Die Erfindung betrifft ein Verfahren zur Herstellung von Benzodiazepin-Derivaten der Formel

worin R und R1 Wasserstoff oder Halogen und R2 Formyl, Niederalkanoyl, Benzoyl oder Niederalkyloxyniederalkanoyl bedeuten, bzw. von Säureadditionssalzen dieser Verbindungen.
Der Ausdruck niederes Alkyl bezeichnet eine geradkettige oder verzweigte aliphatische Kohlenwasserstoffgruppe mit vorzugsweise 1 bis 4 Kohlenstoffatomen, wie z.B. eine Methyl-, Äthyl- oder n-Propylgruppe. Der Ausdruck niederes Alkanoyl bezeichnet den Rest einer aliphatischen Carbonsäure mit vorzugsweise höchstens 5 Kohlenstoffatomen, wie z.B. Acetyl, Propionyl und dgl. Unter den Bezeichnungen niederes Alkoxyniederalkyl und niederes Alkyloxyniederalkanoyl ist ein durch eine niedere Alkyloxygruppe substituierter niederer Alkyl- bzw. Alkanoylrest (beispielsweise ein Methoxymethyl- bzw. Methoxyacetylrest) zu verstehen, wobei der Ausdruck niedere Alkyloxygruppe eine über ein Sauerstoffatom gebundene niedere Alkylgruppe (wie z.B.
Methoxy) bezeichnet. Unter der Bezeichnung Halogen sind die vier Halogene Fluor, Chlor, Brom und Jod zu verstehen, sofern nichts anderes angegeben ist.
Das erfindungsgemässe Verfahren ist dadurch gekennzeichnet, dass man eine Verbindung der Formel

worin R und R1 vorstehend angegebene Bedeutung besitzen und R3 für Wasserstoff, niederes Alkyl, Phenyl oder niederes Niederalkoxyniederalkyl steht, oder ein Salz davon, mit Chromsäure oxidiert. Erwünschtenfalls kann eine erhaltene Verbindung in an sich bekannter Weise zur entsprechenden in 1-Stellung unsubstituierten Verbindung hydrolysiert werden. Eine erhaltene in 1-Stellung unsubstituierte Verbindung kann zur Herstellung von entsprechenden methylierten Derivaten dienen.
Gegebenenfalls kann eine erhaltene Verbindung mit einer Säure in ein Salz übergeführt werden.
Eine besondere Ausführungsform des erfindungsgemässen Verfahrens besteht darin, dass man als Ausgangsprodukt eine Verbindung der Formel II einsetzt, worin R3 Phenyl oder niederes Alkoxyniederalkyl bedeutet, wie z.B. 1 -(2-Aminoäthyl YS-chlor- 2-methoxymethyl-3-phenylindol.
Nach einer weiteren besonderen Ausführungsform des erfindungsgemässen Verfahrens stellt man zunächst eine Verbindung der Formel I her (z.B. I-Acetyl-7-chlor-2,3-dihydro-5- phenyl- 1 H- 1 ,4benzodiazepin oder7-Chlor-2,3-dihydro- 1 methoxyacetyl-5-phenyl-lH-1,4-benzodiazepinX worauf man diese durch Hydrolyse in eine der Formel I entsprechende Verbindung, worin R2 jedoch Wasserstoff bedeutet, überführt, beispielsweise in 7-Chlor-2,3-dihydro-5-phenyl- 1 H- 1 ,4benzodiazepin.
Nach einer dritten besonderen Ausführungsform synthetisiert man zunächst - wie vorstehend beschrieben über eine Verbindung der Formel I - eine der Formel I entsprechende Verbindung, worin R2 jedoch Wasserstoff bedeutet. Die erhaltene Verbindung kann anschliessend zu einer entsprechenden Verbindung methyliert werden, worin R2 jedoch eine Methylgruppe bedeutet, z.B.
7-Chlor-2,3-dihydro- 1 -methyl-5-phenyl- 1 H- l,4benzodiazepin.
