Verfahren zur Herstellung von Benzodiazepin-Derivaten
Die Erfindung betrifft ein neues Verfahren zur direkten Umwandlung von 1 ,4-B enzodiazepin-2-onen der Formel I in 2-Amino-1,4-benzodiazepine der Formel II.

Bisher wurden Verbindungen der Formel I einerseits und Verbindungen der Formel II andererseits durch verschiedenartige Synthesen unter Benützung unterschiedlicher Reagenzien hergestellt. Es bestand seit langem das Bedürfnis nach einer Synthese, bei der Verbindungen der Formel I in Verbindungen der Formel II umgewandelt werden können, um damit die Notwendigkeit der Benützung verschiedener synthetischer Wege für die Herstellung dieser Verbindungen zu vermeiden.
Es wurde gefunden, dass man 1,4-Benzodiazepin-2one der Formel I direkt in 2-Amino-1,4-benzodiazepine der Formel II umwandeln kann, indem man Verbindungen der Formel I mit einer Verbindung der Formel

Auf diese Weise erhält man aus 1,4-Benzodiazepin-2-onen direkt 2-Amino-1,4-benzodiazepine und kann dadurch den Nachteil der verschiedenartigen Syntheserouten zur Her stellung dieser beiden Verbindungen vermeiden. Aus serdem wurde festgestellt, dass die Ausbeuten bei der erfindungsgemässen Reaktion bis zu 70 o/o und höher betragen.

Erwünschtenfalls kann jedoch auch für diese Ausführungsform des erfindungsgemässen Verfahrens ein Säurekatalysator eingesetzt werden.
In den obigen Formeln bedeutet der Ausdruck Alkyl geradkettige und verzweigte Alkylgruppen mit 1-7 Kohlenstoffatomen, wie Methyl, Äthyl, Propyl, Isopropyl und dgl. In ähnlicher Weise bedeutet der Ausdruck Alkoxy Radikale mit 1-7 Kohlenstoffatome wie Methoxy, Äthoxy, Propoxy, Isopropoxy, Isobutoxy usw.
Der Ausdruck Halogen umfasst alle 4 Halogene, d. h.
Jod, Brom, Chlor und Fluor, wobei Chlor, Fluor und Brom bevorzugt sind.
Bevorzugte Ausgangsmaterialien im erfindungsgemässen Verfahren sind 1,4-Benzodiapezin-2-one der Formel I, worin Rt entweder Wasserstoff oder Methyl und Rt entweder Phenyl, 2-Pyridyl oder in 2-Stellung mit einem Halogenatom, wie Fluor oder Chlor substituiertes Phenyl bedeuten, Rs ist zweckmässigerweise entweder Halogen, wie Chlor oder Fluor, oder Trifluormethyl oder Nitro und befindet sich in 7-Stellung des Benzodiazepin-2-on-Ringes. darstellt, ist Rs vorzugsweise ein Chlor- oder Bromatom in 7-Stellung des Benzodiazepin-Ringes.
Bevorzugte organische Amine sind primäre Amine, wie Methylamin, Methylamin, Isopropylamin, Propylamin, Butylamin usw. Obwohl primäre Amine bei der Durchführung des erfindungsgemässen Verfahrens bevorzugt sind, können auch sekundäre Amine eingesetzt werden. Irgend ein konventionelles sekundäres Alkylamin kann verwendet werden, z. B. Diäthylamin, Dimethylamin, N,N-Propylbutylamin, Pyrrolidin, Piper idin usw.
Gemäss einer Ausführungsform des erfindungsgemässen Verfahrens wird ein 1,4-Benzodiazepin-2-on der Formel I mit dem Aminreagens, wie Pyrrolidin, Ammoniak oder einem der genannten primären oder sekundären Alkylamine in Gegenwart eines Säurekatalysators umgesetzt. Bei der Durchführung dieser Reaktion kann irgend ein konventioneller Säurekatalysator eingesetzt werden. Typische Säurekatalysatoren, welche man bei der Durchführung der erfindungsgemässen Reaktion einsetzen kann, sind p-Toluolsulfosäure, Lewis Säuren, wie Titantetrachlorid, Aluminiumtrichlorid, Zinntetrafluorid, Ferrichlorid, Arsentrichlorid, Antimontrichlorid, Bortrifluorid usw. Die bevorzugten Säurekatalysatoren sind p-Toluolsulfosäure und Titantetrachlorid. Die Umwandlung der Verbindungen der Formel I in Verbindungen der Formel II wird zweckmässigerweise in Gegenwart eines inerten organischen Lösungsmittels durchgeführt.
