The present invention relates to a process for preparing new compounds of the formula I (see formula sheet), wherein R1 is an alkyl group with 1 to 10 carbon atoms, a cycloalkyl group with 3 to 8 carbon atoms or the phenyl group.
According to the invention, the new compounds of the formula I, in which Rl has the above meaning, are obtained by lactonizing compounds of the formula II in which Rl is an alkyl group with 1 to 10 carbon atoms, a cycloalkyl group with 3 to 8 carbon atoms or the phenyl group.
Suitable alkyl radicals are preferably the isopropyl and isobutyl groups. Suitable cycloalkyl radicals are preferably the cyclopentyl, cyclohexyl and cycloheptyl groups.
For example, the procedure is that compounds of the formula II, wherein Rl has the above meaning, in the presence of a catalytic amount of an acid, for. B. conc. Sulfuric acid or p-toluenesulfonic acid, lactonized. The water formed in the lactonization can be mixed with an inert solvent, e.g. B. an aromatic hydrocarbon such as benzene, toluene or xylene are continuously distilled off. If you use water-binding acids such as hydrogen chloride or conc. Sulfuric acid, the resulting water does not need to be distilled off.
After the reaction has ended, the compounds of the formula I can be isolated and purified by known methods.
The previously unknown compounds of the formula II used as starting material are prepared by adding compounds of the formula III, in which Rl is an alkyl group having 1 to 10 carbon atoms, a cycloalkyl group having 3 to 8 carbon atoms or a phenyl group, with an acetyl halide in the presence of a Friedel -Crafts catalyst, preferably with tin (IV) chloride, in a solvent which is inert under the reaction conditions, such as benzene or carbon tetrachloride, at 20 to 800, the resulting compounds of the formula IV, wherein Rl is an alkyl group with 1 to 10 carbon atoms, a cycloalkyl group with 3 to 8 carbon atoms or a phenyl group, reacts with sulfur and morpholine, the compounds of formula V thus obtained,
wherein Rl stands for an alkyl group with 1 to 10 carbon atoms, a cycloalkyl group with 3 to 8 carbon atoms or a phenyl group, saponified with alcoholic alkali metal hydroxide solution, the compounds of the formula IV thus obtained, wherein Rl for an alkyl group with 1 to 10 carbon atoms, a cycloalkyl group with 3 to 8 carbon atoms or a phenyl group, treated with a boron trihalide, preferably boron tribromide, or with hydrogen halide, preferably hydrogen bromide.
The compounds of the formula I have not yet been described in the literature. In pharmacological testing, they are characterized by diverse and highly pronounced effects and can therefore be used as remedies. Thus, the compounds of the formula I have an edema-inhibiting and anti-inflammatory effect, as can be shown in the carrageenan edema test on rats. The dose to be used naturally varies depending on the type of administration and the condition to be treated. In general, however, satisfactory results are obtained in test animals at a dose of 1 to 100 mg / kg body weight; if necessary, this dose can be administered in 2 to 3 portions or as a sustained-release form.
For larger mammals the daily dose is 20 to 3,000 mg; For oral administration, a suitable form of administration contains 20 to 500 mg of the active substance, mixed with liquid or solid carrier substances.
The compounds of the formula I also have an antipyretic and analgesic effect.
As medicaments, the new compounds or their water-soluble, physiologically tolerable salts can be administered alone or in a suitable medicinal form with pharmacologically inert auxiliaries.
In the following examples, all temperatures are given in degrees Celsius.

Crystallization of the residue from methylene chloride ether yields 2'-methoxy-4'-phenyl-phenylthionacetic acid morpholide with a melting point of 132-133.
d) 2'-methoxy-4'-phenyl-phenylacetic acid
18.8 g of 2'-methoxy-4'-phenyl-phenylthionacetic acid morpholide are refluxed in 90 ml of 3N-ethanolic potassium hydroxide overnight. The reaction mixture is evaporated, the residue is dissolved in water, the solution must be strongly alkaline. It is then extracted three times with methylene chloride. The organic phases are washed once with water. The aqueous phases are combined and acidified. It is then extracted three times with ethyl acetate and washed once with water. The ethyl acetate phases are combined, dried over anhydrous sodium sulfate and concentrated. Crystallization of the residue from methylene chloride pertrol ether gives 2'-methoxy-4'-phenyl-phenyl acetic acid. M.p. 164-1650.
e) 2'-Hydroxy-4'-phenyl-phenylacetic acid
2 g of 2'-methoxy-4'-phenyl-phenylacetic acid are dissolved in 60 ml of dry methylene chloride and 4 ml of boron tribromide are added dropwise at 0 °. After 30 minutes at room temperature, it is poured onto 150 ml of 2N sodium hydroxide. It is then acidified to pH = 1 with 2N hydrochloric acid and extracted three times with ethyl acetate. The organic phases are washed twice with water, combined, dried over anhydrous sodium sulfate and concentrated. The residue is crystallized from acetone-hexane. The phenylacetic acid obtained in this way melts at 150-151 ".