Für die Überführung von Verbindungen der Formel II oder deren Salze in solche der Formel I oder deren Salze sind die Reaktionsparameter, wie Lösungsmittel, Temperatur, Reaktionszeit und Druck variierbar. So kann die Reaktion in einem inerten organischen oder anorganischen Lösungsmittel vorgenommen werden. Beispiele für derartige inerte organische und anorganische Lösungsmittel sind Eisessig, Wasser, Schwefelsäure, Pyridin, Aceton und Dimethylformamid. Ein besonders bevorzugtes Lösungsmittel ist Eisessig. Der Druck ist kein kritisches Merkmal für die Reaktion. Die Reaktionszeit ist variierbar in Abhängigkeit von der Reaktionstemperatur. Bei höheren Temperaturen werden kürzere Reaktionszeiten benötigt. Günstige Reaktionszeiten sind eine halbe Stunde bis 24 Stunden. Bevorzugt ist eine Reaktionszeit von 1 bis 8 Stunden.
Der Reaktionstemperatur ist ganz allgemein eine obere Grenze gesetzt, nämlich diejenige, bei der eine Zersetzung des Ausgangsmaterials und/oder des Reaktionsproduktes eintritt. Ein für die Reaktion günstiges Temperaturintervall liegt zwischen -10 und 50 . Besonders bevorzugt sind Temperaturen zwischen 20 und 30 . In besonderen Fällen kann die Reaktion auch bei einer Temperatur unterhalb - 10" vorgenommen werden. Temperaturen unterhalb - 50 kommen jedoch kaum in Betracht, da dann die Reaktionsgeschwindigkeit zu klein wird.
Verbindungen der Formel I lassen sich in bekannter Weise zu der Formel I entsprechenden Verbindungen, worin R2 jedoch Wasserstoff bedeutet, hydrolysieren. Diese Verbindungen können in an sich bekannter Weise in l-Stellung methyliert werden.
Verbindungen der Formel I, die basischen Charakter besitzen, lassen sich mit anorganischen wie auch mit organischen Säuren in Salze überführen. Beispiele für derartige Säuren sind Salpetersäure, Schwefelsäure, Salzsäure, Bromwasserstoffsäure, Phosphorsäure, Essigsäure, Zitronensäure, Bernsteinsäure, Benzoesäure, Ameisensäure, p-Toluolsulfonsäure und dergleichen.
Die Ausgangsmaterialien der allgemeinen Formel II sind neue Vorbindungen. Sie können hergestellt werden, indem man von

worin R, R1 und R3 vorstehende Bedeutung besitzen, ausgeht, welche ihrerseits in Analogie zu den Vorschriften in J. Proc.
Roy. Soc. N.S. Wales 71, 475-485 (1938) dargestellt werden können.
Verbindungen der Formel III können durch Einführung einer
2-Aminoäthylgruppe in die entsprechenden Verbindungen der
Formel II übergeführt werden, beispielsweise durch Umsetzung mit
Chloracetonitril und anschliessende Reduktion mit Lithiumaluminiumhydrid oder durch Behandeln mit N-(2-BromäthylS phthalimid und anschliessende Hydrazinolyse.
Die Verfahrensendprodukte besitzen antikonvulsive, muskelrelazierende und tranquillisierende Eigenschaften.
In den nachstehenden Beispielen sind alle Temperaturen in Grad Celsius angegeben.
Beispiel 1:
3 g Chromsäureanhydrid werden in 3 ml Wasser gelöst. Nach Verdünnen mit 30 ml Eisessig werden bei 20-30 3,2 g l-(2-Amino- äthyl > 5-chlor-2-methyl-3-phenylindol-hydrochlorid portionsweise eingetragen. Man rührt bei Raumtemperatur während 7 Stunden, verdünnt das Reaktionsgemisch mit Eiswasser und stellt mit konzentriertem Ammoniak alkalisch. Die Basen werden mehrmals mit Äther extrahiert, die Extrakte werden getrocknet und eingedampft. Nach Chromatographie des Rückstandes an 60 g Kieselgel mit Essigester erhält man nach Kristallisation aus Äther Hexan l-Acetyl-7-chlor-2,3-dihydro-5-phenyl- I H- 1,4-benzo- diazepin vom Schmelzpunkt 165-167".
Ein Gemisch von 0,5 g l-Acetyl-7-chlor-2,3-dihydro-5 phenyl-l H-1,Cbenzodiazepin, 20 ml Äthanol und 2 ml 4n Natronlauge wird während 2 Stunden unter Rückfluss gekocht. Nach dem Einengen im Vakuum versetzt man mit Wasser und saugt das ausgefallene Produkt ab. Umkristallisation aus essigester-Hexan liefert 7-Chlor-2,3-dihydro-5-phenyl- 1 H- 1,4-benzodiazepin vom Schmelzpunkt 168-170".
