Gegenstand der Erfindung ist ein Verfahren zur Herstellung neuer 9,10-Dihydro-9,10-äthano-anthrazene mit dem Kern der Formel

die in 9-Stellung einen gegebenenfalls N-substituierten Aminoalkylrest und in mindestens einer der Stellungen 1 bis 8 eine freie Hydroxylgruppe aufweisen, sowie ihrer Salze.
Im Aminoalkylrest der neuen Verbindungen ist der die Aminogruppe mit dem Äthanoanthrazen-Kern verbindende Alkylenrest vor allem ein niederer gerader oder verzweigter Alkylenrest mit höchstens 5 Kohlenstoffatomen, insbesondere ein solcher, der die Aminogruppe vom Kern durch 1 bis 3, insbesondere 1 oder 3, Kohlenstoffatome trennt, wie der Propylen-(1,3)-, Propylen-(1,2)-, Äthylen-(1,2)- oder Methylenrest.
Die Aminogruppe des Aminoalkylrestes in 9-Stellung kann unsubstituiert oder substituiert sein. Eine substituierte Aminogruppe ist dabei z. B. eine primäre oder sekundäre, d. h. eine mono- oder disubstituierte Aminogruppe. Als Substituenten sind dabei vor allem niedere aliphatische Kohlenwasserstoffreste, die auch durch Heteroatome, wie Sauerstoff-, Schwefel- oder Stickstoffatome unterbrochen und/oder durch Hydroxylgruppen substituiert sein können, oder niedere araliphatische Kohlenwasserstoffreste zu nennen. Als solche Reste kommen vor allem niedere Alkylreste, niedere Alkylenreste, niedere Oxaalkylen-, niedere Azaalkylen- oder niedere Thiaalkylenreste, oder Phenylniederalkylreste in Betracht. Niedere Alkylreste sind dabei beispielsweise Methyl-, Äthyl-, Propyl-, Isopropyl-, gerade oder verzweigte, in beliebiger Stelle verbundene Butyl-, Pentyl-, Hexyl- oder Heptylreste.
Niedere Alkylrenreste, Oxaalkylenreste, Azaalkylenreste und Thiaalkylenreste sind vor allem solche, die mit dem Amino-Stickstoffatom zusammen einen 5-7gliedrigen Ring ergeben, wie z. B. Butylen-(1,4)-, Pentylen-(1,5)-, Hexylen (1,5)-, Hexylen-(1,6)-, Hexylen-(2,5)-, Heptylen-(2,7)-, Heptylen-(2,6)-, 3-Oxapentylen-(1,5)-, 3-Thiapentylen (1,5)-, 2,4-Dimethyl-3-thiapentylen-(1,5)-, 3-Aza-pentylen (1,5)-, 3-Niederalkyl-3-aza-pentylen-(1,5)-, wie 3-Methyl3-aza-pentylen-(1,5)-, 3-Hydroxyniederalkyl)-3-aza-pentylen-(1,5)-, wie 3-iB-Hydroxyäthyl)-3-aza-pentylen-(1,5)-, 3-Oxahexylen-(1,6)- oder 3-Azahexylen-(1,6)-reste. Phenylniederalkylreste sind vor allem Benzyl- oder 1- oder 2-Phenyläthylreste, die auch durch niedere Alkylreste, z. B.
die genannten, niedere Alkoxyreste, z. B. Methoxy oder Äthoxy, Halogenatome, z. B. Fluor-, Chlor oder Bromatome, oder Trifluoromethylgruppen substituiert sein können.
Die Amihogruppe ist demnach beispielsweise eine Benzylamino- oder N-Niederalkyl-benzylaminogruppe, wie N Methyl- oder N-Äthyl-benzylaminogruppe oder eine gegebenenfalls C-niederalkylierte und/oder im Ring ss-einfach ungesättigte Pyrrolidino- oder Piperidinogruppe oder eine gegebenenfalls C-niederalkylierte Piperazino-, N'-Niederalkyloder N' -(Hydroxyniederalkyl)-piperazino-, Thiomorpholinooder Morpholino-gruppe, oder vor allem eine Mono- oder Diniederalkylaminogruppe, wie eine Methylamino-, Äthylamino-, Dimethylamino-, Diäthylamino- oder N-Methyl äthylaminogruppe.
Die neuen Verbindungen können weitere Substituenten enthalten. So können sie an den aromatischen Ringen (Stellungen 18) weitere Substituenten enthalten, wie z. B. niedere Alkylreste, z. B. die genannten, vor allem Methylreste, Halogenatome, z. B. die genannten, vor allem Chlor, und/oder Trifluormethylgruppen. Ferner können die neuen Verbindungen in 10-Stellung substituiert sein, z. B. durch niedere Alkylreste, z. B. die genannten, insbesondere Methyl oder Halogenatome, z. B. die genannten, vor allem Chlor.