Irgend ein organisches Lösungsmittel kann dabei verwendet werden. Typische inerte organische Lösungsmittel sind Diäthyläther, Tetrahydrofuran, Benzol, Hexan, Pentan, Äthylpropyläther und Mischungen davon. Bei der Durchführung dieser Reaktion sind Temperatur und Druck keine kritischen Faktoren; die Reaktion kann bei Raumtemperatur und darunter und bei Atmosphärendruck oder bei erhöhten Temperaturen und/oder erhöhten Drucken durchgeführt werden. Im allgemeinen ist es bevorzugt, dass man die Reaktion bei einer Temperatur zwischen etwa 0 und 25 durchführt. Obwohl man mit Vorteil niedere Temperaturen verwendet, können Temperaturen bis zur Rückflusstemperatur des Lösungsmittels benützt werden.
Falls man 1,4-Benzodiazepin-2-one der Formel I in der vorstehend definierten Weise ohne Benützung eines Säurekatalysators zur Reaktion bringt, so wird diese Reaktion zweckmässigerweise bei erhöhten Temperaturen und Drucken in Gegenwart eines inerten organischen Lösungsmittels durchgeführt. Irgend ein inertes organisches Lösungsmittel der vorstehend beschriebenen Art kann eingesetzt werden. Bevorzugte Lösungsmittel sind Dimethylformamid und Dimethylsulfoxyd. Bei der Durch führung dieser Reaktion ist es im allgemeinen bevorzugt, Temperaturen zwischen etwa 1000 und 1750 C und Drucke zwischen etwa 50 und 75 Atmosphären einzusetzen. Obwohl man diese Reaktion zweckmässigerweise ohne Verwendung eines Säurekatalysators durchführt, kann ein solcher Säurekatalysator erwünschtenfalls verwendet werden.
Da jedoch seine Verwendung keinen zusätzlichen Vorteil mit sich bringt, wird man einen derartigen Säurekatalysator nur in seltenen Fällen einsetzen.

ist und zwar durch Behandlung mit einem Oxydationsmittel. Typische Oxydationsmittel sind Mangandioxyd, Selendioxyd, Silberoxyd, Azodicarbonsäureester usw. Im allgemeinen kann die Reaktion in einem geeigneten organischen Lösungsmittel durchgeführt werden. Typische organische Lösungsmittel sind Benzol, Toluol, Essigsäure, Butanol, Pyridin usw. Bei der Durchführung der Oxydationsreaktion sind Temperaturen und Drucke nicht von Belang. Die Oxydation kann bei Raumtemperatur oder bei erhöhten oder bei niedrigen Temperaturen durchgeführt werden. Im allgemeinen ist es zweckmässig, die Oxydationsreaktion bei Temperaturen zwischen etwa 10 und 400 durchzuführen.
Die Konzentration der Reaktionspartner ist nicht kritisch; das Oxydationsmittel soll jedoch mit Vorteil im stöchiometrischen Überschuss bezogen auf die zu oxydierende Verbindung eingesetzt werden, d. h. ein Überschuss über das stöchiometrische Äquivalent, jedoch weniger als zwei stöchiometrische Äquivalente.
In den folgenden Beispielen sind alle Temperaturen in 2c angegeben.
Beispiel 1
Eine Lösung von 75 g 7-Chlor-1,3-dihydro-5-phenyl-2H-1,4-benzodiazepin-2-on in 250 ml N,N-Dime thylformamid wird bei 10 mit Methylamin gesättigt.
Die Reaktionsmischung wird in ein Druckgefäss gebracht und 67 Atmosphären Stickstoff aufgedrückt. Das Druckgefäss wird sodann 24 Stunden auf 1500 erhitzt, anschliessend abgekühlt und abgeblasen. Die Lösung giesst man in 4 Liter Wasser. Die wässrige Mischung wird mit Chlorwasserstoffsäure auf einen pH-Wert von 6 gebracht und filtriert. Der Niederschlag wird in einem Liter Dichlormethan gelöst und 3 mal mit je 250 ml 3 n Chlorwasserstoffsäure extrahiert. Die sauren Extrakte werden vereinigt, mit 250 ml Dichlormethan gewaschen, mit Ammoniumhydroxyd basisch gestellt und 3 mal mit je 250 ml Dichlormethan extrahiert. Die organischen Schichten werden vereinigt, mit Wasser gewaschen, über wasserfreiem Natriumsulfat getrocknet, filtriert und eingedampft. Der Rückstand wird 3 mal mit je 150 ml siedendem Benzol ausgezogen.
