dans laquelle le symbole R représente un reste acyle contenant de 1 à 6 atomes de carbone, composés intéressant également l'industrie des parfums et arômes pour leurs propriétés organoleptiques remarquables.
La présente invention a pour objet un procédé pour la préparation du composé I telle que défini ci-dessus, caractérisé en ce que a) on traite un alcool insaturé de formule

au moyen d'un peracide organique et soumet ensuite le produit résultant dudit traitement à l'action d'un agent acide pour obtenir un composé de formule

b) soumet finalement le composé hydroxylé ainsi obtenu à une hydrogénation catalytique:

La première étape dudit procédé consiste en l'époxydation du 4-(2,6,6-triméthyl-cyclohex-1-ène-1-yl)-but- cis-3-ène-2-ol (II). Selon une mise en oeuvre particulière du procédé de l'invention, ladite époxydation s'effectue à l'aide d'un peracide organique tel l'acide peracétique, perbenzoïque, monochloroperbenzoique, perphtalique ou performique, dans un solvant chloré comme le chloroforme, le chlorure de méthylène, le trichloroéthylène ou dichloro-éthane. On utilise de préférence l'acide peracétique dans le chlorure de méthylène.
L'acide peracétique peut être préparé in situ par l'action d'eau oxygénée sur de l'acide acétique en présence de petites quantités d'acide sulfurique suivant des méthodes conventionnelles [voir par exemple H. O. House, Modern Synthetic Reactions, 2sema édition, Benjamin Inc., Menlo Park (1972), p. 293].
Des agents tampons comme des sels alcalins d'acides organiques peuvent être incorporés au milieu d'époxydation. On peut employer par exemple le formiate, l'acétate, le propionate, le butyrate, l'oxalate, le citrate ou le tartrate de sodium ou de potassium. L'acétate de sodium est utilisé de préférence.
Le traitement acide du produit résultant de l'action d'un peracide organique sur le composé II, s'effectue au moyen d'un acide fort, minéral ou organique, en présence d'un solvant organique inerte. Comme agent acide on peut utiliser l'acide chlorhydrique, sulfurique ou phosphorique par exemple,
I'acide benzènesulfonique ou p-toluènesulfonique, ou encore une terre de diatomées acide.
Comme solvant organique, on peut utiliser un hydrocarbure aliphatique ou aromatique tel l'hexane, le cyclohexane ou le toluène, ou un éther comme le dioxanne ou le diméthoxyéthane par exemple. On peut également utiliser un hydrocarbure halogéné, tel que le chloroforme, le chlorure de méthylène, le tri- ou le tétrachloro-éthylène par exemple. On utilise de préférence le solvant de l'étape réactionnelle précédente,
I'époxydation, dans le cas présent le chlorure de méthylène.
Le 2,6,10,1 O-tétraméthyl-l -oxa-spiro[4.5]déc-3-ène-6-ol (III) est finalement soumis à une hydrogénation catalytique pour donner le composé spirannique saturé correspondant I.
L'hydrogénation du composé III s'effectue en présence d'un catalyseur métallique, selon les techniques conventionnelles.
On peut utiliser à cet effet un catalyseur tel que l'oxyde de platine, le palladium sur charbon ou le nickel de Raney par exemple, en présence d'un solvant organique inerte tel un alcool comme le méthanol, l'éthanol ou l'isopropanol par exemple, ou un hydrocarbure aliphatique ou aromatique,
I'hexane ou le toluène par exemple. Ladite hydrogénation s'effectue de préférence en présence de palladium sur charbon et d'alcool éthylique.
L'alcool insaturé de formule II, utilisé comme produit de départ dans le procédé ci-dessus, peut être obtenu à partir de p-ionone, selon la méthode décrite dans J. Org. Chem. 38, 1247 (1973).
L'invention a également pour objet l'utilisation du composé
I, obtenu selon le procédé défini précédemment, comme produit de départ pour la préparation des composés spirannique oxygénés de formule V. Ladite utilisation est caractérisée en ce qu'on esterifie le composé I.
L'estérification de l'alcool tertiaire de formule I s'effectue selon les techniques usuelles, en traitant par exemple ledit alcool au moyen d'un halogénure d'acyle, le chlorure de préférence, en présence d'une base organique telle que la N,Ndiméthyl-aniline par exemple.
C'est ainsi que l'acétate de 2,6,10,10-tétraméthyl-1- oxa-spiro[4.5]déc-6-yle est obtenu à partir du 2,6,10,10tétraméthyl-1-oxa-spiro[4.5]décane-6-ol, par traitement de ce dernier au moyen de chlorure d'acétyle, en présence de N,Ndiméthylaniline.
Le formiate de 2,6,10, 10-tétraméthyl-1 -oxa-spiro[4.5]- déc-6-yle par contre, est obtenu en traitant ledit alcool au moyen de formylimidazole, selon la méthode décrite dans
Liebigs Ann. Chem. 655, 95 (1962).
L'invention sera illustrée de façon plus détaillée à l'aide des exemples ci-après. Dans lesdits exemples, les températures sont indiquées en degrés centigrades et les abréviations utilisées possèdent le sens usuel dans l'art.
Exemple I 2,6,10,10-Tétraméthyl-l-oxa-spiro[4.5]décane-6-ol
i) 2,28 g (environ 12 mmole) d'acide peracétique à 40%, contenant environ 2% d'acétate de sodium anhydre, ont été ajoutés goutte à goutte à un mélange, refroidi à 00, de 2,33 g (12 mmole) de 4-(2,6,6-triméthyl-cyclohex-1ène-1-yl)-but-cis-3-ène-2-ol et 1,73 g (18 mmole) d'acétate de sodium anhydre dans 70 ml de chlorure de méthylène. Après addition des réactifs, on a poursuivi l'agitation du mélange réactionnel durant 15 h à température ambiante, puis on l'a filtré.
Le filtrat clair ainsi obtenu a ensuite été successivement traité par deux portions de NaHCO3 à 10% dans H2O, puis au moyen d'une solution aqueuse saturée en NaCI, avant d'être séché sur Na2SO4. Après évaporation des fractions volatiles, on a obtenu 2,49 g d'un résidu qui, après séparation par chromatographie sur colonne (Gel de Silice - Cyclohexane/Acétate d'éthyle 7 : 3), a donné 0,53 g de produit de départ et 1,76 g d'un produit époxydé (rendement de l'époxydation: 90%).
ii) Le produit d'époxydation obtenu ci-dessus dissous dans 50 ml de chlorure de méthylène a été agité durant 24 h à température ambiante, en présence de 0,07 g d'acide ptoluène-sulfonique et sous atmosphère d'azote. Après les traitements de neutralisation, lavage et séchage indiqués précédemment - voir lettre i) -, la distillation du résidu (1150/0,005
Torr) a donné 1,58 g (environ 90%) de 2,6,10,10-tétraméthyl- 1-oxaspiro[4.5]déc-3-ène-6-ol, Eb. 80-90o/0,1 Torr.

