Procédé pour la préparation de dérivés de la bêta-phénylalanine La présente invention est relative à la préparation de nouveaux dérivés de bêta-phénylalanine.

dans laquelle R1 et R2 sont identiques ou différents et représentent un radical méthyle ou éthyle, Ri ou R2 pou vant également représenter l'hydrogène ou un radical acyle comprenant de 1 à 30 atomes de carbone, mais, de pré férence, R1 et R2 sont différents, l'un d'eux représentant de l'hydrogène, l'autre représentant le radical méthyle, M représente de l'hydrogène ou un cation non toxique pharmaceutiquement acceptable (comme par exemple un ion de métal alcalin, un ion ammonium ou un ion am monium substitué)
et X représente du chlore, du brome, de l'iode ou du fluor et de préférence du chlore ou du brome.
Les composés de formule I ainsi que leurs esters alkyliques et alkényliques en Ci à C30 présentent une activité hypotensive qui se prolonge pendant une longue période après leur administration et, de ce fait, ils consti tuent des agents utiles pour l'abaissement de la tension sanguine.
Les composés de formule I, préparés par le procédé selon l'invention, ainsi que leurs esters décrits, peuvent être acylés à l'atome d'azote amino par un agent d'acy- lation comprenant de 1 à 30 atomes de carbone.
Il a été trouvé que des composés préférés tels que 3-méthoxy-4-hydroxy-5-chlorophénylalanine (chlorhydrate), 3-méthoxy-4-hydroxy-5-bromophénylalanine (chlorhydrate), et 3-hydroxy-4-méthoxy-5-bromophénylalanine (chlorhydrate) abaissent la tension sanguine de rats hypertendus (sui vant le procédé décrit par M. Gerlod, A. Hurlimann et C. von Planta dans Helv. Physiol. Acta 24, 58 (l966)) après administration par voie orale ou sous-cutanée à des doses comprises entre 30 et 100 mg/kg.
Les composés de formule I peuvent être administrés par voie orale, intramusculaire, intraveineuse ou sous- cutanée.
Les composés de formule I qui présentent un groupe amino libre sont administrés de préférence sous la forme d'un sel formé avec un acide non toxique, lequel acide peut être un acide inorganique tel que l'acide chlorhy drique, l'acide bromhydrique, l'acide phosphorique ou l'acide sulfurique. Lorsque les composés de formule I sont estérifiés, d'autres sels possibles sont ceux obtenus avec des acides organiques tels que l'acide acétique, l'acide succinique, l'acide maléfique, l'acide fumarique, l'acide tartrique, l'acide citrique, l'acide palmitique, l'acide méthane-sulfonique, l'acide éthane-sulfonique, etc.
Ces nouveaux dérivés de bêta-phénylalanine peuvent être mélangés à des ingrédients pharmaceutiquement acceptables et intégrés dans des formules pharmaceu tiques pour l'administration orale ou parentérale. Les compositions peuvent prendre la forme de comprimés, de poudres, de granulés, de capsules, de suspensions, de solutions, d'émulsions, de solutions et suspensions injec tables, et autres préparations du même genre.

Dans ces formules II et III, R1, R2 et X sont tels que définis plus haut, et M représente de l'hydrogène ou un cation monovalent, par exemple un cation de métal alcalin.
Plus particulièrement, les composés répondant aux formule II et III où R1 est de l'hydrogène et R2 repré sente un radical méthyle ou éthyle sont des composés nouveaux et seront représentés ci-après respectivement par les formules IIb et IIIb.
Suivant la nature et la position respective de R1 et de R2, les 5-halobenzaldéhydes 3,4-disubstitués de for peuvent être obtenus de différentes manières dont certaines sont décrites ci-après à titre d'illustration sous les en-têtes A, B et C.
A. Lorsque R1 représente un radical méthyle ou éthyle et lorsque R, est de l'hydrogène, on peut utiliser comme produit de dépara le benzaldéhyde 3,4-disubstitué correspondant. A cette fin, le benzaldéhyde 3,4-disubstitué peut être tout d'abord halogéné pour fourrer le 5-halo- benzaldéhyde correspondant, la réaction étant effectuée de préférence à la température ordinaire dans un solvant convenable comme par exemple l'acide acétique. Lorsque X désigne du fluor, le 5-fluorbenzaldéhyde 3,4-disubstitué sera obtenu de préférence au départ de 5-bromobenzal- déhyde correspondant par transposition d'halogène.

