Die vorliegende Erfindung betrifft ein Verfahren zur Herstellung von neuen organischen Verbindungen der Formel 1,

worin R1 für Wasserstoff, eine Alkylgruppe oder Hydroalkyl- gruppe mit jeweils höchstens 4 Koblenstoffatomen, R2 für eine Hydroxyalkylgruppe mit höchstens 4 Kohlenstoffatomen oder eine Alkoxyalkylgruppe mit zusammen höchstens 6 Kohlen- stoffatomen, und R3 für Wasserstoff oder Halogen stehen, sowie ihrer Säureadditionssalze.
In den Verbindungen der Formel 1 besitzt die Alkylgruppe höchstens 4 Kohlenstoffatome und kann geradekettig oder verzweigt sein und beispielsweise für Methyl, Aethyl, Propyl, Isopropyl, n-Butyl, Isobutyl oder tert.-Butyl stehen.
Die Hydroxyalkylgruppe besitzt vorzugsweise 24 Kohlen- stoffatome, wobei sich die Hydroxylgruppe vorzugsweise in Endstellung befindet. Sie steht vorzugsweise für Hydroxyäthyl oder eine geradkettige oder verzweigte Hydroxypropyl- oder Hydroxybutylgruppe.
Die Alkoxyalkylgruppe besitzt vorzugsweise zusammen Kohlenstoffatome und steht beispielsweise für Methoxyäthyl, Aethoxyäthyl, Propoxyäthyl, Aethoxypropyl, Methoxypropyl, Propoxypropyl, Methoxybutyl oder Aethoxybutyl.
Halogen bedeutet vorzugsweise Fluor, Chlor oder Brom, insbesondere jedoch Chlor.
Die Verbindungen der Formel 1 können in ihre Salze übergeführt werden und umgekehrt.
Erfindungsgemäss gelangt man zu Verbindungen der Formel I, indem man Verbindungen der Formel II,

Das erfindungsgemässe Verfahren kann, wie nachfolgend beschrieben, durchgeführt werden.
Verbindungen der Formel II, worin die reaktive Gruppe kovalent oder ionisch an das Kohlenstoffatom gebunden ist, und eine Aminogrupe, die gegebenenfalls durch eine oder zwei Alkylgruppen mit 14 Kohlenstoffatomen, insbesondere Methylgruppen, substituiert sein kann, ein Halogenatom, ins besondere Chlor, die Sulfhydrilgruppe, eine Alkoy- oder Alkylthiogruppe mit 1-5 Kohlenstoffatomen, beispielsweise die
Methoxy- oder die Methylthiogruppe, eine gegebenenfalls
Substituenten aktivierte Aralkylthiogruppe, beispielsweise die p-Nitrobenzylthiogruppe oder eine Tosylgruppe sein kann,
werden mit Verbindungen der Formel I[I[H zweckmässigervJeise in einem inerten organischen Lösungsmittel, beispielsweise in
Xylol, Dioxan usw., bei Temperaturen zwischen 500 und
1700 C miteinander umgesetzt. Die Reaktionszeit beträgt ca.
zwischen 2 und 20 Stunden. Falls die die reaktive Gruppe X für eine gegebenenfalls substituierte Aminogruppe steht, erfolgt die Umsetzung zweckmässigerweise in Gegenwart katalytischer
Mengen einer Säure, beispielsweise der p-Toluolsulioil- säure oder Schwefelsäure, oder eines sauer reagierenden
Salzes, wie Ammoniumchlorid,
oder es werden als Ausgangs verbindungen Säureadditionssalze von Verbindungen der
Formel 11 verwendet, worin X für eine gegebenenfalls substi tuierte Aminogruppe steht. Hierbei kaan als Reaktionsmedium auch ein Überschuss von Verbindungen der Formel IHP anstelle eines Lösungsmittels verwendet werden. Vorzugsweise erfolgt die Umsetzung durch Eiitzen des Reaktionsgemisches auf
Siedetemperatur während 2-6 Stunden. Bei der erfindungs gemässen Umsetzung ist die Verwendung der Verbindungen der Formel II in isolierter Form nicht unbedingt erforderlich.
