Description
技术领域
本发明主要涉及一种新的微生物株类干酪乳杆菌(Lactobacillusparacasei)GM-080及其用来刺激IFN-γ分泌和治疗过敏相关疾病的用途。
背景技术
过敏是指对正常无害物质产生免疫介导的不良反应的后天潜力。过敏反应引发例如发痒、咳嗽、气喘、打喷嚏、流泪、炎症及疲劳等症状。一般认为过敏反应包括早期特异性免疫应答与后期炎症反应。据报导,过敏原(例如花粉与尘螨)通过刺激高亲和力免疫球蛋白(IgE)受体来介导早期过敏。例如,当肥大细胞与嗜碱性粒细胞受到过敏原刺激时,将会释放组胺和细胞因子(cytokines)。接着,肥大细胞与嗜碱性粒细胞所释放的细胞因子通过恢复炎症细胞来介导后期过敏。另据报导,嗜酸性粒细胞、巨噬细胞、淋巴细胞、中性粒细胞和血小板的流入开始了恶性炎症循环。此后期过敏扩大了初始免疫应答,其又触发更多炎症细胞的释放(Blease等人,Chemokines and their role inairway hyper-reactivity。Respir Res 2000;1:54-61)。
为治疗该过敏症状,已寻求各种治疗手段。其中,已经使用了抗过敏剂与组胺H受体拮抗剂(抗组胺)。投药组胺拮抗剂以对抗因应答过敏原的存在而由肥大细胞释放之组胺的作用。它们减少了由目标组织上的组胺作用而引起的发红(redness)、发痒及肿胀,且用于防止或缓解多种由肥大细胞脱粒而引起的症状。但是,抗组胺也与诸如降低的敏捷性,减慢的反应次数及嗜睡等不良反应有关(美国专利第6,225,332号)。
同样也有通过调整细胞因子(Cytokines)以治疗过敏的报导。其中,干扰素-γ(IFN-γ)被发现用以抑制Th2淋巴细胞中细胞因子的过表达(over-expression),尤其抑制IL-4的分泌以降低B细胞的增殖。IFN-γ也可刺激Th1的免疫应答和抑制IgE的合成(Sareneva T等人,Influenza A virus-induced IFN-α/β and IL-18 synergisticallyenhance IFN-γ gene expression in human T cells,J Immunol 1998;160:6032-6038;Shida K等人,Lactobacillus casei inhibits antigen-inducedIgE secretion through regulation of cytokine production in murinesplenocyte cultures,Int Arch Allergy Immunol 1998;115:278-287)。因IFN-γ可抑制B细胞增殖和IgE分泌,因此相信IFN-γ对治疗过敏有效。
乳杆菌(其为革兰氏阳性细菌)是普遍使用于工业食品发酵。新近研究表明乳杆菌可刺激细胞之IFN-γ分泌(Contractor NV等人,Lymphoid hyperplasia,autoimmunity and compromised intestinalintraepithelial lymphocyte development in colitis-free gnotobioticIL-2-deficient mice.J Immunol 1998;160:385-394)。某些特殊乳杆菌(例如乳酸双歧杆菌(Bifidobacterium lactis)和短乳杆菌亚种(Lactobacillus brevis subsp.))被发现可刺激来源于小鼠和人类血液中的淋巴细胞的IFN-γ分泌(美国专利公告号:US 2002/0031503A1;美国专利第5,556,785号)。也据报导,乳杆菌可刺激来源自人类或小鼠之淋巴细胞分泌介白素12(IL-12),其为可激活T细胞和NK细胞来分泌IFN-γ的T细胞刺激性细胞因子(Hessle等人,Lactobacilli fromhuman gastrointestinal mucosa are strong stimulators of IL-12production,Clin Exp Immunol 1999;116:276-282)。

具体实施方式
