Die als Ausgangsprodukte verwendeten Polypeptidderivate der Formel II können nach für die Synthese von Verbindungen dieser Art allgemein bekannten Methoden hergestellt werden, wobei die Aminosäuren in der festgelegten Reihenfolge einzeln oder nach vorheriger Bildung kleinerer Peptideinheiten miteinander verknüpft werden. Die Verknüpfung der Aminosäuren und/oder Peptideinheiten erfolgt z.
B. in der Weise, dass man eine Aminosäure mit geschützter a -Aminogruppe und aktivierter terminaler Carboxylgruppe mit einer Aminosäure oder einem Peptid mit freier c(-Aminogruppe und freier oder geschützter terminaler Carboxylgruppe umsetzt oder dass man eine Aminosäure oder ein Peptid mit aktivierter ce-Ami- nogruppe und geschützter terminaler Carboxylgruppe mit einer Aminosäure oder einem Peptid mit freier terminaler Carboxylgruppe und geschützter cc-Aminogruppe umsetzt. Die Aminosäuren und Peptide können während der Synthese auch mit löslichen (Bayer-Methode) oder unlöslichen (Merrifield Methode) hochmolekularen Polymer-Schutzgruppen versehen werden. Das Einbauen des Aminoalkohols in die Polypeptide geschieht auf analoge Weise wie für die Aminosäuren.
Die Carboxylgruppe kann beispielsweise durch Überführung in ein Säureazid, -anhydrid, -imidazolid, -isoxazolid oder einen aktivierten Ester oder durch Reaktion mittels eines Carbodiimids oder N,N'-Carbonyldiimidazols aktiviert werden.
Vorzugsweise wird als Kondensationsmethode die Carbodiimidmethode, die Azidmethode, die Methode der aktivierten Ester und die Anhydridmethode verwendet.
An der Reaktion nicht beteiligte freie, funktionelle Gruppen können beim Aufbau des erfindungsgemässen Peptids durch die von der Synthese langkettiger Peptide her bekannten Schutzgruppen geschützt werden.
Die Umwandlung einer nicht mehr benötigten geschützten Aminogruppe in eine freie Gruppe sowie die Umwandlung einer funktionell abgewandelten Carboxylgruppe in eine freie Carboxylgruppe im Laufe des Verfahrens zur Herstellung der neuen Polypeptide erfolgt nach an sich bekannten Methoden durch Behandlung mit hydrolysierenden bzw. reduzierenden Mitteln.
Die Ausgangsprodukte zur Herstellung der neuen Polypep tidderivate können, sofern sie bisher nicht bekannt waren, nach den für die Peptidchemie bekannten Methoden erhalten werden, wobei die Aminosäuren bzw. der Aminoalkohol einzeln oder nach vorheriger Bildung kleinerer Peptideinheiten miteinander verknüpft werden.
Die durch den Rest Z substituierten Hydroxyphenylalaninderivate können z. B. hergestellt werden, indem man das Hydroxyphenylalanin mit einer entsprechenden Carbonylverbindung umsetzt und das entstandene Addukt durch katalytisch erregten Wasserstoff oder durch komplexe Hydride zum N Alkylderivat reduziert. Letzteres wird auf bekanntem Wege mit einer N-Schutzgruppe, z. B. der tert. Butyloxycarbonylgruppe, versehen und zur Peptidsynthese eingesetzt. Auch kann man ein Hydroxyphenylalanin, dessen Stickstoffatom, phenolische Hydroxylgruppe und Carboxylgruppe geschützt sind, z. B. Boc-Tyr(Boc)-OCH3, in einem inerten Lösungsmittel mit einer starken Base, z. B. NaH, und dem entsprechenden Halogenderivat ZHal zum alkylierten N,O,O'-geschützten Hy droxyphenylalanin, z. B. Boc-N-Z-Tyr(BOC)-OCH3, umsetzen. Dieses Derivat wird zur Peptidsynthese, z. B. nach der Azidmethode, eingesetzt.
Vorgängig können auch die 0 Schutzgruppen entfernt werden.
Das Methioninosulfoxid kann z. B. durch Oxidation des Schwefelatoms des Methioninols zu einer Sulfoxidgruppe hergestellt werden. Diese Oxidation kann nach an sich für die
Umsetzung von Thioäthern in Sulfoxide bekannten Verfahren durchgeführt werden. Vorteilhaft findet die Reaktion in saurer wässriger Lösung oder in einem mit Wasser mischbaren Lösungsmittel statt. Als Säure kann man sowohl eine starke (z. B.
HCI) als auch eine schwache Säure (z. B. Essigsäure) benützen.
Als mit Wasser mischbares Lösungsmittel kann man z. B. Methanol verwenden. Als Oxidationsmittel verwendet man vorzugsweise Wasserstoffperoxid. Das Oxidationsmittel wird in der theoretischen Menge eingesetzt.
Die Polypeptidderivate der Formel I und die physiologisch verträglichen Säureadditionssalze bzw. Komplexe dieser Verbindungen weisen im Tierversuch interessante pharmakodynamische Eigenschaften auf. Sie können daher als Heilmittel verwendet werden. Insbesondere besitzen sie analgetische Eigenschaften.
Die Verbindungen zeigen z. B. eine hohe Affinität zum Opiatrezeptor im Rattenhirn. Die Testierung erfolgt wie beschrieben bei C. B. Pert and S. H. Snyder, Molecular Pharmacology 10, 868 (1974). Die ED,,, d.h. die Konzentration bei der 50% des spezifisch gebundenen [3H]-Naloxans verdrängt werden, liegt bei diesen Verbindungen bei 10-5 bis 10 Mol/Liter.
Die analgetischen Eigenschaften zeigen sich auch im Tail Flick-Test an der Maus mit Dosen von 1 bis 50 mg/kg Körpergewicht i.v. Die neuen Verbindungen können deshalb als Heilmittel, insbesondere zur Linderung von Schmerzzuständen verschiedenster Genese, verwendet werden. Die zu verwendenden Dosen variieren naturgemäss je nach Art der Substanz, der Administration und des zu behandelnden Zustandes. Im allgemeinen werden jedoch bei Testtieren befriedigende Resultate mit einer Dosis von ca. 0,4 bis 60 mg/kg Körpergewicht erhalten. Diese Dosis kann nötigenfalls in 2 bis 4 Anteilen oder auch als Retardform verabreicht werden. Für grössere Säugetiere liegt die Tagesdosis bei etwa 30 bis 350 mg. So enthalten z. B. für orale Applikationen die Teildosen etwa 7,5 bis 175 mg der Verbindungen der Formel I neben festen oder flüssigen Trägersubstanzen.
Die erfindungsgemäss erhaltenen Verbindungen der Formel I können als Heilmittel Verwendung finden. Diese Heilmittel, beispielsweise eine Lösung oder eine Tablette, können nach bekannten Methoden, unter Verwendung der üblichen Hilfs- und Trägerstoffe, hergestellt werden.