Die Verbindungen der Formel 1[, worin R1 für Wasserstoff steht, können zur Herstellung von Verbindungen der Formel 1, worin R1 eine Alkyl- oder Hydroxyalkylgruppe bedeutet, verwendet werden, indem man in Verbindungen der Formel 1, worin R2 für Wasserstoff steht, eine Alkyl- oder Hydroxyalkyl- gruppe einführt.
Die nachfolgende Alkylierung von Verbindungen der Formel 1, worin R1 für Wasserstoff steht, kann, wie folgt, durchgeführt werden:
Verbindungen der Formel 1, worin R1 für eine Alkylgruppe steht, können durch Umsetzung mit reaktionsfähigen Estern entsprechender Alkanole erhalten werden:

Zur Umsetzung mit den reaktionsfähigen Estern entsprechender Alkanole werden Verbindungen der Formel 1, worin R1 für Wassérstoff steht, vorteilhafterrieise in einem inerten organischen Lösungsmittel gelöst, die erhaltene Lösung mit einem säurebindenden Mittel, beispielsweise Kalium- carbonat, und mit den reaktionsfähigen Estern entsprechender Alkanole versetzt, und das so erhaltene lReaktionsgemisch während 14 Stunden auf eine Temperatur zwischen Raum- temperatur (ca. 25" C) und Siedetemperatur des Reaküons- gemisches erhitzt.
Bei dieser Umsetzung können als reaktionsfähige Ester die Ester der Alkanole mit organischen oder anorganischen Säuren, beispielsweise Toluolsulfonsäure oder von Halogenwasserstoffsäure, verwendet werden. Anstelle der säurebindenden Mittel kann ein Überschuss von Verbindungen der Formel 1, worin R1 für Wasserstoff steht, treten. Als inertes organisches Lösungsmittel kommt hierbei beispielsweise Aceton oder Propanol in Betracht.
Zu Verbindungen der Formel 1, worin Ri für eine Alkylgruppe steht, kann man auch durch Umsetzung von Verbindungen der Formel 1, worin Rt für Wasserstoff steht, mit entsprechenden Aldehyden in Gegenwart eines katalytisch erregten Wasserstoffes oder in Gegenwart eines Redukrions- mittels, beispielsweise Ameisensäure, gelangen.
Die reduktive Alkylierung von Verbindungen der Formel 1, worin R1 für Wasserstoff steht, kann beispielsweise so durchgeführt werden, dass man diese Verbindungen der Formel 1 in 90%iger Ameisensäure löst, der erhaltenen Lösung den ent- sprechenden Aldehyd zusetzt und das so erhaltene lReaktions- gemisch während 5-20 Stunden auf Temperaturen zwischen 50 und 1500 C, volzugsweise jedoch auf Siedetemperatur des Reaktionsgemisches, erhitz±.
Die nachfolgende Hydroxyalkylierung von Verbindungen der Formel 1, worin R1 für Wasserstoff steht, kann, wie nachfolgend beschrieben, erfolgen: Verbindungen der Formel 1, worin R1 für Wasserstoff steht, werden mit Monoestern entsprechender Glykole unter Verwendung des für die Alkylierung beschriebenen Verfahrens umgesetzt.
Zur Herstellung von ss-E{ydrosLyalkyl-lDerivaten können Verbindungen der Formel 1, worin R1 für Wasserstoff steht, mit Alkylenoxyden in einem inerten organischen Lösungsmittel, beispielsweise Toluol, bei einer Temperatur zwischen 80 und 1200 C, während 3-7 Stunden umgesetzt werden, wobei die Umsetzung wegen Flüchtigkseit des Alkylen onsids in einem Druckrohr erfolgen soll.
Die erfindungsgemäss erhaltenen Verbindungen der Formel 1 können auf an sich bekannte Weise isoliert und gereinigt werden.
Die als Ausgangsverbindungen verwendeten Verbindungen der Formel IH können, wie nachfolgend beschrieben, hergestellt werden: Verbindungen der Formel II, worin X für Halogen, Alkoxy, Sulfhydril oder für Alkylthio steht, können erhalten werden, indem man auf an sich bekannte Weise Verbindungen der Formel IV,

Diejenigen Verbindungen der Formel II, worin X eine andere Bedeutung als die einer gegebenenfalls mono- oder dialkylierten Aminogruppe, Halogen, Alkoxy-, Sulihydril- oder Alkylthiogruppe besitzt, können unter Verwendung bekannter Verfahren hergestellt werden.

