一种西红花酸对AGEs抑制效果的检测方法
技术领域
本发明属于病理检测领域,具体的涉及一种一种西红花酸对AGEs抑制效果的检测方法。
背景技术
糖尿病(DM)血管病变的发生是一个复杂的病理生理过程,白细胞粘附于血管内皮、穿透血管内膜、聚集于血管壁是DM血管病变的早期事件,并惯穿其整个过程。在糖尿病患者体内有高浓度的晚期糖基化产物(AGEs)蓄积,AGEs通过与内皮细胞上晚期糖基化产物受体(RAGE)结合,诱发氧化应激,释放大量活性氧自由基(ROS),ROS又通过氧化敏感机制激活MAPK、NF-κB、PKC等信号传导物,使内皮细胞分泌粘附分子如细胞间粘附分子(ICAM-1)、E-选择素(E-selectin)、细胞粘附分子-1(VCAM-1)等,这些粘附分子介导了单核细胞/中性粒细胞与血管内皮细胞粘附和跨内皮迁移,导致白细胞在血管内皮下募集和滞留,同时,这些细胞还可分泌致炎因子如TNF-α,IL-2等,进一步加重血管内皮的损伤,促进DM血管病变的发生发展。
我们前期研究发现,西红花酸具有抑制晚期糖基化产物(AGEs)的作用,但抑制效果如何却没有一个有效的手段实现。
发明内容
为克服现有技术中的不足,本发明的目的在于提供一种西红花酸对AGEs抑制效果的检测方法。
为解决上述技术问题,本发明采用了以下技术方案:一种西红花酸对AGEs抑制效果的检测方法,包括以下步骤:
步骤一、细胞分离液的配制;
步骤二、晚期糖基化终产物(AGEs)的制备;
步骤三、牛中性粒细胞与单核细胞的分离与纯化;
步骤四、检测西红花酸对单核细胞和中性粒细胞的影响;
步骤五、利用统计学处理西红花酸对单核细胞和中性粒细胞的影响即可得知西红花酸对AGEs的抑制效果。
本发明的检测方法简单、有效的检测出了西红花酸对AGEs抑制效果。
具体实施方式
一种西红花酸对AGEs抑制效果的检测方法,包括以下步骤:
步骤一、细胞分离液的配制备;
所述细胞分离液包括I液和II液,所述I液用10份33.9%的复方泛影葡胺加上24份9%的聚蔗糖制成;所述II液:由10份50%的复方泛影葡胺加上20份9%的聚蔗糖制成。全部溶液用双蒸水配成所需浓度,过滤除菌,4℃保存备用。
步骤二、晚期糖基化终产物(AGEs)的制备;
将5mg/ml BSA与浓度为50mmol/L的d-葡萄糖溶于含EDTA(0.5mmol/L)的PBS(PH7.4)中,混匀,无菌过滤后,置于37℃,CO2培养箱避光孵育3个月后,用透析膜充分透析以去除未结合的葡萄糖,过滤除菌后4℃存放备用。
步骤三、牛中性粒细胞与单核细胞的分离与纯化;
抽取健康小牛全血,肝素抗凝,用PBS等倍稀释混匀。预先在离心管内依次加入细胞分离液II液与I液,使二液界面清晰,将肝素抗凝血缓慢平铺在梯度分离液上。II液、I液、抗凝血之间的比例为3∶3∶4。室温下(25℃)离心,2500rpm,20min;离心后可见到三种不同的细胞层,小心吸取第一层细胞层(单个核细胞)与第二层细胞层(中性粒细胞),分别移入离心管中,纯化细胞。
步骤四、检测西红花酸对单核细胞和中性粒细胞的影响
传至96孔板的EC融合生长成单层时,分为6组,即control组,AGEs模型组(100g/mlAGEs),葛根素阳性对照组,西红花酸(0.01μM,0.1μM,1μM)组,每组8个复孔。不同剂量西红花酸预孵12h,再换培养液,模型组和药物实验组分别加入100g/mlAGEs,作用24h,37℃温育,然后吸出培养液,PBS洗1次,加入新分离的单核细胞(1×105个/孔)或中性粒细胞(2×105个/孔),37℃,5%CO2培养箱继续培养1h,PBS轻洗去未粘附细胞;加入0.25%虎红,1001/孔。室温作用10min,PBS洗去游离虎红,加入PBS-乙醇(1∶1)200 1/孔,室温作用1h后,用酶标仪于570nm处测吸光值(OD)。
步骤五、利用统计学处理西红花酸对单核细胞和中性粒细胞的影响即可得知西红花酸对AGEs的抑制效果。

步骤一、细胞分离液的配制;
步骤二、晚期糖基化终产物(AGEs)的制备;
步骤三、牛中性粒细胞与单核细胞的分离与纯化;
步骤四、检测西红花酸对单核细胞和中性粒细胞的影响;
步骤五、利用统计学处理西红花酸对单核细胞和中性粒细胞的影响即可得知西红花酸对AGEs的抑制效果。
