Abstract

La presente invención se refiere a células B modificadas por modificación genética y sus usos de las mismas, por ejemplo, para el tratamiento de una variedad de enfermedades y trastornos, que incluyen cáncer, enfermedad cardíaca, enfermedad inflamatoria, enfermedad de desgaste muscular, enfermedad neurológica y similares. En ciertas modalidades, la invención se refiere a una célula B modificada aislada ("célula CAR-B"), capaz de expresar un receptor quimérico ("receptor CAR-B"), en donde el receptor quimérico comprende (a) un dominio extracelular; (b) un dominio transmembrana; y (c) un dominio citoplasmático que comprende al menos un dominio de señalización. En varias modalidades, la invención comprende una célula B modificada aislada, en donde la célula B es capaz de expresar y secretar una carga útil, en donde la carga útil no se expresa naturalmente en una célula B o se expresa a niveles más altos que los expresados naturalmente en una célula B. En varias modalidades, la carga útil es un anticuerpo o fragmento del mismo.
