背景技术
日常生活中反复性疾病的发作困扰着人们的健康,其根原为免疫力低下所致。免疫力是人体自身的防御机制,是人体识别和消灭外来侵入的任何异物(病毒、细菌等);处理衰老、损伤、死亡、变性的自身细胞以及识别和处理体内突变细胞和病毒感染细胞的能力。祖国医学认为免疫力就是指人体的正气,人抵抗疾病全在于正气,所谓“正气存内,邪不可干”,“邪之所凑,其气必虚”。正气就是人体的卫御之气,包括卫气及元气。正气与人体的肾、脾、肺有密切关系。其中,肾为元气之根,脾为正气之源,肺为卫气之本,所以正气的强弱与人体五脏的盛衰密切相关。五脏虚可导致正虚,正虚可使免疫力下降,从而降低抗病力。平衡阴阳,固本正元是提高免疫力的必备条件。
《素问·宝命全角论》中记载:“人生有形,不离阴阳。”人体机能的下降是阴阳失去平衡的表现,正气虚弱,正不胜邪,邪气乘虚而入,阻塞经络,经络不通,气血运行不畅所致,免疫力下降,继而引发各种疾病。西医认为人生病都是细菌感染的,因而采用灭菌、止痛的方法治病,通过抗生素等药物杀灭细菌,已达到消炎、祛病的疗效,对自身免疫力考虑不足;而中医认为阴阳失衡气血不畅是病之根本,在治疗上以扶正祛邪,疏通经络,行气活血为目的,继而调理阴阳平衡,祛除病症。
然而临床上应用中药调理阴阳由于症候复杂,疗程较长,应用不便,使得患者很难遵医调理。鉴于此,申请人总结30余年中药调理经验,研究中草药对提高免疫力的特性,挖掘中医药特性标本兼治及辨证施治的精髓,发明了一种平衡阴阳提高免疫力的药酒。
发明内容
本发明的目的就是针对上述临床中现有治疗存在的问题,而提供一种方便有效的治疗方法,饮用药酒后吸收迅速,可及早发挥药效,因为人体对酒的吸收较快,药物之性——“药力”通过酒的吸收而进入血液循环,能较快地发挥治疗效果。依据中医“阴阳平衡”治疗理论,调理气血、平衡阴阳、祛邪消炎、疏通经络,从而精力充沛,百病不侵。本发明人联系中医理论,反复研究,结合酒的特性,并通过动物和临床试验反复验证,终于研制出了一种平衡阴阳提高免疫力的特制药酒。
本发明调理兼施,通络益气、补精填髓、平衡阴阳,适用于阴阳失衡免疫力低下等引起的疾病,疗效确切,治愈率高,且操作方便。
本发明选择苜蓿为君药,辅以枸杞子、陈皮、大枣、核桃仁4位药进行组合,依据中药相生相克的原则,将这些药物有效的组合使得各药物功效产生协同作用,从而达到开胃健脾,滋补心、肝、脾、肾,平衡阴阳,提高免疫力的功效。其中苜蓿味苦、性平、无毒,有利大小肠,安中,和胃,舒筋活络之功效,且含有丰富的蛋白质、钙、磷、铁、维生素A、维生素B(族)、维生素C、维生素D维生素E维生素K多糖及苜蓿皂甙。苜蓿中多糖成分可作为免疫佐剂,具有增强人体免疫力的功效,苜蓿中的蛋白质、钙、磷、铁等营养成分,可增强人体的营养和免疫力;枸杞子味甘,性平,归肝经、肾经、肺经,有养肝、滋肾、润肺、明目之功能;陈皮味辛、苦,性温,入脾、肺经,有理气燥湿、健脾化痰的作用;大枣味甘性温,归脾、胃经,补脾和胃,益气生津,调营卫,解药毒之效;核桃仁味甘性温,归肾、肺、大肠经,有补肾,温肺,润肠之功效。
本发明药物组分的用量也是经过发明人进行大量实验结果得出,下列各组用量范围都具有较好疗效:本发明药物各组分用量是:苜蓿6~18份、枸杞子1~4份、陈皮1~4份、大枣3~7份、核桃仁4~6份。

