总而言之,现在已经清楚地知道饮食性硒是人类和动物的健康和福利所必需的。一些研究已经证实有机来源的硒比无机来源的硒有更高的生物利用率。商品化可获得的唯一有机硒来源是富硒酵母制剂。在酵母中,硒主要以富含L-硒代甲硫氨酸的蛋白质的形式存在。尽管硒酵母现在已经被广泛地接受作为饮食性硒的来源,但是它的应用有一些缺点。酵母中有机结合的硒的浓度受限于其从富含亚硒酸盐的 培养基形成L-硒代甲硫氨酸的能力。目前酵母中最高可能的硒浓度为2000μg/g干物质。其次,因为酵母中有机结合的硒是通过生物学过程生成的,而所述生物学过程容易受到大规模生产过程中的细微变化的影响,因此硒化合物的确切组成是可变化的,并且是不容易知道的。偶尔,酵母含有不同浓度的无机硒化合物例如亚硒酸盐和硒酸盐。第三,酵母中存在的有机硒化合物是作为胞内蛋白质的一部分。在这些化合物被消化后可吸收之前,酵母的细胞壁必须破裂以便将蛋白质释放到动物的胃肠道内,在这里所述蛋白质可以接受消化酶的蛋白水解作用。只有当蛋白质被水解成单氨基酸或二肽后,硒化合物才能被吸收。从完整的酵母细胞将硒化合物释放为单氨基酸或二肽是不完全的,并高度依赖于消化道中的条件。因为这些缺点,特别需要开发出富硒酵母的替代品用作容易进行生物利用的饮食性硒的来源。本申请人更早期的专利US 6,911,550涉及络盐。本发明的改进涉及某些非常稳定的酯和有机衍生物。
最近,对具有提高的生物利用率的用作人和家畜补充剂的饮食性硒来源的需求有所增加。最近已经可以以合理价格商品化地获得合成的硒代氨基酸。但是,这些氨基酸具有低水溶性,且它们的结晶具有疏水特性,该性能导致低溶解速度。低溶解度和低溶解速度降低了这些化合物在饲喂给动物后的生物利用率。本发明的一个主要目的是鉴定具有提高的生物利用率的硒代氨基酸的衍生物,并制备所述衍生物。
硒和硫一样都是第VIA族元素的成员。存在着不同同素异形形式的硒,其具有-2、0、+2、+4、和+6价氧化态。硒是非金属元素。它可以形成单原子阴离子,并从而可以形成离子键和共价键。在-2价氧化态时,硒与碳取代基形成共价键,并常常可以取代天然存在的化合物中的硫。硒的生物学作用归功于这些天然存在的化合物,在所述化合物中硒以-2价氧化态存在,并常常与碳共价结合作为功能蛋白 质的一部分。已经提出将硒代氨基酸作为硒的饮食性来源。但是,现在认识到动物的营养状态和饮食组成以及消化道内容物都可能会显著地降低这些化合物的生物利用率。因此,需要开发硒代氨基酸的衍生物,这可以提高这些氨基酸的生物利用率。在先前的专利(US6,911,550)中,本申请的发明人描述了具有提高的生物利用率的可逆的硒代氨基酸衍生物。这些可逆的衍生物是硒代氨基酸(例如L-硒代甲硫氨酸)的1∶1锌复合物。本发明的主要目的是制备具有提高的生物利用率的新的不可逆的硒代氨基酸衍生物。通过形成α氨基和/或羧基和保护基团之间的共价键对硒代氨基酸进行化学修饰形成这些新化合物。这些化学稳定的化合物在被动物摄入后被酶促修饰成硒代氨基酸。
本发明的另一个目的是描述制备这些衍生物的方法以及它们作为家畜饲料成分的用途。
发明概述
制备作为人和家畜的补充硒的有效饮食性来源的新的硒代氨基酸衍生物。所述新的衍生物相对于亲代硒代氨基酸具有改善的物理学、化学或生物学特性。这些衍生物相对于硒代氨基酸具有增强的生物利用率和/或增加的稳定性。它们是硒代氨基酸例如L-硒代甲硫氨酸的1∶1复合物。