Zu einer Lösung von 1 g 7-Chlor-2,3-dihydro-5-phenyl-lH- 1,4-benzodiazepin in 10 ml Dimethylformamid werden 0,3 g Natriumhydrid-suspension (50%ig in Mineralöl) zugefügt. Das Reaktionsgemisch wird während 30 Minuten auf 60-70 erwärmt, danach auf Raumtemperatur abgekühlt und mit 0,4 ml Methyljodid versetzt. Man rührt während einer Stunde bei Raumtemperatur, gibt hierauf 1 ml Methanol zu, giesst das Reaktionsgemisch in Wasser und extrahiert dreimal mit Äther. Die Extrakte werden getrocknet und eingedampft. Chromatographie des Rückstandes an 25 g Kieselgel (0,05-0,2 mm) mit Essigester liefert nach Kristallisation aus Äther-Hexan 7-Chlor-53-dihydro-l-methyl- 5-phenyl-1H-1,4-benzodiazepin vom Schmelzpunkt 95-97".
Das Ausgangsmaterial kann folgendermassen hergestellt werden:
Eine Mischung von 67,1 g Phenylaceton, 99 g 4-Chlor phenylhydrazin-hydrochlorid, 500 ml Äthanol und 500 ml l0%iger äthanolischer Salzsäure wird während 4 Stunden unter Rückfluss gekocht. Der nach Einengen im Vakuum verbleibende Rückstand wird zwischen Äther und wässriger Salzsäure verteilt. Die Ätherphase wird mit verdünnter Salzsäure und mit Wasser gewaschen, getrocknet und eingedampft. Der erhaltene Rückstand liefert durch Kristallisation aus 70%igem Methanol 5- Chlor-2-methyl- 3- phenylindol vom Schmelzpunkt 86-87".
36,3 g 5-Chlor-2-methyl-3-phenylindol werden bei 0" portionsweise in eine Suspension von 7,5 g Natriumhydrid in 225 ml Dimethylformamid eingetragen. Nach beendeter Zugabe wird während 40 Minuten bei 0-5" gerührt. Zu der auf - 30" abgekühlten Reaktionsmischung werden darauf 19,5 ml Chloracetonitril zugetropft. Nach beendeter Zugabe lässt man die Temperatur innerhalb einer Stunde auf 20 ansteigen, giesst das Gemisch in 400 ml Eiswasser und extrahiert mehrmals mit Äther. Die Auszüge werden mit Wasser gewaschen, über Natriumsulfat getrocknet und eingedampft. Das erhaltene öl wird in Methanol gelöst und in der Kälte kristallisiert. Man erhält 5-Chlor-2-methyl-3-phenylindol-l-acetonitril vom Schmelzpunkt 127-129".
Zu einer Suspension von 5 g Lithiumaluminiumhydrid in 200 ml Äther werden portionenweise 10 g 5-Chlor-2-methyl-3phenylindol-l-acetonitril zugefügt. Nach der Zugabe wird während 2 Stunden bei Raumtemperatur gerührt und mit 25 ml Wasser hydrolysiert. Das anorganische Material wird abgesaugt und gut mit Benzol ausgewaschen. Das Filtrat wird eingedampft; der Rückstand wird in Äthanol aufgenommen und mit äthanolischer Salzsäure auf pH 5 gebracht. Das auskristallisierte Hydrochlorid wird abgesaugt, mit Äther gewaschen und getrocknet. Man erhält 1 -(2-Aminoäthyl 5-chlor-2-methyl-3-phenylindol-hydro chlorid, welches unter Zersetzung bei 285-290" schmilzt.
Beispiel 2:
1,76 g 1 -(2-Aminoäthyl5-chlor-2-methoxymethyl-3-phenyl- indol-hydrochlorid werden zu einer gerührten Mischung von 1,8 g Chromsäureanhydrid, 2,5 ml Wasser und 20 ml Eisessig zugefügt. Nachdem während 4 Stunden bei Raumtemperatur gerührt wurde, wird mit Eiswasser verdünnt, mit konzentriertem Ammoniak alkalisch gestellt und mehrmals mit Äther extrahiert.