(2)枸杞子、大枣挑选去杂,清洗后置于冷水中浸泡1小时,于蒸煮罐中加热煮沸,90~100℃保持2小时,温度在70℃时用80~120目滤筛过滤,滤液贮存;滤渣经二次提取后,滤液与第一次提取液充分混合,贮存备用,滤渣废弃;
(3)核桃仁、陈皮挑选去杂、清洗,粗粉碎后置于冷水中浸泡1小时,于蒸煮罐中加热煮沸,90~100℃保持2小时,温度在70℃时用80~120目滤筛过滤,滤液贮存;滤渣经二次提取后,滤液与第一次提取液充分混合,贮存备用,滤渣废弃;
(4)充分混合(1)(2)(3)中提取的滤液,置于储藏罐,在地窖中储存沉淀30天后,过80~120目滤筛,过滤后滤液置于新储存罐密封后在地窖储存15天后,过100~140目滤筛,即可得所药酒;
服用时用蒸馏水1∶2稀释药酒,每天60ml/人。
临床试验
为了验证本发明药酒的积极效果,发明人进行了大量的动物实验,现选择部分试验报告如下。
试验例1 本发明药酒大鼠脏器系数的影响
一、一般资料
1、实验动物来源:从河北医科大学动物学部购买SD大鼠80只,体重60~80g,随机分成四组,每组20只,雌雄各半,单笼饲养。
二、试验方法
受试动物设三个剂量组(2.5ml/kg体重、5ml/kg体重、10ml/kg体重)和一个对照组(蒸馏水灌胃),将药酒分别给各剂量组受试动物灌胃,连续30天。试验期间各组动物均喂饲基础饲料,并自由进食、饮水。实验结束后处死大鼠,对肝、肾、脾等进行病理组织学检查(石蜡切片)。
三、试验结果
表1各组动物脏体比值结果(%)
从表1可以看出,所有动物肝、肾、脾等均未见明显病理改变,说明药酒对无毒副作用、安全可靠。
试验例2 本发明药酒对免疫调节的作用
一、一般资料
1、实验动物来源:从河北医科大学动物学部购买清洁级雌性昆明种小鼠88只,体重18~22g,随机分成8组,每组11只,单笼饲养。
二、剂量选择
药酒人体推荐量为人体推荐量为60ml/天/人(浓缩液为6ml/天/人),小鼠的等效剂量相当于人体推荐量的10倍,即每日服用1ml/kg体重为低剂量,在按低计量的2倍和3倍各设一个剂量组,即2ml/天/kg体重(中剂量)和3ml/天/kg体重(高剂量)。对照组给蒸馏水,连续等体积灌胃受试小鼠15天后,测定各项指标。
三、实验仪器与试剂
游标卡尺(精密度0.02mm)、绵羊红细胞(SRBC)、微量注射器(50ul)。
二氧化碳培养箱、恒温水浴、离心机、手术器械、玻片架、200目筛网、豚鼠血清(补体)、Hanks液、SA缓冲液、琼脂糖。
756MC紫外课可见分光光度计、计时器、血色素吸管、印度墨汁、碳酸钠。
四、试验方法:
1.脏器/体重比值测定:
2.迟发型变态反应(DTH)(足跖增厚法)
小鼠腹腔注射2%(v/v)SRBC0.2ml致敏后4天,测量左后足跖厚度,然后在测量部位皮下注射20%(v/v)SRBC(20ul/每鼠),注射后于24小时测量左后足跖部厚度,同一部位测量三次,取平均值。以攻击前后足跖厚度的差值(足跖肿胀度)来表示DTH的程度。
3.脾脏抗体生成细胞检测(Jerne改良玻片法)
取羊血,用生理盐水洗涤三次,每只鼠经腹腔注射2%(v/v)SRBC0.2ml。将SRBC免疫5天后的小鼠处死,取脾,制成细胞悬液。将表层培养基加热溶解后,与等量双倍HanKs液混合,分装小试管,每管0.5ml,再向管内加50ul10%SRBC(v/v,用SA液配制)、10ul脾细胞悬液,迅速混匀后,倾倒于已刷琼脂糖薄层的玻片上,待琼脂凝固后,将玻片水平扣放在片架上,放入二氧化碳培养箱中温育1.5小时,然后用SA缓冲液稀释的补体(1∶10)放入到玻片架凹槽内,继续温育1.5小时后,计数溶血空斑数。
4.小鼠碳粒廓清试验
小鼠尾静脉注射1∶3稀释的印度墨汁,待墨汁注入,立即计时。注入墨汁后,1、10min,分别从内眦静脉丛取血20ul,并将其加到2mlNa2CO3溶液中。用756MC紫外可见分光光度计在600nm波长处测光密度值(OD),以Na2CO3溶液作空白对照。将小鼠处死,取肝脏和脾脏称重。计算吞噬指数a。
5.实验数据用t检验进行统计。
五、试验结果
1.药酒对小鼠体重的影响