B. Lorsque, dans la formule I, R1 représente de l'hydrogène et R2 représente un radical méthyle ou éthyle, on part en général d'un 3-alkoxybenzaldéhyde substitué, ledit alkoxy comprenant de 1 à 4 atomes de carbone.
A cette fin, le 3-alkoxybenzaldéhyde substitué peut d'abord être halogéné pour fournir le 5-halobenzaldéhyde correspondant qui est déalkylé pour donner le 5-halo- protocatechualdéhyde, ce dernier étant ensuite alkylé sélectivement en position 4. Ladite déalkylation peut être effectuée en utilisant l'une ou l'autre méthode connue dans la technique pour une telle réaction, par exemple à l'aide de chlorure d'alu minium anhydre et de pyridine dans du chlorure de méthylène, sons l'influence de la chaleur.
La O-alkylation qui est sélective pour la position 4 s'effectuera dans un solvant inerte en utilisant l'un ou l'autre procédé connu dans la technique, à condition que le rapport molaire agent d'alkylation/aldéhyde soit inférieur à deux. Comme exemples de systèmes sol vant/agent d'alkylation, on peut citer acétone/iodure de méthyle et dioxanne/diazométhane. - Le schéma 2 suivant décrit les aspects particuliers de cette méthode par rapport au processus du schéma 1

En variante, le benzaldéhyde substitué mentionné peut également être obtenu au départ d'un 3-alkoxy- benzaldéhyde substitué dans lequel le groupe alkoxy pré sente plus de deux atomes de carbone, par exemple le 3-isopropoxy-4-hydroxybenzaldéhyde. Dans ce but, ce produit de départ sera d'abord halo géné pour donner le 5-halobenzaldéhyde correspondant qui est ensuite alkylé par exemple à l'aide de diméthyl- ou diéthylsulfate pour fournir le 3-isopropoxy-4-alkoxy- 5-halobenzaldéhyde correspondant.
Ce produit sera déisopropylé, par exemple en ame nant à la température du reflux pendant un temps très court (de une à dix minutes) sa solution dans de l'acide bromhydrique aqueux (48 0/0). Le schéma 3 suivant décrit les particularités de cette méthode par rapport au processus du schéma 1.

Les composés de formule III peuvent alors être pré parés au départ des composés de formule générale II. Cette préparation des composés de formule III, produits de départ pour le procédé selon l'invention, est illustrée dans le schéma 6 suivant. Ce schéma comprend les possi bilités suivantes Un benzaldéhyde substitué de formule II est d'abord amené à réagir avec une quantité stochiométrique d'acé- tylglycine en présence d'un excès d'anhydride acétique. La réaction est effectuée dans un solvant convenable (comme par exemple un mélange alcool/eau) sous l'in fluence de la chaleur (reflux), de préférence en présence d'acétate de sodium, afin d'obtenir une azlactone.
L'azlactone obtenue est alors transformée en dérivé d'acide phénylpyruvique substitué correspondant soit par une base forte, soit par un acide fort (les alcalis étant préférés). La réaction s'effectue sous l'influence de la chaleur dans un solvant inerte comme par exemple l'eau ou un mélange hydroalcoolique, la concentration en alcali étant au moins 10 %.
En variante, on peut utiliser des conditions alcalines relativement douces (concentration en alcali inférieure à 10 %) et on obtient alors intermédiairement un acide 2-acétamido phénylpropénoïque substitué en position 3, lequel est ensuite transformé en acide phénylpyruvique substitué correspondant par un acide minéral ou par une base forte en forte concentration. Le schéma 6 suivant illustre le procédé au départ de 3-alkoxy-4-hydroxy-5- halobenzaldéhyde (formule IIa). Le composé de for mule III qui peut être obtenu ainsi, sera fait réagir selon le procédé revendiqué.