Die heissen Benzolfraktionen werden vereinigt und eingeengt, wobei man nach Kühlen rohes 7-Chlor-2-methylamino5-phenyl-3H-1,4-benzodiazepin erhält. Durch Umkristallisieren des Reaktionsproduktes aus einer Mischung von Dichlormethan und Methanol erhält man 7-Chlor2-methylamino-5-phenyl-3H-1,4-benzodiazepin in Form von gelben Prismen vom Schmelzpunkt 248-249".
Beispiel 2
Eine Lösung von 5 g 7-Chlor-1,3-dihydro-5-phenyl- 2H-1,4-benzodiazepin-2-on, 30 ml Pyrrolidin und 0,1 g p-Toluolsulfosäure-monohydrat in 100 ml Toluol wird gerührt und unter Verwendung eines Wasserabscheiders und eines Trockenaufsatzes zum Rückfluss erhitzt. Nach 22,5 Stunden wird das Destillat verworfen und die zurückbleibende Lösung im Reaktionsgefäss im Vakuum eingedampft. Der Rückstand wird mit einer Mischung von Äther und 3 n Essigsäure extrahiert, die Säureschicht wird filtriert und das Filtrat durch verdünnte Natronlauge alkalisch gestellt. Man erhält rohes 7-Chlor 5-phenyl-2-pyrrolidin-3H-1,4-benzodiazepin. Das rohe Reaktionsprodukt wird aus Isopropanol umkristallisiert und liefert das reine Produkt.
Beispiel 3
Eine Lösung von 5 g (0,0184 Mol) 7-Chlor-1,3-dihydro-5-phenyl-2H-1,4-benzodiazepin-2-on in 125 ml trockenem Tetrahydrofuran wird zu einer Lösung von 15 g Methylamin in 100 ml trockenem Tetrahydrofuran zugesetzt. Die Mischung, welche sich in einem Dreihalskolben befindet, der mit einem Rührer, einem Tropftrichter und einem Trockeneiskühler ausgestattet ist, wird äusserlich in einem Eisbad gekühlt und unter Stickstoff gehalten. Ein Tetrahydrofuran/Titantetrachloridkomplex, welchen man durch Zusatz von 1,8 g (0,01 Mol) Titantetrachlorid zu 60 ml Tetrahydrofuran herstellt, wird im Verlaufe von 20 Minuten langsam zur Reaktionsmischung zugesetzt. Die erhaltene Mischung wird laufend gerührt und die Temperatur auf Raumtemperatur ansteigen gelassen. Nach 4 Stunden stellt man im Dünnschichtchromatogramm fest, dass die Reaktion vollständig ist.
Die Reaktionsmischung wird sodann über Nacht stehen gelassen und anschliessend filtriert. Die Filtrate werden zur Trockene eingedampft und der Rückstand in 500 ml Dichlormethan gelöst. Die organische Lösung wird mit 200 ml verdünntem Ammoniumhydroxyd und sodann 2 mal mit je 100 ml gesättigter Kochsalzlösung gewaschen, über wasserfreiem Natriumsulfat getrocknet, filtriert und eingedampft. Der Rückstand wird aus einer Mischung von Tetrahydrofuran und Hexan kristallisiert und liefert 7-Chlor-2-methylamino 5-phenyl-3H-1,4-benzodiazepin.
