Verbindungen der Formel (II), die als Ausgangsstoffe verwendet werden können, sind beispielsweise 3 ,5-Diamino- benzoesäure, 3,4-Diaminobenzoesäure, 2,4-Diaminobenzoesäure, 2,5-Diaminobenzoesäure sowie deren Alkylester, wo bei die Alkylgruppen 1 bis 4 C-Atome aufweisen, beispieles.
weise 2,4-Diaminobenzoesäuremethylester, 2,4-Diaminobenzoesäureäthylester, 2,4-Diaminobenzoesäurebutylester, 3,5-Diaminobenzoesäuremethylester sowie 2,4-Diaminobenzamid und 3,5-Diaminobenzamid.
Verbindungen der Formel (III) und deren reaktive Derivate, die beim erfindungsgemässen Verfahren verwendet werden können, sind insbesondere Salicylsäure, 2-Alkoxybenzoesäuren mit 1 bis 4 C-Atomen in den Alkoxygruppen, beispielsweise 2-Methoxybenzoesäure, 2-Äthoxybenzoesäure und 2-n -Butoxybenzoesäure; 2-Acyloxybenzoesäuren mit 1 bis 4 C-Atomen im Acylrest, beispielsweise 2-Acetoxybenzoesäure; sowie die entsprechenden Säurehalogenide, Ester, Säureanhydride und auch die gemischten Säureanhydride der obengenannten Säuren mit einer anderen Säure, beispielsweise einer anderen Carbonsäure, der Kohlensäure, Schwefelsäure, einer Sulfonsäure, der Phosphorsäure usw.
Diese Reaktion kann beispielsweise so ausgeführt werden, dass man eine Verbindung der Formel (II) mit einem reaktiven Derivat einer Carbonsäure der Formel (III) bei einer Temperatur von im allgemeinen -10"C bis 50 C, vorzugsweise OOC bis 20"C, während 30 Minuten bis 4 Stunden kon- densiert.
Lösungsmittel, die bei dieser Reaktion verwendet werden können, sind beispielsweise Wasser, Benzol, Toluol, Tetra hydrofuran, Diäthyläther, Dioxan, Dimethylformamid, Chlo roform, Methylenchlorid, Acetonitril, Aceton, Tetrachlorkohlenstoff, Äthylacetat, deren Mischungen usw.
Bei der Reaktion kann man Beschleuniger verwenden, beispielsweise anorganische Basen, wie die Hydroxyde, Carbonate und Acetate von Alkalimetallen oder Erdalkalimetallen, beispielsweise Kaliumacetat, Natriumacetat, Natriumcarbonat,
Kaliumcarbonat, Natriumhydroxyd, Calciumacetat und Calciumcarbonat; sowie tertiäre Amine wie beispielsweise Pyridin, Triäthylamin, Dimethylanilin und Picolin.
Eine Verbindung der Formel (in), deren Aminorest durch Phosphortrichlorid oder einen Ester der chlorphosphorigen Säure aktiviert wurde, kann mit einer Verbindung der Formel (III) bei einer Temperatur zwischen Zimmertemperatur und Rückflusstemperatur des verwendeten Lösungsmittels umgesetzt werden. Vorzugsweise führt man diese Reaktion aus, nachdem man die Hydroxylgruppe der Verbindung (in), worin Rl Hydroxyl bedeutet, geschützt hat. Die Reaktion wird in Gegenwart eines Lösungsmittels vorgenommen, beispielsweise eines neutralen Lösungsmittels wie Benzol, Toluol, Xylol, Dioxan oder Tetrahydrofuran, oder eines basischen Lösungsmittels wie Pyridin, Triäthylamin, Dimethylanilin oder Picolin.
Falls die Reaktion in Gegenwart eines neutralen Lösungsmittels ausgeführt wird, setzt man vorzugsweise ein tertiäres Amin als Beschleuniger zu. Der Zeitbedarf für diese Reaktion beträgt 30 Minuten bis 3 Stunden.
Eine Verbindung der Formel (Il} kann mit einer Verbindung der Formel (III) in einem inerten organischen Lösungsmittel in Gegenwart eines Beschleunigers für die Amidbildung vorgenommen werden, beispielsweise Dicyclohexylcarbodiimid, bei einer Temperatur zwischen Zimmertemperatur und Rückflusstemperatur des verwendeten Lösungsmittels und während einer Zeit von 1 bis 5 Stunden. Falls die Verbindung der Formel (11) eine Hydroxylgruppe aufweist, schützt man diese vorzugsweise vor Beginn der Reaktion. Bei dieser Reaktion können als Lösungsmittel Benzol, Toluol, Tetrahydrofuran, Chloroform, Methylenchlorid, Acetonitril und andere verwendet werden.