本发明提供一种新颖微生物株类干酪乳杆菌GM-080,其能用于治疗过敏。该株GM-080于2004年2月19日保藏于中国典型培养物保藏中心,入藏登记号(accession number)为CCTCC M 204012。
类干酪乳杆菌GM-080是分离自健康人体胃肠道(GI tract)。使从胃、肠或十二指肠取出的组织样品悬浮于在37℃下培养了2天的含100g/mL氨苄西林(ampicillin)的MRS肉汤培养基(broth medium)中,接着在琼脂板上划线接种(streak plating)。于该板上生长的乳杆菌菌落可在显微镜检查之下预先筛过。接着将候选株与脾细胞一起培养。测定在肉汤中由脾细胞如此所产生的IFN-γ的量。接着,GM-080因其IFN-γ高生产力被选中。
GM-080之真菌学特征如下所示:

确定GM-080的16SrDNA序列分析。结果表明GM-080是与其它类干酪乳杆菌菌株高度同源(如图2所示)。此外,种系发生间距树显示于图3。此外,显示了随机扩增的多形态DNA(RAPD分析)。
其表明GM-080属于类干酪乳杆菌,但具有特定的16SrDNA序列。综上所述,GM-080是一种新颖类干酪乳杆菌菌株。
(e)GM-080的细胞壁蛋白:
当与其它熟知的类干酪乳杆菌菌株比较时,GM-080的细胞壁蛋白表明具有相似的图案。GM-080的细胞壁蛋白的SDS-PAGE图案显示于图4。
(f)用于鉴别GM-080的标准化检测系统:
鉴别微生物的标准化检测系统于2003年5月29日所申请的美国专利第10/446,781号中有揭示,其利用了以特定微生物培养与未用特定微生物培养的测试细胞系之间的基因表达差异来作为鉴别用标记。所测试之基因如表2所列。

GM-080在经HCl溶液(pH2.0)处理3小时且接着经胆汁处理4小时之后呈活性。因此GM-080被认为在消化作用中保持活性。GM-080从一健康受检者上分离,而且是安全、天然、无毒的,且符合G.R.A.S.(Generally Regarded as Safe,公认安全级)标准。
此外,GM-080强力粘着于肠上皮细胞。综上所述,GM-080可停留在肠中更长的时间,以起到调整生理机能的作用。通过占据肠上皮细胞的粘着位置,GM-080也会阻碍其它病原细菌粘着至肠。GM-080被视作良好的益生菌。
根据本发明,发现GM-080刺激IFN-γ分泌,且可用于治疗过敏相关疾病。
本发明的一方面提供了一种包含GM-080的组合物。优选将该包含GM-080的组合物用来刺激IFN-γ分泌,其有助于治疗过敏相关疾病。
本文所使用术语“过敏相关疾病”是指其中对一般无害环境抗原产生系统性反应的疾病,该反应起因于抗原与抗体或由较早暴露于该相同抗原所产生的T细胞之间的相互作用。本文所使用的术语“过敏反应”是指因预先存在的抗体或T细胞而对无害环境抗原或过敏原所产生的应答。存在过敏反应的各种免疫机制,但是最普通类型为过敏原到肥大细胞上的IgE抗体之结合,其引发哮喘、枯草热(hay fever),及其它普通过敏反应。过敏相关疾病包括气道高反应性和炎症、异位性皮肤炎、过敏性结膜炎、鼻炎、窦炎、过敏性肺炎、外源性过敏性肺泡炎、荨麻疹、湿疹、过敏性反应、血管性水肿、过敏性偏头痛、特定胃肠失调和哮喘。根据本发明的一个优选实施例,过敏相关疾病为气道高反应性和炎症。另一方面,过敏相关疾病是与诸如花粉、霉菌、动物鳞屑和昆虫的空气传播过敏原(气源性过敏原)有关。
本文所使用术语“气源性过敏原”定义为具有至少以下特征:可引发活性反应素应答的特异抗原分组,且能导过敏感体内明显组织变化的环境曝光水平。气源性过敏原是空气传播粒子,其可引起呼吸、皮肤或结膜过敏。豚草花粉(ragweed pollen)的水溶性部分(例如)影响呼吸和结膜粘膜,而豚草花粉的脂溶性过敏原可导致在暴露皮肤上引起典型接触性皮炎。
选择GM-080以当其与脾细胞和外周血单核细胞(PBMC)在活体外共同培育时具有刺激IFN-γ分泌的能力。此外,在根据本发明的模型中,观察到经气源性过敏原激活并接着经GM-080治疗的动物增加了IFN-γ分泌。此外,治疗后气源性过敏原特异IgE的量得到显著降低。另一方面,过敏原特异IgG的量在治疗前与治疗后并未显示显著差别。另外,支气管肺泡灌洗液(BALF)中的嗜酸性粒细胞计数急剧减少;但是BALF中巨嗜细胞和淋巴细胞计数急剧增加。其证实炎症得到了减轻。
