Abstract

Los ácidos nodulispóricos (NAs) comprenden un grupo de indol diterpenos conocidos por sus potentes actividades insecticidas; sin embargo, la biosíntesis de los NAs por su productor natural, Hypoxylon pulicicidum (Nodulisporium sp.) es excepcionalmente difícil de lograr. La identificación de genes responsables de la producción de NA podría permitir optimizar la vía biosintética para proporcionar acceso a los NAs para aplicaciones comerciales. La obtención de cantidades útiles de NAs usando los métodos de fermentación publicados constituye un desafío, haciendo que los estudios de inactivación génica sean un método indeseable para confirmar la función génica. Alternativamente, la expresión génica heteróloga de genes de H. pulicicidum en una especie huésped más robusta como Penicillium paxilli proporciona un camino para identificar rápidamente la función de los genes que desempeñan un rol en la biosíntesis de NA. En este trabajo, nosotros identificamos la función de cuatro genes metabólicos secundarios necesarios para la biosíntesis del ácido nodulispórico F (NAF) y reconstituimos estos genes en el genoma de P. paxilli para permitir la producción heteróloga de NAF en este hongo.