Le 2,6,10, 10-tétraméthyl- 1 -oxa-spiro[4.5]déc-3-ène-6- ol, qui est un produit nouveau, se caractérise par une odeur boisée et camphrée rappelant par certains côtés celle du patchouli. n peut être par conséquent avantageusement utilisé à titre d'ingrédient parfumant ou aromatisant.
iii) 1,54 g (environ 7 mmole) du composé hydroxylé insaturé obtenu ci-dessus, dissous dans 40 ml d'éthanol, ont été hydrogénés à 10 environ, en présence de 250 g de palladium à 5% sur charbon, après absorption de 155 ml d'hydrogène, le mélange réactionnel a été filtré, évaporé et distillé (115 /0,005
Torr) pour donner 1,51 g de résidu. Le produit ainsi obtenu a ensuite été purifié par chromatographie sur colonne (Gel de
Silice - Cyclohexane/Acétate d'éthyle 7 : 3) et l'on a finalement isolé 1,08 g (55%) de 2,6,10,10-tétraméthyl-1-oxa -spiro[4.5]décane-6-ol, Eb. 58-59 /0,5 Torr.

Exemple 2
Formiate de 2,6,10, 10-tétraméthyl- 1-oxa-
spiro[4.5]déc-6-yle
0,4 g (1,9 mM) de 2,6,10,10-tétraméthyl-1-oxa-spiro- [4.5]décane-6-ol et 0,5 g (5,2 mM) de formylimidazole fraîchement sublimé ont été intimement mélangés avant d'être entreposés durant 48 h à température ambiante. Après extraction du mélange réactionnel à l'éther, lavage, séchage et distilla tion fractionnée de la phase organique, on a recueilli 0,3 g d'un produit ayant Eb. 70-800/0,5 Torr et contenant 60% d'ester désiré. Un échantillon pour analyse a été obtenu par chromatographie en phase gazeuse préparative.