根据本发明,用于过敏治疗的GM-080可为活的或灭活的。GM-080优选是灭活的。例如,活的细菌株可经加热步骤或此项技术中通常所用的其它处理方法所处理,来杀灭乳杆菌成为灭活的菌株。
根据本发明,该乳杆菌菌株可被包括在药物组合物、饮食添加剂、食品、健康食品、医疗食品或其组份中,它们一般为人所服用。在本发明的一个优选实施例中该乳杆菌菌株可以以食品形式来传递,例如以通过牛奶中乳酸发酵而制得的凝结牛奶产品的形式。根据本发明而制得的食品产品可便利地为婴儿或儿童所服用。
本发明的另一方面提供用来刺激患者IFN-γ分泌的方法,该方法包含使该患者服用包含经分离的微生物GM-080的组合物。
仅为达成说明的目的而并无意限制本发明的范围,举出以下实施例。
实施例1:类干酪乳杆菌GM-080的分离
样本:通过内窥镜取得的一片人的胃、肠或十二指肠组织在37℃下,2mL含100μg/mL氨苄西林的乳杆菌MRS肉汤(0881)中培养约2天。将肉汤覆于含CaCO3的MRS琼脂上且在37℃下培育两天。选择生长于板上的单个菌落,并使其经受革兰氏染色。接着选出革兰氏阳性细菌。所有株在37℃下乳杆菌MRS肉汤中培养至固定相(stationary phase),且通过以3000g离心15分钟来收集,并以2mL和1mL的PBS(磷酸盐缓冲盐水,pH7.2)来冲洗。使该等菌株的培养物重新悬浮于1mL的PBS中,接着在95℃下加热30分钟,并接着接受高压蒸锅作用并储存于-20℃PBS中。
脾细胞分离:向来自健康志愿者的5mL血液样本添加5mLFicoll-Hypaque(17-1400-02,Pharmacia)且接着以500g离心30分钟。取得脾细胞。在每一脾细胞样本中,将细胞密度调节至每个样本5×106个细胞。该等脾细胞样本在2mL RPMI 1640(pH7.7)中培育6小时。
外周血单核细胞之分离:向来自健康志愿者的5mL血液样本添加5mL Ficoll-Hypaque(17-1400-02,Pharmacia)且接着以500g离心30分钟。该等外周血单核细胞(PBMC)取自样本的界面,并以PBS冲洗两次。将该等PBMC(105细胞/mL)转移到六孔板的孔中,其中每孔含有pH7.7的2mL RPMI 1640培养基。
刺激IFN-γ分泌:脾细胞或PBMC样本与特定量的革兰氏阳性细菌共同培养。共同培养36小时后,分别收集各个样本中的细胞。使所收集之细胞重新悬浮并以2000rpm离心5分钟。取上层清液用来测定在各个样本中之IFN-γ含量。
IFN-γ含量之测定:通过ELISA确定IFN-γ含量,其包含以下步骤:
-添加30L 2.5μg/mL之纯化小鼠抗人IFN-γ抗体(Cat.No 18181D,USA)至10mL涂覆缓冲剂中(0.1MNa2HPO4,pH9.0),且添加100L抗体溶液至ELISA板的每一孔中;
-在4℃下摇动该板
-以冲洗缓冲剂(PBS中0.05%Tween20)冲洗该板的每个孔;
-添加300μL阻断缓冲剂(PBS中1%BSA)至该板的每一孔中;
-在室温下摇动该板至少2个小时;
-添加100μL脾细胞样品的上层清液至该板的每一孔中;
-在4℃下隔夜摇动该板;
-以冲洗缓冲剂冲洗该板的每个孔;
-在室温下培育该板1小时;
-以冲洗缓冲剂冲洗该板的每个孔;
-添加以稀释缓冲剂稀释(1∶1000)的150μL Streptavidin-AKP至该板的每一孔中;
-在室温下摇动该板1小时;
-以冲洗缓冲剂将该板的每个孔冲洗八次;
-添加200μL底物pNpp至该板的每一孔中;
-在室温下培育该板直至完成底物反应;
-在405nm(意即OD405)下量测该板的每个孔的吸收度。
结果:革兰氏阳性细菌中,选择GM-080以在脾细胞和PBMC中具有刺激IFN-γ分泌的最强能力。
实施例2:16srDNA序列测定
DNA提取:通过使用DNA Stool Mini Kit(cat No.51504)来提取GM-080及其它细菌CCRC12913、CCRC14001和CCRC16100之基因组DNA。根据如下所列的步骤来执行纯化:
-添加1.4mL的ABS缓冲剂至培养物中且使其涡流1分钟;
-在70℃下加热由前述步骤所得的溶液5分钟;
