Abstract
应用人工诱变技术培育出一株851R菌株,用该组菌作为生产菌株,在本发明所设计和使用的培养基中培养,可分别生产出富硒、锌、钙、多糖、多肽及多种维生素的单细胞蛋白的营养液,该营养液对人类防癌延寿具有进一步的开发利用价值。
Description
本发明是关于工业发酵的一种方法,特别是关于利用微杆菌属的人工诱变菌株-一个命名为851R菌的新菌株作为生产菌株以大豆为主要原料进行工业发酵来生产单细胞蛋白营养液的方法。
在当今世界,人们通过各种尝试如采用生物发酵方法生产单细胞蛋白取得成功。杨振华利用抗氨固氮菌生产富硒单细胞蛋白、维生素E和菌肥的方法(中国发明专利86100490 1988.1.)是通过培育一种具有抗氨固氮能力的固氮菌将空气中的氮通过常规的工业发酵生产方法生产出富硒、锌和多种维生素的单细胞蛋白。
本发明的目的是通过培育出的一种851R菌及设计出适合于该菌生长的以大豆为主要原料的培养基,以达到用851R菌作为生产菌株,运用常规的工业发酵的生产方法生产出851R型超级营养液。大豆是蛋白质丰富的食物,但由于大豆中含有蛋白酶抑制剂,所以大豆蛋白很难被人体全部吸收,然而,大豆蛋白经过851R菌利用后,转化为单细胞蛋白营养液,不但成为易被人体吸收的蛋白源,而且该营养液具有健全中枢神经系统、提高机体免疫功能、调节内分泌的功效,该营养液对癌细胞的生长有某些抑制作用。
本发明所涉及的微生物是微杆菌属的变异菌851R菌株(Microbacterium851R),其主要特征为:革兰氏阳性菌,不运动,属氧化型,在牛奶中加热到63℃30分钟,或在牛奶中加热至72℃经15分钟仍能生存,菌体呈桂红色或橘红色,但在38℃左右,菌体颜色变成粉红色,40℃时变成无色。该菌株具有R片段,运用DNA分子杂交技术可检测出851R菌中R片段的存在(该菌种已于1989年5月19日送交武汉中国典型培养物保藏中心保藏,保藏号为CCTCC NO:M89048)。
在本发明所设计或使用的含氮源培养基中,用851R菌作为生产菌种,可生产出富含多糖、多钛、多种微量元素及维生素的单细胞蛋白营养液。该营养液中含有二十余种氨基酸。每100毫升这种营养液中含有:氨基酸593~4172毫克,维生素E0.3~1.0mg,维生素B20.1~0.3mg,维生素PP 0.5~1.0mg,硒1.0~5.0ppm,锌10~20ppm,钼5.0~10ppm,钴5.0~10ppm,锰5.0~15ppm,铜3.0~10ppm,铁50~150ppm等。
本发明所设计及使用的培养基为:
1.含氮源培养基
大豆粉 5~10%
或豆浆(以大豆量计) 5~20%
或各种豆饼、葵花籽饼 5~10%
酵母膏(或酵母粉、蛋白胨、牛肉膏) 0.02~0.5%
碳酸钙 0.05~0.5%
磷酸氢二钾 0.02~0.2%
硫酸镁 0.01~0.08%
氯化钠 0.01~0.08%
钼酸钠 5.0~50ppm
亚硒酸钠 1.5~10ppm
硫酸锌 2.5~50ppm
氯化钴 2.5~20ppm
2.淀粉培养基
干淀粉 1~2%
含有淀粉的新鲜物质(甘薯,马铃薯等) 10~20%或
含有淀粉的干物质(玉米粉等) 1~10%
酵母膏 0.04~0.08%
磷酸氢二钾 0.05~0.1%
硫酸镁 0.02~0.04%
氯化钠 0.02~0.04%
钼酸钠 10~20ppm
硼酸 10~20ppm
亚硒酸钠 5~20ppm
硫酸锌 5~20ppm
氯化钴 5~20ppm
以上两种培养基中还可以适量的增加其他人体必须的微量元素及苯甲酸或苯甲酸钠。
