本发明涉及糖原在制备用于控释葡萄糖的食品制剂中的用途。
特别是,本发明涉及具有平均分子量超过2,000,000道尔顿的糖原在制备用于控释葡萄糖的人工食品制剂中的使用。
本领域状况
包含葡萄糖来源的人工食品制剂是本领域众所周知的。
这些制剂可应用于许多领域。
一个应用领域涉及作为膳食补充剂用于专业或业余运动员的用途。
第二个应用领域涉及当需治疗的个体不能通过正常的胃肠道给药时进行胃肠外给药的用途。
第三个应用领域涉及肠内饲喂的用途,其以液体形式通过插管直接施用入胃或肠。
第四个应用涉及用作能引起饱足感但不提供有效热量摄取的制剂(填充剂)。
本领域已知的人工食品制剂通常通过水解淀粉或其衍生物来获得。淀粉是植物界中分布最广泛的多糖,其包含葡萄糖的聚合物链。淀粉主要包含两种聚合物:直链淀粉(约占重量的20%)和支链淀粉(约占重量的80%)。直链淀粉是一种线性聚合物,其中葡萄糖单元通过α(1→4)糖苷键连接在一起。支链淀粉是分支的聚合物,其具有与直链淀粉结构相似的基链,但是其侧链上每24-30个葡萄糖单位通过α(1→6)键形成分支。
专利申请EP1548033或US 2005/0159329描述了通过酶水解平均分子量在90,000和150,000道尔顿之间的淀粉获得的高度分支的多糖。此外,专利申请EP1369432中描述了平均分子量在3,500和20,000道尔顿之间的相似的多糖。
专利申请EP487187描述了包含低卡路里含量(每100g有160至240Kcal)的麦芽糊精的食品制剂。专利申请EP 514528描述了包含麦芽糊精和β-葡聚糖和/或戊聚糖的可溶性膳食产品。
麦芽糊精是由包含经α(1→4)糖苷键连接在一起的数十个葡萄糖分子的淀粉水解而来的一类物质。取决于淀粉水解的程度(通过化学/物理或酶方式或者通过二者的组合进行),可获得各种类型的麦芽糊精,其区别在于组成它们的葡萄糖分子数目,通常是2至20个单位。基于它们的长度确定右旋糖当量(DE),可以从最小的4-6至最大的36-39。DE值越高,水解程度就越大且链长越短。它们被身体消化的最终结果通常是葡萄糖,但所述过程发生的速率以及随后的能量产生取决于DE值。
含有葡萄糖来源的人工食品制剂包含其它基本营养素,例如用于在例如体力消耗或剧烈的体育活动后大量出汗的那些情形下恢复水/盐平衡。
这些制剂的理想摩尔渗透压浓度应具有与血浆的数值(280-300mOsml/kg)完全相同或稍稍更低的数值,即该能量溶液就血液而言应是低渗的,假定摩尔渗透压浓度影响胃排空速率以及尤其是水和溶解分子的肠吸收。利用麦芽糊精可能产生除了葡萄糖来源外还包含其它营养素的等渗制剂。
包含具有α(1→4)键的低分子量多糖的本领域已知人工食品制剂提供了被迅速吸收和代谢的葡萄糖的快速供给。
这具有两个主要类型的缺点。
第一个缺点在于这些食品制剂不能施用于糖尿病个体,因为它们会导致血液葡萄糖水平快速升高(血糖症)。
第二个缺点在于能量效应不是持久的,除非大量物质被摄取,而由于这些物质的过量摩尔渗透压浓度会造成水被肠回收,这会导致腹泻现象。
为了克服这些缺点,已尝试研究具有较高分子量和更难于水解的键的组合物。
专利申请WO00/32064描述了包含碳水化合物诸如淀粉或淀粉衍生物以及交联多糖诸如纤维素衍生物、树脂、果胶和藻酸盐的混合物的组合物。
专利申请WO2004/023891描述了包含具α(1→6)键之多糖的食品制剂,所述多糖通常是葡聚糖、普鲁兰多糖(pullulan)和alternan,且分子量在300,000和1,000,000道尔顿之间。
专利申请EP153013描述了基于葡聚糖的制剂,即具α(1→6)键且分子量在50,000和1,000,000道尔顿之间的多糖,其很少或没有肠吸收,可用作能引起饱足感的制剂(填充剂)。
