Abstract

技术领域
本发明总体上涉及光学成像、可视化和光疗。更具体而言,本发明涉及将光活性的功能单元结构整合到生物活性的小分子中。
背景技术
说明书通篇中用括号引用出版物。在具体实施方式部分的后面列出了对应于每篇参考文献的完整引用。这些出版物的公开内容通过引用的方式整体纳入本申请中,以便完整地和清楚地描述本发明所属领域的状况。
吸收、发射或散射电磁光谱的可见、近红外(NIR)或长波(UV-A,>300nm)区内光的分子可用于光学层析成像术、光学相干层析成像术、荧光内镜检查术、光声技术、声致荧光技术、光散射技术、激光辅助导引手术(LAGS)和光疗。与荧光现象相关的高灵敏度与核医学的高灵敏度相似,使得器官和组织的可视化成为可能,且没有电离辐射的负面效应。诊断剂和治疗剂(通常称为“半抗原”、“效应物”或“功能单元”),例如荧光团、光敏剂、放射性核素、顺磁剂等,向体内特定位点的靶向递送仍然是各种损伤的诊断、预后和治疗中所迫切需要的(Hassan et al.,Licha et al.,Shah et al.,Vasquez et al.,和Solban et al.)。常规的靶向方法(被称为“生物缀合法”或“侧基设计(pendant design)”)包括将这些试剂连接到靶向体内特定位点的生物活性载体上。在所述生物缀合法中,这两个单元可以独立存在和发挥功能,其中靶向和成像/治疗功能可以是分开的。生物活性载体包括小分子药物、激素、拟肽、酶抑制剂、受体结合剂、受体拮抗剂、受体激动剂、受体调质、DNA结合剂、转录因子、细胞周期机制抑制剂、转导分子、蛋白-蛋白相互作用抑制剂、蛋白-生物大分子相互作用抑制剂、大分子蛋白、多糖、多核苷酸等。在过去的数十年间,所述生物缀合法已经得到了广泛的开发,并且已经在采用中等和大尺寸(大约是分子量>1000道尔顿)的载体时尤其是在肿瘤检测方面取得了适度的成功(Licha et al.和Shah et al.)。这是因为将染料、药物、金属络合物或其他效应物分子连接到大分子载体例如抗体、抗体片段或大的肽上不会显著改变生物活性靶向性质;即所述生物缀合物仍然能够有效地结合受体。然而,该方法确实存在一些严重的限制,因为高分子量生物缀合物向肿瘤细胞的扩散是高度不利的,并且实体瘤中的净正压使得这个问题更加复杂(Jain et al.)。此外,许多染料倾向于在水性介质中形成聚集体,导致荧光淬灭。
因此,需要还具有生物活性靶向能力的光活性小分子。然而,设计小分子生物缀合物时的一个问题是,经常观察到当诊断剂或治疗剂与生物活性靶向载体的大小相似时,所述诊断剂或治疗剂与被靶向的受体的结合受到严重损害(Hunter et al.)。因此,将大的功能单元如染料或光敏剂取代到小分子药物中,是一个难以克服的挑战。为了克服这一难题,已经实施了下述方法(被称为“整合法”或“内在双功能法”),其中将放射性核素金属离子掺入到类固醇或吗啡生物碱骨架中以使得原始药物和对应的放射性核素模拟物的分子拓扑非常相似(Rajagopalan,U.S专利5,330,737;Rajagopalan,U.S专利5,602,236,和Hom et al.)。与上述生物缀合法不同,所述整合单元(例如靶向和成像/治疗)的两个功能是不可分割的。所述整合法是基于这样一种原理,即抗体、酶和受体是多特异性的,并且将会结合任何与天然抗原、底物或配体在拓扑上类似的分子。先前对类固醇模拟物的研究确认了将金属离子整合到天然受体配体中是一种将可用于诊断和治疗的放射性核素选择性递送到靶组织的可行策略(Hom et al.和Skaddan et al.)。这种整合设计将功能单元的单原子同配取代物加入分子骨架中。然而,将大的功能单元如染料或光敏剂取代到小分子药物、肽、伪肽或拟肽中,是一个难以克服的挑战。尽管先前已经报道了核苷向荧光核苷的转化(Miyata et.al.),但是峰值电子光谱(吸收、激发和发射)仍然在UV区。此外,该转化方法限于这种单核苷用途。