-使该溶液涡流约15秒并接着将其以约13,000rpm离心1分钟;
-将上层清液移至一个新的离心管;
-在上层清液中添加InhibitEx锭剂且摇动其使锭剂溶解,接着在室温下培育1分钟;
-使该溶液以约13,000rpm离心3分钟使细菌粘着至InhibitEx;
-将上层清液移至一个新的离心管中且接着以约13,000rpm离心3分钟;
-取200μL上层清液至一个新的离心管并添加蛋白酶K;
-添加200μL缓冲剂AL且使其涡流15分钟以获得均质溶液;
-添加15μL蛋白酶K至该均质溶液中,且使其涡流15秒;
-在70℃下培育该溶液10分钟;
-添加200μL96-100%的乙醇且涡流;
-将该溶液移至QIAamp旋转管柱(spin column)并使其以约13,000rpm离心1分钟;
-将该QIAamp旋转管柱移至一个新的离心管且添加500L缓冲剂AW1,并接着使其以约13,000rpm离心1分钟;
-将该QIAamp旋转管柱移至一个新的离心管且添加500μL缓冲剂AW2,并接着使其以约13,000rpm离心1分钟;
-将该QIAamp旋转管柱移入一个新的离心管且添加200L缓冲剂AE,并接着使其在室温下培育1分钟;且
-使其以约13,000rpm离心1分钟以溶离DNA。
16srDNA片段扩增:用来扩增L区域的引物是根据以下而设计:类干酪乳杆菌16SrRNA V1区域,5’-CAC CGA GAT TCA ACATGG-3’(SEQ ID No.1)和乳杆菌保守(conserved)16SrRNA,5’-CCCACT GCT GCC TCC CGT AGG AGT-3’(SEQ ID No.2)(Ward,L.J.H.和Timmins,M.J.(1999)Differentiation of Lactobacillus casei,Lactobacillusparacasei and Lactobacillus rhamnosus by polymerase chain reaction.Lett.Appl.Microbiol.29:90-92).GM-080、CCRC12913、CCRC14001和CCRC16100的基因组DNA被当作执行PCR反应的模板。该16s rDNAPCR扩增程序如下:(1)95℃下10分钟;(2)95℃下45秒;(3)46℃下45秒;(4)72℃下1分钟;(5)72℃下7分钟;步骤2至5重复30个循环。
16s rDNA序列测定:GM-080、CCRC12913、CCRC14001和CCRC16100的PCR产物接受琼脂糖凝胶电泳(图2)且将其排序。通过ARB序列编辑器(释放8.1)将该等序列对照复合序列比对数据集(multiple sequence alignment dataset)(NCBI blastn,http://www.ncbi.nlm.nih.gov/BLAST)来进行序列比对。其也表明类干酪乳杆菌菌株PB4、AY186046;F31、AF243147;KLB58、AF243168之16s rDNA序列类似于GM-080的16s rDNA序列,如图3所示(随VectorNTITM产生,Inc。)。另外,如图4所示,16s rDNA种系发生间距树随EMBL-EBI ClustalW(http://www.ebi.ac.uk/clustalw)产生。根据该16s rDNA分析,GM-080与类干酪乳杆菌菌株KLB58高度相关,但又与KLB58截然不同。综上所述,GM-080属于类干酪乳杆菌。
实施例3:随机扩增的多形态DNA(RAPD分析)
GM-080、类干酪乳杆菌ATCC25598、25302、335、11582和27216之DNA提取如实施例2所述般执行。
用于随机扩增的引物是5’-ATGTAACGCC-3’(Gardiner,G.、Ross,R.P.、Collins,J.K、Fitzgerald,G.、Stanton,C.之Development of aprobiotic cheddar cheese containing human-derived Lactobacillusparacasei strains.Appl Environ Microbiol.1998;64:2192-2199).
