La présente invention concerne les compositions utilisées aussi bien dans l'art médical ou vétérinaire, que dans la cosmétique ou les soins des parties superficielles du corps humain, et comprenant un extrait d'Öuf de caille.
Par "Öuf de caille", on entend l'Öuf de l'espèce animale Coturnix, domestique ou sauvage, et notamment de la souche B Mina.
Par "extrait", on entend toute fraction ou partie, biologiquement active vis-à-vis de l'homme ou de l'animal, de l'Öuf de caille considéré en tant que produit naturel, obtenue après ouverture de ce dernier et éventuellement différents traitements mécaniques, physiques, chimiques, permettant de séparer, ou enrichir tout constituant dudit Öuf, qu'il s'agisse du blanc et/ou du jaune, ou de tout autre constituant actif ou utile dudit Öuf. De tels extraits sont en général obtenus sous forme homogène, à l'état sec, par exemple par lyophilisation, ou à l'état humide, par exemple sous forme de fluides.
L'effet biologiquement bénéfique de l'Öuf de caille, notamment dans le traitement de différentes affections est connu depuis longtemps. Ceci a par exemple été établi et discuté dans le document français suivant: "La Clinique", N<o> 22, 15 mars 1978, pages 2 à 4, JC Truffier.
Conformément au document EP-A-0 032 344, on a décrit et proposé un médicament par voie orale, consistant uniquement en un extrait d'Öuf de caille, sous forme de lyophilisat.
Conformément au document EP-A-2 477 017, on a décrit et proposé une composition ingérable, comprenant, dans la proportion pondérale de 90 à 97%, un extrait d'Öuf de caille, un hydrolysat ou autolysat de protéines animales, et un acide aminé ayant une action hépatique protectrice.
Conformément au document FR-A-2 538 248, à titre de produit cosmétique, on a déjà proposé une composition comprenant en mélange homogène, un extrait homogène d'Öufs, dans la proportion pondérale de 5 à 75% par rapport au poids total de ladite composition, un support adapté à une application en couche ou film sur les parties superficielles du corps humain, dans la proportion pondérale de 20 à 80% en poids, et différents additifs usuels en cosmétique.
Pour une composition à usage externe et à base d'un extrait d'Öufs de caille, on a découvert qu'en limitant ledit extrait à une quantité demeurant biologiquement efficace, effectivement dosable, mais au plus égale à 1 pour mille en poids du total de la composition, d'une part on diminuait le délai d'action biologique de ladite composition, et d'autre part on limitait de manière importante les effets secondaires de l'extrait d'Öuf de caille.
Par ailleurs, limiter la quantité d'extrait d'Öuf de caille dans la proportion précédemment indiquée permet d'augmenter la stabilité dans le temps des compositions à usage externe considérées par la présente invention.
L'extrait d'Öuf de caille présent dans une composition selon la présente invention peut être dosé selon toutes techniques appropriées, à partir d'une propriété, caractéristique ou activité spécifique dudit extrait, par exemple une activité antiprotéasique, notamment antitripsique dudit extrait.
Préférentiellement, la quantité de l'extrait d'Öuf de caille est comprise entre 0,01 et 0,05, et par exemple entre 0,02 et 0,04 du poids total de la composition.
Selon un mode préféré d'exécution de l'invention, l'extrait de caille est obtenu par lyophilisation de tout ou partie des fluides, dont jaune et blanc, contenus dans l'Öuf de caille.
De manière traditionnelle, une composition selon l'invention comprend différents additifs usuels pour les compositions médicamenteuses à usage topique, ou les compositions cosmétiques. A titre d'exemple, ces additifs sont choisis parmi:
- les produits anti-oxydants, comme le tocopherol
- les agents conservateurs, comme l'acide sorbique,
- les colorants,
- les parfums naturels ou synthétiques,
- les agents bactéricides ou anti-microbiens, notamment les antibiotiques,
- les agents de texture,
- et les tensioactifs.
Le support d'une composition selon l'invention consiste à titre préférentiel en une émulsion homogène ou système comprenant au moins deux phases, par exemple une émulsion du type huile dans eau ou eau dans huile.
Ce support est à titre d'exemple adapté à une application sur la peau et/ou le cuir chevelu du corps humain, et a pour ce faire une forme de présentation choisie dans le groupe comprenant les crèmes, baumes, pommades, huiles, shampooings, mousses et savons.