Etant donné que les composés de formule 1 présen tent un atome de carbone asymétrique, ils peuvent exister sous la forme des isomères D et L aussi bien que sous la forme optiquement inactive DL . A la fin des schémas de réaction cités plus haut, on obtient la forme DL<B> .</B> Les isomères D et L sont obtenus au départ de la forme DL par résolution optique en utilisant l'un ou l'autre procédé connu dans la tech nique, par exemple par formation d'un sel avec une base optiquement active ou par chromatographie sur cellulose (par exemple Whatman CF 11 ou CC 31) et élution à l'aide d'un solvant polaire (par exemple n-butanol saturé en acide chlorhydrique 3 N).
Chacun des isomères ainsi que le stade de résolution optique sont compris dans l'invention.
L'invention est illustrée par les exemples suivants dans lesquels la partie I se rapporte à la préparation préférée des composés de départ, et la partie II au pro cédé selon l'invention.
<I>Exemple I</I> 1. a) Un mélange de 115g de 5-bromovanilline, 58,5 g d'acétylglycine, 82 g d'acétate de sodium et 150 ml d'anhydride acétique est chauffé pendant deux heures à 1100 C.
On y ajoute 1,251 d'un mélange éthanol/eau (1/1) et le milieu réactionnel est maintenu sous agitation pen dant 30 minutes, filtré, lavé avec une autre portion de 500 ml de mélange éthanol/eau et séché sous pression réduite à 400 C pour donner 130 g d'azlactone.
Un échantillon de 50 g d'azlactone obtenue est dis sous dans 500 mi d'hydroxyde de sodium aqueux (2 N) et la solution est chauffée pendant deux heures à 60 700 C. Le pH de la solution est alors amené à 5,5 à l'aide d'acide chlorhydrique (6 N) et la solution est déco lorée sur charbon actif. Après filtration, le pH de la solution est amené à 2 avec de l'acide chlorhydrique (6 N).

Les cristaux obtenus sont filtrés, lavés à l'eau et séchés sous pression réduite pour donner 49 g d'acide 2-acétylamino-3-(3'-méthoxy-4'-hydroxy-5'-bromophé- nyl)-propénoïque.
b) Un échantillon de 25 g de cet acide propénoïque substitué est mis en suspension dans 200 ml d'un mélange acide acétique glacial/acide chlorhydrique (12 N) (30/70) et chauffé pendant six heures à la température du reflux et sous agitation énergique. Le milieu est ensuite refroidi et versé sous agitation dans deux litres d'eau.
Les cristaux obtenus sont filtrés, lavés à l'eau et séchés sous pression réduite à 400 C pour donner 17,9 g d'acide 3-méthoxy-4-hydroxy-5-bromophénylpyruvique, fus. 238-240 C (dec.), Rf = 0,25 ( 0,05) par chroma tographie sur couche mince de Silicagel G (un pro duit de E. Merck, Darmstadt, Rép. Féd. allemande), l'élution étant effectuée à l'aide d'un mélange benzène/ méthanol/acide acétique: 45/8/4 et suivie d'une détec tion à l'aide de chlorure ferrique.
II. Un échantillon de 10 g de l'acide obtenu est dissous dans 100 ml d'hydroxyde d'ammonium concentré et une solution de 350 mg de borohydrure de sodium dans 10 ml d'eau y est ajoutée sous agitation. L'agitation est maintenue pendant deux heures. Après ce temps de réaction, l'hydroxyde d'ammonium en excès est éliminé par évaporation du solvant sous pression réduite.
Le résidu est repris dans de l'eau qui est ensuite évaporée et le résidu est à nouveau repris dans de l'eau. La solution obtenue est acidifiée à l'aide d'acide chlorhydrique concentré jusqu'à pH 1,5.
La solution est maintenue sous agitation pendant une heure et le solvant est alors évaporé, le résidu est repris par deux portions d'eau et, ensuite, par du n-butanol et le solvant est évaporé. Le résidu est repris dans de l'éthanol. La fraction insoluble (sels minéraux) est éli minée par filtration et le filtrat est concentré jusqu'à obtention d'un sirop. Par addition d'éther, on obtient du chlorhydrate de DL-3-méthoxy-4-hydroxy-5-bromo- phénylalanine, Rf = 0,65 0,05 et 0,77 0,05 par chromatographie ascendante sur papier Whatman No 1 , l'élution étant effectuée à l'aide de n-butanol saturé en acide chlorhydrique 3 N.