RAPD结果如图5所示。根据该RAPD分析,GM-080与熟知的类干酪乳杆菌菌株截然不同。综上所述,GM-080是一种新颖类干酪乳杆菌菌株。
实施例4:GM-080细胞壁蛋白提取和分析
根据由Angelis所描述的方法(Angelis,M.D.、Corsetti,A、Tosti,N.、Rossi,J.、Corbo,M.R.、和Gobbetti,M.(2001)Characterization ofNon-Starter Lactic Acid Bacteria from Italian Ewe Cheeses Based onPhenotypic,Genotypic,and Cell Wall Protein Analyses.Appl.Environ.Microbiol.67:2011-2020)来纯化细胞壁蛋白。收获于MRS肉汤()中隔夜培养的细胞,且接着以含0.1M CaCl2之0.05MTris-HCl(pH 7.5)冲洗2次,并使其重新悬浮于10.0的OD600的1mL相同缓冲剂中。以8000×g离心5分钟后,以含0.01M EDTA、0.01MNaCl、和2%(wt/vol)SDS的1.0ml提取缓冲剂(pH8.0)将细胞壁蛋白自离心块中提取出来。在室温下将悬浮液储存60分钟,100℃下加热5分钟,且在4℃以11600×g离心10分钟。通过12%SDS-PAGE来分析上层清液和以Comassie蓝进行染色。
结果如图6所示。GM-080的图案具有三条特异带,P1、P2、和P3,其与先前研究(Angelis等人,2001)所报导的类干酪乳杆菌之图案相似。因此,GM-080被证实属于类干酪乳杆菌。
实施例5:用来鉴别GM-080的标准检测系统
刺激:通过添加新鲜的培养基并培养16小时使Jurkat细胞得到更新。随后,将细胞分为2组,一组用于使用乳杆菌而进行的培养而另一组用于未使用乳杆菌而进行的培养。当细胞浓度达到1×107/10mL时,使用或不使用1×107不同乳杆菌(CCRC 12193、GM-080或CCRC12188)来刺激细胞24小时。。刺激之后,收集细胞并以PBS冲洗2次,并用于RNA分离。
RNA分离和标记:根据制造商说明,使用Trizol Reagent(LifeGaithersburg,Md.)从细胞中提取RNA。充分混合8L的RNA(10μg)和2L寡聚-dT(12-18mer,1g/L),并保持在70℃达10分钟且接着以冰冷却2分钟。在暗处将RNA与反转录标记混合物及3L Cy3-dUTP(1mM)、2L SuperScript III(200U/L)及RNasin(1L)混合。使混合物在50℃下培育2小时用于反转录,且通过添加1.5L 20mM EDTA来终止反应。在标记之后,通过NaOH处理移除RNA且以HCl中和。立即以YM30纯化试剂盒来纯化cDNA。
微阵列制造:许多所选择的基因通过聚合酶链反应扩增并通过260nm分光光谱仪(spectrophotometry)来定量。所有经纯化的PCR产物在50%二甲基亚砜中被调节至0.1μg/μl的浓度,并以双份滴点(spot)于UltraGAPSTM涂覆的载玻片(Inc.,Corning,N.Y.)上。在印刷之后,微阵列在700mJoulesand下UV交联,在室温下储存于干燥器中的载玻片容器中。该基因如上所述列于表2。
微阵列杂交:使经荧光标记的cDNA在100℃下于杂交溶液(5xSSC、0.1%SDS和25%甲酰胺)中变性5分钟,冷却至环境温度,并沉积在载玻片上。在55℃下执行杂交18小时。杂交之后,连续以低严格性(1x SSC和0.1%SDS)、中严格性(0.1x SSC和0.1%SDS)、高严格性(0.1x SSC)缓冲剂进行冲洗,且最终通过压缩N2进行干燥。