R2 obige Bedeutung besitzt und Y für einen Säurerest einer Mineralsäure oder einer oganischen Sulfonsäure mit Ausnahme des Fluorid-Ions steht, umsetzt.
In den Verloindungen der Formel VIII bedeutet Y zweck mässigerweise Chlor, Brom, Jod, Arylsulfonat, z. B. p-Toluolsulfonat oder Alkylsulfonat, z. B. Methansulfonat. Vorzugs- weise steht Y für Jod. Falls R2 für eine Hydroxyalkylgruppe steht, ist es zweckmässig, eine acetalische Schutzgruppe zu verwenden, beispielsweise die Hydrozylgruppe durch eine Tetrahydropyranylätherbildung zu schützen. Die Hydrolyse der Schutzgruppe erfolgt in an sich bekannter Weise, z. B. mit Salzsäure.
Die Umsetzung von Verbindungen der Formel VII mit Ver- bindungen der Formel VIII wird zweckmässigerweise in einem inerten organischen Lösungsmittel, wie Tetramethylharnstoff, Dimethylformamid oder Dimethylsulfoxid, vorzugsweise jedoch Tetramethylharnstoff, durchgeführt. Zweckmässigerweise arbeitet man bei erhöhten Temperaturen, die jedoch nicht sonderlich kritisch sind, aber vorzugsweise zwischen 60 und 1500 C, insbesondere 70 und 1200 C, liegen. Die Umsetzung wird zweckmässigerweise in Gegenwart einer starken Base, wie Alkalimetallhydrid oder -amid, vorzugsweise Alkalisnetallhydrid, durchgeführt. Die bevorzugte Base ist Natriumhydrid. Das Verfahren wird vorzugsweise in einer inerten Atmosphäre, beispielsweise Stickstoffatmosphäre, durchgeführt.
Die Verbindungen der Formeln III, VII und VIII sind bekannt.
Die Verbindungen der Formel 1 zeichnen sich durch günstige pharmakodynamische Aktivitäten aus. Sie können insbesondere als Neuroleptika beispielsweise zur Behandlung von psychotischen Störungen verwendet werden. Diese günstige Wirkung zeigt sich beispielsweise in einem Test, worin die Hemmung der Laufaktivität bei Mäusen p.o. Verabreichung von 1 bis 50 mg Wirksubstanz pro kg Tierkörpergewicht gemäss der von Caviezel und Baillod in Pharm. Acta Helv. (1958)33, 469-484, angegebenen Methode gemessen wird.
Die Verbindungen der Formel I zeichnen sich überdies überraschenderweise durch im Vergleich zu ähnlichen auf dem Markt befindlichen Neuroleptika geringere als Nebenwirkungen zu wertende, pharmakodynamische Effekte, z.B. durch geringere Kreislaufwirkungen sowie geringere anticholinergische Effekte aus.
Für die obige Anwendung kann die zu verabreichende Dosu abhängig von dem verwendeten Wirkstoff, der Verabreichungsart sowie der erwünschten Therapie verschieden sein. Im allgemeinen erhält man zufriedenstellende Resultate mit einer täglichen Dosis von 0,15 mg bis ca. 50 mg pro kg Tierkörpergewicht. Diese Menge kann auch in kleineren Dosen 2 bis 4 mal täglich oder in Retardform verabreicht werden. Bei grösseren Säugetieren soll die täglich zu verabreichende Menge von 10 bis 500 mg betragen. Die für die orale Verabreichung geeignete Form soll von 2 bis ca. 250 mg des Wirkstoffes zusammen mit einem festen oder flüssigen pharmakodynamisch verträglichen Träger- oder Verdünnungsmittel enthalten.
Eine Einheitsdosis, beispielsweise eine zur oralen Verabreichung geeignete Tablette, kann zwischen 5 und 100 mg des Wirkstoffes zusammen mit geeigneten pharmazeutisch indifferenten Hilfsstoffen, wie Lactose, Maisstärke, Talk, Magnesiumstearat usw., enthalten.
Die Verbindungen der Formel I können ebenfalls in Form von deren pharmazeutisch verträglichen Säureadditionssalzen, beispielsweise als Hydrochlorid oder als Maleinat oder Fumarat, verabreicht werden, die den gleichen Grad an Aktivität besitzen wie die freien Basen.