<I>Exemple 2</I> I. a) Une solution de 500 g de vanilline, 250 g de bicarbonate de sodium et 747 g d'iodure de potassium dans 101 d'eau est traitée par 834 g d'iode. Le milieu est maintenu sous agitation pendant quatre heures et abandonné pendant une nuit. Les cristaux obtenus sont traités par une solution aqueuse à 10 % de thiosulfate de sodium, filtrés, lavés à l'eau et séchés sous pression ré duite pour donner de la 5-iodovanilline.
b) Un échantillon de 27,8 g de 5-iodovanilline obte nue, 11,7 g d'acétylglycine, 8,2 g d'acétate de sodium et 30,6 g d'anhydride acétique sont chauffés à 1051, C pen dant quatre heures. Le milieu de réaction est repris avec 125 ml d'un mélange éthanol/eau (1/1) et les cristaux sont filtrés et séchés sous pression réduite.
L'azlactone obtenue est recristallisée de l'acide acé tique (fus. 194-1950 C) et dissoute dans 300 ml d'hy droxyde de sodium aqueux (agitation pendant une heure à 55o C).
La solution est acidifiée jusqu'à pH 5,3 à l'aide d'acide chlorhydrique (6 N), décolorée au charbon actif, filtrée et le pH est amené à 1,5 à l'aide d'une autre frac tion d'acide chlorhydrique (6 N). La solution est alors chauffée sous agitation à 551, C jusqu'à cristallisation.
Les cristaux sont filtrés, lavés à l'eau et séchés sous pression réduite pour donner de l'acide 2-acétamido-3- (3'-méthoxy-4'-hydroxy-5'-iodophényl)-propénoïque, fus. 218,5-220 C.
c) Cet acide est mis en suspension dans 100 ml d'un mélange acide acétique/acide chlorhydrique (12 N) (30/70) et le milieu est chauffé pendant trois heures à 900 C sous agitation.
Après ce temps de réaction, le précipité est filtré, lavé à l'eau et séché sous pression réduite pour donner de l'acide 3-méthoxy-4-hydroxy-5-iodophénylpyruvique, fus. 226-229 C, Rf = 0,22 ( 0,05) par chromatographie sur couche mince de Silicagel G (un produit de E. Merck, Darmstadt, Rép. Féd. allemande) dans le système benzène/méthanol/acide acétique: 45/8/4.
II. Cet acide est dissous dans 50 ml d'hydroxyde d'ammonium concentré et la solution est maintenue sous agitation pendant 15 minutes. On y ajoute 0,250 g de borohydrure de sodium et le milieu est maintenu sous agitation pendant deux heures.
Après ce temps de réaction, le pH de la solution est amené à 1,2 à l'aide d'acide chlorhydrique (6 N) et le solvant est évaporé sous pression réduite. Le résidu est repris à l'aide de trois portions successives d'eau afin d'éliminer l'acide chlorhydrique libre.
Le résidu est repris par du n-butanol, le solvant est évaporé et le résidu est repris par un minimum de n-butanol chaud et traité avec de l'éther anhydre.
Les cristaux sont filtrés, lavés avec de l'éther anhydre et séchés sous pression réduite pour donner du chlorhy- drate de DL-3-méthoxy-4-hydroxy-5-iodophénylalanine, fus. 187-190 C, Rf = 0,35 ( 0,05) par chromatogra phie en couche mince dans le système méthanol/chloro forme/hydroxyde d'ammonium: 20/20/5. <I>Exemple 3</I> I. a) Dans 1 litre de chlorure de méthylène, on met en suspension 231g de 5-bromovanilline et 146,7 g de chlorure d'aluminium anhydre et on y ajoute goutte à goutte 348 g de pyridine anhydre. Le milieu est chauffé pendant 24 heures à 450 C et on le verse ensuite dans 2 litres d'acide chlorhydrique (4 N) à 00 C.
Le précipité est filtré, lavé à l'eau et séché sous pression réduite pour donner de la 5-bromoprotocatéchualdéhyde (fus. 228 2290 C après recristallisation de l'éthanol).
b) Un échantillon de 108,5 g de 5-bromoprotocaté- chualdéhyde, 43,3 g de bicarbonate de sodium, 100 ml d'iodure de méthyle et 700 ml d'acétone (séchée sur carbonate de potassium) sont chauffés au reflux pendant 20 heures.