信号检测和数据分析:经N2干燥的载玻片立即以对每一载玻片相同的光电倍增器激光强度和灵敏度水准(sensitivity level)被扫描于GenePix 4000B扫描仪(AxonInc.)上。获得原始荧光数据(10nm分辨率),并在Microsoft ExcelTM中执行后续加工和数据可视化。为了比较独立的杂交实验的结果,从各个独立点的杂交信号中减去局部背景信号,且接着除以管家基因β肌动蛋白。每一基因的最终表达以双份的平均值来表达。则获得使用乳杆菌培养及未使用乳杆菌培养的Jurkat细胞的基因表达图。选择在使用乳杆菌(CCRC12193、GM-080或CCRC12188)培养之Jurkat细胞较未使用该等菌而培养的Jurkat细胞中向上或向下调节了超过2倍的一组基因。结果如表3所示。该区别表示不同的种或株可开放(turn on)或关闭(turn off)细胞之不同基因。因此,自基因表达概图(gene expressionprofile),指出CCRC 12193、GM-080和CCRC 12188是类干酪乳杆菌,但属于不同株。
实施例6:GM-080至肠上皮细胞的粘着力
在本实施例中,Caco-2细胞被当作上皮细胞。Caco-2细胞具有附着于其上的功能性微绒毛和水解酶,其展示了活体外肠的成熟上皮细胞的相区别的形态学和功能。
细胞:在37℃下,于以5%CO2/95%空气而补充了5%FBS的最低必需培养基(Minimum essential medium,MEM,)中培养Caco-2。为了粘着力分析,在置放于6孔板中之玻璃盖玻片上制备2ml单层Caco-2细胞(3×105细胞/ml)。每隔一天替换培养介质,且该等单层在2周培育后用于粘着力分析。在即将使用之前,将该等单层以PBS冲洗2次并向每一孔添加1.5ml的MEM且在细菌接种前于37℃下培育1小时。
粘着力:以PBS冲洗过1次且被重新悬浮于1.5ml MEM培养基中的1.5ml(4×108CFU/ml)GM-080被添加至Caco-2细胞。在37℃下培育1小时后,将单层细胞以PBS缓冲剂冲洗4次,与3ml甲醇混合且在室温下培育5至10分钟,以PBS冲洗3次,在空气中干燥且经革兰氏染色。在油浸下以显微镜检测(×100)粘着细菌,计数每块盖玻片上的15个随机区域,且测定每个区域的粘着细菌的平均值±SD。
结果:计数之后,有102±23.6个GM-080细菌粘着至Caco-2细胞。因此,根据Jacobsen等人所建立的标准(Jacobsen,C.N.、Nielsen,R.V.、Hayford,A.E.、Moller,P.L.、Michaelsen,K.F.、Paerregarrd,A.、Sandstrom,B.、Tvede,M.和Jakobsen,M.Screeing of probiotic activities offorty-seven strains of Lactobacillus spp.by in vitro techniques anevaluation of the colonization ability of five selected strains in human.Appl.Environ.Microbiol.1999;65:4949-4956),GM-080被认为对Caco-2细胞具有强粘着力。
实施例7:在一环境模拟胃肠道中GM-080及其它乳杆菌的活动
酸:将具有不同pH值(2.0、2.5和3.2)的9ml PBS添加至经隔夜培养的GM-080、植物乳杆菌(L.plantarum)、嗜酸乳杆菌(L.acidophilus)、干酪乳杆菌(L.casei)和保加利亚乳杆菌(L.bulgaricus),且接着进一步在37℃下培养3小时。在培养之后,以9mL pH为7.4之PBS来逐次稀释1mL细胞。估算在经酸处理之前与之后的细胞计数,且如下所列显示于表4中。
胆汁:将具有pH值(2.0)的9ml PBS添加至经隔夜培养的GM-080、植物乳杆菌、嗜酸乳杆菌、干酪乳杆菌和保加利亚乳杆菌,且接着进一步在37℃下培养3小时。在培养之后,使1mL细胞以6,000rpm离心10分钟。离心块(pellet)重新悬浮于100μL PBS(pH7.2)。向该溶液进一步添加含0.3%(w/v)牛胆汁的10mL MRS肉汤。培养该等细胞,并在3、12和24小时取1mL样本。该等样本以9mL pH为7.4之PBS逐次稀释。估算在经胆汁处理之前与之后的细胞计数,且也显示于表4中。其显示了此等乳杆菌在模拟胃肠道之环境中保持活性。

实施例8:动物模型
动物:自台湾“国家实验动物中心”(National Laboratory AnimalCenter)获得雌性BALB/c小鼠,且在光线和温度均受控制的房间中饲养2周。
