技术领域
本发明涉及重组病毒疫苗领域,具体地,本发明涉及一种表达猪干扰素(α、β或γ-干扰素)的重组新城疫弱毒疫苗株,更具体地,涉及一种表达猪干扰素(α、β或γ-干扰素)的重组新城疫LaSota弱毒疫苗株。本发明还公开了制备所述重组新城疫弱毒疫苗株的方法和该重组新城疫弱毒疫苗株在制备猪α、β或γ-干扰素中的应用。
背景技术
干扰素主要是由动物细胞在病毒等诱生剂的作用下诱导产生的。许多动物,包括人、哺乳动物、鸟类、鱼类等都能产生干扰素。根据干扰素的抗原特性和分子结构分成不同的类型。干扰素(IFN)分为I型和II型,I型干扰素基因缺少内含子,包括α、β、δ、ω、τ5种类型,II型仅有γ型1种,含有内含子。目前所发现的猪干扰素包括IFN-α、β、ω、τ和IFN-γ,并且IFN-τ未在其它物种中发现[1]。猪IFN-α、ω,分别是由12个和5个以上的相关功能基因编码的蛋白质家族组成,这些干扰素的各亚型之间同源性很高,天然IFN-α常常是IFN-α、ω功能基因表达产物的混合体,而INF-β、ω和IFN-γ仅由单一基因编码。现在已完成基因克隆、测序和定位的猪干扰素基因主要包括IFN-α、β、ω和IFN-γ。
干扰素是一种高活性、多功能的生物活性蛋白,具有强烈的免疫调节作用,可明显提高机体抗感染能力。它无论对病毒、细菌、立克次氏体、衣原体、原虫,只要寄生在细胞内,均有一定的抑制作用。猪白细胞IFN对哺乳仔猪和断奶仔猪病毒性腹泻或病毒、细菌混合感染引起的腹泻均有良好的预防和治疗效果[2]。IFN具有预防和治疗双重作用,并且无毒副作用,不会产生耐药性,也不存在药物残留。干扰素类产品的开发为防治动物病毒性疾病开辟了新的途径,具有良好的推广应用前景。
负链RNA病毒的反向遗传操作(Reverse genetic)是通过操作病毒基因组cDNA制造新病毒的过程,其基本过程是:①组装完整的病毒基因组(或重组型基因组)cDNA克隆,5’末端精确地缀于T7启动子后,3’末端精确缀于自我剪切的核酸酶序列和T7转录终止信号之前,构成基因组cDNA转录模板;②以基因组cDNA转录模板与启动病毒复制必须的转录相关功能结构蛋白如核蛋白(NP)、磷酸蛋白(P)和聚合酶蛋白(L)的表达质粒(T7启动子)一起,共转染整合表达T7聚合酶的病毒复制许可细胞;③24-72小时后收获培养上清,过滤后继续敏感细胞传代或接种鸡胚尿囊腔救获(rescue)病毒。对基因组cDNA进行突变、缺失或外源基因插入修饰后,通过反向遗传操作系统(reverse genetic system,RGS系统)可获得相应的突变或重组的负链RNA病毒。
申请人之前获得授权的中国专利“新城疫LaSota疫苗株反向遗传操作系统及其应用”(申请号200510097997.8,申请日2005年9月2日)已经建立了一种新城疫LaSota弱毒疫苗株的反向遗传操作系统,并且成功救获了野生型病毒株,并且为进一步开展新城疫病毒活载体疫苗研制及重组新城疫病毒(Newcastle disease virus,NDV)相关基础奠定了坚实的基础。
发明内容
基于猪干扰素在治疗猪病毒性疾病和免疫调节方面广阔的应用前景,本文在已建立的新城疫病毒疫苗株,如新城疫病毒LaSota疫苗株反向遗传操作系统的基础上,构建了表达猪α、β、γ干扰素基因的全基因组克隆,实现了猪α、β、γ干扰素在SPF鸡胚内高效、稳定表达,并对其表达产物进行了抗病毒活性研究,探索猪干扰素产业化应用的可行性。
因此,本发明的一个目的是提供以下各项:
1.一种表达编码猪干扰素的基因的重组新城疫弱毒疫苗株。
2.根据以上1的重组新城疫弱毒疫苗株,其为重组新城疫LaSota弱毒疫苗株。