Die so hergestellte Verbindung der Formel (I) kann, wenn sie einen Ester darstellt, verseift werden, oder man kann sie acylieren, wenn sie eine oder zwei phenolische Hydroxylgruppen enthält. Diese Reaktionen können auf bekannte Weise vorgenommen werden. Beispielsweise kann man den Ester durch Einwirkung eines Hydroxyds oder Carbonats eines Alkalimetalls oder Erdalkalimetalls in Wasser oder einem wässrigen organischen Lösungsmittel bei einer Temperatur zwischen Zimmertemperatur und 10000 während 0,5 bis 30 Stunden verseifen. Die Acylierung der phenolischen Hydroxylgruppen kann mittels eines Säurechlorids oder Säureanhydrids einer niederen Fettsäure bei 0 bis 1000C während 1 bis 10 Stunden geschehen.
Die Verwendung eines Lösungsmittels ist nicht erforderlich, man kann aber auch ein inertes Lösungsmittel wie Tetrahydrofuran, Dioxan oder Aceton verwenden. Ein Beschleunigungsmittel wie Pyridin, Triäthylamin oder Natriumacetat oder auch ein saurer Beschleuniger wie Schwefelsäure oder p-Toluolsulfonsäure kann zweckmässig eingesetzt werden.
Falls das Produkt eine freie Carboxylgruppe aufweist, kann ein pharmakologisch annehmbares Salz für den gleichen Zweck wie das saure Produkt verwendet werden.
Die erfindungsgemäss hergestellten Verbindungen der Formel (I) zeigen charakteristische biologische Wirkungen, insbesondere auf ein System mit Antigen-Antikörper-Reaktion. Beispielsweise wurde bei in-vitro-Versuchen gefunden, dass eine beliebige wasserlösliche Verbindung der Formel (1) schon in sehr kleinen Mengen die Hämolyse roter Blutkör perchen von Schafen (sRBC) inhibiert, welche durch die Reaktion von sRBC und anti-sRBC aus Mäuseserum in Gegenwart eines Komplementärstoffes indiziert wird (Immunhämolyse). Bei Tierversuchen zeigten die Verbindungen der Formel (I) eine starke Inhibierung der anaphylaktischen Reaktion der Haut, und zwar bei oralen Verabreichung in Dosen bis zu 100 mg/kg.
Die Verbindungen der Formel (I) inhibieren nicht nur die obengenannte Immunreaktion, sondern haben ausserdem antipyretische, sedaüve, antiinflammatorische und antiallergische Eigenschaften. Die Verbindungen sind besonders wertvoll zur Behandlung von Krankheiten, welche durch immunologische Störungen verursacht werden, beispielsweise rheumatische Arthritis, Bronchialasthma und Nephritis.
Das erfindungsgemässe Verfahren wird durch die folgenden Versuche und Beispiele weiter erläutert.
Versuch I
Inhibierung wärmeinduzierter Hämolyse
Rote Blutkörperchen (RBC) einer männlichen Wister -Imamichiratte wurden in einem 0,15 M Phosphatpuffer (pH 7,4) zu einer 1 %-igen Suspension von RBC suspendiert. In ein Reagenzglas wurden 5 ml der Suspension eingebracht und dann 0,1 ml einer 10%-igen Lösung von Dimethylsulfoxyd in Äthanol, die eine zu untersuchende Verbindung aufwies, und man liess dann 20 Minuten lang bei 53"C inkubieren. Das Reagenzglas wurde dann in Wasser gekühlt und der Inhalt bei 2000 Ulmin 10 Minuten lang zentrifugiert.
Mittels eines Spektrophotometers Hitachi-124 wurde die optische Dichte der überstehenden Flüssigkeit bei 540 mli gemessen, und der Inhibierungswert der Hämolyse wurde nach folgender Gleichung abgeschätzt:

Versuch 2
Inhibierung von immunreaktion-induzierter Hämolyse
Es wurden die folgenden vier Lösungen hergestellt.
A. Lösung des Antiserums
Handelsübliches Schafblut wurde dreimal mit physiolo gischer Kochsalzlösung gewaschen und bei 2000 U/min 5
Minuten lang zentrifugiert. Ein ml komprimierter sRBC wur de in 10 ml physiologischer Salzlösung suspendiert. Je 0,2 ml der erhaltenen Suspension wurde Mäusen (IV cs 5W) intra venös injiziert, und nach 4 Tagen wurde den Mäusen eine
Blutprobe entnommen. Aus den Proben wurde Serum mit
Anti-sRBC-Antikörpern gewonnen und auf das 40-fache des ursprünglichen Volumens mit einer gelatinehaltigen Vernal pufferlösung mit einem pH von 7,5, die weiterhin Ca+e und
Mg++ enthielt (GVB++) verdünnt, wobei man eine Antiserum -Lösung erhielt.
B. Komplementärlösung
Als Komplementärquelle wurde Meerschweinchenserum verwendet. Dieses Meerschweinchenserum wurde mit GVB++ auf das 40-fache des ursprünglichen Volumens verdünnt, wo bei man eine Komplementärlösung erhielt. Alle Arbeitsgänge zur Herstellung dieser Lösung wurden unter Kühlung vor genommen.
C. sRBC-Suspension sRBC wurde in GAB++ suspendiert. 0,2 ml der Suspension wurde unter Auffüllen auf 3,3 ml mit Wasser vollständig homolysiert. Dann wurde die erhaltene Suspension nach
Vervollständigung der Hämolyse auf eine optische Dichte von
0,455 bei 541 ntli eingestellt.
D. Untersuchungslösung
Eine zu untersuchende Verbindung wurde in GVB++ so aufgelöst, dass man eine Konzentration erhielt, welche das
1,5-fache der beabsichtigten Endkonzentration betrug.
Zu 2 ml der Untersuchungslösung wurden 0,5 ml der An tiserumlösung, 0,25 ml der sRBC-Suspension und 0,25 ml der
Komplementärlösung zugegeben, und das Gemisch wurde bei 37 C 40 Minuten lang inkubiert. Dann wurden zur Reaktionsmischung zur Vervollständigung der Reaktion 0,3 ml einer 3,8%-igen wässrigen Trinatriumcitratlösung gegeben.
Nichthämolysierte sRBC wurden durch 5 minütiges Zentrifugieren bei 2000 U/min vom Gemisch abgetrennt. Die Absorption der überstehenden Flüssigkeit wurde bei 541 mli gemessen.
Eine Vergleichsprobe wurde daneben auf die gleiche Weise wie beschrieben hergestellt, wobei man aber 2 ml GVB++ ohne Untersuchungsverbindung verwendete.
Ausserdem wurde für jeden Versuch eine Blindlösung nach der oben beschriebenen Arbeitsweise hergestellt, wobei jedoch anstelle der 0,25 ml Komplementärlösung 0,25 ml GVB++ verwendet wurden, und man korrigierte die durch die Färbung der Untersuchungslösung hervorgerufenen Messfehler.
Der Hämolyse-Inhibierungswert (%) wurde nach der folgenden Gleichung abgeschätzt:

Beispiel 1
Eine Lösung von 17,5 g 2-Acetoxybenzoylchlorid in 20 ml Dioxan wurde auf einmal zu einem Gemisch von 4,6 g 3,5 -Diaminobenzoesäure, 15,3 g Kaliumacetat und 80 ml Wasser gegeben und das Ganze bei einer Temperatur zwischen 0 und 5 C gerührt. Man rührte bei dieser Temperatur noch eine Stunde weiter, dann bei Zimmertemperatur 30 Minuten lang und bei 50 C 30 Minuten lang. Das Reaktionsgemisch wurde in 200 ml kalte verdünnte Salzsäure gegossen und die ungelösten Substanzen abgetrennt. Diese wurden in etwa 100 ml 1N Natronlauge gelöst und die Lösung mit Aktivkohle behandelt. Dann wurde mit Salzsäure auf ein pH von 3 bis 4 angesäuert, wobei Kristalle ausfielen. Diese wurden abfiltriert und mit Wasser gewaschen.
Nach Umkristallisieren aus Methanol/Wasser erhielt man 7,2 g 3,5-Bis-(salicylamido) -benzoesäure mit einem Schmelzpunkt von 305 bis 3080C (Zersetzung).

Beispiel 2
Eine Lösung von 17,2 g 2-Acetoxybenzoylchlorid in 20 ml Dioxan wurde auf einmal zu einem Gemisch von 4,6 g 3,4 -Diaminobenzoesäure, 15,3 g Kaliumacetat und 70 ml Wasser gegeben und die Mischung bei 0 bis 5 C gerührt, und bei dieser Temperatur wurde 30 Minuten lang weitergerührt.
Zum Gemisch gab man nun eine Lösung von 1 g 2-Acetoxybenzoylchlorid in 3 ml Dioxan. Dieses Gemisch rührte man bei Zimmertemperatur 30 Minuten lang und danach weitere 30 Minuten bei 50 C. Das Gemisch wurde dann in 200 ml kalte verdünnte Salzsäure gegossen und das ungelöste Gemisch von öl und pulvrigem Feststoff abgetrennt. Diese Stoffe wurden in etwa 100 ml 1N Natronlauge gelöst, die Lösung mit Aktivkohle behandelt und dann angesäuert, wobei Kristalle ausfielen. Die Kristalle wurden abfiltriert und mit Wasser gewaschen. Nach Umkristallisieren aus einem Gemisch aus Dioxan und Wasser erhielt man 6,9 g 3,4-Bis-(salicylamido)-benzoesäure mit einem Schmelzpunkt von 305 bis 307 C (Zersetzung).

Beispiel 3
Ein Gemisch aus 3,32 g 2,5-Diaminobenzoesäuremethylester, 5,8 ml Triäthylamin und 20 ml Tetrahydrofuran wurde tropfenweise zu einer Lösung von 8,7 g 2-Acetoxybenzoylchlorid in 150 ml Tetrahydrofuran im Verlauf von 30 Minuten gegeben, währen bei 5 bis 100C gerührt wurde. Nach Beendigung der Zugabe rührte man noch eine Stunde bei Zimmertemperatur nach und goss das Gemisch auf 500 ml Eis und Wasser, wobei Kristalle ausfielen. Diese Kristalle wurden abfiltriert, mit Wasser gewaschen und in Luft getrocknet. Nach Umkristallisieren aus Äthylacetat erhielt man 8,1 g 2,5-Bis-(2'-acetoxybenzamido)-benzoesäuremethylester mit einem Schmelzpunkt von 170 bis 171 0C.