Beispiel 4
Eine Lösung von 5 g (0,0178 Mol) 1,3-Dihydro-7 nitro-5-phenyl-2H-1 ,4-benzodiazepin-2-on in 150 ml trockenem Tetrahydrofuran wird zu einer Lösung von 10 g Methylamin in 100 ml trockenem Tetrahydrofuran zugesetzt. Die erhaltene Lösung wird in der in Beispiel 3 beschriebenen Weise gekühlt und mit einer Lösung von 2,4 g (0,013 Mol) Titantetrachlorid in 50 ml trockenem Benzol behandelt. Die Mischung wird auf Raumtemperatur erwärmt und über Nacht gerührt. Man setzt etwa 10 ml Wasser zu, um das überschüssige Titantetrachlorid zu zerstören, und filtriert die Lösung. Nach Entfernung des Lösungsmittels wird der Rückstand mit 50 ml äthanol behandelt. Der Rückstand wird vom Niederschlag abfiltriert. Nach Filtration wird die zurückbleibende Lösung eingeengt und gibt 2-Methylamino-7nitro-5-phenyl-3H-1,4-benzodiazepin.
Beispiel 5
Eine Lösung von 10 g (0,0348 Mol) 7-Chlor-1,3 dihydro-5-phenyl-2H-1 ,4-benzodiazepin-2-on-4-oxyd in 400 ml trockenem Tetrahydrofuran wird zu einer Lösung von 20 g Methylamin in 200 ml trockenem Tetrahydrofuran zugesetzt. Die erhaltene Mischung wird entsprechend den Angaben in Beispiel 3 gekühlt und dann mit einem Tetrahydrofuran/Titantetrachloridkomplex behandelt, den man aus 4,8 g (0,025 Mol) Titantetrachlorid und 100 ml trockenem Tetrahydrofuran herstellt. Der Zusatz erfolgt im Verlaufe von 20 Minuten.
Die erhaltene Mischung lässt man auf Raumtemperatur erwärmen, über Nacht stehen und filtriert sodann.
Die Filtrate werden zur Trockene eingedampft und das erhaltene Reaktionsprodukt aus Methanol umkristallisiert, wobei man 7- Chlor-2-methylamino-5-phenyl- 3H-1,4-benzodiazepin-4-oxyd in Form von gelben Prismen erhält.
Beispiel 6
Eine Lösung von 10 g (0,034 Mol) 7-Chlor-1,3-di hydro-5-phenyl-2H-1 ,4-benzodiazepin-2-on-4-oxyd in 400 ml trockenem Tetrahydrofuran wird zu einer Lösung von 20 g Dimethylamin in 200 ml trockenem Tetrahydrofuran zugesetzt. Die Lösung wird in der in Beispiel 3 beschriebenen Weise gekühlt und sodann mit einer Lösung von 4,8 g (0,025 Mol) Titantetrachlorid in 100 ml trockenem Benzol behandelt. Die Zusatzzeit beträgt 20 Minuten. Die Reaktionsmischung lässt man sodann auf Raumtemperatur erwärmen, rührt 3 Stunden und setzt sodann 10 ml Wasser zu. Die Lösung wird filtriert und das Lösungsmittel unter vermindertem Druck abgedampft. Der Rückstand wird in 50 ml Athanol gerührt und sodann mit 100 ml 0,5 n Natriumhydroxydlösung zur Entfernung des Ausgangsmaterials kräftig geschüttelt.
Die Mischung wird mit 400 ml Wasser verdünnt und sodann filtriert. Der Niederschlag wird mit Wasser gewaschen und aus Äthanol umkristallisiert und liefert 7-Chlor-2-dimethylamino-5-phenyl-3H-1 ,4-benzodiazepin-4-oxyd.
Beispiel 7
Eine Lösung von 10 g (0,0348 Mol) 7-Chlor-1,3 dihydro-5-phenyl-2H-1 ,4-benzodiazepin-2-on-4-oxyd in 400 ml trockenem Tetrahydrofuran wird bei 0-100 zu erst mit 15 ml Isopropylamin und sodann mit einer Lösung von 4,8 g (0,025 Mol) Titantetrachlorid in 100 ml trockenem Benzol behandelt. Die Zusatzzeit beträgt 20 Minuten. Die Reaktionsmischung lässt man sodann auf Raumtemperatur erwärmen und rührt über Nacht. Man versetzt mit 10 ml Wasser, filtriert und dampft ein. Der Rückstand wird in 500 ml Äthanol gerührt und sodann gut mit 10 ml 5 n Natriumhydroxydlösung zur Entfernung des Ausgangsmaterials geschüttelt. Die Mischung wird mit 400 ml Wasser verdünnt und filtriert.
Das unlösliche Produkt wird auf dem Filter mit Wasser gewaschen und sodann aus Äthanol umkristallisiert, wobei man 7-Chlor-2-isopropylamino-5-phenyl-3H-1,4-benzo- diazepin-4-oxyd erhält.