3.根据以上1或2的重组新城疫弱毒疫苗株,其中所述猪干扰素是猪α、β或γ-干扰素,优选所述猪α、β和γ-干扰素的氨基酸序列分别如SEQID No.1、SEQ ID No.2和SEQ ID No.3所示,更优选编码所述猪α、β和γ-干扰素的基因如SEQ ID No.4、SEQ ID No.5和SEQ ID No.6所示。
4.根据以上1或2的重组新城疫弱毒疫苗株,其为rL-Swine α、rL-Swine β、或rL-Swine γ。
5.根据以上1-4中任何一项的重组新城疫弱毒疫苗株在制备猪干扰素中的应用。
6.一种生产以上1-4中任何一项的重组新城疫弱毒疫苗株的方法,该方法包括:
(1)构建转录质粒,该转录质粒包括其中插入编码猪干扰素的基因的所述新城疫弱毒疫苗株的基因组cDNA序列;
(2)构建一个或多个转录辅助质粒,该转录辅助质粒包括编码所述新城疫弱毒疫苗株的核蛋白(NP)的cDNA序列、编码所述新城疫弱毒疫苗株的磷酸蛋白(P)的cDNA序列、和编码所述新城疫弱毒疫苗株的大聚合酶蛋白(L)的cDNA序列;
(3)将所述转录质粒和转录辅助质粒共转染所述新城疫弱毒疫苗株复制许可的宿主细胞,培养转染后的宿主细胞;
(4)收获上清液,过滤后继续敏感细胞传代或接种鸡胚尿囊腔救获重组病毒株。
7.根据以上6的方法,其中所述重组新城疫弱毒疫苗株是重组新城疫LaSota弱毒疫苗株。
8.根据以上6或7的方法,其中所述转录质粒是pBRN-FL-SwIFNα、pBRN-FL-SwIFNβ、或pBRN-FL-SwIFNγ(基因序列分别见SEQ ID No.7、SEQ ID No.8、SEQ ID No.9,质粒图谱分别见图5-7)。
9.根据以上6或7的方法,其中所述转录辅助质粒是质粒pBSNP、pBSP或pBSL。
10.根据以上6-9中任何一项的方法,其中所述宿主细胞是BHK-21。
11.根据以上6-10中任何一项的方法制备的重组新城疫弱毒疫苗株,优选为rL-Swine、rL-Swine β、或rL-Swine γ。
12.一种生产猪干扰素的方法,该方法包括:
1)将根据以上1-4中任何一项的重组新城疫弱毒疫苗株接种敏感细胞或接种鸡胚尿囊腔;
2)收获细胞上清液或鸡胚尿囊液。
13.重组新城疫(Newcastle disease virus)LaSota弱毒疫苗株rL-Swineα的SPF鸡胚接种尿囊液,其保藏号为CGMCC No.2615。
14.重组新城疫(Newcastle disease virus)LaSota弱毒疫苗株rL-Swineβ的SPF鸡胚接种尿囊液,其保藏号为CGMCC No.2628。
15.重组新城疫(Newcastle disease virus)LaSota弱毒疫苗株rL-Swineγ的SPF鸡胚接种尿囊液,其保藏号为CGMCC No.2616。
新城疫病毒LaSota株作为表达载体,分别构建表达猪α、β、γ-干扰素(Swine interferon-α,Swine interferon-β,Swine interferon-γ)完整开放阅读框的全基因组质粒pBRN-FL-SwIFNα、pBRN-FL-SwIFNβ、或pBRN-FL-SwIFNγ,转染BHK-21细胞获得表达SwIFN-α、β、γ重组新城疫病毒。重组新城疫(Newcastle disease virus)LaSota弱毒疫苗株rL-Swineα、rL-Swine β和rL-Swine γ接种的SPF鸡胚的鸡胚尿囊液于2008年7月31日保藏于中国微生物菌种保藏管理委员会普通微生物中心,其保藏号分别为CGMCC No.2615、CGMCC No.2628和CGMCC No.2616。