La solution est alors évaporée jusqu'à siccité et le résidu est repris par du chloroforme. La solution obtenue est traitée avec de l'acide chlorhydrique sec. Le milieu est filtré et la solution est versée sur 200 g de Silicagel Merck 7729 (un produit de E. Merck, Darmstadt, Rép. Féd. allemande), décolorée sur charbon actif et concen trée jusqu'à cristallisation. Après filtration, on obtient de la 5-bromo-isovanilline (fus. 116,5-117,5 C).
c) Un échantillon de 32,4 g de 5-bromo-isovanilline, 16,4 g d'acétylglycine, 22,9 g d'acétate de sodium et 27 ml d'anhydride acétique sont chauffés pendant deux heures à 1100 C. Le milieu de réaction est versé dans 800 ml d'eau. Le précipité obtenu est filtré, lavé à l'eau et séché sous pression réduite à 40 C sur pentoxyde de phosphore.
Après recristallisation de l'acide acétique glacial, fil tration, lavage à l'éther éthylique et séchage, on obtient l'azlactone, fus. 210-211 C, Rf = 0,66 ( 0,05) par chromatographie sur couche mince de Silicagel G (un produit de E.Merck, Darmstadt, Rép. Féd. alle mande) dans le système benzène/dioxanne/acide acéti que: 90/25/4.
d) Un échantillon de 20g d'azlactone obtenue est dissous dans. 250 ml d'hydroxyde de sodium aqueux (2 N) et la solution est chauffée pendant une heure à 70o C sous agitation.
Le pH de la solution est amené à 1,5 à l'aide d'acide chlorhydrique (6 N) et le milieu réactionnel est chauffé pendant une heure à 700 C. Les cristaux obtenus sont filtrés, lavés à l'eau et séchés à 400 C sous pression ré duite pour donner de l'acide 2-acétamido-3-(3'-hydroxy- 4'-méthoxy-5'-bromophényl)-propénoïque, fus. 218- 219 C, Rf = 0,25 ( 0,05) par chromatographie sur couche mince de Silicagel G (un produit de E. Merck, Darmstadt, Rép. Féd. allemande) dans le système ben- zène/méthanol/acide acétique: 45/8/4, la détection étant réalisée à l'aide de chlorure ferrique.
e) Un échantillon de 12 g d'acide 2-acétamido-3- (3'- hydroxy- 4'- méthoxy - 5'- bromophényl) - propénoïque obtenu est chauffé pendant 5 heures à 950 C dans un mélange de 30 ml d'acide acétique et 70 ml d'acide chlor hydrique (12N).
Le milieu chaud est filtré sur charbon actif et la cristallisation se produit par refroidissement.
Après filtration, les cristaux sont lavés à l'eau et séchés à 400 C sous pression réduite pour donner de l'acide 3-hydroxy-4-méthoxy-5-bromophénylpyruvique, fus. 203-205 C, Rf = 0,23 ( 0,05) par chromatogra phie sur couche mince dans le système benzène/métha nol/acide acétique: 45/8/4, la détection étant réalisée à l'aide de chlorure ferrique.
II. Un échantillon de 5 g d'acide phénylpyruvique substitué obtenu est dissous dans 50 ml d'hydroxyde d'ammonium concentré et la solution est maintenue sous agitation pendant 1/4 d'heure. On y ajoute 0,5 g de boro- hydrure de sodium et le milieu est maintenu sous agi tation pendant quatre heures.
Le pH de la solution est amené à 1,5 à l'aide d'acide chlorhydrique (6 N) et le solvant est évaporé. Le résidu est repris par plusieurs portions d'eau que l'on évapore ensuite pour éliminer l'acide chlorhydrique libre.
Le résidu est repris par deux portions de butanol, lequel est ensuite évaporé et enfin par un minimum de butanol chaud duquel la cristallisation se produit par refroidissement.
Après filtration, lavage à l'éther et séchage sous pression réduite, on obtient du chlorhydrate de DL-3- hydroxy-4-méthoxy-5-bromophénylalanine, fus. 151- 153 C, Rf = 0,53 ( 0,05) par chromatographie sur couche mince de Silicagel G (un produit de E. Merck, Darmstadt, Rép. Féd. Allemande) dans le système métha nol/chloroforme/hydroxyde d'ammonium : 20/20/5, la détection étant réalisée à l'aide de chlorure ferrique.