发明内容

本发明的一个方面涉及一种无光活性的生物活性分子的整合光活性的类似物的生产方法。所述方法包括用光活性官能团置换无光活性的生物活性分子的无光活性的官能团。

具体实施方式
本发明涉及无光活性的生物活性小分子的整合光活性的类似物(下文中称为“整合光活性的类似物”或简称为“类似物”)的制备方法和用途。整合光活性的类似物可能会通过下述方法来制备,即用具有类似大小和分子拓扑的光活性部分置换无光活性的生物活性小分子的无光活性官能团。所述整合光活性的类似物可被给予患者并且用作生物光学诊断造影剂和/或光疗剂。在一个实施方案中,所述整合光活性的类似物靶向患者体内的特定组织、细胞、受体等。在一个实例中,所述类似物靶向患者体内的患病组织、细胞、受体等。
本发明的整合光活性的类似物的吸收、激发和发射最大波长在350nm或更高波长的近红外(NIR)或可见光谱中。这一点对患者的诊断或治疗处理是有利的,因为当可见光或NIR光被用在生物光学诊断和治疗程序中时它们不太可能会损害组织。相反,波长低于350nm的紫外(UV)光可以导致组织损伤。350nm或更大的较长波长光还能够更深地穿透组织,从而允许对低于350nm的UV波长无法到达的目的组织进行诊断或治疗程序。在一个实施方案中,所述整合光活性的类似物的吸收、激发和发射最大波长介于约400nm和约900nm之间。
两种用于将结构和功能部分整合到单个分子类似物中的通用方法包括:(a)将一种已知的生物活性分子转化为整合光活性的类似物;(b)将一种光活性体转化到为一种具有生物活性的整合了光活性的类似物。在任一方法中,所获得的分子均拥有光活性和生物功能的基本性质。根据结构和功能,本发明的整合光活性的类似物可能会被描述为“整合的荧光团”、“整合的生色团”、“整合的光敏剂”等。所述用于设计整合光活性的类似物的通用方法主要包括:(a)选择所需的生物活性分子;(b)鉴别含有可置换部分(例如芳族或杂芳族部分)的分子区域;和(c)用具有类似大小的光活性官能团置换所述部分,或者对所述部分进行修饰以使其具有光活性。所获得的本发明的整合了光活性的类似物可同时用于诊断和治疗应用。
整合光活性的类似物的合成和用途可以多种方式来加以实施。在一个实施方案中,选择具有已知或所需结构和功能的分子。例如,所选的光活性分子可能会靶向患者体内的特定组织、细胞、蛋白、受体等。鉴别所述分子的分子结构中的无光活性官能团,并且用光活性光能团置换以产生整合光活性的类似物。将所获得的整合光活性的类似物以诊断有效量给予患者,以检测所述患者体内的该光活性分子。在所述类似物与其靶位点结合了一段时间之后,使患者的整个身体或靶组织暴露于具有350-1200nm波长的光之下。在一个实例中,使患者的整个身体或靶组织暴露于具有400-900nm波长的光之下。然后检测由于所述整合光活性的类似物的吸收和激发而从该患者身上发射出的光。由于所述整合光活性的类似物的靶向性质,可以通过评估从该患者身上发出的光的位置和强度作出诊断。