采用RT-PCR方法,证实收获的鸡胚尿囊液内的重组病毒含有相应外源基因。将重组病毒尿囊液经紫外线照射灭活新城疫病毒,利用表达增强型绿色荧光的重组水疱性口炎病毒(VSV-EGFP)在猪肾细胞(PK)和牛肾细胞(MDBK)细胞系上测定rL-Swine α,rL-Swine β,rL-Swine γ抗病毒活性,表达SwIFN的重组新城疫病毒均能有效抑制VSV-EGFP在PK和MDBK上的复制,rL-Swine α、rL-Swine β、rL-Swine γ接种SPF鸡胚所收尿囊液抗病毒活性高达2×106~2×107IU/mL。结果表明:SwIFN-α,SwIFN-β,SwIFN-γ在重组新城疫病毒rL-Swineα,rL-Swineβ,rL-Swineγ接种的SPF鸡胚尿囊液中获得良好表达,并具有高效而稳定的抗病毒活性。

实施例1猪α、β、γ干扰素在新城疫病毒LaSota疫苗株中的高效稳定表达及抗病毒活性检测
材料和方法
1材料
表达增强型绿色荧光蛋白报告基因的重组水疱性口炎病毒VSV-EGFP商购自中国农业科学院哈尔滨兽医研究所[10]。BHK-21细胞(ATCCCCL-10)、PK(15)细胞(ATCC CCL-33)、MDBK(NBL-1)细胞(ATCCCCL-22)由本实验室传代培养,培养基为含10%胎牛血清的DMEM(Dulbecco′s Modified Eagle Media);SPF鸡胚商购自中国农业科学院哈尔滨兽医研究所;表达T7聚合酶的痘病毒vTF7-3(ATCC,VR-2153)购自ATCC;新城疫LaSota克隆株rLaSota(按照ZL200510097997.8制备)由本实验室保存;rLaSota反向遗传操作系统基因组转录模板质粒pBRN-FL-PmeI(参见:葛金英,温志远,王永,等。表达绿色荧光蛋白重组新城疫病毒LaSota疫苗株的构建[J].微生物学报,2006,46(4):547~551)(病毒基因组转录质粒pBRN-FL-PmeI,含有新城疫病毒的基因组全长cDNA,pBRN-FL-PmeI的序列信息见SEQ ID No.10,质粒图谱见图4)。
如ZL200510097997.8或参考文献[11]所述,将表达新城疫病毒(基因组序列:GenBank登录号AY845400)的核蛋白(NP)、磷酸蛋白(P)及大聚合酶蛋白(L)基因的开放阅读框架(ORF)cDNA分别紧接着克隆在pBlueScript载体(Clontech)质粒T7启动子下游,分别构建转录辅助质粒pBSNP,pBSP和pBSL(序列分别如图8至10所示)。
2表达猪α、β、γ干扰素基因的全基因组克隆的构建
参照genebank登录号EU364896、EF104599、EU249804委托上海捷瑞生物工程有限公司合成猪干扰素α、β、γ,合成的片段经EcoRV酶切位点连入同样经此酶处理的pBlueScript载体(Clontech)上,质粒命名为pBS-SwIFNα、pBS-SwIFNβ、pBS-SwIFNγ。
以质粒pBS-SwIFNα、pBS-SwIFNβ、pBS-SwIFNγ为模板,通过PCR引物在干扰素基因ORF的5′端引入PmeI限制酶识别序列和NDV自身聚合酶L识别的转录终止序列GE(TTAAGAAAAAA)、转录起始序列GS(ACGGGTAGAA)及增强基因表达的Kozak序列(GCCACC),在干扰素基因ORF的3′端引入PmeI限制酶识别序列(所用引物见表1);为了确保重组基因组cDNA全长总碱基数仍保持6的倍数,在引物GS和Kozak序列之间加入适当数量碱基进行调整。PCR末端经PmeI酶切插入同样经PmeI处理的pBRN-FL-PmeI[4]载体内,表达猪α、β、γ干扰素的重组基因组全长cDNA克隆命名为pBRN-FL-SwIFNα、pBRN-FL-SwIFNβ、pBRN-FL-SwIFNγ(图1,质粒图谱见附图1,2,3)。利用引物NDV-3#PF:5′-gtccgtgcattgatcatg-3′和各基因PCR所用下游引物鉴定方向。