Beispiel 8
Eine Mischung von 10 g (0,0348 Mol) 7-Chlor-1,3 dihydro-5-phenyl-2H-1 ,4-benzodiazepin-2-on-oxyd und 15 ml Piperidin in 400 ml trockenem Tetrahydrofuran wird entsprechend den Angaben in Beispiel 3 gekühlt und sodann mit einer Lösung von 4,8 g (0,025 Mol) Titantetrachlorid in 100 ml trockenem Benzol behandelt. Die Zusatzzeit beträgt 20 Minuten. Die Mischung lässt man sodann auf Raumtemperatur erwärmen und rührt über Nacht. Man versetzt mit 10 ml Wasser und filtriert die Reaktionsmischung. Das Lösungsmittel wird entfernt, der Rückstand in 50 ml Äthanol gerührt und sodann gut mit 100 ml 0,5 n Natriumhydroxydlösung geschüttelt. Die Mischung wird filtriert und der Niederschlag mit Wasser gewaschen.
Durch Umkristallisieren aus einer Mischung von Dichlormethan und Hexan erhält man 7-Chlor-5-phenyl-2-piperidino-3H-1,4-benzodiazepin-4-oxyd in Form von gelben Prismen.
Beispiel 9
Eine Lösung von 10 g (0,348 Mol) 7-Chlor-1,3 dihydro-5-phenyl-2H-1 ,4-benzodiazepin-2-on-4-oxyd in 400 ml trockenem Tetrahydrofuran wird in einem Drei halskolben, welcher mit einem Trockeneiskühler und einem Tropftrichter ausgerüstet ist, gerührt. Das Reaktionsgefäss wird von aussen in einem Eisbad gekühlt.
Man leitet Ammoniakgas in die Lösung bis flüssiger Ammoniak im Trockeneiskühler rasch kondensiert. Sodann versetzt man tropfenweise im Verlaufe von 20 Minuten mit einer Lösung von 4,8 g (0,025 Mol) Titantetrachlorid in 100 ml Benzol. Das Kühlbad wird entfernt und die Reaktionsmischung auf Raumtemperatur erwärmen gelassen. Nach dem Rühren über Nacht versetzt man mit 10 ml Wasser und filtriert die Lösung.
Durch Entfernen der Lösungsmittel erhält man einen Rückstand, der mit 50 ml Äthanol aufgeschlämmt und sodann heftig mit 100 ml einer 0,5 n Natriumhydroxydlösung geschüttelt wird. Die erhaltene Mischung wird mit 400 ml Wasser verdünnt und der Niederschlag durch Filtration entfernt und mit Wasser gewaschen. Durch Umkristallisieren aus Methanol erhält man 2-Amino-7-chlor-5-phenyl-3H-1,4-benzodiazepin- 4-oxyd.
Beispiel 10
Zu einer Suspension von 25 g (0,0916 Mol) 7 Chlor-1,3,4,5-tetrahydro-5-phenyl-2H-1,4-benzodiaze- pin-2-on, gelöst in 300 ml trockenem Tetrahydrofuran (unter Stickstoffatmosphäre) setzt man eine Lösung von 30 g Methylamin in 300 ml trockenem Tetrahydrofuran zu. Die erhaltene Mischung wird in einem Eisbad gekühlt und unter Rühren im Verlaufe von 10 Minuten mit einer Lösung von 12 g (0,0632 Mol) Titantetrachlorid in 100 ml Benzol versetzt. Man entfernt sodann das Eisbad und versetzt nach 4 Stunden mit 200 ml Wasser. Die Lösung wird sodann filtriert und der Niederschlag mit Tetrahydrofuran gewaschen. Die vereinigten Filtrate werden zur Trockene eingedampft.
Das erhaltene Öl kristallisiert aus Methanol/iither und gibt festes 7-Chlor-4,5-dihydro-2-methylamino-5-phenyl-3H1,4-benzodiazepin in Form von weissen Plättchen vom Schmelzpunkt 175-1 80a.