Die mit Wasser gewaschenen und über Natriumsulfat getrockneten Auszüge werden eingedampft. Chromatographie des Rückstandes an 50 g Kieselgel mit 10% Äthanol in Essigester liefert nach Kristallisation aus Äther-Hexan 7-Chlor-2,3-dihydro- 1-methoxy- acetyl-5-phenyl-1H-l,4benzodiazepin vom Schmelzpunkt 140-141".
Die Verseifung von 7-Chlor-2,3-dihydro- 1-methoxyacetyl-5- phenyl-lH-1,4-benzodiazepin nach der im Beispiel 1, Absatz 2, angegebenen Vorschrift liefert 7-Chlor-2,3-dihydro-5-phenyl IH- 1,4-benzodiazepin.
Das Ausgangsmaterial kann wie folgt hergestellt werden:
In eine Suspension von 20 g Lithiumaluminiumhydrid in 400 ml Tetrahydrofuran werden bei. 10-20" 60 g 5-Chlor-3-phenylindol-2-carbonsäureäthylester eingetragen. Nach beendeter Zugabe lässt man während 15 Minuten rühren und hydrolysiert das Gemisch bei 10-20 durch vorsichtiges Zutropfen von 100 ml Wasser. Man verdünnt mit Äther, trennt vom anorganischen Material ab und dampft das Filtrat ein, zuletzt unter Zusatz von Benzol. Kristallisation des Rückstandes aus Benzol-Hexan liefert 5-Chlor-3-phenylindol-2-methanol, das aus Methylenchlorid Hexan umkristallisiert wird und dann bei 123-125" schmilzt.
Eine Lösung von 12,8 g 5-Chlor-3-phenylindol-2-methanol in 300 ml Methanol wird mit 10 Tropfen konzentrierter Schwefelsäure versetzt und während 15 Minuten auf dem Dampfbad erwärmt. Nach Neutralisation mit Triäthylamin wird eingedampft und der Rückstand zwischen Äther und Wasser verteilt. Die mit Wasser gewaschene und über Natriumsulfat getrocknete Ätherphase wird eingedampft und das erhaltene Rohprodukt aus Benzol-Hexan kristallisiert. Man erhält 5-Chlor-2-methoxymethyl3-phenylindol. Eine chromatographisch gereinigte Probe zeigt nach Kristallisation aus Benzol-Hexan einen Schmelzpunkt von 123-124".
Ein Gemisch von 13,6 g 5-Chlor-2-methoxymethyl-3-phenyl- indol, 3,75 g Natriumhydrid und 100 ml Dimethylformamid wird bei 0-5" während 30 Minuten gerührt. Hierauf kühlt man auf - 40" ab, gibt 7,5 ml Chloracetonitril zu und lässt die Temperatur innerhalb von einer Stunde auf Raumtemperatur anteigen. Durch Zutropfen von 100 ml Wasser unter gleichzeitigem Kühlen kristallisiert das Produkt aus dem Reaktionsgemisch. Die Kristalle werden abgesaugt, mit Wasser gewaschen, getrocknet und aus Methanol-Methylenchlorid umkristallisiert. Man erhält 5-Chlor-2 methoxymethyl-3-phenylindol- 1-acetonitril vom Schmelzpunkt 126-128".
Zu einer Suspension von 1 g Lithiumaluminiumhydrid in 40 ml Äther werden 3,1 g 5-Chlor-2-methoxymethyl-3-phenylindoll-acetonitril zugefügt. Nach der Zugabe wird während einer Stunde bei Raumtemperatur gerührt. Das nach üblicher Aufarbeitung anfallende Rohprodukt wird in Äther aufgenommen und durch Zugabe von methanolischer Salzsäure in sein Hydrochlorid über geführt. Man erhält 1 -(2-Aminoäthyl 5-chlor-2-methoxymethyl- 3-phenylindol-hydrochlorid, das nach Umkristallisation aus Wasser-Methanol bei 207-212 unter Zersetzung schmilzt.

UNTERANSPRÜCHE
1. Verfahren nach Patentanspruch 1, dadurch gekennzeichnet, dass man eine erhaltene Verbindung der Formel I zur entsprechenden in 1-Stellung unsubstituierten Verbindung hydrolysiert.