过敏原纯化:尘螨过敏原Der p 5被表达于包含PGEX-2T表达载体作为重组Der p 5-谷胱甘肽S-转移酶融合蛋白的大肠杆菌中,其可通过谷胱甘肽琼脂糖结合色谱法进行纯化。培养及引入能表达所要过敏原的特异大肠杆菌株。收集细菌并以TBS(pH 7.5)冲洗,且添加0.1M苯甲基磺酰氟。通过添加Dnase I、Tween 20和溶解酵素,和通过冰冻-解冻方法使该等细胞破裂。添加EDTA至该混合溶液,且通过离心作用来移除残留物以获得含重组Der p 5-谷胱甘肽S-转移酶融合蛋白的上层清液。使该上层清液接受谷胱甘肽琼脂糖亲和管柱(affinitycolumn)作用以吸收该融合蛋白。接着在4℃下以TBS缓冲剂来冲洗该管柱,且接着以在Tris碱(pH8.0)中经还原的谷胱甘肽冲洗,以将该蛋白质自管柱分离出来。通过SDS-PAGE来估算该蛋白质的分子量,并且分析浓度。
致敏作用:通过腹腔内注射10μg之Der p 5与4mg氢氧化铝,小鼠被自动致敏。初始致敏后14和21天,让小鼠暴露在0.1%经纯化之Der p 5的气溶胶(aerosol)下30分钟,以执行吸入挑战。
治疗:将经致敏的小鼠分为三组进行实验。对A组小鼠在两周期间喂以MRS肉汤10次以作为对照组。使B组小鼠在两周期间服用干酪乳杆菌10次,且每次服用109CFU细菌。因为干酪乳杆菌已被证实对抑制IgE分泌有效,所以将B组作为阳性对照。使C组小鼠两周内服用GM-08010次,且每次服用109CFU细菌。
实施例9:IgG和IgE分泌
Der p5特异IgG和IgE之测定:最后一次吸入挑战后18小时,自尾部取出500μL血液样本。将该血液样本保持在室温下1小时且接着使其接受离心作用。将血清储存在-80℃下。通过ELISA测定Der p5特异IgG2a和IgE的量。通过于涂覆缓冲剂(0.1M NaHCO3,pH9.6)中被稀释为10μg/mL浓度的200μL经纯化的Der p5涂覆具有96个孔之蛋白质高结合板(Protein high-binding plate)。在4℃下隔夜培育之后,以PBS-Tween 20冲洗该等板,且接着添加300μL阻断缓冲剂(3%BSA)。在室温下摇动2小时后,再次以PBS-Tween20冲洗该等板。血清以1∶10稀释以用于IgG量测,且以1∶4稀释以用于IgE量测。在室温下摇动该样本2小时。在4℃下隔夜培育后,以PBS-Tween 20冲洗该等板,且添加200μL生物素化大鼠抗小鼠IgE单克隆抗体、或大鼠抗小鼠IgG mAb。在室温下摇动该样本2小时,且接着以PBS-Tween 20冲洗。接着添加200μL链酶亲合素-碱性磷酸酶(Streptavidin-alkaline phosphatase)(1∶1000)且在室温下摇动该样本1小时。经6次冲洗后,通过添加200μL磷酸酶底物对磷酸硝基苯酯、二钠盐(pNPP)(N-2770,USA)而开始颜色反应。板在405nm微板自动读取器(Taiwan)中来读取。
数据分析:为了分析IgE和IgG含量之改变,执行单因素ANOVA分析重复量测,以比较组间区别。在变化分析之后,将Duncan多范围测试用于区分实验组与对照组间的区别。将p<0.05的值被用以表示统计学上的显著性差异。
结果:结果如图7所示。其证实经GM-080治疗的动物血清中的IgE分泌剧烈降低,且仅为未经治疗的动物血清中IgE分泌中的25%。另一方面,经GM-080治疗的动物血清中IgG分泌升高至2倍。因为IgG分泌代表Th1 T细胞反应,所以GM-080旨在消除与过敏相关疾病相关联的IgE分泌。
实施例10:支气管肺泡灌洗液细胞计数
样本制备:经致敏18小时之后,通过由气管切开术(tracheostomy)而引入的聚乙烯管,以5×0.5ml等分试样的0.9%无菌盐水来灌洗小鼠。离心(4℃下,500g,10分钟)灌洗液,且使细胞离心块重新悬浮于1ml PBS溶液中。自经列伊氏染色(Leu’s stain)染色的细胞离心器制剂制成分化之细胞计数。
数据分析:为了分析细胞计数的改变,执行单因素ANOVA分析重复量测,以比较组间区别。在变化分析之后,将Duncan多范围测试用于区分实验组与对照组间的区别。将p<0.05的值用以表示统计学上的显著性差异。