<I>Exemple</I> 4 (base libre) Une solution de 1 g de chlorhydrate de DL-3-mé- thoxy-4-hydroxy-5-bromophénylalanine (exemple 1) dans 50 ml d'eau est traitée en une fois par 2,5 ml de résine échangeuse d'ions anionique Amberlite LA2 (un pro duit liquide de Rohm and Haas Co., Philadelphia, Pa, USA) dans 60 ml de benzène.
Le milieu est maintenu sous agitation pendant 24 heu res. La couche aqueuse est alors lavée deux fois avec du benzène et la couche organique est lavée deux fois à l'eau.
Les fractions aqueuses sont réunies et le solvant est évaporé pour donner de la DL-3-méthoxy-4-hydroxy-5- bromophénylalanine sous forme de base libre, pureté par titration acide/base : 96,4 0/0. <I>Exemple 5</I> (base libre) Dans 50 ml d'eau, on dissout 16,33 g de chlorhy- drate de DL-3-méthoxy-4-hydroxy-5-bromophénylala- nine (exemple 1). Par addition de triéthylamine dans de l'alcool, on amène le pH de la solution à 5,1. La solu tion est filtrée et le filtrat est évaporé jusqu'à siccité.
Le résidu est traité par deux portions d'éthanol, lequel est ensuite évaporé.
Le résidu est trituré d'abord avec deux portions de chloroforme afin d'éliminer le chlorhydrate de triéthyl- amine et ensuite avec de. l'acétone. La solution est filtrée et le filtrat est évaporé jusqu'à siccité.
Le résidu est cristallisé de l'eau chaude pour donner de la DL-3-méthoxy-4-hydroxy-5-bromophénylalanine, base libre, pureté par titration acide/base : 95,20/o, humidité 2,4 0/0.
<I>Exemple</I> 6 (purification) Une solution de 1 g de chlorhydrate de DL-3-mé- thoxy-4-hydroxy-5-bromophénylalanine dans 25 ml d'éthanol anhydre est saturée à 00 C par de l'acide chlor hydrique gazeux. La solution est alors chauffée pendant une heure à 800 C. Le solvant est évaporé et le résidu est repris avec deux portions d'éthanol anhydre et précipité en versant la solution dans de l'éther anhydre pour don ner du chlorhydrate de DL-3-méthoxy-4-hydroxy-5- bromophénylalanine, fus. 194-197 C, pureté par titration acide/base : 100 0/0.
<I>Exemple 7</I> (utilisation) A une solution de 1 g de chlorhydrate de DL-3- méthoxy-4-hydroxy-5-bromophénylalanine dans 33,6 ml d'hydroxyde de sodium aqueux (N), on ajoute goutte à goutte et sous agitation 2,4 ml d'anhydride acétique. La réaction est exothermique et la couleur du milieu passe d'orange à jaune.
Le milieu est maintenu sous agitation à température ordinaire pendant 17 heures et il est ensuite filtré. Le pH du filtrat est amené à 2,3 à l'aide d'acide chlorhy drique et le solvant est évaporé par distillation azéotro pique. Le résidu est repris avec du chlorure de méthy lène. Après filtration, la solution est traitée au charbon actif, filtrée, concentrée jusqu'à faible volume et on y ajoute de l'hexane pour obtenir un précipité qui est filtré et séché sous pression réduite. On obtient ainsi de la DL-3-méthoxy-4-acétoxy-5-bromo-N-acétylphényl- alanine.
Lorsque ce produit est examiné par chromatographie sur couche mince de Kieselgel G (un produit de E. Merck, Darmstadt, Rép. Féd. allemande) dans le système propanol/eau : 75/25, la détection étant réalisée à l'aide de pourpre de bromocrésol, on obtient un seul spot, la réponse à la ninhydrine et au chlorure ferrique étant négative.
<I>Exemple 8</I> (utilisation) A une solution de 50 g de chlorhydrate de DL-3- méthoxy-4-hydroxy-5-bromophénylalanine dans 500 ml d'hydroxyde de sodium aqueux (N), on ajoute 0,5 g d'hydrosulfite de sodium et, lentement, de l'anhydride acétique jusqu'à pH 7, l'addition d'anhydride acétique est ensuite maintenue tout en maintenant le pH aux environs de 8 à l'aide d'hydroxyde de sodium aqueux (N).
