB反应比制造商推荐多温育10分钟来确定相邻正常组织的真实背景染色。然后载玻片用蒸馏水洗涤三次两分钟。加入苏木精复染液并且载玻片温育五分钟。载玻片再次用水洗涤两分钟三次,用100%乙醇脱水4次,每次2分钟,并然后用4种变化的二甲苯清洗,每次2分钟。在显微镜下观察组织,显现为棕色沉淀并且核染上轻微的蓝色的评估为阳性反应。
对多种肿瘤类型进行染色。图6所示的载玻片是具有代表性的切片,其显示了中度分化的T2N1M0乳腺侵袭性导管癌,其用3G7单克隆抗体作为一抗染色。载玻片的染色范围为很深的黑色沉淀物到类似于在载玻片上的同种型对照(未显示)的很浅。OFA/iLRP的表达已在迄今测试的所有肿瘤类型中见到。
结果表明,单克隆抗体适合于IHC染色方案,并可用于一系列不同的实验和诊断应用。一种潜在的应用是用于OFA/iLRP治疗的适用性或作为筛查/诊断测试。由于OFA/iLRP是专门针对保守区域设计的,这些抗体应对所有的物种交叉反应。
实施例7
OFA/iLRP单克隆抗体对细胞活力的影响
本实验的目的是测定针对OFA/iLRP设计的单克隆抗体是否对细胞活力有任何影响。由于OFA/iLRP的性质,预期设计为破坏OFA|OFA向LR转换的或抑制其它蛋白|蛋白相互作用的单克隆抗体将具有活性。
所有细胞在37℃的湿润室中(Mediatech,Inc.Manassas,VA)生长于含L-谷氨酰胺、100I.U.青霉素、100μg/ml链霉素和10%胎牛血清的RPMI1640。DU-145细胞从美国典型培养物保藏中心(ATCC,Manassas,VA)获得并遵循标准方案在培养基中生长。DU145细胞生长至75到85%密度之间,遵循标准方案收集,并用修改的Neubauer brightline血球计计数,以400,000细胞/ml悬浮。
单克隆抗体(2C6和3G7)溶解于完全培养基。稀释至适当浓度后,分装50μl到96孔测定板中,其用层粘连蛋白/巢蛋白复合物(50μg/ml)包被或为未处理的(透明底黑色)(Corning生物科学,Corning,NY),并分装50μl细胞(20,000细胞/孔),过夜培养。将20μl的CellTiter-蓝((Promega,Madison,WI))加入到细胞,并且在37℃额外温育两小时。遵循标准荧光方案在0.001秒的积分时间下于DTX-880(Beckman公司)上读取细胞。为确定半胱天冬蛋白酶活性是否被诱导,使用ApoOne测定法(Promega,Madison,WI)测定半胱天冬蛋白酶3/7活性。数据输出至excel并随后至Prism 5.0(GraphPad软件公司)中作图,并使用单因素ANOVA来分析其与背景对照(用于肽的稀释剂)之间的统计学差异。为确定对照组(0)与处理之间的差异,进行了Dunnets事后检验。任何p<0.05(*)被视为是显著的。
所有的DU 145细胞培养于未包被或包被有层粘连蛋白/巢蛋白的96孔板上。2C6或3G7存在时,对细胞活力存在影响(图7A和B)。当通过单因素ANOVA进行统计学分析时,尽管在较高浓度下,2C6和3G7抗体引起相比于对照组下降的活力。但是,考虑到3G7(0.32mg/ml)和2C6(1.29mg/ml)之间的浓度差异,3G7似乎有更高的活性。在ApoOne半胱天冬蛋白酶3/7测定中没有看到任何组有显著的差异。
对比与单独的PBS,当DU 145细胞于任一种抗体存在时生长,似乎对细胞活力具有显著的影响。不存在半胱天冬蛋白酶3/7活化而在ApoOne测定中没有见到显著的变化。这表明OFA/iLRP抗体可能具有用作抗癌治疗的能力。
可以在不偏离本发明的精神和范围下,做出如本文前面所阐述的许多修改和变化,并因此只应施加如所附权利要求书所表明的限制。

Claims (24)
1.分离的抗体,其特异性结合癌胚胎抗原/未成熟层粘连蛋白受体蛋白(OFA/iLRP)的区域,其中所述区域是免疫原性的。
2.权利要求1的分离的抗体,其中所述免疫原性区域位于OFA/iLRP的二聚化区域。
3.权利要求1的分离的抗体,其中所述OFA/iLRP的区域包括多肽序列(a)FFREPRLLWTDPR、(b)VTDPRADHQPLTE、(C)YRDPEEIEKEEQ或(d)FPTEDWSAQPATED。
4.权利要求1的分离的抗体,其中所述OFA/iLRP的区域与提高免疫原性应答的肽缀合。
5.权利要求1的分离的抗体,其中所述OFA/iLRP与匙孔槭血蓝蛋白、卵清蛋白或血清白蛋白缀合。
6.权利要求1的分离的抗体,其中所述抗体为单克隆抗体。
7.权利要求6的分离的抗体,其中所述单克隆抗体为2C6或3G7。
8.权利要求6的分离的抗体,其中所述区域包括多肽序列FREPRLLWTDPRC或YRDPEEIEKEEQC。
9.包含权利要求1的抗体的药物组合物。
10.权利要求9的组合物,其中所述抗体为单克隆抗体。

(b)使得所述抗体与OFA/iLRP结合并与OFA/iLRP形成夹心结构;和
(c)用OFA/iLRP特异性抗体检测样品中OFA/iLR的表达。
14.权利要求13的方法,其中所述抗体的至少一种结合OFA/iLRP和成熟LRP。
15.权利要求14的方法,其中识别OFA/iLRP和成熟的LRP的抗体作为捕获抗体,而OFA/iLRP特异性抗体作为检测抗体。
16.权利要求13的方法,其中所述第一抗体是3G7,而所述第二抗体是2C6。
17.检测样品中癌症的方法,其包括:
(a)使样品与OFA/iLRP特异性抗体接触;
(b)使样品与生物素标记的二抗接触;和
(c)用链霉抗生物素蛋白检测样品中的OFA/iLRP,
其中在样品中检测到OFA/iLRP代表存在癌症。
18.测定样品中OFA/iLRP的量的方法,其包括:
(a)使OFA/iLRP特异性抗体与荧光团缀合;
(b)使缀合的抗体与样品接触;和
(c)用荧光偏振测定样品中OFA/iLRP的量。
19.测定血液样品中OFA/iLRP的量的方法,其包括:
(a)使血液样品与OFA/iLRP特异性抗体接触,和
(b)用流式细胞术测定样品中OFA/iLRP的量。
20.治疗OFA/iLRP阳性癌症的方法,其包括以足够治疗癌症的量给患有OFA/iLRP阳性癌症的对象施用OFA/iLRP特异性抗体。
21.权利要求20的方法,其中所述抗体与具有抗癌特性的胶体连接。
22.权利要求19的方法,其中所述抗体与化疗剂缀合。
23.权利要求19的方法,其中所述抗体与蛋白质缀合。
24.检测对象中的OFA/iLRP阳性癌症的方法,其包括:
(a)使OFA/iLRP特异性抗体与不透射线的染料缀合;