结果:结果如图8所示。BALF中血液细胞类型组成(contribution)代表了炎症程度。此外,过敏性哮喘的主要症状是气管慢性炎症和嗜酸性粒细胞浸润(infiltration)。其证实经GM-080治疗的动物的BALF中嗜酸性粒细胞自5%剧烈降低至1%。另一方面,经GM-080治疗的动物的BALF中巨噬细胞和淋巴细胞显著上升。
实施例11:支气管肺泡灌洗液中IFN-γ分泌
样本制备:经致敏24小时之后,通过由气管切开术而引入的聚乙烯管,以5×0.5ml等分试样的0.9%无菌盐水来灌洗小鼠。离心(4℃下,500g,10分钟)灌洗液,且让上层清液接受IFN-γ定量分析,如实施例1所描述。
结果:结果如图9所示。其显示喂以GM-080的动物在BALF中产生了约100pg/mL的IFN-γ。另一方面,对照组在BALF中仅产生了20至40pg/mL的IFN-γ。GM-080对抑制过敏性炎症是有效的。
实施例12:灭活GM-080用于治疗过敏
灭活GM-080的制备:在喂食小鼠之前,使冻干的GM-080粉末悬浮于蒸馏水中并接受高压蒸锅作用(121℃下,15分钟)。
小鼠和致敏作用:自“国家实验动物饲养研究中心”(NationalLaboratory Animal Breeding and Research Center)(台湾台北)购得雌性BALB/c小鼠(6-8周大)。将所有动物个别地保持在温度(24±2℃)和湿度(60±5%)受到控制的笼中,且在无特异病原(specific-pathogen-free)条件下保持12小时日/夜循环。使BALB/c小鼠腹腔内注射(i.p.)被吸附至4mg氢氧化铝的10g重组屋尘螨(Dermatophagoides pteronyssinus)过敏原Der p5-6xHis融合蛋白。三周内以每天每鼠107、109和1011CFU GM-080的量来喂养该等小鼠。对该等小鼠在第14天刺激(boost)以相同剂量的过敏原作为致敏作用,且在致敏21天后,以于PBS中稀释之0.1%Der p5-6xHis来挑战该等小鼠。吸入挑战在与一DeVilbissTM pulmosonic喷雾器(Model2512;Corp.,Somerset,PA)相连的1L腔室中执行,该喷雾器生成气溶胶薄雾。18小时后,通过尾部静脉出血来收集血清,且通过ELISA测定IgE,如实施例9中所描述。
结果:结果如表10所示。其显示经尘埃过敏Der p-5挑战的BALB/c小鼠较未试验的(naive)组(p<0.05)具有显著提高的血清IgE含量。此暗示可成功建立过敏性致敏的小鼠模型。在每日以不同剂量的GM-080喂养21天后,GM-080组中血清IgE较对照组已显著降低(p<0.05)。结果显示灭活GM-080可通过减少过敏原特异性IgE来降低过敏应答。
尽管已经说明且描述本发明的实施例,但所属技术领域的技术人员可作出不同的修正和改良。本发明并不欲受限于所说明的特定形式,而所有未脱离本发明之精神和范畴内的修正均属于在随附权利要求书中所界定的范畴之内。

1.一种经分离的微生物株类干酪乳杆菌(Lactobacillus paracasei)GM-080,其保藏于中国典型培养物保藏中心,保藏登记号为CCTCC M204012。
2.一种包含根据权利要求1所述的微生物株之组合物。
3.根据权利要求2所述的组合物,其中所述微生物株是活的或经灭活的。
4.根据权利要求3所述的组合物,其中所述微生物株是经灭活的。
5.根据权利要求2所述的组合物,其是药物组合物、饮食添加剂、食品或其组份的形式。
6.微生物株类干酪乳杆菌GM-080在制备刺激IFN-γ分泌的药物中的应用,其中所述微生物株类干酪乳杆菌GM-080保藏于中国典型培养物保藏中心,保藏登记号为CCTCC M 204012。
7.微生物株类干酪乳杆菌GM-080在制备治疗过敏相关疾病的药物中的应用,其中所述微生物株类干酪乳杆菌GM-080保藏于中国典型培养物保藏中心,保藏登记号为CCTCC M 204012。
8.根据权利要求7所述的应用,其中所述过敏相关疾病是选自由以下组成的组:气道高反应性和炎症、异位性皮肤炎、过敏性结膜炎、鼻炎、窦炎、过敏性肺炎、外源性过敏性肺泡炎、荨麻疹、湿疹、过敏性反应、血管性水肿、过敏性偏头痛、特定胃肠失调和哮喘。
9.根据权利要求8所述的应用,其中所述过敏相关疾病是气道高反应性和炎症。
10.根据权利要求7所述的应用,其中所述过敏相关疾病是与暴露于气源性过敏原有关。