所述整合光活性的类似物还可以用于治疗性地处理遭受病症折磨的患者,所述病症显示出可被所述类似物靶向的患病组织或细胞。在一个实施方案中,所述类似物靶向与目的组织或细胞相连的整联蛋白受体。在另一个实施方案中,所述类似物为靶向微管蛋白聚合剂的光活性抑制剂。在另一个实施方案中,所述类似物为靶向血管破裂剂的光活性抑制剂。在另一个实施方案中,所述类似物为靶向激酶例如β-RAF激酶的光活性抑制剂。
在将所述整合光活性的类似物给予患者之后,所述类似物靶向并且结合所述目的组织、细胞、蛋白或受体。将用于将所述整合光活性的类似物光碎片化/光激发为活性种的合适波长的光给予到患者体内结合的类似物所处的区域。由对所述整合光活性的类似物的光碎片化/光激活所产生的活性种会损害或杀死位于所述结合的类似物附近的患病组织或细胞,从而有益地治疗该患者的病症。
所述整合光活性的类似物的开发包括选择靶向特定组织、器官、损伤、细胞等的合适的生物活性分子。这些生物活性分子包括但不限于各种整联蛋白的选择性拮抗剂,如α2bβ3a、αvβ3、α5β1和αvβ5拮抗剂。被靶向的整联蛋白与许多健康状况有关,如血管发生、CV血管发生、再狭窄、血管障碍、骨质疏松症、癌症、乳癌、卵巢癌、黑素瘤、肿瘤、血栓形成、炎症等,对所述健康状况的诊断或治疗处理是非常需要的。
一旦选择了生物活性小分子,则鉴别位于该分子上的无光活性部分并且用光活性部分对其进行置换。所述分子的任何部分均可以被光活性部分置换,只要所述取代不会导致所获得的光活性类似物的生物活性或生物活性靶向性质的实质性损失。例如,位于靶向特定组织、受体等的分子上的无活性部分可以被光活性部分置换,只要所获得的光活性类似物还优先地靶向特定的组织、细胞、受体等。
同样靶向特定组织、器官、受体等的小分子的片段和/或衍生物也可以被修饰或合成以产生本发明的光活性分子。小分子的片段或衍生物的光活性类似物同样可用于诊断和光疗用途。
本发明的光活性部分的非限制性实例包括但不限于烯烃类、苯类、萘类、萘醌类、芴类、蒽类、蒽醌类、菲类、并四苯类、萘二酮类、吡啶类、喹啉类、喹嗪、喹噁啉类、奎尼丁、蝶啶、异喹啉类、吲哚类、异吲哚类、吡咯类、咪唑类(imidiazoles)、噁唑类、噻唑类、吡唑类、吡嗪类、嘌呤类、苯并咪唑类、呋喃类、苯并呋喃类、二苯呋喃类、咔唑类、吖啶类、吖啶酮类、菲啶类、噻吩类、苯并噻吩类、硫芴类、氧杂蒽类、氧杂蒽酮类、黄酮类、蒽环类抗生素;甘菊环类和氮杂甘菊环类(azaazulenes)、靛青类、苯并卟啉类、方酸类、咕啉类、香豆素类和花青类。可以通过以下方式将这些光活性部分用化学方法转化为具有生物活性的光活性小分子(例如受体结合剂),所述方式即将官能团添加到所述光活性部分上,所述官能团可使所获得的小分子具有生物活性或生物靶向性质。
本发明的光活性部分还包括可用在光疗程序中的反应性物种(或中间产物)。光疗部分包括但不限于自由基、卡宾、氮宾、单线态氧等。可以引入到小分子中以便合成光疗类似物的I型光反应性部分的实例包括但不限于叠氮化物、偶氮化物、重氮化合物、亚磺酸酯、噻二唑类、过氧化物,以及在照射时形成的自由基或活泼中间体。可以引入到小分子中以便合成光疗类似物的II型光反应性部分的实例包括但不限于酞菁、卟啉、延长的卟啉和苯并卟啉。这可以通过用化学方法将酞菁、卟啉、延长的卟啉和/或苯并卟啉体系转化为具有生物活性的物质(例如受体结合剂)来完成。这可以通过将能够使所获得的小分子具有生物活性或生物靶向性质的官能团添加至所述部分上来实施。
在一个实施方案中,本发明的生物活性小分子同时包括光活性部分和光反应性部分。