Die neuen Verbindungen besitzen wertvolle pharmakologische Eigenschaften, insbesondere eine zentralhemmende Wirkung. So weisen sie neben einer cocainantagonistischen Wirkung insbesondere einen Antagonismus gegenüber psychomotorischen Stoffen, wie z. B. Mescalin, auf, wie sich im Tierversuch, z. B. an Mäusen bei oraler Gabe in Dosen von 10 bis 100 mg/kg, zeigt, und besitzen eine Hemmwirkung auf die spinale Reflexübertragung und eine histaminolytische Wirkung. Die neuen Verbindungen können daher als beruhigende und insbesondere psychotrnpe, wie antidepressive bzw.
tranquillisierende Mittel Verwendung finden. Sie besitzen einen grösseren therapeutischen Index und eine längere Wirkungsdauer als die bekannten Äthanoanthrazene. Sie können auch als Zusatzstoffe zu Tierfutter verwendet werden, da sie eine bessere Nahrungsverwertung und eine Gewichtszunahme dieser Tiere bewirken. Weiter können die neuen Verbindungen als Ausgangs- oder Zwischenprodukte für die Herstellung anderer wertvoller Verbindungen dienen.
Besonders hervorzuheben sind die Verbindungen der Formel

worin n für 1 oder 3 steht, R1 einen niederen, Alkylrest, insbesondere den Äthylrest oder vor allem den Methylrest, oder ein Wasserstoffatom bedeutet, und R2 einen niederen Alkylrest, insbesondere den Äthylrest oder vor allem den Methylrest darstellt, vorzugsweise diejenigen Verbindungen der obigen Formel, worin der Rest OH in 2-Stellung steht, speziell das 2-Hydroxy-9-(dimethylaminomethyl)-9, 10-dihydro-9, 10- äthano-anthrazen und vor allem das 2-Hydroxy-9-(methyl aminomethyl)-9, 1 0-dihydro- 9,10-äth 9,10-Åathanoanthrazen, welches beispielsweise in Form seines Hydrochlorids bei oraler Gabe an der Maus eine deutliche mescalinantagonistische Wirkung in einer Dosis von 10 mg/kg und eine cocainantagonistische Wirkung in einer Dosis von 10 bis 30 mg/kg besitzt.
Das erfindungsgemässe Verfahren zur Herstellung der neuen Verbindungen ist dadurch gekennzeichnet, dass man in einem 9,10-Dihydro-9,10-äthano-anthrazen, das in 9-Stellung einen Aminoalkylrest und in mindestens einer der Stellungen 18 eine durch einen durch Reduktion abspaltbaren Rest substituierte Hydroxylgruppe aufweist, oder in einem Salz davon, die Hydroxylgruppe(n) durch Reduktion freisetzt.
Ein durch Reduktion abspaltbarer Rest ist in erster Linie ein a-Aralkylrest, wie ein Benzylrest, oder ein a-Aralkoxycarbonylrest, wie ein Carbobenzoxyrest. Die Abspaltung eines solchen Restes erfolgt vorzugsweise. durch Reduktion mit katalytisch erregtem Wasserstoff, wie Wasserstoff in Gegenwart eines Hydrierungskatalysators, wie eines Palladium- oder Platinkatalysators.
Der durch Reduktion abspaltbare Rest kann auch ein ss-Halogenalkylrest oder ein ,8-Halogenalkoxycarbonylrest, wie z. B. ein 2-Jodäthoxycarbonylrest oder ein 2,2,2-Tribromoder 2,2,2-Trichlor-äthoxycarbonylrest sein. Zur Abspaltung kommt als Reduktionsmittel vor allem nascierender Wasserstoff in Betracht, wie er z. B. durch Einwirkung von Metallen bzw. Metallegierungen auf wasserstoffabgebende Mittel, wie
Carbonsäuren, Alkohole oder Wasser, erhältlich ist. Vor allem verwendet man Zink oder Zinklegierungen in Essigsäure oder Äthanol. Ferner kommen auch Chrom-II-verbindungen, wie Chrom-II-chlorid oder Chrom-II-acetat, in Betracht.
Der durch Reduktion abspaltbare Rest kann aber auch ein Acylrest sein. Die reduktive Freisetzung der Hydroxylgruppe(n) erfolgt dann z. B. durch Behandeln mit einem komplexen Metallhydrid, wie Lithiumaluminiumhydrid.