3重组病毒的拯救
BHK-21细胞接种于六孔板内生长达50~80%单层时,转录质粒pBRN-FL-SwIFNα、pBRN-FL-SwIFNβ、pBRN-FL-SwIFNγ分别与转录辅助质粒pBSNP、pBSP和pBSL共转染预先感染vTF7-3的BHK-21细胞。转染上清经0.22um孔径滤器过滤后接种9~11天的SPF鸡胚尿囊腔;接种后的SPF鸡胚继续培养3-5天,取鸡胚尿囊液50μl按常规进行新城疫病毒的血凝(HA)和血凝抑止(HI)试验,详细操作方法参见文献[4]。收获HA及HI试验结果阳性尿囊液,-70℃冻存,并按常规方法分别于9-10日龄鸡胚尿囊腔接种进行传代、病毒滴度测定[4]。重组新城疫LaSota弱毒疫苗株rL-Swine α、rL-Swine β和rL-Swineγ接种的SPF鸡胚尿囊液于2008年7月31日保藏于中国微生物菌种保藏管理委员会普通微生物中心,其保藏号分别为CGMCC No.2615、CGMCC No.2628和CGMCC No.2616。
4重组病毒接种鸡胚尿囊液抗病毒活性检测
分别取rL-Swineα、rL-Swine β、rL-Swineγ以及rLaSota接种的SPF鸡胚尿囊液2ml放入平皿中,置于紫外灯下照射2h,每间隔20min摇动一次,以灭活NDV病毒。经紫外线灭活的尿囊液接种SPF鸡胚以检测病毒灭活是否彻底,每胚100μL,共接3枚,接种后的鸡胚置37℃孵化器内孵化,3天后检测鸡胚尿囊液是否HA阳性。HA阴性样品分装,-70℃保存备用。
用VSV-EGFP-PK和VSV-EGFP-MDBK细胞系统测定rL-Swineα、rL-Swineβ、rL-Swineγ的抗病毒活性,具体为:取灭活后重组病毒rL-Swineα、rL-Swine β、rL-Swine γ鸡胚尿囊液,每个样品100μL,用含5%胎牛血清的DMEM进行10倍系列稀释,在生长于96孔细胞培养板上的PK和MDBK细胞上加入2×101~2×108稀释的尿囊液,同时取相同稀释倍数的灭活rLaSota鸡胚尿囊液作为对照组,于37℃作用24h。吸出上清后,接种VSV-EGFP,剂量为200PFU(噬斑形成单位)/孔感染细胞,感作1h后换完全培养液继续培养,同时设空白对照组,24h后荧光显微镜观察结果。以空白对照组为对照,将减少50%VSV-EGFP感染率的样品的最高稀释度定为1个病毒抑制单位(IU)。
5重组病毒尿囊液抗病毒稳定性检测
取灭活后重组病毒rL-Swine α、rL-Swine β、rL-Swineγ鸡胚尿囊液分装,100μL/管,一部分放4℃冰箱长期保存,每间隔1周取出一份进行抗病毒活性检测;另一部分放-70℃冰箱,取冻融1次、2次、3次样品检测抗病毒活性单位的变化,以检测反复冻融对抗病毒活性的影响。
结果
1表达猪α、β、γ干扰素基因的全基因组克隆的构建及鉴定
通过PCR引物在干扰素基因SwIFNα、SwIFNβ、SwIFNγ的ORF的5′端和3′端引入PmeI限制酶识别序列(所用引物见表1);由于插入的外源基因为PmeI单切连入pBRN-FL-PmeI载体,因此克隆质粒经PmeI酶切的产物应有两条,一条为载体,约为18kb,另一条为外源基因及其所携带的GE/GS等长约500~600bp;而经鉴定引物进行PCR扩增,应得到比相应外源基因大200bp左右的条带,产物约700~800bp,电泳结果显示与预期相符(结果未显示)。证明表达猪α、β、γ干扰素的重组基因组全长cDNA克隆构建成功,构建示意图见图1。

