5. Verfahren nach Anspruch 1, dadurch gekennzeichnet, dass man die Umsetzung bei einer Temperatur von 20 bis 50"C ausführt.
6. Verfahren nach Anspruch 1, dadurch gekennzeichnet, dass das 'molare Verhältnis des Oxidationsmittels zur Verbindung der Formel III das 3- bis 1 5fach beträgt.

Dieses Verfahren kann jedoch ausschliesslich auf solche Verbindungen angewandt werden, welche in der 1-Stellung des Benzodiazepin-Ringes keine Substituenten tragen und kann nicht für Verbindungen Anwendung finden, welche an der 1 Stellung Substituenten, wie eine Methylgruppe, aufweisen.
Umfangreiche Untersuchungen zur Überwindung dieser Nachteile führten zu einem Verfahren, das allgemeiner für die Herstellung von Dihydrobenzodiazepin-D erivaten anwendbar ist. worin R1, R2, X und Y der obigen Definition entsprechen, mit einem Oxidationsmittel, die Entformylierung an der 4-Stellung des Benzodiazepin-Ringes und die Oxidation der Kohlenstoff-Stickstoff-Bindung in der 4,5-Stellung gleichzeitig ein und führt zu den Benzodiazepin-2-on-Derivaten der Formel I, auch dann, wenn Substituenten in der 1-Stellung des Benzodiazepin-Ringes vorhanden sind.
Die Dihydrobenzodiazepin-2-on-Derivate der Formel I zeigen eine starke Zentralnervensystem-dämpfende Wirkung, wie sedierende Wirkung, antikonvulsive Wirkung, muskelrelaxierende Wirkung oder hypnotische Wirkung und sind daher äusserst wertvoll als Sedativa, Antikonvulsiva, Muskel Relaxantien und Tranquilizers.
Es ist daher ein Ziel der Erfindung, ein in industriellem Massstab vorteilhaftes Verfahren zur Herstellung von Verbindungen der Formel I, wie sie oben definiert sind, zu schaffen.
Weitere Ziele ergeben sich aus der nachfolgenden Beschreibung.
Um diese Zielsetzungen zu erfüllen, schafft die vorliegende Erfindung ein Verfahren zur Herstellung eines Dihydrobenzodiazepin-2-ons der Formel

In der vorliegenden Beschreibung bedeutet der Ausdruck C1-C4-Alkyl ein gerad- oder verzweigtkettiges Alkyl mit 1 bis 4 Kohlenstoffatomen, wie Methyl, Äthyl oder Propyl. Der Ausdruck Halogenalkyl umfasst vorzugsweise ein gerad- oder verzweigtkettiges Cl-C4-Alkyl, das durch ein Halogenatom (z.B. Chlor, Fluor und Brom) substituiert ist, z.B. 2,2,2-Trifluoräthyl. Der Ausdruck Cycloalkylalkyl umfasst vorzugsweise C3-CrCycloalkyl-Cl-C4-Alkyl, beispielsweise Cyclopropylmethyl, Cyclopropyläthyl, Cyclohexylmethyl und dergleichen.
Der Ausdruck Alkoxyalkyl umfasst vorzugsweise ein C1-C4 -Alkoxy-Cl-C4-Alkyl, beispielsweise Methoxymethyl, Methoxyäthyl, Äthoxymethyl, Äthoxyäthyl und dergleichen. Der Ausdruck Acyloxyalkyl umfasst vorzugsweise ein C2-C5-AI- kanoyloxy-C1-C4-Alkyl, beispielsweise Acetoxymethyl und Acetoxyäthyl. Der Ausdruck Halogen bedeutet Chlor, Brom, Jod oder Fluor.
Das erfindungsgemässe Verfahren wird durch Umsetzung der 4-Formylbenzodiazepin-2-on-Derivate der Formel III mit einem geeigneten Oxidationsmittel durchgeführt. Als Oxidationsmittel ist Chromsäure besonders bevorzugt, die Erfindung ist jedoch selbstverständlich nicht darauf beschränkt.
Der bevorzugte Anteil des Oxidationsmittels zur Verbindung der Formel III ist ein 3- bis 15fach molarer, istjedoch nicht darauf beschränkt.
Im allgemeinen verläuft die Umsetzung vorteilhafterweise in Gegenwart eines Lösungsmittels. Zu Lösungsmitteln gehören beispielsweise Wasser, Aceton, Tetrachlorkohlenstoff, Chloroform, Essigsäure, Schwefelsäure, Tetrahydrofuran, Dioxan, Methanol und Äthanol oder sie können in passender Weise aus anderen inerten Lösungsmitteln gewählt werden, welche die Umsetzungnicht stören. Die Menge des eingesetzten Lösungsmittel hängt von der Art der Ausgangsmaterialien ab und beträgt vorzugsweise das 5- bis 40fache Gewicht, bezogen auf die Verbindung III.
Die Umsetzung verläuft im allgemeinen bei Raumtemperatur, die bevorzugte Temperatur istjedoch 20 bis 50"C. Der Ablauf der Umsetzung kann durch Erwärmen oder Kühlen gelenkt werden. Die Reaktionszeit hängt von den Ausgangsmaterialien und Reaktionstemperaturen ab und im allgemeinen ist die Umsetzung in etwa 5 Stunden beendet.
Die Tetrahydro-4-formylbenzodiazepin-2-on-Derivate der Formel III, welche erfindungsgemäss als Ausgangsmaterial verwendet werden, können leicht durch Behandeln von Di hydro-4-formylbenzodiazepin-D erivaten der Formel