具体实施方式
由于现有可用于饲料添加剂的硒来源的令人不满意的性能,因此有必要开发具有提高的生物利用率的硒代甲硫氨酸的衍生物。所述新的衍生物所需特性包括:
1.所述衍生物必须是容易生物利用的硒来源。
2.所述衍生物必须比亲代化合物更稳定。
3.所述衍生物的物理学特性例如溶解度、溶解速度和气味比亲代化合物更为有利。
4.可以通过利用商品化可获得的试剂并以合理的代价从亲代化合物容易地制备所述衍生物。
5.认识到所有含硒化合物都有着窄的安全性范围,所述衍生物必须和亲代化合物一样安全。
6.所述衍生物在瘤胃内容物中必须是稳定的,从而使得它可以用作反刍动物的硒来源。
发现其他商品化可获得的硒代氨基酸(例如甲基-L-硒代半胱氨酸)具有与L-硒代甲硫氨酸相似的不需要的物理学特性。因此,也制备了这些硒代氨基酸的衍生物。发现这些衍生物具有与硒代甲硫氨酸相似的物理学特性。
一组硒代氨基酸衍生物是简单的脂肪族酯例如甲酯、乙酯、丙酯和异丙酯。在这个组中,异丙酯是优选的化合物。在存在适当的催化剂或偶联剂的情况下,通过硒代氨基酸和异丙醇的反应可以容易地制备这些酯。这些催化剂或偶联剂包括浓硫酸和亚硫酰氯。通常以盐酸化物的形式分离出氨基酸酯。L-硒代甲硫氨酸异丙酯盐酸化物易溶于水,并且在固体状态以及在溶液中都比L-硒代甲硫氨酸明显更为稳定。这些衍生物比亲代硒代氨基酸具有更高的脂溶性,并可以在pH>5.0时通过被动扩散自肠内容物中被快速地吸收。
第二组研究的衍生物是硒代氨基酸的N-琥珀酰衍生物。通过硒代氨基酸与琥珀酐的反应可以容易地获得这些化合物。这些化合物是部分解离的酸,因为硒代氨基酸的α-氨基受到掩蔽。可以分离这些化合物并容易地纯化成其盐的形式。可以制备钾盐、钠盐、钙盐或镁盐。N-琥珀酰L-硒代甲硫氨酸的钠盐易溶于水。其在固体状态或在溶液中都比L-硒代甲硫氨酸明显更为稳定。这些衍生物比亲代硒代氨基酸具有更高的脂溶性,并且可以在pH<3.0时通过被动扩散自胃肠内 容物中被快速地吸收。
第三组研究的衍生物是硒代氨基酸的N-氨甲酰衍生物和乙内酰脲衍生物。通过L-硒代甲硫氨酸和氰酸钾于90℃在水溶液中反应可以获得N-氨甲酰L-硒代甲硫氨酸。在3N盐酸中加热N-氨甲酰衍生物提供L-硒代甲硫氨酸乙内酰脲。N-氨甲酰衍生物的水溶性较高,并且其溶液也比亲代硒代氨基酸更为稳定。乙内酰脲水溶性较低,但是比亲代硒代氨基酸更为稳定。
上述化合物都是硒代氨基酸的可逆衍生物。在经动物消化之后,预计它们可通过酶催化反应容易地转化为亲代硒代氨基酸。例如,预期通过存在于血液以及其他组织例如肝脏中的酯酶可以容易地水解L-硒代甲硫氨酸异丙酯。酯在pH为7.4的血浆中的非酶促水解也是可能的。通过血浆和肝脏中的酰胺酶可以酶促水解N-琥珀酰衍生物。
可以将本发明所描述的硒代氨基酸衍生物添加到固体或液体饲料中用作容易利用的硒来源。添加的化合物的量将取决于所要供应的动物。对于猪和家禽,日粮中要补充0.05-2.00ppm的硒,优选0.1-0.3ppm的硒。对于牛,饲料中要补充0.05-10mg的硒/头/天,优选2-7mg的硒/头/天。
提供以下实施例,以举例说明获得这些化合物的实用方法、它们的特性、以及它们作为动物营养中硒的来源的应用。
