Abstract
本发明公开了一种全血DNA的提取方法,解决了目前提取全血DNA时提取率不高的缺点,本发明的步骤为:将血置于离心管中,加入含有蛋白酶K的缓冲液进行酶切,离心,加入酚,再离心,然后用酚-氯仿混合物提取2~3次,即得DNA;本发明所用的蛋白酶K的缓冲液为含有20mmol/L Tris.Cl(pH8.0)、200mmol/L EDTA(pH8.0)、1%SDS、240ug/ml蛋白酶K的水溶液。本发明具有简单、有效、提取率高等优点。

本发明涉及一种全血DNA的提取方法。
背景技术
目前,提取全血DNA的方法一般为:首先用红细胞裂解液破碎红细胞,然后通过离心使白细胞沉淀,然后反复洗涤以弃去血红蛋白,虽然这样能提取出纯度较高的DNA,但是在破碎、离心过程中有相当数量的白细胞亦遭受破坏,尤其是冻存多年的陈旧血液;由于数量不等的白细胞随着血红蛋白一起被丢弃,因而造成了DNA的大量损失;有些血液样本资源十分宝贵,来源有限,不可再生,丢失会造成无可估量向的损失,其后果不可挽回。因此,急需一种既能保证纯度、提取率又高的方法来提取全血DNA。
发明内容
针对上述现有技术的不足,本发明提供了一种提取率高的全血DNA的提取方法。
本发明是通过以下技术方案实现的:
一种全血DNA高产出率的提取方法,包括以下步骤:
(1).取冻融处理的ACD抗凝血,置于离心试管中,加入含有240ug/ml蛋白酶K的缓冲液,振荡混匀,置于50℃水浴20~28小时,水浴过程中缓慢摇动离心管;
(2).酶切完成后,向离心管中加入等体积的pH为8.0的重蒸饱和酚,盖紧管盖,反复颠倒或用DNA混合仪转动混匀,直至两相混合均匀;
(3).把离心管置于13000rpm下离心5分钟,轻轻移动试管,用移液器将上清液体移至另一个尖底离心管中;
(4).重复步骤(2)(3);
(5).向上述离心管中加入酚,再加入与酚等体积的氯仿或直接加入事先混好的1∶1的酚-氯仿混合物,盖紧管盖,反复颠倒或用DNA混合仪转动混匀,至两相混合均匀;
(6).用移液器将上清液移至另一个尖底试管中;
(7).向上述溶液中加入氯仿,盖紧管盖,反复颠倒或用DNA混合仪转动混匀,至两相混合均匀;
(8).将上清液转移至尖底试管中,加入-20℃的无水乙醇,颠倒混匀;
(9).把离心管置入高速离心机,以13000rpm离心8分钟,打开试管盖,将试管口朝下,轻轻地倒掉上清液;
(10).加入70%乙醇,清洗一次,把离心管放入高速离心机以13000rpm离心8分钟,打开试管盖,反转试管口朝下,轻轻倒掉上清液;
(11).将尖底管倒扣于吸水纸上,吸干残留液体后平卧,室温晾干;
(12).晾干后加入双蒸水,37℃保温或常温下过夜,使DNA溶解;
(13).将溶解后的DNA进行浓度测定,然后置于-20℃冰冻保存。
所述含有240ug/ml蛋白酶K的缓冲液为含有20mmol/L Tris.Cl(pH8.0)、200mmol/L EDTA(pH8.0)、1%SDS、240ug/ml蛋白酶K的水溶液。
本发明的提取方法省略了可致白细胞丢失的裂解红细胞过程,利用特制DNA提取溶液直接对全血成份进行消化处理,然后对消化液用酚、酚-氯仿混合液以及氯仿进行抽提,减少了白细胞的损失,大大提高了DNA的产量,基本做到无损失全量提取DNA。在有些情况下,DNA是一种极为宝贵的、不可再生的资源,尤其是那些已经去世的具有宝贵研究价值的遗传病患者,所留下的血液样本量极为有限,若采用常规的DNA的提取方法提取DNA,由于提取率低,会造成不可估量的损失,而采用本发明的方法则可取得较好的效果。本发明具有效果好、操作简单等优点。
具体实施方式
实施例:提取全血DNA,步骤如下:
(1).取冻融处理的ACD(柠檬酸、柠檬钠、葡萄糖)抗凝血100ul,置于1.5ml离心试管中,加入400ul含有240ug/ml蛋白酶K的缓冲液,振荡混匀,置于50℃水浴24小时,进行酶切,水浴过程中缓慢摇动离心管。
(2).酶切完成后,含有血液的混浊的溶液会变成清澈透明的红葡萄酒状,这时向离心管中加入等体积(约500ul)的pH为8.0重蒸饱和酚,盖紧管盖,反复颠倒20次左右或用DNA混合仪转动混匀10分钟,直至两相混合均匀,切忌使用振荡器或用力过猛。
(3).把离心管置于13000rpm下离心5分钟,轻轻移动试管,避免搅动界面,用1ml吸头的移液器将上清液体移至另一个1.5ml的尖底离心管中,为了最大限度回收DNA,可以稍稍吸带下层的酚。
(4).重复步骤(2)(3)。
(5).向上述离心管中加入250ul的酚,再加入250ul氯仿或直接加入500ul事先混好的1∶1的酚-氯仿混合物,盖紧管盖,反复颠倒20次左右或用DNA混合仪转动混匀10分钟,直至两相混合均匀。
(6).用1ml吸头的移液器将上清液移至另一个1.5ml的尖底试管中,不要吸入下层的酚。
(7).向上述溶液中加入500ul氯仿,盖紧管盖,反复颠倒20次左右或用DNA混合仪转动混匀10分钟,直至两相混合均匀。
(8).将上清液转移至1.5ml的尖底试管中,加入1ml-20℃的无水乙醇,颠倒混匀,这时会看到呈棉絮状析出的DNA。
(9).把离心管置入高速离心机,以13000rpm离心8分钟,打开试管盖,将试管口朝下,轻轻地倒掉上清液。
(10).加入1ml70%乙醇,清洗一次,把离心管放入高速离心机以13000rpm离心8分钟,打开试管盖,反转试管口朝下,轻轻倒掉上清液。
(11).将尖底管倒扣于吸水纸上,吸干残留液体后平卧,室温晾干10-15分钟。
(12).晾干后加入双蒸水(ddH2O)100-500ul,37℃保温3小时或常温下过夜,使DNA溶解。
(13).将溶解后的DNA进行浓度测定,然后置于-20℃冰冻保存。
所述含有240ug/ml蛋白酶K的缓冲液为含有20mmol/L Tris.Cl(pH8.0)、200mmol/LDTA(pH8.0)、1%SDS、240ug/ml蛋白酶K的水溶液。
采用传统酚~氯仿提取法提取全血DNA,与本发明方法提取相同血样DNA,1ml血得到DNA的量的比较:
应用独立样本T检验,两总体样本均为II型糖尿病病人血样,统计结果如下:

设置置信水平为α=0.05,临界置信水平为0.000,远小于设置的置信水平,因此两种方法提取DNA平均1ml血得到DNA的量有显著性差异。由数据可知,用本发明的提取方法的提取率高,几乎可达100%。由表中数据可知,本发明的方法所提取的DNA与传统方法在OD260/280的比值方面略有下降,然而这并不影响所获得的DNA分子在常规的分子生物学操作的实验结果,因此这种变化是可以忽略的。
所用样本数据见下表1、2。

Claims (2)
1.一种全血DNA的提取方法,其特征在于:包括以下步骤:
(1).取冻融处理的ACD抗凝血,置于离心试管中,加入含有240ug/ml蛋白酶K的缓冲液,振荡混匀,置于50℃水浴20~28小时,水浴过程中缓慢摇动离心管;
(2).酶切完成后,向离心管中加入等体积的pH为8.0的重蒸饱和酚,盖紧管盖,反复颠倒或用DNA混合仪转动混匀,直至两相混合均匀;

(10).加入70%乙醇,清洗一次,把离心管放入高速离心机以13000rpm离心8分钟,打开试管盖,反转试管口朝下,轻轻倒掉上清液;
(11).将尖底管倒扣于吸水纸上,吸干残留液体后平卧,室温晾干;
(12).晾干后加入双蒸水,37℃保温或常温下过夜,使DNA溶解;
(13).将溶解后的DNA进行浓度测定,然后置于-20℃冰冻保存。
2.根据权利要求1所述的一种全血DNA的提取方法,其特征在于:所述缓冲液为含有20mmol/LTris.Cl、200mmol/L EDTA、1%SDS、240ug/ml蛋白酶K的水溶液。