Zu einer Lösung von 2,0 g (0,007 Mol) 7-Chlor4, 5-dihydro-2-methylamino-5-phenyl-3H-1 ,4-benzodiazepin in 50 ml trockenem Benzol setzt man eine Lösung von 1,2 g (0,007 Mol) Diäthylazodicarboxylat in 10 ml trockenem Benzol zu. Die erhaltene Lösung wird 1 Stunde zum Rückfluss erhitzt, gekühlt, mit 50 ml verdünntem Ammoniumhydroxyd extrahiert und filtriert, wobei man 7-Chlor-2-methylamino-5-phenyl-3H-1,4benzodiazepin in Form von weissen Prismen erhält.

The reaction mixture is placed in a pressure vessel and 67 atmospheres of nitrogen are injected. The pressure vessel is then heated to 1500 for 24 hours, then cooled and blown off. The precipitate is dissolved in one liter of dichloromethane and extracted 3 times with 250 ml of 3N hydrochloric acid each time. The acidic extracts are combined, washed with 250 ml of dichloromethane, made basic with ammonium hydroxide and extracted 3 times with 250 ml of dichloromethane each time. The organic layers are combined, washed with water, dried over anhydrous sodium sulfate, filtered and evaporated. The residue is extracted 3 times with 150 ml of boiling benzene each time.
The hot benzene fractions are combined and concentrated, crude 7-chloro-2-methylamino5-phenyl-3H-1,4-benzodiazepine being obtained after cooling. By recrystallizing the reaction product from a mixture of dichloromethane and methanol, 7-chloro-2-methylamino-5-phenyl-3H-1,4-benzodiazepine is obtained in the form of yellow prisms with a melting point of 248-249 ".
Example 2
A solution of 5 g of 7-chloro-1,3-dihydro-5-phenyl-2H-1,4-benzodiazepin-2-one, 30 ml of pyrrolidine and 0.1 g of p-toluenesulfonic acid monohydrate in 100 ml of toluene is stirred and heated to reflux using a water separator and drying attachment. After 22.5 hours the distillate is discarded and the remaining solution in the reaction vessel is evaporated in vacuo. The residue is extracted with a mixture of ether and 3N acetic acid, the acid layer is filtered and the filtrate is made alkaline with dilute sodium hydroxide solution. Crude 7-chloro 5-phenyl-2-pyrrolidine-3H-1,4-benzodiazepine is obtained. The crude reaction product is recrystallized from isopropanol and gives the pure product.
Example 3
A solution of 5 g (0.0184 mol) of 7-chloro-1,3-dihydro-5-phenyl-2H-1,4-benzodiazepin-2-one in 125 ml of dry tetrahydrofuran is added to a solution of 15 g of methylamine in 100 ml of dry tetrahydrofuran were added. The mixture, which is located in a three-necked flask equipped with a stirrer, a dropping funnel and a dry ice condenser, is cooled externally in an ice bath and kept under nitrogen. A tetrahydrofuran / titanium tetrachloride complex, which is prepared by adding 1.8 g (0.01 mol) of titanium tetrachloride to 60 ml of tetrahydrofuran, is slowly added to the reaction mixture over the course of 20 minutes. The mixture obtained is stirred continuously and the temperature is allowed to rise to room temperature. After 4 hours, a thin-layer chromatogram shows that the reaction is complete.
The reaction mixture is then left to stand overnight and then filtered. The filtrates are evaporated to dryness and the residue is dissolved in 500 ml of dichloromethane. The organic solution is washed with 200 ml of dilute ammonium hydroxide and then twice with 100 ml of saturated sodium chloride solution each time, dried over anhydrous sodium sulfate, filtered and evaporated. The residue is crystallized from a mixture of tetrahydrofuran and hexane and gives 7-chloro-2-methylamino 5-phenyl-3H-1,4-benzodiazepine.
Example 4
A solution of 5 g (0.0178 mol) of 1,3-dihydro-7 nitro-5-phenyl-2H-1,4-benzodiazepin-2-one in 150 ml of dry tetrahydrofuran becomes a solution of 10 g of methylamine in 100 ml of dry tetrahydrofuran added. The solution obtained is cooled in the manner described in Example 3 and treated with a solution of 2.4 g (0.013 mol) of titanium tetrachloride in 50 ml of dry benzene. The mixture is warmed to room temperature and stirred overnight. About 10 ml of water are added to destroy the excess titanium tetrachloride and the solution is filtered. A solution of 10 g (0.0348 mol) of 7-chloro-1,3 dihydro-5-phenyl-2H-1,4-benzodiazepin-2-one-4-oxide in 400 ml of dry tetrahydrofuran becomes a solution of 20 g of methylamine in 200 ml of dry tetrahydrofuran were added. The mixture obtained is cooled as described in Example 3 and then treated with a tetrahydrofuran / titanium tetrachloride complex which is prepared from 4.8 g (0.025 mol) of titanium tetrachloride and 100 ml of dry tetrahydrofuran. The addition takes place over the course of 20 minutes.