实施例1:L-硒代甲硫氨酸异丙酯盐酸化物(化合物1)的制备。
向1000ml圆底烧瓶内加入150ml异丙醇。将烧瓶放置在冰水浴中,并仔细地逐滴加入浓硫酸(43.208g,工业级,最小纯度93%),并持续搅拌。持续搅拌下仔细加入L-硒代甲硫氨酸(66.962g,0.338摩尔)。将Soxhlet提取管连接到烧瓶的顶部。将带有充满3A分子筛的烧结盘的玻璃提取筒放入到提取管内。加入异丙醇填满提取管。连接回流冷凝器和提取管。通过加热罩使混合物加热,引起异丙醇的轻 微回流。在回流下加热反应混合物48小时。停止加热,并将烧瓶放置于冰水浴中。持续混合下缓慢地加入氢氧化铵溶液。形成了大量的白色沉淀。过滤混合物,并用异丙醇洗涤沉淀。在减压条件下浓缩合并的滤液和洗涤液,得到稠油。将残渣溶解在100ml乙酸乙酯中。将乙酸乙酯溶液转移到分液漏斗中,并用稀释的氢氧化铵溶液和盐水(Brine)相继进行抽提。在无水硫酸镁上干燥乙酸乙酯抽提液,过滤,在减压条件下除去溶剂,得到黄色的稠油(42.337g,产率52.61%)。将油溶解在200ml异丙醇中,并加入浓盐酸(20g)。在减压条件下浓缩混合物,将残渣溶解在最小量的乙酸乙酯中。逐滴加入无水乙醚直到出现浑浊。将混合物在冰箱内存放4天。过滤出白色结晶沉淀,并用无水乙醚洗涤。

利用UV/Vis检测器于210nm通过HPLC分析含有1mg/ml L-硒代甲硫氨酸异丙酯盐酸化物的水溶液,并通过Rheodyne Loop注射器向柱头上注射20μl样品。使用250×4.6mm Discovery Cyano柱(Supelco),以1ml/min的0.1%乙酸作为流动相。L-硒代甲硫氨酸异丙酯盐酸化物的滞留时间为4.467分。L-硒代甲硫氨酸在这一系统中的滞留时间为4.167分。用L-硒代甲硫氨酸异丙酯盐酸化物获得了占超过99%的检测器响应的单峰。这一系统可用于测定预混合物中的L- 硒代甲硫氨酸异丙酯盐酸化物。
实施例2:N-琥珀酰L-硒代甲硫氨酸(化合物2)的制备。
给250ml 3颈圆底烧瓶装配温度计、回流冷凝器和加料漏斗。将乙酸乙酯(75ml)放入烧瓶内。在研钵内将琥珀酐(12.404g)精细地碾成粉末,并将其添加至烧瓶内的乙酸乙酯中。用磁性搅拌器搅拌混合物,直到所有固体全部溶解。加入L-硒代甲硫氨酸(19.630g,0.1摩尔)。稀释硫酸(用5份水稀释1份浓硫酸得到1.0ml溶液)。在回流并持续搅拌的条件下加热混合物1小时。过滤热的澄清溶液。当过滤液冷却时,形成白色结晶沉淀。沉淀称重为24.92g(产率84.14%)。

实施例3:N-氨甲酰L-硒代甲硫氨酸(化合物3)的制备
给250ml 3颈圆底烧瓶装配温度计、回流冷凝器和加料漏斗。将水(40ml)放入烧瓶内。将氰酸钾(9.735g,0.115摩尔)加入烧瓶内的水中,用磁性搅拌器搅拌冷却的混合物直至所有固体全部溶解。加入L-硒代甲硫氨酸(19.815g,0.1摩尔)。在回流并剧烈搅拌的条件下加热混合物。使内部温度达到94℃,然后下降到80-85℃。将反应混合物于80-85℃保持2小时。将所获得的澄清溶液冷却至室温。在持续搅拌的条件下,缓慢地加入盐酸(11.272g,0.115摩尔)。形成了大块白色结晶沉淀,减压过滤。沉淀物称重为20g(产率83.65%)。