因此本领域中已知的包含具α(1→6)键之多糖的人工食品制剂已知提供了葡萄糖的逐渐释放,只要链长不是太长即可,在此情形下吸收剧烈减少。
糖原是主要为动物来源的多糖,它主要包含经α-1-4葡糖苷键连接并且每5-10个葡萄糖单位具有经α-1-6葡糖苷键形成的分支的D-葡萄糖分子。糖原分支的数目和程度根据其获取来源之动物物种而变化。天然糖原的分子量在106-107道尔顿数量级。在自然界中,糖原常常与蛋白质结合,即糖原蛋白,这种一种涉及细胞糖原合成过程的酶。
商品化糖原衍生物的质量来自或多或少量存在的蛋白质残余物(从表示为ppm的氮量方面来测量)和还原糖。专利EP 654048描述了具有低氮和还原糖含量且分子量约2,500,000道尔顿的高质量糖原衍生物。
糖原在化妆品部门被用作软化剂(如JP-A-87-178 505中所述)和保湿剂(如JP-A-88-290 809中所述),在食品部门被用作添加剂,以及在滴眼液中被用作润湿剂和润滑剂(如专利WO99/47120中所述)。
发明简述
令人惊奇的是,申请人已发现糖原、尤其是专利EP654048中所述的糖原能在模拟胃肠道的体外系统中提供与获自较低分子量的麦芽糊精类型的其他多糖相似的葡萄糖逐渐释放。
本发明因此涉及糖原在制备用于进行葡萄糖控释的人工食品制剂中的应用。
申请人已发现,尽管有着超过了2,000,000道尔顿的高分子量,但在模拟胃肠道的体外系统中,上述糖原通过胃肠道中存在的酶被酶降解并允许葡萄糖在跨越20-24小时的期间被逐渐释放。
除此之外,申请人还观察到,因为糖原具有超过2,000,000道尔顿的高分子量,对于被摄取的等量葡萄糖而言,其摩尔渗透压浓度较施加麦芽糊精而言要低得多,这避免了本领域已知的伴随腹泻后果的水回收问题。
申请人还认识到,在糖原经胃肠道运输期间观察到的葡萄糖逐步且持续释放可有利的提供用合适剂量糖原作为糖尿病个体之食物或饮料的可能性。
另一方面,本发明还涉及适于葡萄糖控释的人工食品制剂,其中包含糖原和至少一种其它的可食用组分,优选至少一种营养素。
申请人已发现,本发明的人工食品制剂具有比本领域中已知的含麦芽糊精的制剂更好的可口性。
尤其是申请人观察到本发明的人工食品制剂尝起来甜度稍低,在味觉上讨人喜欢并且不会太腻。
有利的是,本发明的人工食品制剂包含至少一种选自以下的营养素:碳水化合物、蛋白质、氨基酸及衍生物、脂质、磷脂、维生素和矿物盐。
在进一步的方面,本发明还涉及用于葡萄糖的肠道或胃肠外给药的水性制剂,其中包含糖原和至少一种另外的药用可接受赋形剂。
申请人还发现糖原溶液的低摩尔渗透压浓度使得可能制备具有比包含蔗糖或低分子量多糖之已知溶液的葡萄糖含更高的高葡萄糖含量等渗水性制剂(300mOsm/kg)。
申请人还发现,与葡萄糖慢释放的其它多糖不同,上述糖原溶液即使在高多糖浓度情况下也具有接近生理数值的pH值。
发明详述
特别是,本发明涉及糖原在制备适于葡萄糖控释的人工食品制剂中的用途。
本发明中所用糖原获自可提取自动物或真菌的天然糖原。软体动物,尤其是蚌类(贻贝和Mytilus gallus provincialis),是特别有效的糖原来源,因为它们可低成本地大量获取并且包含相当量的糖原(平均占重量的2.5%-3.9%)。其它天然的糖原来源包括其它双壳类软体动物诸如蛤、牡蛎、一些种类的腹足动物或海螺诸如指甲履螺(舟螺)以及富含糖原的脊椎动物器官诸如肝脏和肌肉。
有利的是本发明中所用糖原具有高于2,000,000道尔顿的分子量,优选在2,000,000和5,000,000道尔顿之间。
优选地,本发明中所用糖原具有相对于键的总数而言在5%至15%之间、优选在8%和12%之间的α-1-6葡糖苷键百分数。