2重组病毒的拯救及鉴定
全基因组转录质粒pBRN-FL-SwIFNα、pBRN-FL-SwIFNβ、pBRN-FL-SwIFNγ分别与转录辅助质粒pBSNP、pBSP和pBSL共转染预先感染vTF7-3的BHK-21细胞。转染上清接种后SPF胚继续培养3-5天,取鸡胚尿囊液50μL按常规进行新城疫病毒的血凝(HA)和血凝抑止(HI)试验。收获HA及HI试验结果阳性尿囊液,-70℃冻存。同时进行RT-PCR鉴定,PCR产物测序结果(数据未显示)证明重组病毒基因组内含有相应的外源基因片段,至此成功拯救出表达猪干扰素α、β、γ的重组新城疫病毒,并分别命名为rL-Swine α、rL-Swineβ和rL-Swineγ。
3rL-Swineα、rL-Swine β和rL-Swine γ抗病毒活性检测
收获的重组病毒尿囊液经紫外线灭活,进行10倍系列稀释,稀释后的样品处理细胞24h,然后按每孔200PFU的接种量感染VSV-EGFP,24h后经荧光显微镜观察结果。2×103倍稀释的rL-Swine α和2×104倍稀释的rL-Swine β可以完全抑制VSV-EGFP的复制,无荧光出现(图2A,D),2×103倍稀释孔的rL-Swine γ仅有数个发荧光细胞,可90%以上抑制VSV-EGFP的复制。2×104倍稀释rL-Swine α和rL-Swine γ以及2×105倍稀释孔的rL-Swine β可部分抑制VSV-EGFP的复制,荧光亮度约为对照组的一半(图2B,E,H),未加干扰素样品的对照孔出现大量荧光(图2L),VSV-EGFP在PK细胞中大量复制。同样2×102倍稀释的rLaSota病毒接种SPF鸡胚尿囊液组也出现大量荧光(图2K),不能抑制VSV-EGFP的复制,但是10倍稀释时可以非特异性的部分抑制VSV-EGFP在PK细胞中的复制(图2J)。测得rL-Swine α、rL-Swine β在PK细胞上的抗病毒活性为2×106IU/mL,rL-Swine γ抗病毒活性为2×105IU/mL。
经同样处理的MDBK细胞接种VSV-EGFP 24h后经荧光显微镜观察结果。2×106倍稀释的rL-Swine α和rL-Swine β以及2×105倍稀释的rL-Swineγ可以抑制90%的VSV-EGFP在MDBK细胞上的复制(图3A,D,G),2×106倍稀释孔的rL-Swine β仅有数个发荧光细胞。2×107倍稀释rL-Swine α和rL-Swine β以及2×106倍稀释孔的rL-Swine γ可部分抑制VSV-EGFP的复制,荧光亮度约为对照组的一半(图3B,E,H),2×108倍稀释rL-Swine α和rL-Swine β以及2×107倍稀释孔的rL-Swine γ与未加干扰素样品的对照孔一样出现大量荧光(图2C,F,I),几乎不能抑制VSV-EGFP在MDBK细胞中大量复制。测得rL-Swine α、rL-Swine β在MDBK细胞上的抗病毒活性为2×108IU/mL,rL-Swine γ抗病毒活性为2×107IU/mL。
4重组病毒尿囊液抗病毒稳定性检测
rL-Swine α、rL-Swine β、rL-Swine γ接种的鸡胚尿囊液经反复冻溶3次后,利用VSV-EGFP-PK细胞系统测定抗病毒活性无明显下降;4℃保存可长达10天其抗病毒活性无明显下降,仍具有高达106TCID50/mL抗病毒活性。可以在传统冷藏条件下长途运输而不影响使用。