Die Verabreichung von Verbindungen der Formel 1 bzw.
von deren Salzen kann entweder oral in Form von Tabletten, Granulaten, Kapseln oder Dragees, oder parenteral in Form von Injektionslösungen erfolgen.
Aus Gründen der leichten Herstellung ünd günstigen Verabreichung werden feste Zubereitungen bevorzugt, insbesondere hartgefüllte Kapseln und Tabletten.
Eine vorzugsweise Tablettenzubereitung besteht aus 10 mg 5 -(2-Methoxyäthyl) - 8-chlor-1 1 - (4-methyl-1 -piperazinyl) - 5H- dibenzo[b,e][1,4]diazepin, 140 mg Lactose, 5 mg Maisstärke, 5 mg Talk und 0,1 mg Magnesiumstearat.
Vom pharmakodynamischen Standpunkt bevorzugte Verbindungen der Formel I sind diejenigen, worin R1 für Methoxy äthyl, Methyl oder Hydroxyäthyl, R2 für Methoxyäthyl oder Hydroxyäthyl und R3 für Halogen, insbesondere Chlor, stehen.
In den nachfolgenden Beispielen sind die Temperaturen in Grad-Celsius angegeben. Die Raumtemperatur beträgt ca.
20-30" C.
- Beispiel 5-(2-Methoxyäthyl)-8-chlor-11- (4-methyl-1-piperazinyl)- 5H- dibenzo[b, e] [1,4]diazepin
6,05 g (0,02 Mol) 5-(2-Methoxyäthyl)-8-chlor-10,11-di- hydro- SH-dibenzo[b,e] [1 ,4]diazepin-1 1 -on, 30 mlPhosphoroxychlorid und 1 ml N,N-Dimethylanilin werden während 3 Stunden zum Sieden (Rückfluss) erhitzt. Die erhaltene Lösung wird im Vakuum zur Trockne eingedampft, der Rückstand zweimal mit Xylol nachgetrocknet und mit 20 ml Dioxan und 25 ml N-Methylpiperazin während 6 Stunden zum Sieden (Rückfluss) erhitzt. Die Lösung wird sodann möglichst weitgehend eingeengt und der Rückstand zwischen Ammoniakwasser und Aether verteilt. Die Aetherphase wird zweimal mit Wasser gewaschen und zur Abtrennung der basischen Anteile mit verdünnter Essigsäure erschöpfend extrahiert.
Die Base wird aus den vereinigten Extrakten durch Zugabe von Natriumhydroxid freigesetzt, in Chloroform aufgenommen, die Chloroformphase mit Wasser gewaschen, über wasserfreiem Natriumsulfat getrocknet und zur Trockne eingedampft. Der Rückstand wird in Aether aufgenommen, an basischem Aluminiumoxid chromatographiert und aus Aether/Petroläther umkristallisiert, wobei man das 5-(2-Methoxyäthyl)-8-chlor-1 1-(4-methyl- 1- piperazinyl)- SH-dibenzo [b,e] [1,4]diazepin in Form von gelbenKristallenvomSmp.160-161 C erhält.
Beispiel 2
Unter Verwendung des im Beispiel 1 beschriebenen Ver fahrens, und ausgehend von 5-[2-(2-Tetrahydropyranyl- oxy)äthyl]-8-chlor-10, 1 1-dihydro-SH-dibenzo[b,e] [1,4]dia zepin-11-on, wobei man jedoch die Extraktion der basischen
Anteile mit verdünnter Salzsäure durchführt und die Salzsäure
Extrakte zur Abspaltung der 2-Tetrahyd}opyranylgruppe während 1+ Stunden auf 60 C erwärmt. Die Aufarbeitung wie im Beispiel 1 beschrieben führt zum 5-(2-Hydroxyäthyl)-8 chior-1 1 - (4-methyl-1 -piperazinyl)- SH-dibenzo[b,e] [1,4jdiazepin vom Smp. 1571500 C.