In erhaltenen Verbindungen kann man im Rahmen der Definition der Endstoffe Substituenten einführen, abwandeln oder abspalten.
So kann man beispielsweise in erhaltene N-unsubstituierte oder N-monosubstituierte Aminoalkylgruppen Substituenten einführen, z. B. die oben genannten. Die Einführung erfolgt in üblicher Weise, z. B. durch Umsetzen mit einem reaktionsfähigen Ester eines entsprechenden Alkohols. Ein reaktionsfähiger Ester ist vor allem ein Ester mit einer starken organischen oder anorganischen Säure, wie besonders einer Halogenwasserstoffsäure, wie Chlor-, Brom- oder Jodwasserstoffsäure, der Schwefelsäure oder einer Arylsulfonsäure, z. B. der p-Toluolsulfonsäure. Die Substituenteneinführung kann auch reduktiv erfolgen, z. B. durch Umsetzen mit einem entsprechenden Aldehyd oder Keton unter reduzierenden Bedingungen, d. h. in Gegenwart von katalytisch erregtem Wasserstoff oder Ameisensäure.
In erhaltenen Verbindungen, die am Aminoalkyl-Stickstoffatom einen a-Aralkylrest, wie einen Benzylrest, aufweisen, kann man diesen durch Reduktion abspalten. Die Reduktion erfolgt in üblicher Weise, z. B. wie oben für den O-ständigen a-Aralkyl- bzw. a-Aralkoxycarbonylrest beschrieben.
Die Erfindung betrifft auch diejenigen Ausführungsformen des Verfahrens, bei denen man einen Ausgangsstoff unter den Reaktionsbedingungen bildet oder gegebenenfalls in Form eines Salzes und/oder Racemates oder optischen Antipoden verwendet.
Die genannten Reaktionen können in üblicher Weise in An- oder Abwesenheit von Verdünnungs-, Kondensationsund/oder katalytischen Mitteln, bei erniedrigter, gewöhnlicher oder erhöhter Temperatur, gegebenenfalls im geschlossenen Gefäss durchgeführt werden.
Je nach den Verfahrensbedingungen und Ausgangsstoffen erhält man die Endstoffe in freier Form oder in der ebenfalls in der Erfindung inbegriffenen Form ihrer Säureadditionssalze. So können beispielsweise basische, neutrale oder gemischte Salze, gegebenenfalls auch Hemi-, Mono-, Sesquioder Polyhydrate davon erhalten werden. Die Säureadditionssalze der neuen Verbindungen können in an sich bekannter Weise in die freie Verbindung übergeführt werden, z. B. mit basischen Mitteln, wie Alkalien oder Ionenaustauschern.
Anderseits können die erhaltenen freien Basen mit organischen oder anorganischen Säuren Salze bilden. Zur Herstellung von Säureadditionssalzen werden insbesondere solche Säuren verwendet, die zur Bildung von therapeutisch verwendbaren Salzen geeignet sind. Als solche Säuren seien beispielsweise genannt:
Halogenwasserstoffsäuren, Schwefelsäuren, Phosphorsäuren, Salpetersäure, Perchlorsäure, aliphatische, alicyclische, aromatische oder heterocyclische Carbonoder Sulfonsäuren, wie Ameisen-, Essig-, Propion-, Bernstein-, Glykol-, Milch-, Äpfel-, Wein-, Zitronen-, Ascorbin-, Malein-, Hydroxymalein- oder Brenztraubensäure; Phenylessig-, Benzoe-, p-Aminobenzoe-, Anthranil-, p-Hydroxybenzoe-, Salicyl- oder p-Aminosalicylsäure, Embonsäure, Methansulfon-, Äthansulfon-, Hydroxyäthansulfon-, Äthylensulfonsäure; Halogenbenzolsulfon-, Toluolsulfon-, Naphthalinsulfonsäure oder Sulfanilsäure; Methionin, Tryptophan, Lysin oder Arginin.
Diese oder andere Salze der neuen Verbindungen, wie z. B. die Pikrate, können auch zur Reinigung der erhaltenen freien Basen dienen, indem man die freien Basen in Salze überführt, diese abtrennt und aus den Salzen wiederum die Basen freimacht. Infolge der engen Beziehungen zwischen den neuen Verbindungen in freier Form und in Form ihrer Salze sind im Vorausgegangenen und nachfolgend unter den freien Verbindungen sinn- und zweckmässig, gegegenenfalls auch die entsprechenden Salze zu verstehen.