Comme déjà indiqué, une composition selon l'invention peut être utilisée comme médicament, en particulier à usage topique, et par exemple comme composition dermatologique. Dans ce dernier cas, et de manière préférée, la composition dermatologique comporte une quantité thérapeutiquement efficace de l'extrait d'Öuf de caille pour le traitement d'une affection cutanée non infectieuse, choisie dans le groupe incluant l'eczéma, notamment atopique, le prurit de tout ou partie du corps humain, le psoriasis et les allergies.
Une composition selon l'invention peut être aussi utilisée à titre de composition cosmétique ou pour les soins d'hygiène des parties externes du corps humain.
On décrit ci-après, à titre d'exemple, un procédé complet d'obtention d'une composition selon l'invention à partir d'Öufs de caille.
1. Obtention d'un extrait d'Öuf de caille, entier, c'est-à-dire rassemblant le jaune et le blanc, sous forme homogène et à l'état sec.
On utilise des Öufs de caille de toutes espèces et provenances, par exemple de l'espèce B Mina, produits par des élevages placés sous surveillance sanitaire et vétérinaire. Les Öufs frais mis en Öuvre peuvent être embryonnés ou non.
Les Öufs de caille sont préalablement traités de manière à exclure de leurs coquilles tout germe pathogène, par exemple par immersion dans un ou plusieurs bains de solutions aseptiques.
Les Öufs sont ensuite cassés mécaniquement et déversés dans une cuve maintenue sous agitation. Après avoir séparé les coquilles, on procède à une filtration de manière à obtenir, sous forme fluide, un mélange de blanc et jaune d'Öufs de caille. Ce mélange est stérilisé par tout moyen approprié, préservant les propriétés biologiques essentielles de ses constituants.
Le mélange stérilisé, sous forme fluide, est ensuite lyophilisé avec tout matériel approprié, pour obtenir une poudre homogène d'Öufs de caille entiers.
2. Préparation d'une forme aqueuse intermédiaire de l'extrait d'Öuf de caille.
De manière à favoriser l'incorporation homogène de l'extrait d'Öufs de caille dans le support défini ci-après, on prépare une suspension homogène en milieu aqueux de la poudre précédemment obtenue, de la manière suivante:
- on prélève 18,75 mg de la poudre précitée,
- on dispose d'un excipient, soluble en phase aqueuse, constitué par 37,5 mg de stéarate de magnésium, 973,5 mg de cellulose microcristalline, de 187,5 mg de croscarmellose sodique, de 6318,75 mg de lactose,
- et on mélange dans 100 ml d'eau pure stérile, la quantité d'extrait d'Öufs de caille précitée, et 7,5 g de l'excipient ayant la composition définie précédemment, pour obtenir 107,5 g d'un liquide laiteux (A).
3. Préparation du support adapté à une application en couche ou film sur les parties superficielles du corps humain.
Ce support (B) est constitué par une phase aqueuse (b), et une phase huileuse (b min ).
La phase aqueuse (b) est une eau stérile osmosée. Elle représente en général entre 43 et 70% en poids du support (B), et dans le présent exemple, est égale à 215,875 g.
La phase huileuse ou grasse (b min ) comporte les produits et quantités suivants:
- huile de jojoba: 2 à 4% en poids du support (B), et dans le présent exemple 7,85 g,
- un complexe d'huiles végétales extraites d'abricot et d'avocat: 2 à 3% du support (B), et dans le présent exemple 11,775 g,
- beurre de karité: 2 à 6% du support (B), et dans le présent exemple 11,775 g,
- des esters d'acides gras extraits de soja: 8 à 10% du support (B), et dans le présent exemple 35,325 g,
- des émulgateurs extraits de céréales et de soja, comprenant des lécithines et des protéines: 16 à 34% du support (B), et dans le présent exemple 109,8 g.
Dans ces conditions, la phase huileuse (b min ) représente 30 à 57% en poids du support (B), et dans le présent exemple 176,5 g.
Par des moyens traditionnels, on associe les phases (b) et (b min ) sous la forme d'une émulsion huile dans eau, qui constitue le support (B). Le rapport pondéral entre les phases (b) et (b min ), résultant des proportions précédemment définies, permet une bonne hydratation de la peau.
Cette émulsion est stabilisée en ajoutant un agent naturel, à la fois anti-bactérien et anti-oxydant, consistant en un extrait de romarin, dosé à 0,02% de la composition obtenue à l'issue de l'étape suivante, soit 0,1 g dans le présent exemple.