表2~3结果表明,经口给予小鼠一定剂量的药酒15天后,其体重与对照组比较无显著性差异(免疫二组的中、高剂量组动物各有一只于中期意外死亡)。

由表4可见,经口给予小鼠不同剂量的药酒15天,各组动物的脏器系数与对照组比较均无显著性差异。

由表5可见,经口给予小鼠不同剂量的药酒15天,中剂量组的足跖肿胀度与对照组比较有显著性差异(P<0.05)。

由表6可见,经口给予小鼠不同剂量的药酒15天,其中、高剂量组的抗体生成细胞数与对照组比较有显著性差异。

由表7可见,经口给予小鼠不同剂量的药酒15天,其高剂量组的单核-巨噬细胞吞噬指数与对照组比较有显著性差异。
六、小结
经口给予小鼠不同剂量的药酒15天,能明显提高小鼠的脾脏抗体生成细胞数、小鼠足跖肿胀度及单核-巨噬细胞吞噬功能。根据《保健食品功能学评价程序和检验方法》的规定,可判定药酒具有免疫调节作用。
结论
经过动物试验结果显示,此种中药药酒对阴阳失衡等所致的免疫力低下具有显著疗效,服用方便,且在试验检测过程中末发现明显不良反应和毒副作用。
Claims (4)
1.一种平衡阴阳提高免疫力的药酒,其特征在于它是由以下重量份的原药浸泡制成:苜蓿6~18份、枸杞子1~4份、陈皮1~4份、大枣3~7份、核桃仁4~6份。
2.根据权利要求1所述的药酒,其中原料药的用量为:苜蓿12份、枸杞子2.5份、陈皮2.5份、大枣5份、核桃仁5份。
3.权利要求1或2所述药酒的制备方法,它包括下列步骤:
a)称取各原料药苜蓿、枸杞子、陈皮、大枣、核桃仁,备用;
b)将所述重量配比的苜蓿去杂粉碎成末,过20~40目筛,选40~60°优质白酒68份,浸泡容器中的原药粗末25~35天后,过80~120目滤筛,得滤液1,贮存备用;
c)枸杞子、大枣挑选去杂,清洗后置于冷水中浸泡1小时,于蒸煮罐中加热煮沸,90~100℃保持2小时,温度在70℃时用80~120目滤筛过滤,滤液贮存;滤渣经二次提取后,滤液与第一次提取液充分混合,得滤液2,贮存备用,滤渣废弃;
d)核桃仁、陈皮挑选去杂、清洗,粗粉碎后置于冷水中浸泡1小时,于蒸煮罐中加热煮沸,90~100℃保持2小时,温度在70℃时用80~120目滤筛过滤,滤液贮存;滤渣经二次提取后,滤液与第一次提取液充分混合,得滤液3,贮存备用,滤渣废弃;
e)充分混合滤液1、滤液2、滤液3即得药酒主要成分滤液4;
f)将滤液4置于储藏罐在地窖中储存沉淀25~35天后,过80~120目滤筛,过滤后滤液置于新储存罐密封后在地窖储存15天后,过100~140目滤筛,即可得权利要求1或2所述药酒。
4.根据权利要求1所述的药酒,使用时以蒸馏水1∶2稀释,每天60ml/人。