Je nach Wahl der Ausgangsstoffe und Arbeitsweisen können die neuen Verbindungen, sofern sie mindestens ein asymmetrisches Kohlenstoffatom besitzen, als Racemate oder als optische Antipoden vorliegen.
Erhaltene Racemate lassen sich nach bekannten Methoden, beispielsweise durch Umkristallisation aus einem optisch aktiven Lösungsmittel, mit Hilfe von Mikroorganismen, oder durch Umsetzen mit einer mit der racemischen Verbindung Salze bildenden optisch aktiven Säure und Trennung der auf diese Weise erhaltenen Salze, z. B. aufgrund ihrer verschiedenen Löslichkeiten, in die Diastereomeren, aus denen die Antipoden durch Einwirkung geeigneter Mittel freigesetzt werden können, zerlegen. Besonders gebräuchliche optisch aktive Säuren sind z. B. die D- und L-Formen von Weinsäure, Di-o-Toluylweinsäure, Apfelsäure, Mandelsäure, Camphersulfonsäure oder Chinasäure. Vorteilhaft isoliert man den wirksameren der beiden Antipoden.
Zweckmässig verwendet man für die Durchführung der erfindungsgemässen Reaktionen solche Ausgangsstoffe, die zu den eingangs besonders hervorgehobenen Endstoffen führen.
Die Ausgangsstoffe sind bekannt oder können, falls sie neu sind, nach an sich bekannten Methoden erhalten werden.
Die neuen Verbindungen können z. B. in Form pharmazeutischer Präparate Verwendung finden, welche sie in freier Form oder gegebenenfalls in Form ihrer Salze, besonders der therapeutisch verwendbaren Salze, in Mischung mit einem z. B. für die enterale oder parenterale Applikation geeigneten pharmazeutischen organischen oder anorganischen, festen oder flüssigen Trägermaterial enthalten.
Die neuen Verbindungen können auch in der Tiermedizin, z. B. in einer der oben genannten Formen oder in Form von Futtermitteln oder von Zusatzmitteln für Tierfutter verwendet werden.
In den folgenden Beispielen sind die Temperaturen in Celsiusgraden angegeben.
Beispiel 1
9,0 g 2-Benzyloxy-9-(methylaminomethyl)-9, 10-di- hydro-9-10-äthano-anthrazen werden in 100 ml Äthanol gelöst und nach Zugabe von 1 g Palladiumkohle (10-proz.) bei Raumtemperatur hydriert. Nach beendeter Wasserstoffaufnahme filtriert man den Katalysator ab und dampft das Filtrat im Vakuum ein. Den Rückstand löst man in 2n Natronlauge und filtriert die ungelösten Anteile ab. Das Filtrat wird nun durch Zugabe von 2-n Salzsäure sauer gestellt und nochmals filtriert. Auf Zugabe von Natriumhydrogencarbonat zum sauren Filtrat fällt das 2-Hydroxy-9-(methylaminomethyl)-9,10di-hydro-9,10-äthano-anthrazen der Formel

aus, das nach Umkristallisation aus Isopropanol bei 120 bis 125 schmilzt. Das Hydrochlorid schmilzt bei 261-262 .
Das als Ausgangsprodukt verwendete 2-Benzyloxy-9 (methylaminomethyl)-9,10-dihydro-9,10-äthano-anthrazen kann auf folgende Weise hergestellt werden:
29 ml 65prozentige Salpetersäure werden bei 30-35 zu 103 ml Acetanhydrid getropft. Die so erhaltene Lösung gibt man anschliessend langsam zu einer Suspension von 90 g 9-Cyano-9,10-dihydro-9,10-äthano-anthrazen und rührt 12 Stunden bei Zimmertemperatur. Der ausgefallene Niederschlag wird dann filtriert und mit Methanol gewaschen. Man erhält 2-Nitro-9-cyano-9,10-dihydro-9,10-äthano-anthrazen vom F. 2150.
Zu einer Lösung von 156 g 2-Nitro-9-cyano-9,10-dihydro9,10-äthano-anthrazen in 1500 ml Dimethylformamid gibt man 16 g Raney-Nickel und hydriert bei 40". Nachdem 40 Liter Wasserstoff aufgenommen worden sind, wird der Katalysator abfiltriert und das Lösungsmittel im Vakuum eingedampft. Zum Rückstand gibt man 80 ml Isopropanol und 80 ml Petroläther, worauf das 2-Amino-9-cyano-9,10-di- hydro-9,10-äthano-anthrazen auskristallisiert. Die Verbindung schmilzt nach Umkristallisation aus Methanol-Wasser bei 133-137".