4. Obtention de la composition finale.
Le liquide laiteux (A), constituant la forme aqueuse intermédiaire de l'extrait d'Öufs de caille, est incorporé par tout moyen traditionnel au support (B), et on obtient ainsi 500 g d'un produit fini stable, de couleur blanche et d'aspect fluide, dont l'étalement se fait facilement sans graisser la peau.
il résulte des quantités et compositions exemplifiées précédemment que ce produit fini comporte 0,0375 en poids de l'extrait d'Öufs de caille.
La composition du produit finalement obtenu peut être complétée par des vitamines, telles que vitamines A et E, des oligo-éléments comme par exemple du zinc, du sélénium.
Ce produit peut être conditionné de manière appropriée, par exemple en pots, flacons ou dans d'autres systèmes permettant de le dispenser, comportant par exemple des pompes et tubes.
La composition obtenue, comme décrit précédemment, a fait l'objet d'essais cliniques sur 21 personnes souffrant de différentes affections cutanées non infectieuses, avec des résultats positifs et favorables obtenus pour 16 d'entre elles. Quelques cas sont décrits ci-après:
Cas N<o> 1: une femme de 87 ans souffrait d'un eczéma ancien sur la partie antérieure d'une jambe. L'application de la composition selon l'invention, deux fois par jour, a entraîné une disparition des lésions au troisième jour d'application. L'application de la composition s'est poursuivie jusqu'à maintenant, et on n'a noté aucune récidive.
Cas N<o> 2: Une femme de 25 ans présentait des problèmes cutanés apparaissant lors de périodes de stress, avec des lésions érythémateuses, chaudes et prurigineuses. L'application d'une composition selon l'invention sur les lésions a entraîné une disparition des symptômes en moins de 30 minutes.
Cas N<o> 3: Un homme de 66 ans souffrait de lésions cutanées avec peau sèche et desquamante, et surtout avec prurit apparaissant de façon spontanée et aggravée lors de la manipulation d'objets poussérieux ou d'herbes, avec des rougeurs cutanées.
L'application d'une composition selon l'invention a entraîné une disparition du prurit en quelques minutes, avec une disparition des rougeurs en moins de 15 minutes. Le prurit qui apparaissait plusieurs fois par jour est devenu de moins en moins fréquent, et était calmé systématiquement par une nouvelle application.
Cas N<o> 4: Un homme de 25 ans souffrait d'un psoriasis relativement ancien, localisé sur les flancs et la partie latérale du haut des cuisses, et qui résistait aux thérapeutiques traditionnelles. L'application de la composition selon l'invention a entraîné une diminution des lésions au deuxième jour, et une disparition du prurit. Après plusieurs jours d'application, l'amélioration est devenue progressive et persistante, en supprimant la desquamation. Au bout de 3 semaines, on a noté une stabilisation du résultat.
Cas N<o> 5: Un enfant de 12 ans souffrait d'une lésion cutanée avec un placard rouge, un peu cartonné, inflammatoire, au niveau du creux poplité et de partie interne du genou droit. L'origine de cette lésion était indéterminée. Au premier jour d'application d'une composition selon l'invention, on a observé une régression des signes inflammatoires locaux, avec une persistance d'une zone érythémateuse localisée. Au deuxième jour et au troisième jour, la lésion initiale avait quasiment disparue, mais différents signes cutanés rappelant un peu une allergie solaire sont apparus sur la face interne de la cuisse. Au quatrième jour, ces signes ont quasiment disparus. Au cinquième jour, la peau a repris son caractère normal.
The present invention relates to compositions used both in the medical or veterinary art, as in cosmetics or the care of surface parts of the human body, and comprising an extract of quail egg.
By "quail egg" is meant the egg of the Coturnix animal species, domestic or wild, and in particular of the strain B Mina.
By "extract" is meant any fraction or part, biologically active with respect to humans or animals, of quail egg considered as a natural product, obtained after opening the latter and possibly various mechanical, physical, chemical treatments, making it possible to separate or enrich any constituent of said egg, whether it is white and / or yellow, or any other active or useful constituent of said egg. Such extracts are generally obtained in homogeneous form, in the dry state, for example by lyophilization, or in the wet state, for example in the form of fluids.
