本发明涉及医药技术领域,是一种溶胀率高、可生物降解的止血粉。其由羧甲基壳聚糖、海藻酸钠、氯化钙为主要成份制备而成,是一种具有多皱折表面、可快速吸水溶胀的微球,本发明止血粉制备方法简单,生产成本低廉,止血效果好。

一种可生物降解的止血粉
技术领域
本发明涉及医药技术领域,是一种溶胀率高、止血效果好并可生物降解的止血粉。
背景技术
出血是战伤中第一位的死亡原因,是创伤中第二位的死亡原因。出血还是创伤病人手术室死亡的主要原因。快速、有效地控制出血可明显减少因出血导致的死亡。
粉状止血制剂不受创面大小及部位的限制,有其独到的优点,增加可生物降解特性后将进一步拓宽其使用范围。目前国外研制的可生物降解止血粉如ARISTATM,是一种用土豆淀粉提纯加工的、具有多微孔表面结构的多聚糖微球,粒径为10-200μm,其能快速吸收血液中的水份,浓缩凝血因子和血小板,促进血液凝结。止血效果较好,但价格昂贵。目前国内尚无类似产品。
发明内容
本发明提供一种可生物降解的止血粉,也是一种多聚糖微球,但其主要成分为羧甲基壳聚糖和海藻酸钠。壳聚糖和海藻酸是两种不同止血机制的生物多聚糖,都具有生物可降解性和良好的组织相容性,但它们各自的止血效果有限。本发明用这两种止血机制不同的生物材料,制备出具有多皱折表面、可快速吸水溶胀的微球,取得良好的止血效果。
本发明止血粉由羧甲基壳聚糖、海藻酸钠、氯化钙以及水、液体石蜡和吐温80制备而成,其中氯化钙为交联剂,吐温80乳化剂。各成份配比为:
羧甲基壳聚糖  1~2g          海藻酸钠  1~2g
氯化钙        0.8~1.6g      液体石蜡  250~1000ml
吐温80        18~75ml
制备方法:
1.制备水相溶液:按配比将羧甲基壳聚糖和海藻酸钠各自溶于100~200ml蒸镏水配制成水溶液。取等量羧甲基壳聚糖溶液和海藻酸钠溶液,充分搅拌混和,制成水相溶液;
2.制备油相溶液:按配比将液体石蜡和吐温80混匀,制成油相溶液;
3.制备氯化钙溶液:将氯化钙溶于25~50ml蒸镏水即可;
4.制备水凝胶微球:将水相溶液加入油相溶液,快速搅拌乳化,再加入氯化钙溶液充分搅拌,交联形成水凝胶微球,静置沉淀,弃上清液;
5.洗涤脱水,制备干粉:将沉淀用石油醚洗涤,去除液体石蜡和吐温80,再用乙醇进行递度脱水,最后干燥成粉。
用AccuSizer780/SIS(美国PSS公司)粒径测定系统测量止血粉粒径,在10-100μm之间。扫描电镜显示,颗粒呈球状结构,表面布满皱折。用创面渗出液模拟体液环境,颗粒溶胀率在3分钟时达到280%,在兔脾出血模型中,止血效果明显优于云南白药。
具体实施方式
现结合实施例,对本发明作详细描述:
实施例:将1g海藻酸钠和1g羧甲基壳聚糖分别溶入100ml蒸馏水中。将羧甲基壳聚糖溶液加入海藻酸钠溶液,充分搅拌混合。然后加入500ml的液体石蜡+37.5ml吐温80组成的乳化液中快速搅拌30分钟,搅拌速度为800-1000转/分。再加入2%氯化钙溶液40ml,充分搅拌,交联形成水凝胶微球,静置沉淀,弃上清。分别用100ml石油醚洗涤3次,再分别用35%、70%、100%递度酒精脱水、真空干燥成粉。
本发明止血粉,制备方法简单,生产成本低廉,吸水溶胀率高,使用方便,止血效果好。

Claims (1)

1.一种可生物降解的止血粉,其特征在于由羧甲基壳聚糖、海藻酸钠、氯化钙以及液体石蜡和吐温80制备而成,各成份配比为:
羧甲基壳聚糖1~2g      海藻酸钠1~2g
氯化钙0.8~1.6g        液体石蜡250~1000ml
吐温80 18~75ml
制备方法如下:
(1)制备水相溶液:按配比将羧甲基壳聚糖和海藻酸钠各自溶于100~200ml蒸镏水配制成水溶液,取等量羧甲基壳聚糖溶液和海藻酸钠溶液,充分搅拌混和,制成水相溶液;
(2)制备油相溶液:按配比将液体石蜡和吐温80混匀,制成油相溶液;
(3)制备氯化钙溶液:将氯化钙溶于25~50ml蒸镏水配制成氯化钙溶液;
(4)制备水凝胶微球:将水相溶液加入油相溶液,快速搅拌乳化,再加入氯化钙溶液充分搅拌,交联形成水凝胶微球,静置沉淀,弃上清液;
(5)洗涤脱水,制备干粉:将沉淀用石油醚洗涤,以去除液体石蜡和吐温80,再用乙醇进行递度脱水,最后干燥成粉。