Exemple 3
Acétate de 2,6,10,10-tétraméthyl-1-oxa-
spiro[4.5]déc-6-yle
5,9 g (0,075 M) de chlorure d'acétyle ont été ajoutés en 30 min, sous agitation, à un mélange maintenu à 200 de 2,12 g (0,01 M) de 2,6,10,10-tétraméthyl-1-oxa-spiro [4.5]décane-6-ol et 10,9 g (0,09 M) de N,N-diméthylaniline.
Après avoir été entreposé durant 2 jours à température ambiante, le mélange réactionnel a été chauffé à reflux pendant 3 h. Après avoir été traité avec 50 ml d'éther, le mélange réactionnel a été filtré et le filtrat obtenu versé sur de la glace et acidifié au moyen de H2S04 à 10% dans H20. La phase organique a ensuite été lavée au moyen d'une solution saturée en NaHCO3 jusqu'à neutralité, finalement séchée, évaporée et distillée selon les techniques usuelles pour donner 2,5 g d'un produit ayant Eb. 90-lOOo/0,1 Torr. Après recristallisation dans un mélange éthanol-eau, on a isolé 1,9 g (75 %) d'ester désiré, F. 55-560.

The present invention relates to a process for the preparation of compound I as defined above, characterized in that a) treating an unsaturated alcohol of formula

by means of an organic peracid and then subjects the product resulting from said treatment to the action of an acidic agent to obtain a compound of formula

b) finally subjects the hydroxylated compound thus obtained to catalytic hydrogenation:

The first step of said process consists of the epoxidation of 4- (2,6,6-trimethyl-cyclohex-1-ene-1-yl) -but-cis-3-en-2-ol (II). According to a particular implementation of the process of the invention, said epoxidation is carried out using an organic peracid such as peracetic, perbenzoic, monochloroperbenzoic, perphthalic or performic acid, in a chlorinated solvent such as chloroform, methylene chloride, trichlorethylene or dichloroethane. Preferably, peracetic acid is used in methylene chloride.
Peracetic acid can be prepared in situ by the action of hydrogen peroxide on acetic acid in the presence of small amounts of sulfuric acid according to conventional methods [see for example HO House, Modern Synthetic Reactions, 2nd edition, Benjamin Inc., Menlo Park (1972), p. 293].
Buffering agents such as alkali salts of organic acids can be incorporated into the epoxidation medium. For example, sodium or potassium formate, acetate, propionate, butyrate, oxalate, citrate or tartrate can be used. Preferably sodium acetate is used.
The acid treatment of the product resulting from the action of an organic peracid on compound II is carried out by means of a strong, mineral or organic acid, in the presence of an inert organic solvent. As acidic agent, hydrochloric, sulfuric or phosphoric acid can be used for example,
Benzenesulfonic or p-toluenesulfonic acid, or alternatively an acidic diatomaceous earth.
As organic solvent, it is possible to use an aliphatic or aromatic hydrocarbon such as hexane, cyclohexane or toluene, or an ether such as dioxane or dimethoxyethane, for example. It is also possible to use a halogenated hydrocarbon, such as chloroform, methylene chloride, tri- or tetrachloro-ethylene, for example. The solvent from the previous reaction step is preferably used,
Epoxidation, in this case methylene chloride.
The 2,6,10,1 O-tetramethyl-1 -oxa-spiro [4.5] dec-3-en-6-ol (III) is finally subjected to catalytic hydrogenation to give the corresponding saturated spiran compound I.
The hydrogenation of compound III is carried out in the presence of a metal catalyst, according to conventional techniques.
A catalyst such as platinum oxide, palladium on carbon or Raney nickel for example, in the presence of an inert organic solvent such as an alcohol such as methanol, ethanol or isopropanol can be used for this purpose. for example, or an aliphatic or aromatic hydrocarbon,
Hexane or toluene for example. Said hydrogenation is preferably carried out in the presence of palladium on carbon and ethyl alcohol.
The unsaturated alcohol of formula II, used as a starting material in the above process, can be obtained from p-ionone, according to the method described in J. Org. Chem. 38, 1247 (1973).
A subject of the invention is also the use of the compound
I, obtained according to the process defined above, as starting material for the preparation of the oxygenated spiran compounds of formula V. Said use is characterized in that the compound I.
The esterification of the tertiary alcohol of formula I is carried out according to the usual techniques, for example by treating said alcohol by means of an acyl halide, preferably the chloride, in the presence of an organic base such as N, Ndimethyl-aniline for example.
Thus, 2,6,10,10-tetramethyl-1-oxa-spiro [4.5] dec-6-yl acetate is obtained from 2,6,10,10tetramethyl-1-oxa-spiro [4.5] decan-6-ol, by treatment of the latter with acetyl chloride, in the presence of N, Ndimethylaniline.
The formate of 2,6,10,10-tetramethyl-1 -oxa-spiro [4.5] - dec-6-yl on the other hand, is obtained by treating said alcohol by means of formylimidazole, according to the method described in
Liebigs Ann. Chem. 655, 95 (1962).
The invention will be illustrated in more detail with the aid of the examples below. In said examples, the temperatures are indicated in degrees centigrade and the abbreviations used have the usual meaning in the art.
Example I 2,6,10,10-Tetramethyl-1-oxa-spiro [4.5] decan-6-ol
i) 2.28 g (about 12 mmol) of 40% peracetic acid, containing about 2% anhydrous sodium acetate, was added dropwise to a mixture, cooled to 00, of 2.33 g ( 12 mmol) of 4- (2,6,6-trimethyl-cyclohex-1ene-1-yl) -but-cis-3-en-2-ol and 1.73 g (18 mmol) of anhydrous sodium acetate in 70 ml of methylene chloride. After addition of the reagents, the reaction mixture was continued to stir for 15 h at room temperature, then filtered.
The clear filtrate thus obtained was then successively treated with two portions of 10% NaHCO3 in H2O, then with a saturated aqueous solution of NaCl, before being dried over Na2SO4. After evaporation of the volatile fractions, 2.49 g of a residue were obtained which, after separation by column chromatography (silica gel - cyclohexane / ethyl acetate 7: 3), gave 0.53 g of product of starting point and 1.76 g of an epoxidized product (epoxidation yield: 90%).
ii) The epoxidation product obtained above dissolved in 50 ml of methylene chloride was stirred for 24 h at room temperature, in the presence of 0.07 g of ptoluenesulphonic acid and under a nitrogen atmosphere. After the neutralization, washing and drying treatments indicated above - see letter i) -, the distillation of the residue (1150 / 0.005
Torr) gave 1.58 g (about 90%) of 2,6,10,10-tetramethyl-1-oxaspiro [4.5] dec-3-en-6-ol, Eb. 80-90o / 0.1 Torr.