该菌种可以用发酵罐进行通气培养,这种罐为一般发酵工业中常用的发酵罐,它带有搅拌和通气装置。可以依需要对所培养的培养物进行搅拌,使其所处的条件一致,也可以按需要通入经过滤的无菌空气,以满足细菌等微生物在发酵过程中对氧气的需要。将851R菌作为生产菌株,经过在种子培养基(本发明所设计和/或使用的培养基)中繁殖。即由小种、中种到大种的繁殖培养,再接种到装有预先经过消毒灭菌的生产用培养基(本发明所设计和/或使用的培养基)中培养36~64小时,在培养过程中通入过滤的无菌空气,通气量为1∶0.5~1.0,搅拌速度180~260转/分,培养温度28~40℃,培养结束后经高温灭菌收获培养液。培养液可直接分装而成产品,也可按其需要用不同的方法制成各种系列产品,如培养液经离心沉淀制成851可乐饮料,培养液经浓缩制成851营养精,培养液经烘干或喷雾干燥成粉剂,进而制成胶囊或片剂。培养液还可直接代替水或鸡蛋加入面粉而制成面包、饼干等食品,培养液亦可用于化妆品的生产。
实施例1
在2.5吨发酵罐中培养,种了罐为0.5吨,投料量为40%。采用5%黄豆粉培养基,其中黄豆粉含量为5.0%,酵母膏0.04%,磷酸氢二钾0.05%,硫酸镁0.02%,碳酸钙0.02%,氯化钠0.02%,钼酸钠10ppm,亚硒酸钠2.5ppm,硫酸锌10ppm,氯化钴2.5ppm,及水。接菌量按1%,种子罐为2立升,菌龄为24小时,(小种→中种→大种,培养24小时作为种子培养物)培养温度30℃,搅拌速度260转/分,通气量1∶0.8。种子在种子罐中培养结束后接入2.5吨大罐,培养温度30℃,搅拌速度260转/分,通气量1∶08,培养45小时结束培养。终止培养后采用高温灭菌,培养液经流水线装瓶,再经高温高压灭菌即成为产品,每100毫升培养液中,干物质含量为3.7%,蛋白质含量为2.06%,脂肪含量0.23%,碳水化合物为0.75%及维生素、无机盐等详见下表:
实施例2
在8吨发酵罐中培养,投料量为40%,即培养基量为3.2吨。采用10%黄豆粉培养基,其中黄豆粉含量为320kg,酵母膏1.28kg,磷酸氢二钾1.6kg,硫酸镁640g,碳酸钙640g,氯化钠640g,钼酸钠32g,亚硒酸钠8g,硫酸锌32g,氯化钴8g,及水。接菌量按1%,菌龄为24小时,(小种→中种→大种,培养24小时作为种子培养物)培养温度30℃,搅拌速度220转/分,通气量1∶0.8。种子在种子罐中培养结束后接射8吨大罐,培养温度30℃,搅拌速度220转/分,通气量1∶0.8,培养48小时结束培养。终止培养后采用高温灭菌,培养液经流水线装瓶,再经高温高压灭菌即成为产品,每100毫升培养液中含有天冬氨酸543mg,谷氨酸710mg,丝氨酸160mg,甘氨酸280mg,组氨酸65mg,精氨酸226mg,苏氨酸232mg,丙氨酸359mg,脯氨酸126mg,酪氨酸101mg,缬氨酸283mg,蛋氨酸53mg,异亮氨酸282mg,亮氨酸309mg,苯丙氨酸148mg,赖氨酸212mg,光氨酸38mg,总计为4127mg。
Claims (1)
1、一种利用细菌生产单细胞营养液的工业发酵生产方法,其特征是采用经人工诱变微杆菌(Microbacterium)而得到的一株命名为851R菌株(Microbacterium 851R,1989年5月19日已寄存于中国武汉,中国典型培养物保藏中心,编号为CCTCC NO.M89048)作为生产菌株,在本发明所设计的以大豆为主的含氮源培养基和本发明所使用的淀粉培养基(大豆为主的含氮源培养基的成分为:大豆粉5~10%或豆浆(以大豆计)5~20%;或各种豆饼、葵花籽饼5~10%,酵母膏(或酵母粉、蛋白胨、牛肉膏)0.02~0.5%,碳酸钙0.05~0.5%,磷酸氢二钾0.02~0.2%,硫碳酸镁0.01~0.08%,氯化钠0.01~0.08%,钼酸钠5~50ppm,亚硒酸钠1.5~10ppm,硫酸锌2.5~50ppm,氯化钴2.5~20ppm;淀粉培养基的成分为:干淀粉1~2%,含有淀粉的新鲜物质(甘薯,马铃薯等)10~20%或含有淀粉的干物质1~10%,酵母膏0.04~0.08%,磷酸氢二钾0.05~0.1%,硫酸镁0.02~0.04%,氯化钠0.02~0.04%,钼酸钠10~20ppm,硼酸10~20ppm,亚硒酸钠5~20ppm,硫酸锌5~20ppm,氯化钴5~20ppm,以上两种培养基还可以适当的增加其他人体必需的微量元素)中发酵培养生产营养液。