当前,猪的病毒性疾病危害严重,而目前尚无特效药物可以治疗病毒性疾病。尽管一些化学合成的抗病毒药物如盐酸金刚烷胺、利巴韦林等对少数病毒病有一定的作用,但由于这些药物在人医上也广泛应用,为防止药物残留及耐药性的产生,目前已禁止在兽医临床上应用;而干扰素具有的生物学活性,及其速效多能的特点使其在防治病毒性传染病,抑制肿瘤生长,促进增重等方面具有重要意义;因此,研究开发的猪基因工程可有效控制当前对我国养猪业危害严重的病毒性疾病,因此,重组猪干扰素具有广阔的应用前景。
目前人重组干扰素和细胞因子产品已广泛用于人类医学临床,但动物干扰素和细胞因子的应用在我国兽医临床中才刚刚起步。正常情况下,动物体内干扰素和细胞因子基因处于封闭状态,只有在病毒感染或诱导下才表达干扰素基因,但是表达量很小,因此单靠病毒感染自身诱导的干扰素对病毒性疾病和细胞内寄生菌和寄生虫病的治疗作用是微乎其微的;而且由于其表达量低,也非常难以提取纯化。因此,在考虑使用干扰素防治动物病毒性疾病时,最好采用基因工程技术生产;采用基因工程技术原核表达技术生产干扰素和细胞因子具有活性效价高、产量大、成本低廉、生产速度快、易于规模化生产等优点,因此可以在生产实践中应用。
近年来分子生物学的迅速发展,干扰素的研究也日益深入,特别是PCR技术的广泛应用,极大地推动了动物干扰素基因的研究,动物干扰素基因的克隆与表达成为研究热点,采用基因工程的方法研制重组干扰素已成为干扰素制剂研究的重点。最初以在大肠杆菌中进行原核表达猪α干扰素为主,表达纯化的蛋白经过复性具有明显的抗病毒生物学活性[5]。证明了原核表达PoIFN-α的可行性及将基因工程原核表达的PoIFN-α真正用于生产实践中作为抗病毒制剂的可行性。但原核表达蛋白经常以包涵体的形式出现,需要经过变性、复性的过程,复性不完全还可能导致干扰素的生物学活性不能正常发挥,而且纯化过程费时、费力,生产成本高限制了干扰素的大规模应用。万建青等[6]利用酵母表达系统表达猪干扰素,尝试真核表达干扰素取得了成功。秦立廷等[7]尝试利用杆状病毒表达系统表达猪γ干扰素,重组杆状病毒感染的昆虫细胞培养上清具有高达105TCID50/mL个抗病毒活性单位。杜以军等[8]利用猪IFN-α的成熟蛋白基因构建了重组腺病毒质粒pAd-poIFN-α转染HEK-293A细胞,滴度为107TCID50/mL。并经RT-PCR证明目的基因在mRNA水平上的有效表达;在PK-15细胞上可以检测到较强的抗猪口蹄疫病毒活性。国内外研究资料显示重组猪干扰素对水泡性口炎病毒(VSV)、流感病毒[9]、传染性胃肠炎病毒(TGEV)[10]、猪繁殖与呼吸障碍综合征病毒(PRRSV)[11]、口蹄疫病毒(FMDV)[12]均有明显的抑制作用。
新城疫病毒反向遗传操作系统的成功建立使NDV作为活载体的应用得到了广泛研究。2001-2002年,Palese.P.[13]等相继构建表达H1亚型流感病毒HA免疫原基因的重组NDV B1株和表达H7亚型流感病毒HA免疫原基因的重组NDVB 1株。2004年,Samal S.[14]等采用LaSota疫苗株为活载体,构建了表达传染性法氏囊病毒(IBDV)变异株GLS-5的重组新城疫病毒,免疫雏鸡可有效抵御IBDV变异株及新城疫强毒的攻击。2007年,Ge J[15]等以LaSota疫苗株为活载体,构建表达H5亚型高致病力禽流感病毒HA基因,免疫雏鸡可100%抵抗H5亚型高致病力禽流感病毒的致死性攻击。