一旦已经生成整合光活性的类似物,则将其给予个体。给予适宜量的时间以使所述类似物与患者体内的靶组织或细胞等结合。应该理解的是,本发明的化合物和组合物的给药量可由具有合理医学判断的本领域主治医师加以确定。用于任何特定患者的具体有效剂量水平取决于多种因素,包括待治疗的障碍、障碍的严重程度;所采用的具体化合物的活性;所采用的具体组合物,患者的年龄、体重、健康状况、性别及饮食。对所述整合光活性的类似物的检测可通过本领域中已知的光学荧光、吸收或光散射方法使用侵入性或非侵入性探针如内窥镜、导管、夹式耳环、手带、头带、表面线圈、指探针等(Muller et al.)来实现。可以使用平面成像、光学层析成像、光学相干层析成像、内窥镜、光声、声致荧光、共聚焦显微或光散射设备来实现成像。
与上述诊断程序类似,可以将所述整合光活性的类似物给予个体以进行治疗。在将所述整合光活性的类似物给予患者之后,给予所述类似物适宜量的时间以使其与患者体内的靶组织或细胞等结合。任选地按照上文所述对该患者进行成像以确定所述类似物在该患者体内的结合部位。一旦确定所述类似物与一个或多个靶位点结合,则用波长和强度足以导致所述整合光活性的类似物光碎片化的光照射该患者。光碎片化通常导致所述类似物的均裂,导致产生自由基中间体的形成。然后,所产生的自由基会损害所述已经结合了所述整合光活性的类似物的一个或多个靶位点的患病组织或细胞,从而治疗性地处理患者的病症。
在一个实施方案中,所述无光活性部分为位于所述无光活性分子上的芳族或杂芳族部分,所述芳族或杂芳族部分可用光活性的芳族或杂芳族部分置换。在另一个实例中,无光活性的芳族或杂芳族部分可用在环结构内具有与所述无光活性部分相同数目原子的芳族或杂芳族部分置换。在另一个实例中,所述无光活性的芳族或杂芳族部分用吡嗪、甘菊环或氮杂甘菊环部分置换。

制剂
可以本发明的整合光活性的试剂配制为用于肠内(口腔或直肠)、非消化道、局部、经皮肤或皮下给药。局部、经皮肤和表皮递送还可以包括气雾剂、乳膏、凝胶、乳液、溶液或悬液。向皮肤内的递送以及通过皮肤的递送可以用已知的方法和试剂(如经皮肤渗透促进剂,例如“氮酮”、N-烷基环酰胺(N-alkylcyclic amide)、二甲亚砜、长链脂肪酸(C10)等)来增强(Gennaro)。
用于制备可药用制剂的方法可以使用本领域中已知的方法(Gennaro)来实现。使用任何整合的光活性试剂以及可药用的缓冲液、表面活性剂、赋形剂、触变剂、调味剂、稳定剂或透皮增强剂来制备制剂。如果本发明的化合物为水溶性的,则可以给予溶于生理盐水的溶液。如果所述化合物不是水溶性的,则可以将所述化合物溶于生物相容性油(例如豆油、鱼油、维生素E、亚麻子油、植物油、甘油酯、长链脂肪酯等)溶解,用含有表面活性化合物(例如植物或动物磷脂;卵磷脂;长链脂肪盐和醇;聚乙二醇酯和醚等)的水乳化,作为局部用乳膏、悬液、水/油乳液或水/油微乳液来给药。
所述整合光活性的试剂还可以被囊封在胶束、脂质体、纳米颗粒、壳交联纳米颗粒、树枝状聚合物、树枝状晶体(dendron)、微胶囊或其他有序的微颗粒中,并且通过上述任何途径来给药。还可以用化学方法将所述整合光活性的试剂缀合到纳米颗粒、壳交联纳米颗粒、树枝状聚合物或树枝状晶体上,以同时实现整合的光电子效应和多价生物效应。这些制剂还可能会在体内增强所述试剂的稳定性。包囊方法包括去污剂透析、冷冻干燥、成膜或注射(Janoff et al.)。制备脂质体及在其中包囊各种分子的方法在本领域中是公知的(Braun-Falco et al.和Lasic et al.)。
剂量