The resulting mixture is allowed to warm to room temperature, stand overnight and then filtered.
The filtrates are evaporated to dryness and the reaction product obtained is recrystallized from methanol, 7-chloro-2-methylamino-5-phenyl-3H-1,4-benzodiazepine-4-oxide being obtained in the form of yellow prisms.
Example 6
A solution of 10 g (0.034 mol) of 7-chloro-1,3-dihydro-5-phenyl-2H-1,4-benzodiazepin-2-one-4-oxide in 400 ml of dry tetrahydrofuran is a solution of 20 g of dimethylamine in 200 ml of dry tetrahydrofuran were added. The solution is cooled in the manner described in Example 3 and then treated with a solution of 4.8 g (0.025 mol) of titanium tetrachloride in 100 ml of dry benzene. The additional time is 20 minutes. The reaction mixture is then allowed to warm to room temperature, stirred for 3 hours and then 10 ml of water are added. The solution is filtered and the solvent is evaporated under reduced pressure. The mixture is diluted with 400 ml of water and then filtered. The precipitate is washed with water and recrystallized from ethanol and yields 7-chloro-2-dimethylamino-5-phenyl-3H-1,4-benzodiazepine-4-oxide.
Example 7
A solution of 10 g (0.0348 mol) of 7-chloro-1,3 dihydro-5-phenyl-2H-1,4-benzodiazepin-2-one-4-oxide in 400 ml of dry tetrahydrofuran is added at 0-100 treated first with 15 ml of isopropylamine and then with a solution of 4.8 g (0.025 mol) of titanium tetrachloride in 100 ml of dry benzene. Example 8
A mixture of 10 g (0.0348 mol) of 7-chloro-1,3-dihydro-5-phenyl-2H-1, 4-benzodiazepin-2-one oxide and 15 ml of piperidine in 400 ml of dry tetrahydrofuran is according to the information cooled in Example 3 and then treated with a solution of 4.8 g (0.025 mol) of titanium tetrachloride in 100 ml of dry benzene. Example 9
A solution of 10 g (0.348 mol) of 7-chloro-1,3 dihydro-5-phenyl-2H-1, 4-benzodiazepin-2-one-4-oxide in 400 ml of dry tetrahydrofuran is in a three-necked flask which is equipped with equipped with a dry ice condenser and a dropping funnel, stirred. Example 10
To a suspension of 25 g (0.0916 mol) of 7 chloro-1,3,4,5-tetrahydro-5-phenyl-2H-1,4-benzodiazepin-2-one, dissolved in 300 ml of dry tetrahydrofuran ( under a nitrogen atmosphere) a solution of 30 g of methylamine in 300 ml of dry tetrahydrofuran is added. The mixture obtained is cooled in an ice bath and, with stirring, a solution of 12 g (0.0632 mol) of titanium tetrachloride in 100 ml of benzene is added over the course of 10 minutes. The ice bath is then removed and, after 4 hours, 200 ml of water are added. The solution is then filtered and the precipitate washed with tetrahydrofuran. The combined filtrates are evaporated to dryness.
The oil obtained crystallizes from methanol / iither and gives solid 7-chloro-4,5-dihydro-2-methylamino-5-phenyl-3H1,4-benzodiazepine in the form of white platelets with a melting point of 175-180a.
To a solution of 2.0 g (0.007 mol) of 7-chloro4, 5-dihydro-2-methylamino-5-phenyl-3H-1,4-benzodiazepine in 50 ml of dry benzene, a solution of 1.2 g ( 0.007 moles) of dietary azodicarboxylate in 10 ml of dry benzene. The resulting solution is refluxed for 1 hour, cooled, extracted with 50 ml of dilute ammonium hydroxide and filtered, 7-chloro-2-methylamino-5-phenyl-3H-1,4-benzodiazepine being obtained in the form of white prisms.