2,6,10,10-tetramethyl-1 -oxa-spiro [4.5] déc-3-en-6-ol, which is a new product, is characterized by a woody and camphoric odor reminiscent in some ways that of patchouli . It can therefore be advantageously used as a perfuming or flavoring ingredient.
iii) 1.54 g (approximately 7 mmol) of the unsaturated hydroxy compound obtained above, dissolved in 40 ml of ethanol, were hydrogenated to approximately 10, in the presence of 250 g of 5% palladium on carbon, after absorption of 155 ml of hydrogen, the reaction mixture was filtered, evaporated and distilled (115 / 0.005
Torr) to give 1.51 g of residue. The product thus obtained was then purified by column chromatography (gel of
Silica - Cyclohexane / Ethyl acetate 7: 3) and finally 1.08 g (55%) of 2,6,10,10-tetramethyl-1-oxa -spiro [4.5] decane-6- was isolated ol, Eb. 58-59 / 0.5 Torr.

Example 2
2,6,10,10-tetramethyl-1-oxa- formate
spiro [4.5] dec-6-yl
0.4 g (1.9 mM) of 2,6,10,10-tetramethyl-1-oxa-spiro- [4.5] decan-6-ol and 0.5 g (5.2 mM) of freshly sublimated formylimidazole were thoroughly mixed before being stored for 48 hours at room temperature. After extracting the reaction mixture with ether, washing, drying and fractional distillation of the organic phase, 0.3 g of a product having Eb. 70-800 / 0.5 Torr and containing 60% of the desired ester. A sample for analysis was obtained by preparative gas chromatography.

Example 3

spiro [4.5] dec-6-yl
5.9 g (0.075 M) of acetyl chloride were added over 30 min, with stirring, to a mixture maintained at 200 of 2.12 g (0.01 M) of 2,6,10,10-tetramethyl -1-oxa-spiro [4.5] decan-6-ol and 10.9 g (0.09 M) of N, N-dimethylaniline.
After being stored for 2 days at room temperature, the reaction mixture was heated under reflux for 3 h. After being treated with 50 ml of ether, the reaction mixture was filtered and the resulting filtrate poured onto ice and acidified with 10% H2SO4 in H2O. The organic phase was then washed with a saturated NaHCO3 solution until neutral, finally dried, evaporated and distilled according to the usual techniques to give 2.5 g of a product having Eb. 90-100o / 0.1 Torr. After recrystallization from an ethanol-water mixture, 1.9 g (75%) of the desired ester were isolated, mp 55-560.