Nach dem erfindungsgemässen Verfahren werden beispielsweise die nachfolgenden Dihydrobenzodiazepin-2-on -Derivate hergestellt: 5-Phenyl-7-chlor- 1 ,3-dihydro-2H- 1,4-benzodiazepin-2-on, l-Methyl-5-phenyl-7-chlor- 1,3-dihydro-2H- l,4-benzodia- zepin-2-on, I-Cyclopropylmethyl-5-phenyl-7-chlor- 1,3-dihydro-2H- 1,4 -benzodiazepin-2-on, 1 -M ethyl-5-( o-chlorphenyl)-7-chlor- 1,3- dihydro-2H- 1,4-benzodiazepin-2-on, l-Methyl-5-(o-fluorphenyl)-7-chlor- 1 ,3-dihydro-2H- 1,4-benzodiazepin-2-on, I-Methyl-5-phenyl-7-nitro- 1 ,3-dihydro-2H- 1 ,4-benzodiazepin- -2-on.
Die vorliegende Erfindung wird durch das nachfolgende Beispiel erläutert.
Beispiel
In 20 ml Eisessig löst man 1,0 g 1-Methyl-4-fonmyl-5-phe- nyl-7-chlor-1,3,45-tetrahydro-2H-1,4-benzodiazepin-2-on, fügt der erhaltenen Lösung unter Kühlen tropfenweise eine Lösung von 2,0 g Chromsäureanhydrid in 1,6 ml Wasser zu und lässt dann das Gemisch während 24 Stunden unter Rühren bei 25"C reagieren.
Der Reaktionslösung werden 50 ml Eiswasser zugefügt und das Gemisch wird unter Kühlen mit einer 30%igen wässrigen Ätznatron lösung neutralisiert und dann mit Methylenchlorid extrahiert. Der organische Extrakt wird über Natriumsulfat getrocknet und das Lösungsmittel unter vermindertem Druck entfernt. Den so erhaltenen Rückstand löst man in Tetrachlorkohlenstoff, extrahiert mit 5%Der wässriger Chlorwasserstoffsäure. neutralisiert die Chlorwasserstoffschicht mit wässrigem Ammoniak und extrahiert neuerlich mit Methylenchlorid. Der organische Extrakt wird über Natriumsulfat getrocknet und das Lösungsmittel unter vermindertem Druck entfernt. Der so erhaltene Rückstand wird aus Äther kristallisiert und ergibt 0,4 g 1 Methyl-5-phenyl-7-chlor- l,3-dihydro-2H- l,4-benzo- diazepin-2-on (Fp. 123 bis 1260C).
Die Tetrachlorkohlenstoffschicht wird mit wässrigem Ammoniak neutralisiert. über Natriumsulfat getrocknet und das Lösungsmittel wird unter vermindertem Druck entfernt. Der Rückstand wird aus Äther und Äthylacetat kristallisiert und ergibt 0,2 g des Ausgangsmaterials.
Das als Ausgangsmaterial in der obigen Umsetzung verwendete 1 -M ethyl-4förmyl-5-phenyl-7-chlor- 1,3,4,5-tetra- hydro-2H- 1,4-benzodiazepin-2-on wird in folgender Weise hergestellt:
A. In 150 ml Dioxan löst man 5,0 g l-Methyl-4-formyl-5- -phenyl-7-chlor-2,3,4, 5-tetrahydro- 1 H- 1,4-benzodiazepin, fügt der erhaltenen Lösung unter Kühlen tropfenweise eine Lösung von 3,67 g Kaliumpermanganat in 67,5 ml Wasser zu und rührt dann das Gemisch während 6 Stunden bei 25 C.