Unter Verwendung des in den vorhergehenden Beispielen beschriebenen Verfahrens und entsprechender Ausgangsverbindungen gelangt man zu folgenden Verbindungen der Formel I: a) 5-(2-Methoxyäthyl)-1 1 - (4-methyl-1-pierazinyl)- SH-di benzo[b,e] [1,4]diazepin b) 5-(2-Hydroyäthyl)-1 1- (4-methyl-1-piperazinyl)- SH-di- benzo[b,ej [1,4]diazepin
Zu dem im Verfahren des Beispiels 1 als die Ausgangsverbindung verwendeten 5-(2-Methoxyäthyl)-8-chlor-10,11- di hydro-5H-dibenzo[b,e] [1,4]diazepin-11-on kann man wie folgt gelangen:

Ein Gemisch von 30,6 g 2-(2-Nitro-4-chlor-phenyl)-amino- benzoesäuremethylester und 120 ml Tetramemylharnstoff wird in Stickstoffatmosphäre mit 3,0 g Natriumhydrid versetzt und das erhaltene Gemisch während 1 Stunde bei Raumtemperatur gerührt. Hierbei entsteht ein tief violett gefärbtes Anion.
Dem Gemisch werden anschliessend 2-Methoxy-äthyl-ptoluolsulfonat zugesetzt und das Rühren während 1 Stunde bei Raumtemperatur und dann 5 Stunden bei 600 C fortgesetzt, wobei die Farbe auf gelb umschlägt. Anschliessend fügt man 1,5 g Natriumhydrid zu (wobei neuerdings Violettfärbung eintritt) sowie 18 g 2-Methoxy-äthyl-p-toluolsulfonat und rührt bei 60 C während 15 Stunden. Der Zusatz von Natriumhydrid und 2-Methoxy-äthyl-p- toluolsulfonat wird so lange fortgesetzt, bis keine Violettfärbung mehr auftritt.
Danach wird überschüssiges Natriumhydrid durch Zusatz von wenig Methanol zerlegt und das Reakrionsproduht zwischen Aether und viel Eiswasser verteilt. Die Aetherphase wird mit verdünnter Chlorwasserstoffsäure, Wasser, und verdünnter wässeriger Natriumcarbonat-Lösung gewaschen und danach eingeengt. Nach Klärung in Aetherlösung an Aluminiumoxid erhält man den N-2-Methoxyäthyl-2-nitro- 4-chlordiphenylamin-2'- carbonsäure-methylester als rotes Harz.
Dieses wird direkt für die nachfolgende Umsetzung verwendet.
34 g N-2-Methoxy-äthyl-2-nitro-4- chlor-diphenylamin-2'carbonsäure-methylester werden in 250 ml Essigester gelöst.
Die Lösung wird mit ca. 3 g Raney-Nickel versetzt und anschliessend unter Normalbedingungen hydriert. Nach der Aufnahme der theoretischen Menge Wasserstoff 6,381/0 C, 760 Torr.) ist die Lösung entfärbt. Nach Absaugen des Katalysators und Nachwaschen mit Methanol wird das Filtrat zur Trockne eingeengt. Der hierbei erhaltene rohe N-2-Methoxy äthyl-2-amino-4- chlor-diphenylamin-2'- carbonsäure-methyl ester wird mit Xylol im Vakuum nachgetrocknet.
Ein Gemisch von 31 g des rohen N-2-Methozy-äthyl-2- amino-4-chlor-diphenylamin-2 ,- carbonsäure-methylesters,
4,7 g granuliertem Natriumamid und 250 ml absolutem Dioxan wird während 4 Stunden unter Stickstoffatmosphäre zum Sieden erhitzt. Danach wird im Vakuum zur Trockne eingeengt.
Dem Rückstand werden zuerst vorsichtig Methanol und danach
Eiswasser zugesetzt, bis das Lactam vollständig ausgefallen ist.
Danach wird bei Raumtemperatur weitergeführt, bis der Nie derschlag krilinelig und damit gut filtrierbar wird. Der Nieder schlag wird abfiltriert, mit Wasser nachgewaschen und im Vakuum getrocknet. Nach Umkristallisieren aus Aceton/ Aether/Petgoläther in Gegenwart von Tierkohle erhält man reines 2-Methozyäthyl-8-chlor- 10,1 1-dihydro-SH-dibenzo[b,e] [1,4]diazepin-11-on.
Zu den in den übrigen Beispielen verwendeten Ausgangs verbindungen gelangt man in analoger Weise durch Ersatz von
2-Methoxyäthyl-p- toluol-sulfonat durch äquivalente Anteile von Aethylenchlorhydrin-tetrahydro-pyranyläther, den man auf an sich bekannte Weise durch Umsetzung von Aethylen cblorhydrin mit 2,3-Dihydropyran erhält.