2. Verfahren nach Patentanspruch I, dadurch gekennzeichnet, dass man bei der Oxidation Eisessig, Schwefelsäure, Pyridin, Aceton oder Dimethylformamid als Lösungsmittel verwendet.
3. Verfahren nach Patentanspruch I, dadurch gekennzeichnet, dass man eine Verbindung der Formel II, worin R3 Phenyl oder Niederalkyloxyniederalkyl bedeutet, oder ein Salz davon oxidiert.
4. Verfahren nach Patentanspruch I, dadurch gekennzeichnet, dass man eine Verbindung der Formel II, worin R Chlor, R1 Wasserstoffund R3 Methyl oder Methoxymethyl bedeuten, oxidiert.
5. Verfahren nach Unteranspruch 1 und 4, dadurch gekennzeichnet, dass man die erhaltene Verbindung der Formel I, worin R Chlor, R1 Wasserstoff und R2 Acetyl oder Methoxyacetyl bedeuten, hydrolysiert.
6. Verfahren nach Patentanspruch I oder einem der Unteransprüche 1-5, dadurch gekennzeichnet, dass man eine erhaltene Verbindung ihit einer Säure in ein pharmazeutisch verwendbares Säureadditionssalz überführt.
PATENTANSPRUCH II
Verwendung von gemäss dem Verfahren nach Unteranspruch 1 erhaltenen, in l-Stellung unsubstituierten Benzodiazepin-Derivaten zur Herstellung von entsprechenden in 1-Stellung methylierten Derivaten durch Methylierung.

The term lower alkyl denotes a straight or branched chain aliphatic hydrocarbon group having preferably 1 to 4 carbon atoms, e.g. a methyl, ethyl or n-propyl group. The term lower alkanoyl denotes the radical of an aliphatic carboxylic acid with preferably at most 5 carbon atoms, e.g. Acetyl, propionyl and the like. The terms lower alkoxy-lower alkyl and lower alkyloxy-lower alkanoyl are to be understood as meaning a lower alkyl or alkanoyl radical (for example a methoxymethyl or methoxyacetyl radical) substituted by a lower alkyloxy group, the expression lower alkyloxy group being a lower one bonded via an oxygen atom Alkyl group (such as

Examples of such inert organic and inorganic solvents are glacial acetic acid, water, sulfuric acid, pyridine, acetone and dimethylformamide. A particularly preferred solvent is glacial acetic acid. The pressure is not a critical characteristic of the reaction. The reaction time can be varied depending on the reaction temperature. Shorter reaction times are required at higher temperatures. Favorable reaction times are half an hour to 24 hours. A reaction time of 1 to 8 hours is preferred.
The reaction temperature is generally set an upper limit, namely that at which decomposition of the starting material and / or of the reaction product occurs. A favorable temperature range for the reaction is between -10 and 50. Temperatures between 20 and 30 are particularly preferred. Compounds of the formula III can by introducing a

In the examples below, all temperatures are given in degrees Celsius.
Example 1:
3 g of chromic anhydride are dissolved in 3 ml of water. After dilution with 30 ml of glacial acetic acid, 3.2 g of 1- (2-amino-ethyl> 5-chloro-2-methyl-3-phenylindole hydrochloride) are added in portions at 20-30. A mixture of 0.5 g of 1-acetyl-7-chloro-2,3-dihydro-5 phenyl-1 H-1, Cbenzodiazepine, 20 ml of ethanol and 2 ml of 4N sodium hydroxide solution is refluxed for 2 hours. The starting material can be produced as follows:
A mixture of 67.1 g of phenylacetone, 99 g of 4-chloro phenylhydrazine hydrochloride, 500 ml of ethanol and 500 ml of 10% ethanolic hydrochloric acid is refluxed for 4 hours. The residue remaining after concentration in vacuo is partitioned between ether and aqueous hydrochloric acid. The ether phase is washed with dilute hydrochloric acid and with water, dried and evaporated. The residue obtained gives 5-chloro-2-methyl-3-phenylindole with a melting point of 86-87 "by crystallization from 70% methanol.