本发明中所用糖原可在获自提取过程后直接使用或可在随后的纯化步骤中进行处理。
如先前已提及的,商品化糖原衍生物的质量来自或多或少量的蛋白质残余物(从表示为ppm的氮含量方面来测量)和还原糖的存在。
就本发明的目的而言,优选使用低还原糖和氮含量的糖原衍生物。优选用于本发明的商品化产物的例子是Sigma-Aldrich生产和配销的糖原衍生物。
优选本发明中所用糖原衍生物包含少于1%重量、更优选少于0.25%重量的还原糖,该含量用F.D.Snell和Snell(″Colorimetric Methodsof Analysis″,New York,1954,Vol.III,p.204)的方法测量。
依照Kjeldahl方法测量,优选本发明中所用糖原衍生物包含少于3,000ppm的氮,更优选少于1,000、进一步优选少于100ppm的氮。
优选的,本发明中所用糖原衍生物是PolglumytTM糖原,其是由A.C.R.A.F.S.p.A.Rome,Italy生产和配销并依照专利EP 654048B1中所述纯化步骤获取的具低还原糖的去蛋白糖原的商品名。
有利的是,本发明的所用糖原衍生物具有超过2,000,000道尔顿、优选2,000,000和5,000,000道尔顿之间的分子量,以及占总键数5%和15%之间、优选8%和12%之间的α-1-6葡糖苷键百分数。
依照本发明适于控释葡萄糖的食品制剂包含糖原和至少一种其它可食用组分,优选至少一种营养素。
有利的是,本发明的食品制剂包含至少一种选自以下的营养素:碳水化合物、蛋白质、氨基酸及衍生物、脂质、磷脂、维生素和矿物盐。
优选的,糖原的存在满足本发明食品制剂中碳水化合物的摄取量。然而本发明的食品制剂可任选的除糖原之外还包含其它类型的碳水化合物。
本发明食品制剂中所用的蛋白质可获自不同的天然来源,诸如牛奶蛋白质、鸡蛋蛋白质或血液蛋白质。所述蛋白质也可以肽的水解形式或个体氨基酸存在。优选至少一半的蛋白质内容以完整的蛋白质提供。
本发明食品制剂中所用的脂质可以是含12至18个碳原子的饱和和未饱和脂肪酸的甘油三酯。优选使用长链多不饱和脂肪酸诸如ω-3、ω-6和ω-9脂肪酸的甘油三酯。特别优选油酸,亚油酸(LA),α-亚麻酸(ALA),花生四烯酸,二十碳五烯酸(EPA),二十二碳六烯酸(DHA)和二十二碳五烯酸(DPA)的甘油三酯。磷脂可优选卵磷脂或其相等物。卵磷脂可以是动物或植物来源的,且主要包含磷酸,胆碱,脂肪酸,甘油,糖脂,甘油三酯和磷脂诸如磷脂酰胆碱,磷脂酰乙醇胺和磷脂酰肌醇。
本发明食品制剂中所用的维生素没有特别的限制,可以是任何已知维生素,诸如B族(B1,B2,B3,B5,B6,B8,B9和B12)和C族的水溶性维生素,或A,D,E和K族的脂溶性维生素。还可使用假维生素诸如胆碱、邻氨基苯甲酸,硫辛酸,生物黄酮素,泛醌和甲基蛋氨酸。制剂内的维生素含量通常表示为每日推荐允许量的百分数或RDA%。
通常基于每日需求而将矿物盐划分为大量元素(超过100mg)、微量元素(1至100mg)和寡元素(oligoelement)(少于1mg)。大量元素是包含钙、氯,磷,镁,钾,钠和/或硫的盐。微量元素是包含铜,锌,氟,碘,硒,铬,钴,锰,钼,硅,镍,钒的盐。寡元素(oligoelement)是包含锡,镍,锗,钒和钨的盐。
食品制剂的形式可以是完全的食品、食品增补剂、供胃肠道施用例如经鼻胃管和鼻肠管进行肠内饲喂施用的营养液、供胃肠外给药的营养液、或者适于糖尿病个体的食品或增补剂。
完全的食品包含就物质和能量摄取而言满足使用者每日需求所必需的所有营养物质。因而所述制剂必须包含包括糖原计占重量30%至70%之间的碳水化合物、占重量10%至30%之间的蛋白质和占重量20%至40%之间的脂质。
