Die Verbindungen der Formel I können in Form von zwei Isomerengruppen vorliegen, nämlich die Verbindungen mit der Konfiguration 8R und diejenigen mit der Konfiguration 8S. Die Konfiguration 8R ist bevorzugt.
Erfindungsgemäss gelangt man zu den neuen Verbindungen der Formel I und ihren Salzen, indem man ein Säureadditionssalz einer Verbindung der Formel II,

Das erfindungsgemässe Verfahren kann analog zu
bekannten Methoden für die Kondensation zur Herstellung von analogen cyclischen Peptidergotalkaloiden erfolgen.
Ein geeignetes Säureadditionssalz einer Verbindung der Formel II ist das Hydrochlorid.
Als reaktionsfähige, funktionelle Derivate einer Verbindung der Formel III können beispielsweise das Säurechlorid, das Säureazid oder das gemischte Anhydrid mit Schwefelsäure oder Trifluoressigsäure eingesetzt werden.
Man kann auch das Additionsprodukt einer Verbindung der Formel III mit Dimethylformamid oder Acetamid und Thionylchlorid, Phosgen oder Oxalylchlorid verwenden.
Vorzugsweise wird die Reaktion in Gegenwart von Triäthylamin oder Pyridin durchgeführt. Geeignete Lösungsmittel sind z.B. Chloroform, Methylenchlorid, Dimethylformamid oder Acetonitril.
Die Umsetzung kann bei Temperaturen zwischen -30" bis +20"C durchgeführt werden.
Bei der Kondensation zwischen den Verbindungen der Formeln II und III bleibt die Konfiguration in Stellung 8 erhalten.
Aus den freien Basen der Verbindungen der Formel I lassen sich in bekannter Weise Säureadditionssalze herstellen und umgekehrt. Geeignete Salze schliessen das Hydrochlorid und das Methansulfonat ein.

Die Verbindungen der Formel I und ihre Säureadditions salze sind in der Literatur bisher nicht beschrieben worden.
Sie weisen im Tierversuch interessante pharmakologische
Eigenschaften auf und können daher als Heilmittel verwendet werden.
Insbesondere zeigen diese Verbindungen eine Dopaminrezeptoren-stimulierende Wirkung. Die dopaminergen Eigenschaften konnten an Ratten, bei denen durch eine 6-Hydroxydopamin-Injektion in die substantia nigra eine unilaterale Verletzung der nigro-neostriatalen Dopaminbahn erzeugt wurde, mit Dosen zwischen etwa 0,05 bis 3 mg/kg festgestellt werden [Methode nach U. Ungerstedt Acta physiol. scand.
Suppl. 367, 69-93 (1973]. Nach Verabreichung des Wirkstoffes war eine deutliche Aktivierung dadurch erkennbar, dass die Ratten in Richtung der nicht denervierten Seite rotierten.
Die dopaminergen Eigenschaften der neuen Verbindungen wurden im Apomorphinstereotypie-Test an der Ratte nach Verabreichung von 30 mg/kg i.p. bestätigt.
Die neuen Substanzen können aufgrund ihrer dopaminergen Eigenschaften zur Behandlung von Parkinsonismus Anwendung finden.
Ferner besitzen die neuen Verbindungen eine Prolaktinsekretions-hemmende Wirkung. So hemmen sie bei der Ratte die Implantationen nach subkutaner Applikation von Dosen zwischen 0,01 und 1 mg/kg und die Laktation mit Dosen von 1 bis 10 mg/kg p.o. [Lit.: Experientia 34, 1330(1978)].
Schliesslich weisen die Verbindungen der Formel I gefässtonisierende Aktivität, zudem zentrale Effekte auf und beeinflussen periphere wie auch zentrale Kreislaufparameter. Sie können daher bei der Behandlung der Migräne und orthostatischer Beschwerden bzw. für Thromboseprophylaxe Verwendung finden.
Für die oben genannten Anwendungen variiert die zu verwendende Dosis selbstverständlich je nach verwendeter Substanz, Art der Administration und der gewünschten Behandlung. Im allgemeinen werden aber befriedigende Resultate mit Dosen von ungefähr 0,1 bis 10 mg/kg Körpergewicht erreicht; die Administration kann mit einer Dosis täglich vorgenommen werden oder nötigenfalls in mehreren Teildosen erfolgen. Für grössere Säugetiere liegt die Tagesdosis im Bereich von etwa 0,5 bis 100 mg der Substanz, geeignete Dosierungsformen für z.B. orale Anwendungen enthalten.im allgemeinen ungefähr 0,3 bis 100 mg wirksame Substanz neben festen oder flüssigen Trägersubstanzen oder Verdünnungsmitteln.
Als Heilmittel können die Verbindungen der Formel I bzw.
ihre physiologisch verträglichen Salze allein oder in geeigneter Arzneiform mit pharmakologisch indifferenten Stoffen verabreicht werden.
Die nachfolgenden Beispiele erläutern die Erfindung.
Temperaturangaben erfolgen in Celsiusgraden und sind unkorrigiert.
Beispiel
2-Methyl-9,10-dihydro-p-ergokryptin
60 ml abs. Dimethylformamid werden unter Rühren auf -20" gekühlt und tropfenweise mit einer Lösung von 2,7 g Oxalylchlorid in 8,5 ml Acetonitril versetzt. Anschliessend trägt man 5,0 g (17,6 mMol) trockene 2-Methyl-9,l0-dihydro- lysergsäure ein, wobei unter vorübergehender Lösung ein grauweisser Niederschlag ausfällt. Nach 30 Minuten Rühren bei 0 verdünnt man unter starkem Kühlen mit 18 ml abs.
Pyridin und trägt 3,18 g (8,8 mMol) (2R, 5S, l0aS, 1 ObS)-2-Amino-5-sek.-butyl- 1 Ob-hydroxy-2-isopropyl-ocata hydro-3 ,6-dioxo-8H-oxazolo[3,2-a]pyrrolo[2, 1 -c]pyrazin-hy- drochlorid ein, rührt kräftig während 2 Stunden und lässt die Temperatur langsam von - 10 bis 0 steigen. Zur Aufarbeitung wird unter starker Kühlung mit 36 ml Citrat-Puffer pH 4 verdünnt und mit 2N-Soda-Lösung alkalisch gestellt. Nach Extraktion mit Methylenchlorid, Trocknen und Eindampfen des Extraktes wird das Rohprodukt an der 50fachen Menge Kieselgel chromatographiert, wobei 2-Methyl-9,10-dihydro- ss-ergokryptin mit 3% Methanol in Methylenchlorid eluiert wird und aus Methylenchlorid/Äther kristallisiert. Smp.
187-190 C (Zers.); [a]20= -3,8 (c=0,4 in Chloroform).
Die folgenden Verbindungen der Formel I können analog dem in Beispiel 1 beschriebenen Verfahren hergestellt werden.