实施例4:L-硒代甲硫氨酸乙内酰脲(化合物4)的制备。
给250ml 3颈圆底烧瓶装配温度计、回流冷凝器和加料漏斗。将水(40ml)放入到烧瓶内。将N氨甲酰L-硒代甲硫氨酸(11.969g,0.05摩尔)加入烧瓶内的水中,边冷却边用磁性搅拌器搅拌混合物。缓慢加入盐酸(14.599g,0.15摩尔)。在回流并剧烈搅拌的条件下加热混合物2小时。过滤澄清的热溶液,然后冷却到室温。形成大块白色结晶沉淀,并在减压下过滤。沉淀称重为8.72g(产率78.88%)。

实施例5:比较亚硒酸钠和N-琥珀酰L-硒代甲硫氨酸(化合物2)对泌乳奶牛的组织硒含量以及全血谷胱甘肽过氧化物酶活性的影响:
制备用于泌乳奶牛现场研究的三种预混料。其中一种预混料不含额外的硒来源,因此被用作安慰剂。第二种预混料含有亚硒酸钠,以及第三种预混料含有N-琥珀酰L-硒代甲硫氨酸(化合物2)。通过混 合一定量的硒来源和充足量的精制糖制备每种预混料,使其含有250ppm硒。每种预混料通过在制剂过程中掺入食用色素溶液进行颜色鉴定,并用随机选择的字母命名。把预混料提供给不知情的动物营养学家,即他们不知道每种预混料中的硒来源。这样就可以避免由于对饲养实验结果的解读所带来的任何可能的偏差。
每天给30只泌乳奶牛饲喂其中一种预混料。测定这些硒来源对组织硒含量和全血谷胱甘肽过氧化物酶活性的影响。在最初的8周消除期内,给所有奶牛都饲喂未添加硒的全混合日粮。每日定量供应一定量的预混料,以提供7.5mg硒。持续处理8周,然后经过4周的消除期。在首个消除期(0周)前1周开始每周1天收集乳样,持续试验的整个20周。在给样品脱脂后,测定所获得的乳清中的硒含量。表1给出了0、8、12、16和20周时乳清中的硒浓度。在首个消除期(0周)前1周开始每周1天(4周的间隔期,持续整个试验)收集血液样品(表1中的8、12、16和20周)。将血液样品抽到无微量元素的含抗凝剂的真空试管内。分析全血等分试样的硒含量以及谷胱甘肽过氧化物酶活性。离心剩余的血液等分试样收获血浆,并测定血浆样品中的硒含量。在首个消除期(0周)前1周开始每周1天(4周的间隔期,持续整个试验)通过活体解剖取得肝脏样品(表1中的8、12、16和20周)。分析肝脏样品中的硒含量。表1给出了试验的结果。表1中的结果显示在限制硒摄入8周(Wk 8)后,乳清、血浆和肝脏中的硒浓度都要明显低于消除期开始前(Wk 0)的硒浓度。给奶牛饲喂不含补充硒的混合日粮(安慰剂)造成硒浓度的轻微增高,但是没有完全恢复到Wk 0时的基础水平。但是,饲喂补充了亚硒酸钠或N-琥珀酰L-硒代甲硫氨酸(化合物2)的日粮造成这些组织中的硒浓度的进行性和显著提高(Wk 12和Wk 16)。在第2个消除期结束时(Wk 20),所有组织中的硒浓度都显著降低。响应日粮硒摄入的变化而发生的硒浓度的显著变化说明这些组织是泌乳奶牛的日粮硒状态的敏感指示物。 值得注意的是,化合物2比亚硒酸钠引起这3种组织中硒浓度在统计学意义上显著更高的增加,说明N-琥珀酰L-硒代甲硫氨酸是比亚硒酸钠具有更高生物利用率的日粮硒的来源。
响应日粮硒摄入的变化而发生的全血中硒浓度和谷胱甘肽过氧化物酶活性(GPX)的变化相对于乳清、血浆和肝脏中的改变更不灵敏。这说明这些参数不是泌乳奶牛的硒状态的有用指示物。