除此之外,所述制剂必须能提供每日2000至2900kcal,并且可以是固体形式,适于溶解或分散在水或其它饮料里,或者是液体,以可立即使用的形式或作为浓缩物。针对特殊的情形(膳食或运动配方)可提供较低或较高的能量摄取量。
食品增补剂只包含就蛋白质和能量摄取量而言满足使用者每日需求所需要的某些营养物质。因而所述制剂能提供每日少于1500kcal,优选100至1000kcal之间。此外,在此情况下,所述制剂可以是如上所述的固体或液体形式,适于添加至正常的饮食中或作为正常饮食的组分。
本发明的食品制剂可进一步包含传统的食品添加剂以改善其外观、愉悦度和保存,诸如着色剂、防腐剂、抗氧化剂、酸性调节剂、增稠剂、稳定剂、乳化剂、增味剂、调味料、润湿剂和甜料。
以下例子用于举例说明本发明而非对它进行限制。
实施例1
在实验室中制备模拟消化系统的模型,包括第一阶段与唾液α-淀粉酶一起保温,第二阶段与胃蛋白酶一起保温,以及第三阶段与胰酶、淀粉葡糖苷酶和pH接近7的牛胆汁一起保温。
将6g底物在带螺纹盖的250ml Pyrex瓶内溶于100ml磷酸盐缓冲液中。
将溶液温度提高至37℃,然后加入0.1ml的人α-淀粉酶溶液(溶液A)。产生的溶液在恒温至37℃的水浴中保温,伴随着磁力搅拌。
用2.50ml的1M HCl溶液将所述溶液调节至pH=2,然后加入0.25ml配制于NaCl溶液中的Sigma P7012胃蛋白酶悬浮液(溶液B)。产生的溶液在37℃保温30分钟,再次伴随使用磁力搅拌。
用8.67ml的1M NaHCO3溶液将所述溶液调节至pH=6.9,然后加入2ml配制于25mM CaCl2中的胰酶和淀粉葡糖苷酶溶液(溶液C)和2.4g的牛羊混合胆汁(Bovine-Ovine Bile)(Sigma B8381)。产生的溶液在37℃保温5分钟,伴随着搅拌。
将溶液转移至透析管(混合纤维素酯,截留值3500)中,置于USPXIII溶解装置的1000ml容器中,其中包含约900ml制备如下的温度为37℃的缓冲液。

用PolglumytTM糖原和麦芽糊精DE 16.5-19.5作为底物一式双份进行测试。6g PolglumytTM糖原样品相当于5.220g葡萄糖,而6g麦芽糊精DE 16.5-19.5样品相当于5.748g葡萄糖。
磷酸盐缓冲液和溶液A,B和C具有下表1中的组成。

用两个商品化试剂盒分析样品以确定所释放的葡萄糖的量:SigmaGAGO20葡萄糖(GO)检验试剂盒和Sigma GAHK20葡萄糖(HK)检验试剂盒(二者均由Sigma-Aldrich Co.供应)。
第一个测试是基于D-葡萄糖通过葡萄糖-氧化酶氧化成D-葡萄糖酸和过氧化氢的过程。释放的过氧化氢在过氧化物酶存在下与o-邻联茴香胺反应形成褐色的氧化产物,该产物在硫酸存在下产生粉色的氧化产物。在540nm检测的颜色强度与葡萄糖浓度成比例。
所获结果总结于下表2-5。

第二个测试是基于在存在ATP下经由己糖激酶的葡萄糖磷酸化。葡萄糖-6-磷酸随后在NAD(烟酰胺腺嘌呤二核苷酸)存在下被氧化成6-磷酸葡萄糖酸,一个由葡萄糖-6-磷酸脱氢酶催化的反应。在所述氧化过程中,等摩尔量的NAD被还原成NADH。随之发生的在340nm的吸收值的增加与葡萄糖浓度成正比例。
所获结果总结于下表6-9中。

实施例2
用双蒸水以逐步增加的浓度(按重量计算为10,14,18,22,26和30%)制备6份PolglumytTM糖原和麦芽糊精DE 16.5-19.5溶液。
分析所得溶液以确定粘度,pH,导电率和摩尔渗透压浓度。结果总结于下表10和11中,连同溶解时间。用装备了2°/55mm圆锥板几何体的Bohlin Gemini 150流变计进行粘度测量。用Knauer渗压计进行摩尔渗透压浓度测量。在分析之前将所述溶液用0.2μm Millipore过滤器过滤。