Beispiel 4
5,3 g des Methylesters der 2,5-Bis-(2'-acetoxybenzamido)- -benzoesäure, erhalten gemäss Beispiel 3, löste man in 150 ml Methanol auf. Zur Lösung gab man 2,5 g Natriumcarbonat, gelöst in 40 ml Wasser, und rührte dann das Gemisch 30 Minuten lang bei Zimmertemperatur. Dann wurde es in 500 ml Wasser gegossen, welches mit 10 ml Essigsäure angesäuert war, wobei Kristalle ausfielen. Diese wurden abfiltriert, mit Wasser gewaschen, getrocknet und aus Äthylacetat umknstallisiert. Man erhielt 3,8 g 2,5-Bis-(salicylamido)-benzoesäuremethylester mit einem Schmelzpunkt von 243 bis 244 C.

Beispiel 5
Eine Lösung von 4 g Natriumhydroxyd in 10 ml Wasser wurde zu einem Gemisch von 5,3 g des nach Beispiel 3 erhaltenen Methylesters der 2,5-Bis-(2'-acetoxybenzamido) -benzoesäure und 100 mi Methanol gegeben. Das Gemisch wurde bei Zimmertemperatur 20 bis 30 Stunden lang gerührt und dann in 500 ml Wasser gegossen, welches 10 ml Essigsäure enthielt. Die ausgefallenen Kristalle wurden abfiltriert, mit Wasser gewaschen und aus einem Gemisch von Dioxan und Wasser umkristallisiert. Man erhielt 3,5 g 2,5-Bis-(salicyl amido)-benzoesäure mit einem Schmelzpunkt von 280 bis 283"C (Zersetzung).

Beispiel6
Ein Gemisch aus 6,64 g 2,4-Diaminobenzoesäuremethylester, 11,5 ml Triäthylamin und 50 ml Tetrahydrofuran wur dètropfenweise im Verlauf von 34 bis 1 Stunde zu einer Lösung von 17,4 g 2-Acetoxybenzoylchlorid in 300 ml Tetrafuran gegeben, während bei 5 bis 10 C gerührt wurde. Es wurde bei Zimmertemperatur 2 Stunden nachgerührt. Das Reaktionsgemisch wurde in 1,5 1 verdünnte Salzsäure gegossen. Die ausgefallenen Kristalle wurden abfiltriert, mit Wasser gewaschen, getrocknet und aus Äthylacetat umkristallisiert. Man erhielt 15 g des Methylesters der 2,4-Bis-(2'-acetoxybenzamido)-benzoesäure mit einem Schmelzpunkt von 143 bis 145"C.

Beispiel 7
Nach der Arbeitsweise aes Beispiels 4 wurde der Methylester der 2,4-Bis-(2'-acetoxybenzamido)-benzoesäure mit Hilfe von Natriumcarbonat hydrolysiert. Die erhaltenen Kristalle wurden aus einem Gemisch von Aceton, Dioxan und Wasser umkristallisiert, und man erhielt den Methylester der 2,4-Bis -(salicylamido)-benzoesäure mit einem Schmelzpunkt von 259 bis 2600C in 82%-ige Ausbeute.

Beispiel 8
Nach der Arbeitsweise des Beispiels 5 wurde gemäss Beispiel 6 erhaltener 2,4-Bis-(2'-acetoxybenzamido)-benzoesäure- methylester mittels Natronlauge hydrolysiert. Nach Umkristallisieren des Verseifungsproduktes aus einem Gemisch von Dioxan und Wasser erhielt man in 85%-iger Ausbeute 2,4 -Bis-(salicylamido)-benzoesäure mit einem Schmelzpunkt von 250 bis 252"C (Zersetzung).

Beispiel 9
Eine Lösung von 3,86 ml Phosphoroxychlorid in 20 ml Tetrahydrofuran wurde tropfenweise im Verlauf von 1 bis 2 Stunden zu einem Gemisch von 3,3 g 2,4-Diaminobenzoe- säuremethylester, 6,7 g 2-Methoxybenzoesäure, 1,8 ml Tri äthylamin und 100 ml Tetrahydrofuran unter Rühren bei 10 bis -5"C gegeben, wonach bei Zimmertemperatur eine