Après addition de l'anhydride acétique (36,25 ml), le pH est amené à 10,8 à l'aide d'hydroxyde de sodium aqueux (dont la quantité totale employée est 602,5 ml). La température est maintenue entre 30 et 400 C pendant toute la durée de réaction.
Le milieu est maintenu pendant quatre heures à la température ordinaire et ensuite acidifié à l'aide d'acide chlorhydrique (6 N). La cristallisation du composé N- acétylé commence à une valeur de pH d'environ 4 mais l'acidification est continuée jusqu'à pH 1,4 et la cristal lisation est effectuée à ce pH pendant 15 heures à 4,, C.
Le précipité obtenu est filtré, lavé à l'eau froide et séché sur pentoxyde de phosphore sous pression réduite à 500 C pour donner de la DL-3-méthoxy-4-hydroxy-5- bromo-N-acétylphénylalanine, fus. 222-223 C (dec.). Lorsque ce produit est examiné par chromatographie sur couche mince de Kieselgel G (un produit de E. Merck, Darmstadt, Rép. Féd. allemande) dans le système propanol/eau : 75/25, la détection étant réalisée soit à l'aide de pourpre de bromocrésol, soit à l'aide de chlorure ferrique, on obtient un seul spot, la réponse à la ninhydrine étant négative.
<I>Exemple 9</I> (résolution optique) a) A 31,6 ml d'une solution éthanolique (4,3 M) de L(-)ephédrine diluée dans 84,5 ml d'acétone, on ajoute 45g de DL-3-méthoxy-4-hydroxy-5-bromo-N-acétyl- phénylalanine.
Après addition subséquente de 90 ml d'acétone, on obtient une solution par chauffage sous agitation. La solution est maintenue pendant cinq jours à température ordinaire.
Les cristaux obtenus sont filtrés, lavés à l'acétone, séchés, réduits en poudre et repris dans 400 ml d'acé tone. La suspension est maintenue sous agitation pendant une heure et est ensuite filtrée.
Les cristaux réunis sont dissous sous agitation dans 80 ml d'éthanol anhydre chaud. Par addition lente de 360 ml d'éther sous agitation et refroidissement ultérieur on obtient un précipité que l'on abandonne pendant quatre heures à température ordinaire avant filtration, lavage à l'éther et séchage sous pression réduite.
On obtient ainsi le sel de L(-)ephédrine de (-)3- méthoxy -4-hydroxy-5-bromo-N-acétylphénylalanine, [a]25 = - 660 (c = 1 dans l'éthanol), fus. 128-131 C.
b) Dans 1075 ml d'un mélange eau/éthanol (25/75), on dissout 21,5 g de sel de L(-)ephédrine de (-)3-mé- thoxy-4-hydroxy-5-bromo-N-acétylphénylalanine.
La solution est versée sur une colonne de 5 cm de diamètre de Amberlite IR 124 (un produit de Rohm & Haas, Philadelphia, Pa, USA) sous forme H+, la vitesse d'écoulement étant ajustée à 5 ml/minute.
L'élution est effectuée avec le même mélange solvant. L'éluat est concentré jusqu'à faible volume, repris dans de l'éthanol, traité au charbon actif et filtré.
Le solvant est évaporé sous pression réduite à 401, C pour donner de la (-)3-méthoxy-4-hydroxy-5-bromo-N- acétylphénylalanine, [a]25 =-400 (c = 1 dans l'éthanol). <I>Exemple 10</I> Une suspension de 11,5 g de (-)3-méthoxy-4-hydroxy- 5-bromo-N-acétylphénylalanine dans 230 ml d'acide chlorhydrique (1,2 N) est saturée en anhydride sulfureux. Une solution est obtenue après 30 minutes de chauffage sous agitation à 1100 C (température du bain) mais le chauffage et l'agitation sont encore maintenus pendant 31/s heures afin d'obtenir une hydrolyse complète.