表17中所述的结果清楚显示了包含约5%D-葡萄糖的制剂已经是等渗溶液,5%的麦芽糊精(DE 16.5-19.5)溶液具有55mOsm/kg的摩尔渗透压浓度(与葡萄糖相比接近1/5),而5%的PolglumytTM糖原溶液则具有少于1mOsm/kg的摩尔渗透压浓度值(比葡萄糖低约300倍)。
因而,如表17中所述,含PolglumytTM糖原的制剂允许制备具有较高葡萄糖含量和/或较高基本营养成分(例如维生素、氨基酸、矿物盐等)含量的制剂。
Claims (24)
1.包含糖原和至少一种其它可食用组分、用于控释葡萄糖的人工食品制剂,其特征在于所述糖原具有高于2,000,000道尔顿的分子量,并且其中α-1-6葡糖苷键占总键数的5%至15%。
2.依照权利要求1的食品制剂,其特征在于所述糖原具有2,000,000至5,000,000道尔顿的分子量。
3.依照权利要求1或2的食品制剂,其特征在于所述糖原中α-1-6葡糖苷键占总键数的8%至12%。
4.依照权利要求1或2的食品制剂,其特征在于所述糖原包含重量少于1%的还原糖。
5.依照权利要求4的食品制剂,其特征在于所述糖原包含重量少于0.25%的还原糖。
6.依照权利要求1或2的食品制剂,其特征在于所述糖原包含少于3,000ppm的氮。
7.依照权利要求6的食品制剂,其特征在于所述糖原包含少于1,000ppm的氮。
8.依照权利要求1或2的食品制剂,其特征在于所述葡萄糖控释是由此50%的葡萄糖在不少于1小时的时间内释放。
9.依照权利要求8的食品制剂,其特征在于所述葡萄糖控释是由此50%的葡萄糖在不少于3小时的时间内释放。
10.依照权利要求8的食品制剂,其特征在于所述葡萄糖控释是由此50%的葡萄糖在不少于5小时的时间内释放。
11.依照权利要求8的食品制剂,其特征在于所述葡萄糖控释是由此80%的葡萄糖在不少于6小时的时间内释放。
12.依照权利要求11的食品制剂,其特征在于所述葡萄糖控释是由此80%的葡萄糖在不少于9小时的时间内释放。
13.依照权利要求11的食品制剂,其特征在于所述葡萄糖控释是由此80%的葡萄糖在不少于12小时的时间内释放。
14.依照权利要求1或2的食品制剂,其特征在于所述可食用组分是营养素。
15.依照权利要求14的食品制剂,其特征在于所述营养素选自碳水化合物、蛋白质、氨基酸和衍生物、脂质、磷脂、维生素和矿物盐。
16.依照权利要求14的食品制剂,其特征在于所述制剂包含包括所述糖原计重量占30%至70%的碳水化合物,重量占10%至30%的蛋白质和重量占20%至40%的脂质。
17.依照权利要求1或2的食品制剂,其特征在于所述制剂是固体形式或水溶液。
18.依照权利要求1或2的食品制剂,其特征在于所述食品制剂选自完全的食品、食品增补剂、供胃肠道施用的营养液或供胃肠外施用的营养液。
19.依照权利要求1或2的食品制剂,其特征在于所述食品制剂选自适于糖尿病个体的食品或增补剂。
20.糖原在制备用于控释葡萄糖的人工食品制剂中的用途,其特征在于所述糖原具有高于2,000,000道尔顿的分子量,并且其中α-1-6葡糖苷键占总键数的5%至15%。
21.依照权利要求20的用途,其特征在于所述糖原具有权利要求1至13中任一项的特征。
22.依照权利要求20或21的用途,其特征在于所述食品制剂是固体形式或水溶液。
23.依照权利要求20或21的用途,其特征在于所述食品制剂选自完全的食品、食品增补剂、供胃肠道施用的营养液或供胃肠外施用的营养液。
24.依照权利要求20或21的用途,其特征在于所述食品制剂选自适于糖尿病个体的食品或增补剂。