36.3 g of 5-chloro-2-methyl-3-phenylindole are added in portions at 0 "to a suspension of 7.5 g of sodium hydride in 225 ml of dimethylformamide. When the addition is complete, the mixture is stirred at 0-5" for 40 minutes. 19.5 ml of chloroacetonitrile are then added dropwise to the reaction mixture, which has cooled to -30 ". 10 g of 5-chloro-2-methyl-3phenylindole-1-acetonitrile are added in portions to a suspension of 5 g of lithium aluminum hydride in 200 ml of ether. After the addition, the mixture is stirred for 2 hours at room temperature and hydrolyzed with 25 ml of water. The inorganic material is sucked off and washed out well with benzene. The filtrate is evaporated; the residue is taken up in ethanol and brought to pH 5 with ethanolic hydrochloric acid. The crystallized hydrochloride is filtered off with suction, washed with ether and dried. 1 - (2-Aminoethyl 5-chloro-2-methyl-3-phenylindole hydrochloride, which melts with decomposition at 285-290 ", is obtained.
Example 2:
1.76 g of 1- (2-aminoethyl5-chloro-2-methoxymethyl-3-phenylindole hydrochloride are added to a stirred mixture of 1.8 g of chromic anhydride, 2.5 ml of water and 20 ml of glacial acetic acid Hours was stirred at room temperature, is diluted with ice water, made alkaline with concentrated ammonia and extracted several times with ether.

The saponification of 7-chloro-2,3-dihydro- 1-methoxyacetyl-5-phenyl-1H-1,4-benzodiazepine according to the instructions given in Example 1, paragraph 2, gives 7-chloro-2,3-dihydro- 5-phenyl IH-1,4-benzodiazepine.
The starting material can be made as follows:
In a suspension of 20 g of lithium aluminum hydride in 400 ml of tetrahydrofuran are at. 10-20 "60 g of 5-chloro-3-phenylindole-2-carboxylic acid ethyl ester added. After the addition is complete, the mixture is stirred for 15 minutes and the mixture is hydrolyzed at 10-20 by carefully adding 100 ml of water dropwise. A solution of 12.8 g of 5-chloro-3-phenylindole-2-methanol in 300 ml of methanol is mixed with 10 drops of concentrated sulfuric acid and heated on the steam bath for 15 minutes. After neutralization with triethylamine, it is evaporated and the residue is partitioned between ether and water. The ether phase, washed with water and dried over sodium sulfate, is evaporated and the crude product obtained is crystallized from benzene-hexane. 5-chloro-2-methoxymethyl3-phenylindole is obtained. A chromatographically purified sample has a melting point of 123-124 "after crystallization from benzene-hexane.
A mixture of 13.6 g of 5-chloro-2-methoxymethyl-3-phenylindole, 3.75 g of sodium hydride and 100 ml of dimethylformamide is stirred at 0-5 "for 30 minutes. It is then cooled to -40", add 7.5 ml of chloroacetonitrile and allow the temperature to rise to room temperature within an hour. The product crystallizes from the reaction mixture by adding dropwise 100 ml of water with simultaneous cooling. The crystals are filtered off with suction, washed with water, dried and recrystallized from methanol-methylene chloride. 5-chloro-2-methoxymethyl-3-phenylindole-1-acetonitrile with a melting point of 126-128 "is obtained.
3.1 g of 5-chloro-2-methoxymethyl-3-phenylindoll-acetonitrile are added to a suspension of 1 g of lithium aluminum hydride in 40 ml of ether. After the addition, the mixture is stirred for one hour at room temperature. The crude product obtained after customary work-up is taken up in ether and converted into its hydrochloride by adding methanolic hydrochloric acid. 1 - (2-Aminoethyl 5-chloro-2-methoxymethyl-3-phenylindole hydrochloride is obtained, which, after recrystallization from water-methanol, melts at 207-212 with decomposition.

SUBCLAIMS

3. The method according to claim I, characterized in that a compound of the formula II in which R3 is phenyl or lower alkyloxy-lower alkyl, or a salt thereof, is oxidized.
4. The method according to claim I, characterized in that a compound of the formula II in which R is chlorine, R1 is hydrogen and R3 is methyl or methoxymethyl is oxidized.
5. The method according to dependent claims 1 and 4, characterized in that the compound of formula I obtained, in which R is chlorine, R1 is hydrogen and R2 is acetyl or methoxyacetyl, is hydrolyzed.
6. The method according to claim I or one of the dependent claims 1-5, characterized in that a compound obtained is converted ihit an acid into a pharmaceutically usable acid addition salt.