Après ce temps de réaction, la solution est décolorée à l'aide de charbon actif et filtrée. La solution est con centrée jusqu'à faible volume, reprise dans de l'eau et évaporée jusqu'à siccité par distillation azéotropique. Le résidu est repris dans de l'éther anhydre. Après filtration et séchage sous pression réduite à 400 C, on obtient du chlorhydrate de (+)3-méthoxy-4-hydroxy-5-bromophé- nylalanine, [a]25 = + 10 (c = 1 dans l'eau).
<I>Exemple 11</I> (utilisation) Dans 100 ml d'hydroxyde de sodium aqueux (N), on dissout 15g de chlorhydrate de DL-3-méthoxy-4- hydroxy-5-bromophénylalanine comme obtenu à la fin de l'exemple 1. On y ajoute goutte à goutte et sous agitation 15 ml de chlorure de palmitoyle dissous dans 30 ml de tétra- hydrofuranne fraîchement distillé. Pendant l'addition, le pH du milieu est ajusté à une valeur comprise entre 6,5 et 7,5 à l'aide d'hydroxyde de sodium aqueux (N). Lorsque l'addition est terminée, le pH est amené à 8.
Pendant les 5 heures suivantes, l'agitation est maintenue et le pH du milieu est lentement amené à 9 par addition d'hydroxyde de sodium aqueux (N) et le milieu est ensuite abandonné pendant une nuit à température ordinaire.
La solution est concentrée sous pression réduite en présence d'une petite quantité d'hydrosulfite de sodium jusqu'à 1/s du volume initial et extraite avec deux por tions d'éther, le pH étant amené à 2 par addition d'acide chlorhydrique (N).
Les fractions organiques sont réunies, décolorées au charbon actif, filtrées, séchées sur sulfate de sodium anhydre et concentrées sous pression réduite. Par addition d'hexane, on obtient un précipité qui est filtré sur verre fritté et séché sous pression réduite à 400 C pour donner de la 3-méthoxy-4-hydroxy-5-bromo-N-palmitoylphényl- alanine, fus. 104-1070 C.
<I>Exemple</I> 12 (résolution optique) Dans<B>150</B> ml d'eau, on met en suspension 4,5 g de chlorhydrate de DL-3-méthoxy-4-hydroxy-5-chlorophé- nylalanine avec 400 ml de solution tampon à pH 7,4 (une solution de 23,9 g de phosphate disodique (12 aq) dans un litre d'eau, avec ajustage du pH à 7,4 par addi tion de phosphate monopotassique).
Le pH de la solution (6,5) est ajusté à 7,4 par addi tion de phosphate disodique et, après filtration, on ajoute 100 ml de solution tampon telle que définie ci-dessus et 0,15 g de L-aminoacide oxydase (un produit de Calbio- chem, Los Angeles, California, USA). Le milieu est maintenu sous agitation à 37 C pendant 43 heures.
Après ce temps de réaction, le pH du milieu est amené à 1,65 par addition d'acide chlorhydrique<B>(6N),</B> on y ajoute du charbon actif et le milieu est chauffé à 500 C pendant 1/z heure et filtré. Le filtrat est traité avec de la Amberlite LA-2 (un produit de Rohm and Haas Co., Philadelphia, Pa, USA) diluée dans du benzène.
La phase aqueuse est séparée, lavée au benzène, acidifiée à l'aide d'acide chlorhydrique (6 N) et le sol vant est évaporé. Le résidu est dissous dans de l'éthanol chaud, la solution (après filtration) est concentrée par évaporation du solvant, traitée par de l'eau jusqu'à éli mination de l'éthanol, extraite à l'éther éthylique et déco lorée au charbon actif.
Après filtration, la solution est alcalinisée et le sol vant est évaporé.
Le résidu est repris dans du méthanol. Après filtra tion, le solvant est évaporé et le résidu est mis en suspen sion dans de l'éthanol, filtré, lavé à l'éthanol et à l'éther, mis en suspension dans un minimum d'eau et traité par 20 ml d'acide chlorhydrique (N).
La solution obtenue est décolorée, le solvant est éva poré et le résidu est repris dans un minimum d'éthanol, dilué dans de l'éther et la solution est versée dans de l'éther éthylique anhydre pour donner un précipité qui est lavé à l'éther anhydre et le chlorhydrate de D(+)3- méthoxy-4-hydroxy-5-chlorophénylalanine est séché sous pression réduite à 501, C.