100 g dieses Amins werden zu 875 ml in Salzsäure gegeben, wonach man bei Zimmertemperatur unter Rühren eine Lösung von 28 g Natriumnitrit in 55 ml Wasser zutropft.
Nachdem das Reaktionsgemisch noch 1 Stunde bei Raumtemperatur gerührt worden ist, wird die Diazoniumlösung filtriert und mit 42 ml konzentrierter Schwefelsäure versetzt.
Man rührt noch 1 Stunde bei 95" und extrahiert dann nach Abkühlen auf Zimmertemperatur mit Äther. Den Äther schüttelt man mit 2n Natronlauge aus und trennt den alkalischen Extrakt ab. Durch Ansäuren dieses alkalischen Auszugs mit konzentrierter Natronlauge scheidet sich das 2-Hydroxy 9-cyano-9,10-dihydro-9,10-äthano-anthrazen aus, das man mit Äther extrahiert und das nach dem Verdampfen des Lösungsmittels als hellgelbes, zähflüssiges Öl zurückbleibt.
46 g dieses Öls werden zu einer Lösung von Natrium äthylat in Äthanol - hergestellt durch Lösen von 4,6 g Natrium in 400 ml Äthanol - gegeben, und anschliessend tropft man 36 g Benzylbromid zu und rührt 4 Stunden bei Raumtemperatur. Das ausgeschiedene Natriumbromid wird abfiltriert, und das Filtrat wird im Vakuum eingedampft. Es verbleibt das 2-Benzyloxy-9-cyano-9,10-di-hydro-9,10-äthano- anthrazen, das nach Reinigung durch Chromatographie und Umkristallisation aus Äthanol bei 98-100 schmilzt.
Zur Hydrolyse des Nitrils werden 91 g 2-Benzyloxy-9cyano-9,10-dihydro-9,10-äthano-anthrazen mit 25 g Kaliumhydroxyd in 450 ml Äthylenglykol während 20 Stunden auf 1900 erhitzt. Man gibt hierauf 1000 ml Wasser zu und filtriert die ungelösten Anteile ab. Das Filtrat wird durch Zugabe von konzentrierter Salzsäure angesäuert, wobei die 2-Benzyloxy 9,1 0-dihydro-9, 10-äthano-9-anthrazen-carbonsäure ausfällt, die bei 195-199 schmilzt.
Zur Überführung in das Säurechlorid werden 60 g 2-Benzyloxy-9,10-dihydro-9,10-äthano-9-anthrazencarbonsäure während 90 Minuten mit 90 ml Oxalylchlorid unter Rückfluss erwärmt. Hierauf wird das überschüssige Oxalylchlorid eingedampft, und es verbleibt das rohe Säurechlorid, das direkt weiter verwendet wird.
60 g des rohen Säurechlorids löst man in 300 ml Methylenchlorid und leitet bei Zimmertemperatur Monomethylamin ein. Nach 2 Stunden werden 200 ml 2-n Natronlauge zugegeben, und die Methylenchlorid-Schicht wird abgetrennt, mit Wasser gewaschen und eingedampft. Es verbleibt das 2-Ben zyloxy-9,10-dihydro-9,10-äthano-9-anthrazencarbonsäure- methylamid, das nach Umkristallisation aus Isopropanol bei 170-172" schmilzt.
12 g dieses Amids werden in 100 ml Tetrahydrofuran gelöst, zu 12 g Lithiumaluminiumhydrid in 100 ml Tetrahydrofuran getropft und während 9 Stunden auf 60 erwärmt. Hierauf kühlt man ab, gibt hintereinander 15 ml Wasser, 15 ml 15prozentige Natronlauge und 45 ml Wasser zu. Den ausgefallenen Niederschlag filtriert man ab und dampft das Filtrat ein. Es verbleibt das 2-Benzyloxy-9-(methylaminomethyl)- 9,10-dihydro-9,10-äthano-anthrazen, dessen Hydrochlorid bei 210-213" schmilzt.
Beispiel 2
In analoger Weise, wie im Beispiel 1 beschrieben, kann man aus der entsprechenden 2-Benzyloxyverbindung oder der entsprechenden 2-Benzyloxy-carbonyloxyverbindung das 2 -Hydroxy-9-(y-methylaminopropyl) -9,10-dihydro-9,10- äthano-anthrazen, F. 215-220 , das 2-Hydroxy-9-(y-di methylaminopropyl)-9' 1 0-dihydro-9, 1 0-äthano-anthrazen und das 2-Hydroxy-9-(dimethylaminomethyl)-9, 10-dihydro- 9,10-äthano-anthrazen, oder ihre Salze, z. B. Hydrochloride, erhalten.