The biologically beneficial effect of quail egg, especially in the treatment of various ailments, has been known for a long time. This was for example established and discussed in the following French document: "La Clinique", N <o> 22, March 15, 1978, pages 2 to 4, JC Truffier.
In accordance with document EP-A-0 032 344, an oral medication has been described and proposed, consisting solely of an extract of quail egg, in the form of a lyophilisate.
In accordance with document EP-A-2 477 017, an ingestible composition has been described and proposed, comprising, in the proportion by weight of 90 to 97%, an extract of quail egg, a hydrolyzate or autolysate of animal proteins, and a amino acid with protective hepatic action.
In accordance with document FR-A-2 538 248, as a cosmetic product, a composition has already been proposed comprising, in a homogeneous mixture, a homogeneous extract of eggs, in the proportion by weight of 5 to 75% relative to the total weight of said composition, a support suitable for application as a layer or film on the surface parts of the human body, in the proportion by weight of 20 to 80% by weight, and various usual additives in cosmetics.
For a composition for external use and based on an extract of quail eggs, it has been discovered that by limiting said extract to a quantity which remains biologically effective, effectively dosable, but at most equal to 1 per thousand by weight of the total. of the composition, on the one hand the time of biological action of the said composition was reduced, and on the other hand the side effects of the quail egg extract were significantly limited.
Furthermore, limiting the quantity of quail egg extract in the proportion indicated above makes it possible to increase the stability over time of the compositions for external use considered by the present invention.
The quail egg extract present in a composition according to the present invention can be assayed according to any suitable techniques, from a specific property, characteristic or activity of said extract, for example an antiprotease, in particular antitripsic activity of said extract.
Preferably, the amount of quail egg extract is between 0.01 and 0.05, and for example between 0.02 and 0.04 of the total weight of the composition.
According to a preferred embodiment of the invention, the quail extract is obtained by lyophilization of all or part of the fluids, including yellow and white, contained in the quail egg.
Traditionally, a composition according to the invention comprises various usual additives for medicinal compositions for topical use, or cosmetic compositions. By way of example, these additives are chosen from:
- antioxidants, such as tocopherol
- preservatives, such as sorbic acid,
- dyes,
- natural or synthetic perfumes,
- bactericidal or anti-microbial agents, in particular antibiotics,
- texture agents,
- and surfactants.
The support for a composition according to the invention preferably consists of a homogeneous emulsion or system comprising at least two phases, for example an emulsion of the oil in water or water in oil type.
This support is, for example, suitable for application to the skin and / or scalp of the human body, and has for this purpose a form of presentation chosen from the group comprising creams, balms, ointments, oils, shampoos, mosses and soaps.
As already indicated, a composition according to the invention can be used as a medicament, in particular for topical use, and for example as a dermatological composition. In the latter case, and preferably, the dermatological composition comprises a therapeutically effective amount of quail egg extract for the treatment of a non-infectious skin condition, chosen from the group including eczema, in particular atopic, pruritus of all or part of the human body, psoriasis and allergies.
A composition according to the invention can also be used as a cosmetic composition or for the hygiene care of the external parts of the human body.
The following describes, by way of example, a complete process for obtaining a composition according to the invention from quail eggs.
1. Obtaining an extract of quail egg, whole, that is to say combining yellow and white, in a homogeneous form and in the dry state.
Quail eggs of all species and origins are used, for example of the species B Mina, produced by farms placed under sanitary and veterinary supervision. The fresh eggs used can be embryonated or not.
The quail eggs are previously treated so as to exclude from their shells any pathogenic germ, for example by immersion in one or more baths of aseptic solutions.
The eggs are then broken mechanically and poured into a tank kept under agitation. After having separated the shells, one proceeds to a filtration so as to obtain, in fluid form, a mixture of white and yellow quail eggs. This mixture is sterilized by any appropriate means, preserving the essential biological properties of its constituents.
The sterilized mixture, in fluid form, is then lyophilized with any suitable material, to obtain a homogeneous powder of whole quail eggs.
2. Preparation of an intermediate aqueous form of the quail egg extract.
In order to favor the homogeneous incorporation of the quail egg extract into the support defined below, a homogeneous suspension of the powder previously obtained is prepared in an aqueous medium, as follows:
- 18.75 mg of the above-mentioned powder are taken,
- there is an excipient, soluble in the aqueous phase, consisting of 37.5 mg of magnesium stearate, 973.5 mg of microcrystalline cellulose, 187.5 mg of croscarmellose sodium, 6318.75 mg of lactose,
- And the quantity of abovementioned quail egg extract is mixed in 100 ml of sterile pure water, and 7.5 g of the excipient having the composition defined above, to obtain 107.5 g of a liquid milky (A).