Nach dem Abkühlen der Reaktionslösung werden 15,5 ml Ameisensäure zugefügt und das Gemisch dann filtriert und mit Dioxan gewaschen. Das Filtrat wird mit 0,5 g Aktivkohle behandelt, mit wässrigem Ammoniak neutralisiert und dann in eine organische und eine wässrige Schicht getrennt. Die wässrige Schicht wird weiter mit Chloroform extrahiert und die vereinigte organische Schicht mit einer gesättigten wässrigen Natriumchloridlösung gewaschen. Die erhaltene organische Schicht wird über Natriumsulfat getrocknet und das Lösungsmittel wird unter vermindertem Druck entfernt. Den so erhaltenen Rückstand reinigt man mittels Chromatographie (Chloroform und 70 g Silikagel) und erhält 4,3 g l-Methyl-4- -formyl-5-phenyl-7-chlor- l,3,4,5-tetrahydro-2H- I ,4-benzo- diazepin-2-on.
Das Produkt wird aus Isopropylalkohol umkri stallisiertund ergibt farblose Prismen (Fp. 162 bis 164"C).
B. Man erhitzt eine Mischung aus 1 ml Essigsäureanhydrid und 0,5 ml 100%igerAmeisensäure während 20 Minuten unter Rühren bei 53 bis 56"C, fügt nach dem Abkühlen der erhaltenen Lösung 1 ml Ameisensäure und dann 0,2 g l-Methyl-5- -phenyl-7-chlor- 1 ,3,4,5-tetrahydro-2H- 1 ,4-benzodiazepin-2- on zu und rührt das erhaltene Gemisch während 4 Stunden bei 3 bis 8 C. Der Reaktionslösung wird eine kleine Menge Wasser zugefügt und das Gemisch mit einer 30%igen wässrigen Ätznatronlösung neutralisiert. Die ausgeschiedenen Kristalle werden abfiltriert, mit Wasser gewaschen und getrocknet und ergeben 0,21 g l-Methyl-4-formyl-5-phenyl-7-chlor- 1,3,4,5-tetrahydro-2H- 1,4-benzodiazepin-2-on.
Umkristallisieren aus Isopropylalkohol führt zu farblosen Prismen (Fp. 162 bis 164 C). Die so erhaltenen Kristalle stimmen in jeder Hinsicht mit der nach dem obigen Verfahren A erhaltenen Probe überein.

In the present description the term C1-C4-alkyl means a straight or branched chain alkyl with 1 to 4 carbon atoms, such as methyl, ethyl or propyl. The present invention is illustrated by the following example.

1.0 g of 1-methyl-4-formyl-5-phenyl-7-chloro-1,3,45-tetrahydro-2H-1,4-benzodiazepin-2-one is dissolved in 20 ml of glacial acetic acid, adds the solution obtained with cooling dropwise a solution of 2.0 g of chromic anhydride in 1.6 ml of water and then the mixture is allowed to react for 24 hours with stirring at 25 ° C.