包含本发明的整合光活性的类似物的组合物可能以单剂量或以多次剂量给药以实现有效的诊断或治疗目的。给药后,所述整合光活性的类似物在靶组织处积聚,并且使所选择的靶位点暴露于具有足够功率和强度的光下以进行诊断和/或治疗。这类剂量可在大泛围内变化,取决于所采用的具体整合光活性的类似物、待检查的器官或组织、在临床步骤中所采用的仪器、治疗所达到的功效等。所述化合物的剂量变化范围为约0.1mg/kg体重-约500mg/kg体重,通常为约0.5mg/kg体重-约2mg/kg体重。对于非消化道给药,无菌溶液或悬液包括浓度范围为约1nM-约0.5M的所述整合光活性的试剂。在另一个实例中,所述无菌溶液或悬液包括浓度范围为约1μM-约10mM的所述整合光活性的试剂。

缩写和定义
为有助于理解本发明,下文中定义了多个术语:
“生物活性小分子”是指分子量通常低于1000道尔顿并且与特定生物靶标(如受体、酶等)结合的分子。
“诊断有效量”是指所讨论的物质的这样的量,即该量在大多数患者体内足够检测到细胞靶组织——如果其存在于被给药的患者体内。术语“有效量”还指所述物质的给药量为仅会在被给药个体体内导致轻度副作用或不会导致副作用,或者按照被给予所述物质的疾病的严重程度从医疗和医药角度来看所述副作用是可耐受的。

“光活性的官能团”或“光活性部分”是指在350-1200nm的波长范围内显示出吸收、激发和发射最大值的任何官能团或部分。这些官能团或部分包括但不限于荧光团、生色团、光敏剂和光反应性部分,其中“荧光团”、“生色团”、“光敏剂”和“光反应性”部分具有本领域中公知的含义。
“光反应性部分”是指在用350-1200nm波长的光激发时经历光化学反应以产生能够导致组织损伤的活性物种的分子部分。
“治疗有效量”是指能够在单独使用每种试剂治疗时实现改善病理状态严重程度和发病频率的目标,同时避免通常与替代疗法相关的有害副作用的每种试剂的量。
“处理”是指任何过程、作用、应用、疗法等,其中向个体包括人提供医疗救助,目的是直接或间接改善个体的病症或者减慢个体病理状态的进展。

实施例1——3,6-二氨基-N2,N5-双(4-甲脒苯甲基)吡嗪-2,5-二甲酰胺的制备

R17和R18各自独立地为氢、C1-C6烷基、C1-C6酰基、C1-C6烷氧基羰基、C1-C6羟烷基、C1-C6聚羟烷基、C1-C6羧基烷基或C1-C6氨基烷基。
18.权利要求17的方法,其中:
R11和R12各自独立地为氢或C1-C6烷基;
Y为-N3或-SOAr;
R13-R16独立地选自氢、C1-C6烷氧基羰基、-F、-CN、-NO2和-NR17R18;并且
R17和R18各自独立地为氢或C1-C6烷基。
22.权利要求1的方法,其中所述整合光活性的类似物具有下述通式:
整合的α2bβ3a光疗剂
其中R29和R30独立地选自氢、C1-C6烷基、C1-C6酰基、C1-C6烷氧基羰基、C1-C6羟烷基、C1-C6聚羟烷基、C1-C6羧基烷基和C1-C6氨基烷基;并且X5为包含与供电子基团连接的I型光反应性基团的取代基。
23.前述权利要求任一项的整合光活性的类似物,用于医疗诊断或光疗程序中的用途。
24.权利要求23的整合光活性的类似物,其中所述医疗诊断程序包括在将所述整合光活性的类似物给药至患者之后,使所述患者暴露于波长为约350nm-约1200nm的光之下。
25.权利要求23的整合光活性的类似物,其中所述医疗诊断程序包括在将所述整合光活性的类似物给药至患者之后,使所述患者暴露于波长为约400nm-约900nm的光之下。