3. Preparation of the support adapted to a layer or film application on the surface parts of the human body.
This support (B) consists of an aqueous phase (b), and an oily phase (b min).
The aqueous phase (b) is sterile osmosis water. It generally represents between 43 and 70% by weight of the support (B), and in the present example, is equal to 215.875 g.
The oily or fatty phase (b min) comprises the following products and quantities:
- jojoba oil: 2 to 4% by weight of the support (B), and in the present example 7.85 g,
a complex of vegetable oils extracted from apricot and avocado: 2 to 3% of the support (B), and in this example 11.775 g,
- shea butter: 2 to 6% of the support (B), and in this example 11.775 g,
esters of fatty acids extracted from soya: 8 to 10% of the support (B), and in the present example 35.325 g,
- emulsifiers extracted from cereals and soya, comprising lecithins and proteins: 16 to 34% of the support (B), and in the present example 109.8 g.
Under these conditions, the oily phase (b min) represents 30 to 57% by weight of the support (B), and in the present example 176.5 g.
By traditional means, phases (b) and (b min) are combined in the form of an oil-in-water emulsion, which constitutes the support (B). The weight ratio between phases (b) and (b min), resulting from the previously defined proportions, allows good hydration of the skin.
This emulsion is stabilized by adding a natural agent, both anti-bacterial and anti-oxidant, consisting of an extract of rosemary, dosed at 0.02% of the composition obtained at the end of the next step, ie 0 , 1 g in this example.
4. Obtaining the final composition.
The milky liquid (A), constituting the intermediate aqueous form of the quail egg extract, is incorporated by any traditional means into the support (B), and 500 g of a stable, white finished product are thus obtained. and fluid in appearance, spreading easily without greasing the skin.
it follows from the quantities and compositions exemplified above that this finished product contains 0.0375 by weight of the quail egg extract.
The composition of the product finally obtained can be supplemented with vitamins, such as vitamins A and E, trace elements such as, for example, zinc, selenium.
This product can be appropriately packaged, for example in jars, flasks or in other systems for dispensing it, comprising for example pumps and tubes.
The composition obtained, as described above, was the subject of clinical trials on 21 people suffering from different non-infectious skin conditions, with positive and favorable results obtained for 16 of them. Some cases are described below:
Case N <o> 1: an 87-year-old woman suffered from old eczema on the front part of a leg. The application of the composition according to the invention, twice a day, caused the lesions to disappear on the third day of application. Application of the composition has continued to date, and no recurrences have been noted.
Case N <o> 2: A 25 year old woman presented skin problems appearing during periods of stress, with erythematous, hot and itchy lesions. The application of a composition according to the invention to the lesions led to a disappearance of the symptoms in less than 30 minutes.
Case N <o> 3: A 66-year-old man suffered from skin lesions with dry and scaling skin, and especially with pruritus appearing spontaneously and aggravated when handling dusty objects or herbs, with skin redness.
The application of a composition according to the invention led to a disappearance of the pruritus in a few minutes, with a disappearance of the redness in less than 15 minutes. The pruritus which appeared several times a day became less and less frequent, and was systematically calmed by a new application.
Case N <o> 4: A 25-year-old man suffered from relatively old psoriasis, located on the flanks and the lateral part of the upper thighs, and which resisted traditional therapies. The application of the composition according to the invention resulted in a reduction in lesions on the second day, and a disappearance of the pruritus. After several days of application, the improvement became gradual and persistent, removing the flaking. After 3 weeks, the result stabilized.
Case N <o> 5: A 12-year-old child suffered from a skin lesion with a red cardboard, a little cardboard, inflammatory, at the level of the popliteal fossa and the internal part of the right knee. The origin of this lesion was unknown. On the first day of application of a composition according to the invention, a regression of the local inflammatory signs was observed, with a persistence of a localized erythematous zone. On the second day and on the third day, the initial lesion had almost disappeared, but various skin signs somewhat reminiscent of a solar allergy appeared on the inner side of the thigh. By the fourth day, these signs were almost gone. On the fifth day, the skin returned to its normal character.