NDV作为载体具有无可比拟的优势,LaSota疫苗株具有良好的安全性和外源基因遗传稳定性,生产成本低廉,表达的外源蛋白产品不仅接近天然状态,而且无须纯化,重复性好便于产业化。为此,在前人研究的基础上,利用新城疫病毒反向遗传操作系统,在新城疫病毒基因组骨架上成功克隆了猪α、β、γ干扰素,并拯救出稳定表达猪α、β、γ干扰素的重组新城疫病毒-rL-Swine α、rL-Swine β和rL-Swine γ。利用VSV-EGFP-PK细胞系统测定rL-Swine α、rL-Swine β、rL-Swine γ接种的鸡胚尿囊液具有高达106TCID50/mL抗病毒活性,经反复冻溶或4℃长期保存其抗病毒活性无明显下降。此外,利用MDBK细胞测定rL-Swine α、rL-Swine β和rL-Swine γ的抗病毒活性,结果证明猪干扰素可以诱导牛细胞产生抗病毒反应,而且活性高达108TCID50/mL,可以同时作为猪、牛通用干扰素应用。以新城疫病毒LaSota株为载体表达猪α、β、γ干扰素既具有高水平抗病毒活性,又便于大规模生产,具有光明的应用前景。

Claims (15)
1.一种表达编码猪干扰素的基因的重组新城疫弱毒疫苗株。
2.根据权利要求1的重组新城疫弱毒疫苗株,其为重组新城疫LaSota弱毒疫苗株。
3.根据权利要求1或2的重组新城疫弱毒疫苗株,其中所述猪干扰素是猪α、β和γ-干扰素,优选所述猪α、β和γ-干扰素的氨基酸序列分别如SEQ ID No.1、SEQ ID No.2和SEQ ID No.3所示,更优选编码所述猪α、β和γ-干扰素的基因如SEQ ID No.4、SEQ ID No.5和SEQ ID No.6所示。
4.根据权利要求1或2的重组新城疫弱毒疫苗株,其为rL-Swine α、rL-Swine β、或rL-Swineγ。
5.根据权利要求1-4中任何一项的重组新城疫弱毒疫苗株在制备猪干扰素中的应用。
6.一种生产权利要求1-4中任何一项的重组新城疫弱毒疫苗株的方法,该方法包括:
(1)构建转录质粒,该转录质粒包括其中插入编码猪干扰素的基因的所述新城疫弱毒疫苗株的基因组cDNA序列;
(2)构建一个或多个转录辅助质粒,该转录辅助质粒包括编码所述新城疫弱毒疫苗株的核蛋白(NP)的cDNA序列、编码所述新城疫弱毒疫苗株的磷酸蛋白(P)的cDNA序列、和编码所述新城疫弱毒疫苗株的大聚合酶蛋白(L)的cDNA序列;
(3)将所述转录质粒和转录辅助质粒共转染所述新城疫弱毒疫苗株复制许可的宿主细胞,培养转染后的宿主细胞;
(4)收获上清液,过滤后继续敏感细胞传代或接种鸡胚尿囊腔救获重组病毒株。
7.根据权利要求6的方法,其中所述重组新城疫弱毒疫苗株是重组新城疫LaSota弱毒疫苗株。
8.根据权利要求6或7的方法,其中所述转录质粒是pBRN-FL-SwIFNα、pBRN-FL-SwIFNβ、或pBRN-FL-SwIFNγ。
9.根据权利要求6或7的方法,其中所述转录辅助质粒是质粒pBSNP、pBSP或pBSL。
10.根据权利要求6-9中任何一项的方法,其中所述宿主细胞是BHK-21。
11.根据权利要求6-10中任何一项的方法制备的重组新城疫弱毒疫苗株,优选为rL-Swine α、rL-Swine β、或rL-Swine γ。
12.一种生产猪干扰素的方法,该方法包括:
1)将根据权利要求1-4中任何一项的重组新城疫弱毒疫苗株接种敏感细胞或接种鸡胚尿囊腔;
2)收获细胞上清液或鸡胚尿囊液。
13.重组新城疫LaSota弱毒疫苗株rL-Swine α的SPF鸡胚尿囊液,其保藏号为CGMCC No.2615。
14.重组新城疫LaSota弱毒疫苗株rL-Swine β的SPF鸡胚尿囊液,其保藏号为CGMCC No.2628。
15.重组新城疫LaSota弱毒疫苗株rL-Swine γ的SPF鸡胚尿囊液,其保藏号为CGMCC No.2616。
